Background: This study examined the role of neutrophilc oxidative stress in an $O_2-induced$ acute lung injury (ALI). Methods: For 48 h, experimental rats were exposed to pure oxygen (normobaric hyperoxia) in a plastic cage. Forty-eight hours after $O_2$ breathing, the rats were sacrificed and the parameters for ALI associated with neutrophilic oxidative stress were assessed Results: Normobaric pure oxygen induced ALI, which was quite similar to ARDS. The $O_2-induced$ neutrophilic oxidative stress was identified by confirming of the increase in lung myeloperoxidase, BAL neutrophils, malondialdehyde (MDA), cytosolic phospholipase $A_2$ ($cPLA_2$) activity in the lung, histological changes and BAL cytospin morphology. Conclusion: In part, ALI-caused by oxygen is affected by neutrophils especially by the generation of free radicals.

The immunomodulatory activity of PJ-P, a polysaccharide fraction extracted from Paeonia japonica, were reported in our previous paper. In the present study, we investigated that PJ-P inhibited cancer growth through activation of macrophages. The activities of peritoneal macrophage to induce tumor necrosis factor (TNF)-$\alpha$, interleukin-1 (IL-1)$\beta$, interleukin-6 (IL-6) and interleukin-12 (IL-12) as well as to ingest fluorescence-latex microbeads were enhanced by treatment of PJ-P. Direct cytocidal activity of PJ-P against cancer cells was not shown. However, in vitro, peritoneal macrophages treated with PJ-P had an activity to kill cancer cells. Furthermore, PJ-P significantly prolonged the survival of mice implanted intraperitoneally with B16F0 mel-anoma cells. These results suggest that PJ-P could be a useful immunomodulator and assistant of anti-tumor agent.

