The anti-inflammation effects of A.C.C. extracts on the LPS-induced Raw 264.7 cell (LPS로 유도한 Raw 264.7 세포에서 A.C.C. 추출물의 항염증 효과)

  • This study was conducted to evaluate the anti-inflammatory activity and clinical efficacy of a sample (A.C.C. extracts) obtained by distillation extraction of 14 herbal medicines including Phellodendron bark. To confirm this, the amount of nitric oxide (NO) produced by the cells in RAW 264.7 cells stimulated by lipopolysaccharide (LPS) and the changes in the production of inflammatory cytokines such as tumor necrosis factor (TNF)-${\alpha}$, interleukin(IL)-$1{\beta}$ and IL-6 were determined. The results showed that A.C.C. extracts strongly inhibited the production of NO and inflammatory cytokines increased by LPS without cytotoxicity. In addition, A.C.C. extracts showed strong bacterial reduction rates of 99.9% in Pseudomonas aeruginosa, Staphylococcus aureus, MRSA (Methicillin-resistant Staphylococcus aureus), Candida albicans and Streptococcus mutans. These findings indicate that A.C.C. extracts are effective ingredients with a strong antimicrobial effect together with an anti-inflammatory effect. In addition, when A.C.C. extracts were applied to infants and toddlers who were suffering from diaper rash, itching, and perspiration symptoms, symptoms of rash, atopy, rash, itching, and heat rash were improved. After the lapse of time, it was visually confirmed that it was considerably relaxed. These findings confirm that A.C.C. extracts comprise a clinically effective anti-inflammatory and anti-bacterial agent that alleviates symptoms such as diaper rash and fever and may therefore be an effective alternative to inflammatory diseases.

Enhancement of Anti-inflammatory Activities of Fermented Scutellaria baicalensis Extracts using Lactobacillus rhamnosus (유산균 발효를 통한 황금 추출물의 항염증 효과 증진)

  • This study was performed to investigate the anti-inflammatory activities of fermented Scutellaria baicalensis extracts using Lactobacillus rhamnosus. The extracts were WE (water extract at $100^{\circ}C$ for 24 h), EE (70% ethanol extract at $60^{\circ}C$ for 24 h), FWE (fermented and water extract at $60^{\circ}C$ for 24 h), FEE (fermented and 70% ethanol extract at $60^{\circ}C$ for 24 h). The cytotoxicity of the extracts was in the range of 11.2 ~ 15.6 % at 1.0 mg/mL concentratioin. The FEE showed the lowest activity at 1.0 mg/mL concentratioin. Compared to the WE, hyaluronidase inhibitory activity contents in the FEE increased to 9.2% at 1.0 mg/mL concentratioin. Nitric oxide production of WE, EE, FWE and FEE at 1.0 mg/mL concentration was mesured as 7.6, 7.9, 6.9, 6.4 ${\mu}M$, respectively. $PGE_2$ secretion of the human fibroblast of the FEE were lower than 810 pg/mL. Our results suggested that the extracts from fermentation process after 70% ethanol extraction had relatively high anti-inflammatory activities and that the Scutellaria baicalensis could be more extracted in FEE than others.

Anti-Oxidative and Anti-Inflammatory Activities of Carpinus pubescens Burkill Extract in RAW 264.7 Cells (RAW 264.7 세포에서 Carpinus pubescens Burkill 추출물의 항산화 및 항염증 활성)

  • In this study, to evaluate the anti-oxidative and anti-inflammatory effects of Carpinus pubescens Burkill ethanol extract (CPEE), we performed the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, reactive oxygen species (ROS) inhibition, and nitric oxide (NO) scavenging assays and an analysis of the related protein expressions. CPEE showed high DPPH radical scavenging activity and effectively increased ROS inhibition activity dose-dependently. Furthermore, CPEE induced the expression of the anti-oxidative enzyme heme oxygenase 1 and its upstream transcription factor, nuclear factor-E2-related factor 2, in RAW 264.7 cells. CPEE was associated with a reduction in NO production, which was induced by lipopolysaccharide treatment in a dose-dependent manner. The expression of inducible nitric oxide synthase (iNOS), an upstream regulator of NO production, was also inhibited. Taken together, these results suggest that CPEE has anti-oxidative and anti-inflammatory activities and could be useful as a potential anti-oxidant and antiinflammatory agent.

