For the biological production of hydrogen, a new fermentative hydrogen-producing bacterium, Enterobacter sp. SNU-1453, was isolated from a domestic landfill. During the culture of this bacterium, pH significantly decreased with the accumulation of various organic acids, and consequently this inhibited the production of hydrogen. It was found that the metabolic flux in this bacterium depended on the pH and affected the hydrogen production. A butanediol pathway was dominant during the fermentation when pH was not controlled. By controlling the pH at 7 this pathway can be shifted to a mixed acid pathway, which is favorable to the production of hydrogen.
The analytical response properties of two types of continuous flow-through electrode system as fulfide ion detectors are examined and directly compared their reponse characteristics under the optimal conditions. In both detection systems, observed peak potentials are logarithmically related to the sulfide ion concentration and at least twenty samples per hour can be determined. In the pH electrode method, the pH of the flowing recipient stream leaving the dialyzer was monitored. The designed system involves the use of continuous flow gas dialyzer in conjunction with the tubular polymer membrane electrode. In this method, optimal experimental conditions are recipient of mixture of $5.0 {\times} 10^{-5} M NaOH + 5.0 {\times} 10^{-3} M$ NaCl and diluent of 0.10 M $H_2SO_4$, and all flow rates of recipient stream, diluent stream, and sample are 1.0 ml/min. In the sulfide ion electrode method, a commercially available sulfide ion-selective electrode was used to detect sulfide ion in the flow-through cell. The optimal flow rates of sulfide anti-oxidant buffer (3.5 g ascorbic acid and 7.6 g $Na_2EDTA$ dissolved in 1.0 M NaOH solution 1 l) and sample were 1.4 ml/min and 1.0 ml/min, respectively.
In the submerged fermentation of fungi, it was known pH had significant effect on the citric acid production. Various growth conditions were applied with different buffer on citric acid production by Aspergillus niger NRRL 567 grown on peat moss to find the optimum pH and most effective buffer solution. The initial pHs of different buffer solutions significantly influenced on the citric acid production and A. niger NRRL 567 produced citric acid more efficiently at high pHs. A phosphate buffer and a carbonate buffer with pH 8.6 and pH 10.0 were identified as suitable buffer solutions for citric acid production. The maximal citric acid production of 564.3 g/kg solid substrate was achieved employing carbonate buffer at pH 10.0.

The aim of this study was to investigate the efficiency of temperature, light intensity and pH on the growth rate of Chlorella vulgaris (C. vulgaris). The size of C. vulgaris (FC-16) was $3-8{\mu}m$, having round in shape. The cells of C. vulgaris (FC-16) was cultured in the Jaworski's Medium with deionized water. To evaluate the efficiency of temperature, light intensity and pH on the growth rate of C. vulgaris, six different fractions of temperature ($10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, $35^{\circ}C$), various light intensities ($100-800{\mu}Em^{-2}s^{-1}$) and seven different fractions of pH (3, 4, 5, 6, 7, 7.5, 9) were prepared. The growth rate of C. vulgaris cultivation was approximately 5.2 to 5.5 times faster, the concentration of Chlorophyll a was also 5 to 5.5 times higher, and cell volume per unit area was 14% higher at $25^{\circ}C$ to $30^{\circ}C$ than those at $10^{\circ}C$. Therefore, the optimal temperature for cultivation of C. vulgaris was estimated $25^{\circ}C$ to $30^{\circ}C$. The growth rate of C. vulgaris increased slowly up to 5 days, exploded after 5 days until 15 days, and then stoped after that. The optimum cultivation period of C. vulgaris was estimated as 15 days. The optimum pH for the growth rate of C. vulgaris was determined pH 7 to 7.5.