Background: Peripheral blood monocytes are important immune effector cells that play a fundamental role in cellular immunity. In addition to their antigen-presenting and phagocytic activities, monocytes/macrophage produce a vast array of regulatory and chemotactic cytokines. Interleukin-8(IL-8), a potent neutrophil-activating and chemotactic peptide, is produced in large quantities by mononuclear phagocytes and may be an important mediator of local and systemic inflammation. Overexpression by IL-8 of such inflammation may be an important step of tissue injury frequently seen in inflammatory reaction. So it could be hypothesized that the agents which block the production of IL-8 can decrease the inflammatory reaction and tissue injury. To evaluate this, we described the effect of Dexamethasone, $PGE_2$, Indomethacin and Interferon-$\gamma$(IFN-$\gamma$) on IL-8 mRNA and protein expression from LPS-stimulated human peripheral blood monocytes(PBMC). Method: PBMC was isolated from healthy volunteers. To evaluate the effect of Dexamethasone, $PGE_2$ & Indomethacin, these drug were treated for 1 hour before and after LPS stimulation and IFN-$\gamma$ was only treated I hour before the LPS stimulation. Northern blot analysis for IL-8 mRNA and ELISA for immunoreactive IL-8 protein in culture supernatant were performed. We repeated above experiment three times for Northern blot analysis and two times for ELISA and got the same result. Results: 1) Pre- and post-treatment of Dexamethasone suppressed both the LPS stimulated IL-8 mRNA expression and IL-8 protein release in PBMC. 2) IFN-$\gamma$ pre-treatment suppressed the IL-8 mRNA expression and IL-8 protein release in unstimulated cells. 3) In LPS stimulated cells, IFN-$\gamma$ suppressed the IL-8 mRNA expression but IL-8 protein release suppression was not observed. 4) $PGE_2$ and Indomethacin exert no effect on the LPS-stimulated IL-8 mRNA and protein expression in concentration used in this experiment ($PGE_2;10^{-6}M$, Indomethacin; $10{\mu}M$). Conclusion: One of the mechanism of antiinflammatory action of Dexamethasone can be explained by the suppressing effect of IL-8 production in some extent and by this antiinflammatory effect, dexamethasone can be used to suppress local and systemic inflammation mediated by IL-8.
Background : Though mononuclear phagocytes serve as the final effectors in killing intracellular Mycobacterium tuberculosis, the bacilli readily survive in the intracellular environment of resting cells. The mechanisms through which cellular activation results in the intracellular killing is unclear. In this study, we sought to explore an in vitro model of a low-level infection of human mononuclear phagocytes with MAC and $H_{37}Ra$ and determine the extent of the lymphocyte dependent cytotoxicity of human monocytes and alveolar macrophages. Materials and Methods : The peripheral monocytes were prepared using the Ficoll gradient method from PPD positive healthy people and tuberculosis patients. The alveolar macrophages were prepared from PPD positive healthy people via a bronchoalveolar lavage. The human mononuclear phagocytes were infected at a low infection rate (bacilli:phagocyte 1:10) with MAC(Mycobacterium avium) and Mycobacterium tuberculosis $H_{37}Ra$. Non-adherent cells(lymphocyte) were added at a 10:1 ratio. After 1,4, and 7 days culture in $37^{\circ}C$, 5% CO2 incubator, the cells were harvested and inoculated in a 7H10/OADC agar plate for the CFU assay. The bacilli were calculated with the CFU/$1{\times}10^6$ of the cells and the cytotoxicity was expressed as the log killing ratio. Results : The intracellular killing of MAC and $H_{37}Ra$ within the monocyte was greater in patients with tuberculosis compared to the PPD positive controls (p<0.05). Intracellular killing of MAC and $H_{37}Ra$ within the alveolar macrophage appeared to be greater than that within the monocytes of the PPD positive controls. There was significant lymphocyte dependent inhibition of intracellular growth of the mycobacteria within the monocytes in both the controls and tuberculosis patients and within the macrophages in the controls(p<0.05). There was no specific difference in the virulence between the MAC and the $H_{37}Ra$. Conclusion : This study is an in vitro model of a low-level infection with MAC and $H_{37}Ra$ of human mononuclear phagocytes. The intracellular cytotoxicity of the mycobacteria within the phagocytic cells was significantly lymphocyte dependent. During the 7 days culture after the intracellular phagocytosis, the actual confinement of the mycobacteria was observed within the monocytes of tuberculosis patients and the alveolar macrophages of the controls as in the case of adding lymphocytes.

The objective of the current study was to determine the effects of arabinoxylane and polysaccharide peptide (PSP) on the immune cell functions. Both arabinoxylane and PSP increased plaque forming cell (10-15%) and rosette forming cell (10-30%) formation. Stimulation of macrophage with PSP (18%) and arabinoxylane (22%) resulted in increased phagocytic effects. Both arabinoxylane and PSP induced the tumor suppressive effect in mice injected sarcoma-180 cell intracutaneously. When the mice injected intraperitoneal cavity with sarcoma 180 cells, survival ratio was increased in the mice fed on arabinoxylane and PSP. The ratio of PCA was slightly decreased in the mice fed on PSP, especially fed on arabinoxylane than in the control mice. The concentrations of blood histamine were slightly decreased in the mice fed on arabinoxylane and PSP. These results suggest that the capacity of arabinoxylane and PSP seems to act as a potent immunomodulator causing augmentation of immune cell activity, and with the absence of notable side-effects, arabinoxylane and PSP could be used as a biological response modifier having possible therapeutic effects against immunological disorders.