Anti-cancer and Anti-inflammatory Effects of Mung Bean and Soybean Extracts (녹두 및 대두추출물의 항암 및 항염증 활성)

  • The quinone reductase (QR) inducing activities of mung bean and soybean solvent extracts were compared using murine hepatoma cells (Hepa 1c1c7). The mung bean extracts (ethylacetate and ethanol) showed higher chemoprevention index values (7.88-8.22) than those of soybean extracts (2.9-5.2) from four different cultivars. The mung bean extracts also had significantly higher inhibitory effects (47-62% at 100 ${\mu}g$/mL) than the soybean extracts (15-42% at 100 ${\mu}g$/mL) against the production of nitric oxide and prostaglandin E2 in lipopolysccharide stimulated macrophage RAW264.7 cells without cytotoxicity. Among seven recovered fractions of mung bean ethanol extract obtained by C 18 silica flash column chromatography, the most non-polar fraction exhibited the highest chemoprevention index of 10.4.

Anti-inflammatory Effect of Scopoletin in RAW264.7 Macrophages (대식 세포인 Raw264.7 cell에서 scopoletin의 항염증 효과)

  • Scopoletin is a component of several plant such as Erycibe obtusifolia, Aster tataricus, Foeniculum vulgare and Brunfelsia grandiflora. It was reported to have anti-angiogenesis and anti-allergy effects. In this study, the anti-inflammatory effect of scopoletin was investigated in Raw264.7 cells, mouse macrophages. The effects of scopoletin on phagocytosis and nitric oxide (NO) production were investigated in lipopolysaccharide (LPS)-induced inflammatory responses. It was observed that scopoletin exerted inhibitory effects on both phagocytosis and NO production. In addition, scopoletin decreased the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) which were related to NO and prostaglandin E2 (PGE2) production. In particular, the expression of pro-inflammatory cytokines such as interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α). The expression levels of IL-1β, IL-6 were remarkably decreased by treatment with scopoletin. Furthermore, the content of TNFα produced by macrophage was decreased in the presence of scopoletin at 8 hr. These results indicate that the anti-inflammatory effect of scopoletin could exert by inhibiting the expression of pro-inflammatory cytokines in Raw264.7 cells stimulated with LPS. The above results suggest scopoletin could be a new remedial agent for anti-inflammation through inhibition of iNOS, COX-2, IL-1β, IL-6 and TNF-α expressions as well as supression of phagocytosis and NO production.

Anti-Inflammatory and Anti-allergic Effects of Gnaphalium affine Extract (떡쑥 추출물의 항염증 및 항알러지 효과)

  • Gnaphalium affine D. DON (GA) has been used as a vegetable as well as a folk medicine in East Asia. The antioxidant and anti-complementary activity of GA extract (GAE) has also been reported. However, little is known about its anti-inflammatory and anti-allergic effect and mechanism of action. In this study, we evaluated the inhibitory effects of GAE on the production of inflammatory mediators such as NO, $PGE_2$, TLR4, eotaxin-1 and histamine. Our results suggest that GAE inhibits the production of NO and $PGE_2$ by inhibiting transcriptional activation via the involvement of iNOS and COX-2. The LPS-induced expression of Toll-like receptor 4 (TLR4) was also attenuated. In addition, GAE inhibited A23187-induced histamine release from MC/9 mast cells. It also inhibited the production of eotaxin-1 induced by IL-4. Collectively, these results suggest that GAE may have considerable potential as a cosmetic ingredient with anti-inflammatory and anti-allergic properties.

Research Trends on the Therapeutic Potential of Cordycepin, an Active Ingredient of the Insect Fungus Cordyceps spp., for the Prevention of Sarcopenia (동충하초(Cordyceps spp.)의 유효 생리활성 성분인 cordycepin의 근감소증 예방에 대한 연구 동향)

  • Sarcopenia, a geriatric and multifactorial syndrome characterized by progressive systemic skeletal muscle disorder, may be associated with many comorbidities. Sarcopenia caused by a decrease in muscle mass and muscle strength is accompanied by the aggravation of various pathological conditions, and as life expectancy increases, its prevalence will continue to increase in the future. During the aging process, chronic oxidative stress and increased inflammatory responses act as major contributors to skeletal muscle loss. In addition, disruption of autophagy and apoptosis signals associated with dysfunction of mitochondria, which are essential for energy metabolism, accelerates the loss of muscle proteins. The pharmacological effect of cordycepin, a major physiologically active substance in the genus Cordyceps, which has been widely used for the prevention and treatment of various diseases for a long time, is directly related to its antioxidant and anti-inflammatory actions. In this review, we present the correlation between apoptosis, autophagy, protein catabolism, and satellite cell activity important for muscle regeneration using cordycepin for the prevention and treatment of sarcopenia. Although there have been few studies so far on the use of cordycepin for sarcopenia, previous studies suggest that cordycepin may contribute to inhibiting the age-related weakening of mitochondrial function and blocking the breakdown of muscle proteins. In addition, the protective effect of cordycepin on muscle cell damage is considered to be closely related to its antioxidant and anti-inflammatory activities. Therefore, it is considered that more continuous basic research is needed, focusing on the molecular biological mechanism of cordycepin, which is involved in the anti-aging of muscle cells.

