  • This study implicated that the CBT-AK5 purified from Lactobacillus casei (LAFTI L26) which showed antitumor activity in ICR mice. Hence, ICR mice were inoculated intraperitoneally Sarcoma-180 as well as CBT-AK5. Then we observed the life span and tumor increment of those ICR mice. Here our studies showed effect on two different way of treatment as intraperitoneally and orally treated in Sarcoma-180 infected ICR mice. We found that intraperitoneally treatment of Sarcoma-180 and CBT-AK5 is more effective than orally fed. The life span of the ICR mice were highly reduced after the inoculation of Sarcoma-180. Those effects like increment of body weight, the growth of ascites and solid were inhibited significantly after the treatment of CBT-AK5 in Sarcoma-180 infected ICR mice. Finally these studies suggested that CBT-AK5 isolated form Lactobacillus casei showed excellent antitumor activity against Sarcoma-180 infected ICR mice.

  • To obtain basic information on the potential use of Camellia japonica leaf as a raw material in functional food, leaf antioxidant and anticancer activities were investigated. The radical-scavenging activity of various solvent fractions from the leaf, as shown by the DPPH radical test, increased in a dose-dependent manner, with the water fraction showing the highest activity. The reducing power of various solvent fractions from the leaf was also dose-dependent, and, again, the water fraction showed the highest reducing power. The water fraction showed strong antioxidant activity in the linoleic acid test and was also capable of scavenging nitrite in a dose-dependent manner. Proportions of 92.15% and 95.61% of available nitrite were scavenged by the water and butanol fractions, respectively, at levels of $1,000{\mu}g/mL$. Both butanol and water fractions exhibited strong inhibitory effects on the growth of human lung and colon cancer cells. The total phenolic contents of the butanol and water fractions were 216.26 mg/g and 220.68 mg/g, respectively. High-performance liquid chromatography (HPLC) showed that quercetin and epicatechin were the predominant phenolic compounds in the water fraction. The activities of this fraction are attributable to the presence of these phenolic compounds, particularly quercetin and epicatechin.

  • The use of mushrooms has immense potential in many diverse applications. Until now, more than 3,000 species are consumed around the world, and more than 100 have shown promising clinical activity against cancer and other chronic diseases. Sparassis latifolia (formerly S. crispa) is an edible mushroom that harbors ${\beta}$-glucan reported to possess immunostimulatory and anticancer properties. However there have been no reports on the anticancer activity of hydrophobic fractions of S. latifolia. In this study, the anticancer activities of S. latifolia extract and hydrophobic fractions were investigated using AGS (stomach cancer), A529 (lung cancer), and HepG2 (liver cancer) cell lines. In cytotoxicity results of A529 cells, fractions of A2, A3, A4, A6, A7, A8, A9, and A10 in all 12 fractions show low $IC_{50}$ values. For HepG2 cells, A7 fraction results in the lowest $IC_{50}$ value while A7, A8, and A11 fractions show low $IC_{50}$ values in AGS cells. S. latifolia extract lead to low cell viability in cancer cells, compared to positive control of paclitaxel. A compound with molecular weight of 181 were detected using HPLC-MS but not identified yet. As a result, the hydrophobic fractions of S. latifolia EtOH extract would be a possible candidate as natural anticancer agents in the future.

  • Millettia erythrocalyx, a species of plant in the Fabaceae family, is widely distributed in the tropical and subtropical regions of the world, such as the Indies, China, and Thailand. The antiviral activity of flavonoids from M. erythrocalyx has been reported; however, the antioxidative and anticancer activities of M. erythrocalyx remain unclear. In this study, we evaluated the antioxidative and anticancer effects of ethanol extract of M. erythrocalyx (EEME) and the molecular mechanism of its anticancer activity in human hepatocellular carcinoma HepG2 cells. EEME exhibited significant antioxidative effects, with a concentration at 50% inhibition ($IC_{50}$) value of $2.74{\mu}g/ml$, as measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay; moreover, it inhibited cell proliferation in a dose-dependent manner in HepG2 cells. Cell cycle analyses showed that EEME induced HepG2 cell accumulation in the subG1 phase in a dose-dependent manner. EEME also induced apoptosis of HepG2 cells, with increases in apoptotic cells and apoptotic bodies, as detected by Annexin V and 4,6-diamidino-2-phenylindole (DAPI) staining, respectively. Treatment with EEME resulted in increased expression of First apoptosis signal (Fas), a death receptor, and Bcl-2-associated X protein (Bax), a proapoptotic protein, and the activation of caspase-3, 8, and 9, resulting in the cleavage of poly (Adenosine diphosphate-ribose) polymerase (PARP). Collectively, these results suggest that EEME may exert an anticancer effect in HepG2 cells by inducing apoptosis via both the intrinsic and extrinsic pathways.

  • This study aims to evaluate the antitumor effect of Adonis multiflora, one of the plants in the Ranunculaceae, on mice to which hepatoma cells were transplanted and to suggest its possibility as a candidate natural substance to replace antitumor drugs. We performed the MTT assay to assess the extract had a decrease in the growth rate of hepatoma cells depending on concentration. In particular, 100 ㎍/㎖ of the extract showed 40% of growth retardation rate. We assessed the autophagy activity to identify the inhibitory autophagy mechanism of tumor cells in the extract. This proved that the activity increases more as the concentration of the extract is higher. We conducted the Western blot test to confirmed the expression of two proteins LC3 and p62. The expression of p62 was in inverse proportion to the concentration of the extract whereas LC3-Ⅱ increased more as the concentration of the extract was higher. This showed that an increase in the autophagy relies on the conentration of the extract. We performed a test to discover the influence of the extracts on hepatoma cells transplanted to mice. The test proved that the extract triggers a significant decrease in the growth rate of tumor cells. Compared to the start of the test, the size of tumor cells with 50, 100 and 200 ㎎/㎏ of the extract respectively increased by 4, 3.7 and 3.5 times whereas in the controlling group by 6.3 times. The size of tumor cells in benign tumor controlling group increased by 3.1 times. This showed a significant decrease in the growth rate of tumor cells compared to the controlling group. We carried out the experiment of influence of the extract on the expression of two proteins LC3 and p62 in the tumor tissue transplanted into mice. The experiment showed that LC3-II increases more as the concentration of the extract is higher. However, there was a rapid decrease in p62 with 200 ㎎/㎏ of the extract compared to the controlling group. In this study, we proved that the autophagy activity of Adonis multiflora extract inhibits the growth of hepatoma cells by in vitro and in vivo experiments. In conclusion, the inhibitory autophagy mechanism of tumor cells in the extract can be used as a new treatment of antitumor.