암 발생의 원인 중 80% 이상이 식사습관과 환경오염에 있는 것으로 인정되고 있다. 음식물, 담배, 술, 대기오염, 스트레스, 자외선이 그 대표적인 원인물질로 꼽을 수 있으며 그 중에서도 매일 섭취하는 음식물이 가장 중요한 발암요인으로 지적되고 있다. 대장암과 유방암의 발생에 대한 Wynder 등의 역학조사에서도 식사습관이 암 발생에 중요함을 시사 하고 있다. 동물성 단백질과 지방의 다량 섭취가 대장암과 유방암의 발생을 증가 시키고 섬유질이 풍부한 곡류와 야채의 섭취는 대장암 발생을 억제한다는 상관관계가 밝혀졌다. 그러나 우리가 늘 섭취하는 음식물 자체는 대장암과 유방암을 유발하는 기능이 거의 없다. 섭취된 음식물이 암을 일으키려면 장내 부패 미생물의 분해작용에 의하여 발암물질로 변환되는 과정이 필요하다. 그 발암물질들이 장관으로 흡수 자극함으로써 암을 유발할 수 있다. 반대로 일부장내 미생물들은 장내 발암물질들을 무독화 하거나 숙주의 면역기능을 증강 시킴으로써 암 발생을 억제할 수도 있다는 사실을 간과해서는 안 될 것이다. Mitsuoka는 장내 미생물이 암을 유발하는 중요한 요인이라고 강조하였다. Veer 등은 유산균 발효유를 많이 섭취하면 유방암 발생이 억제됨을, 국제암연구위원회는 섬유질을 많이 섭취하고 있는 핀란드 쿠피오의 거주자들에게는 덴마크의 코펜하겐에 거주하는 사람들에 비하여 대장암 발생율이 1/4에 불과하고 분변내 유산간균수는 100배 높은 사실을 역학조사에서 밝혔다. 이 외에 유산균과 발효유제품의 항암효과에 대한 실험결과들이 많이 발표되었다. 여기에서는 유산균의 항암효과에 대한 지금까지의 관련 자료들을 요약, 정리하여 고찰하고자 한다.높은 당 함량을 나타냈으며, T-AS는 70.3%의 당과 7.8%의 단백질로 구성 되었다. GLG 대부분의 분획들은 60~93%의 glucose로 구성된 다당류 이었으며, 주로 $\beta$-glucose로 구성된 다당류 이었다. 아미노산은 Asp 및 Glu의 산성 아미노산과 Ala, Leu 등의 함량이 높게 나타났으며, 비알칼리 추출물에서 Ser과 Thr의 함량이 높게 나타났다. 다당류 T-AS는 평균 분자량이 2,000 kD와 12kD에서 주 peak를 나타냈으며, 수용성 분획의 평균 분자량은 12kD이고 비수용성 분획은 36~2,000 kD의 평균 분자량 분포를 갖는 것으로 나타났다. IR과 NMR 분석 결과 890 cm-1에서 흡수 peak를 나타내어 $\beta$-(1,3)0glucan과 $\beta$-(1,6)-glucan의 구조를 갖는 다당류로 확인 되었다. T-AS 분획은 C:H:O:N의 함량비가 38.9:5.7:49.6:1.84%이며, 이 물질의 융점은 163 $^{\circ}C$로 연한 갈색을 나타낸다. 분리된 GLG의 항암활성 기전 규명을 위해, in vivo 항암실험, 항보체 활성능, 항체 생성능, serum protein 분비능, 대식세포의 탐식능과 활성능 및 세포간 물질 분비 등의 상관관계를 조사하였다. 다당류 GLG 분획물들 가운데 항보체의 활성이 높았던 분획은 sarcome 180에 대한 항암 활성이 높게 나타났다. 다당류 T-AS의 보체 활성화 기작은 classical과 alternative complement pathway의 양 경로를 통해 활성화 되었다.
The olive flounder, Paralichthys olivaceus is susceptible to viral hemorrhagic septicaemia virus (VHSV) at $15^{\circ}C$ but no mortality at $20^{\circ}C$ even though the virus can grow well in vitro at $20^{\circ}C$. Thus, we designed an experiment to know immune response of olive flounder against VHSV when the host reared at $15^{\circ}C$ or $20^{\circ}C$. cDNA microarray analysis was performed to compare the gene expression patterns of the kidney cells between the host reared at $15^{\circ}C$ or $20^{\circ}C$. The expression of MHC class I, IL-8, myeloperoxidae and endonuclease G-like having function for the antigen presentation and chemokine-factor were up-regulted both the $15^{\circ}C$ and $20^{\circ}C$ during VHSV infection. MHC class II gene existing on antigen-presenting cells and B cell lymphocytes, immunoglobulin (Ig) genes and phagocytosis related genes were down-regulated at $15^{\circ}C$ but highly expressed at $20^{\circ}C$. It can be thought that innate immune related antigen presentation by MHC class I and phagocytosis reaction against VHSV are efficiently occur both the temperature but macrophage or B cell related antigen presentation via MHC class II fails to induce downstream immune reactions (adaptive immunity) to make antibody, and it can be one of the reason that causes high mortality only at $15^{\circ}C$.