Anti-Inflammatory and Antioxidative Effects of Acaiberry in Formalin-Induced Orofacial Pain in Rats (흰쥐의 악안면 통증에서 아사이베리의 항염증 및 항산화 효과)

  • Acaiberry (Euterpe oleracea Mart.) is widely diffused in amazon and is known that has high antioxidant capacity and potential anti-inflammatory activities. The aim of this study was to evaluate analgesic effects of acaiberry in formalin-induced orofacial pain through p38 mitogen-activated protein kinases (p38 MAPK) and nicotinamide adenine dinucleotide phosphate 4 (NOX4) pathway. Rats were divided into 5 groups (n=6); formalin (5%, $50{\mu}L$), formalin after saline (vehicle) or acaiberry (16, 80, 160 mg/kg, intraperitoneally). The nociceptive response was investigated all of groups, p38 MAPK or NOX4 were analysed at dose of 80 mg/kg of acaiberry in rat's medulla oblongata and adrenal gland. Results indicated that acai berry produced analgesic effect in a dose-dependent manner and significantly reduced formalin-induced nociceptive response at 15~40 min. Acaiberry (80 mg/kg) decreased the increased p38 MAPK activation and NOX4 expression in medulla oblongata and adrenal gland. Based on these results, acaiberry is believed to be useful for modulation of orofacial pain and its treatments because of its anti-inflammatory and antioxidative effects.

The Relationship Between the NF-${\kappa}B$ Activity and Anti-inflammatory Action of Surfactant in the Acute Lung Injury of Rats (백서의 급성폐손상에서 surfactant의 항염증작용과 호중구의 NK-${\kappa}B$ 활성과의 관계)

  • Background : The therapeutic effects of surfactants on acute lung injury derive not only from their recruiting action on collapsed alveoli but also from their anti-inflammatory action in the alveolar sapce. This study evaluated the anti-inflammatory action of a surfactant in an acute lung injury model of rats by neutrophils were recollected from the BAL fluid and the NF-${\kappa}B$ activity of the neutrophilic nuclear protein was evaluated. Methods : Male Sprague-Dawley rats weighing approximately 300 gram were divided into 3 groups, which consisted of 6 rats respectively. In the control group, normal saline(3ml/kg) was instilled into the trachea twice with 30 minute interval. In two other groups, acute lung injury was induced by the intra-tracheal instillation of LPS(5mg/kg). Thirty minutes later, either a surfactant(ST group; 30mg/kg) or normal saline(NT group: 3ml/kg) was instilled via the trachea. Twenty-four hours after the LPS instillation, the BAL fluid was retrieved to measure the WBC count and cytokine(IL-$1{\beta}$ and IL-6) levels. The neutrophils were isolated from the BAL fluid and the nuclear protein was extracted to evaluate the NF-${\kappa}B$ activity using a eletrophoretic mobility shift assay(EMSA). Results : The WBC count of the BAL fluid of the ST group($3,221{\pm}1,914{\times}10^3/{\mu}l$) was higher than that of the control group($356{\pm}275{\times}10^3/{\mu}l$)(p<0.05) and lower than that of the NT group($5,561{\pm}1,757{\times}10^3/{\mu}l$)(p<0.05)). The BAL fluid level of IL-$1{\beta}$ from the NT group($2,064{\pm}1,082pg/ml$) was higher than those of the ST group($360{\pm}234pg/ml$)(p<0.05) and the control group(0pg/ml)p<0.05) and control group($49{\pm}62pg/ml$)(p<0.05). The NF-${\kappa}B$ activity of the neutrophilic nuclear protein in the ST group and NT group was similar. Conclusion : The surfactant, attenuates the alveolar inflammation in the acute lung injury of rats model. However, its anti-inflammatory action does no't appear to be mediated by the inhibition of NF-${\kappa}B$ activity.

Development of Plant Anti-inflammatory Agents : Comparison of Anti-inflammatory and Analgesic Activities of Extracts from Lonicera japonica (식물성 항염증제의 개발 : 인동추출물에 대한 항염증 및 진통작용의 비교)

  • For developing plant anti-inflammatory agents, extracts from Lonicera japonica were obtained and evaluated for their anti-inflammatory and analgesic activities using acute/chronic inflammatory models and writhing tests. It was found that the extracts with a modified extraction method showed higher anti-inflammatory and analgesic activities than those of the extracts based on the ancient literatures.