The immunomodulating effects of polysaccharide extracted from Ganoderma lucidum(PSG) on macrophage were evaluated using murine macrophage cell line ATCC TIB 71 cells or peritoneal exudate cells of BALB/c mice. The cell were incubated with various content of PSG for 24 hours at 0.5% $CO_2$ incubator under varying experimental conditions. PSG stimulated the non-specific activites of macrophage such as mitotic activity and expression of surface interleukin-2 receptors by dose-dependent pattern with statistic significance(p<0.001): however, PSG had little immunoregulatory effects on cytokines derived from peritoneal macrophages of BALB/c mice. There were no significant changes in the se-cretion of interleukin-6, interleukin-6, or tumor necrosis factors(Tn) of PSG treated cells compared to the control group. But PSG increased secretion of cytokines(IL-1 and TNF) when the cells were primed and trigged with BCG and IFN-${\gamma}$.
An epulis was occurred on gingiva of 11-year old female dog, Yorkshire terrier. Our case had feature of ossifying epulis but there were a few multi-nucleated giant cells (MGCs). MGCs had osteoclast-like appearance and giant cell epulis usually appears at the site of tooth extraction. Therefore, we suggest that appearance of MGCs in our case may be due to phagocytosis pre-formed osteoid/bone or our case may be mixed epulides of ossifying and giant cell epulis by mixed stimulation of chronic gingivitis and trauma and in flammation by tooth extraction. Thus, MGCs have possibility enough to appear in ossifying epulis, but ossifying epulis accompanying MGCs has not been reported. Therefore, our case may deserves an attention as an unique case and will be helpful to study pathogenesis of giant cell containing lesion of the jaw.

This study investigated the antibacterial effects of GR ethanol extracts (GRE), sodium chlorate (SC) and a combination of GRE and SC (GS) on Brucella abortus (B. abortus). The antibacterial activities of GRE, SC and GS towards B. abortus were evaluated by incubating B. abortus with GRE, SC and GS. Following treatment with GRE, SC and GS, B. abortus survival and intracellular proliferation in macrophages were monitored. In the cellular cytotoxicity assay, GRE, SC and GS are not cytotoxic at concentrations less than $400{\mu}g/ml$, 15 mM and 0.6GS (1 of GS, GRE $1,000{\mu}g/ml$ + SC 30 mM), respectively. The viability of B. abortus was markedly decreased in a dose-dependent manner in all treatment groups. In addition, B. abortus intracellular proliferation within macrophages was significantly reduced in cells treated with GRE ($400{\mu}g/mL$), SC (15 mM) and 0.5GS (GRE $500{\mu}g/mL$ + SC 15 mM) after 48 hr-incubation (GRE, p < 0.01; SC and 0.5GS, p < 0.001). Especially, in the treatment of GS, the synergistic effect of GRE and SC treatment on B. abortus in macrophage was observed. In conclusion, GS is useful as an antibacterial candidate against B. abortus, and can be applied in the field of meat and milk hygiene.

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