Nonliving Absidia coerulea and Thraustochitrium sp. were used as biosorbents to remove lead and cobalt that are one of representative pollutant in wastewater and radioactive liquid waste. The optimum pH range for maximum lead and cobalt removal was increased 6.5~11.4 and 8.6~12.0 for Absidia coerulea and 4.2~10.5 and 8.9~11.6 for Thraustochitrium sp. to compared to biosorbent-free control, pH of 8.4~11.2 and 10.5~11.5, respectively. With 1 g biosorbent/L at initial solution pH 5.0. Absidia coerulea and Thraustochitrium sp. took up lead from aqueous solutions to the extent of 104 and 125 mg/g biomass, respectively, whereas Absidia coerulea and Thraustochitrium sp. at initial pH 6.0 took up only 2 and 20 mg/g biomass of cobalt, respectively. For initial 500 mg Pb/L at initial pH 5.0. optimum amount of biosorbent for maximum lead uptake was 0.2 g/L for Absidia coerulea and Thraustochitrium sp., whereas optimum 3.0 g biosorbent/L was needed for initial 200 mg Co/L at initial pH 6.0. Absidia coerulea and Thraustochitrium sp. had higher adsorption capacity for lead than that of cobalt.
The optimum nutritional and cultural conditions of polygalacturonase by Ganoderma lucidum in liquid culture were studied. The optimal temperature, pH, and the duration of culture for production of the enzyme was $30^{\circ}C$, 5.5 and 14 days, respectively. The maximal production of the enzyme was obtained in a synthetic medium containing 10 g of pectin, 10 g of soluble starch, 1 g of yeast extract, 2 g of peptone, 1 g of phenylalanine, 2 g of $KH_2PO_4$, 0.2 g of $MgSO_4{\cdot}7H_2O$, 0.05 g of $CaCI_2$ and 100 g of $thiamin{\cdot}HCI$ in 1000 ml of distilled water.
The culture conditions for the enhanced mycelial of Clavicorona pyxidata DGUM 29005 were investigated. The optimal temperature and pH for the mycelial growth were $24^{\circ}C$ and 5.0, respectively. It was shown that trehalose was the best supplement of carbon sources in Czapek-Dox medium as a minimal medium for enhanced mycelial growth. In general, inorganic nitrogen sources were better than organic ones for mycelial growth. Calcium nitrate was the best out of the inorganic nitrogen test. The appropriate phosphorous and vitamin were $Na_2HPO_4$ and p-aminobenzoic acid, respectively. After the mycelial of C. pyxidata DGUM 29005 was cultivated at $24^{\circ}C$ for 20 days in MEM broth(pH 5.0), the specific activities of both exomycelial and endomycelial enzymes were determined. Among the exomycelial enzyme assayed, the specific activity of laccase was much higher than those of other enzymes. However, little or no enzyme activities of ${\alpha}$-amylase, chitinase, lipase and pretense were found.
A halophilic bacterium, H. subglaciescola DH-1, grew at 2.0 M salinity, but did not grow at 0.8 M salinity when cultivated at higher temperature ($40^{\circ}C$) than optimum ($30^{\circ}C$). When the cell extract of strain DH-1 was heated at $50^{\circ}C$ for 60 min in the absence of NaCl, isocitrate dehydrogenase and malate dehydrogenase lost their activities, but when it was heated in the presence of 2.0 M NaCl, the activity was maintained. Meanwhile, the cell extract of E. coli did not catalyze the reduction of $NAD^+$ to NADH coupled with the oxidation of isocitrate and malate at higher salinities than 1.0 M. The pH range for DH-1 was 7 to 10, and that for E. coli was 5 to 9. DH-1 was not grown in conditions with sodium salts other than NaCl.

The strain FJ1 isolated from a rotten wood showed high activity to hydrolysis of cellulosic materials. The strain produced largely enzymes related in hydrolysis of cellulose and hemicellulose, such as CMCase, xylanase, ${\beta}-glucosidase$ , and avicelase. The culture conditions(pH, temperature, inoculation concentration) and substrate specificity to various cellulosic materials were examined to elevate productivity of the enzymes. The enzyme activities of CMCase and xylanase were 13.5U/ml and 24.3U/ml in agitation culture using Mandel's medium, respectively. The high activity of the enzymes was earned when mixed cellulosic materials of rice straw, sawdust, and pulp as substrates, indicating that the strain FJ1 could use crystalline substrates.
This study was carried out to optimize the fermentation conditions for direct alcohol fermentation of xylose by Pichia stipitis CBS 5776. The best cell growth and the ethanol production were obtained under 0.05 VVM aeration and 300rpm agitation at $30^{\circ}C$ using 100 g/l xylose medium of the initial pH 5.0. In the above condition, the maximum specific growth rate and maximum cell concentration were 0.14hr-1 and $1.3 \times109$ cells/ml, respectively. Pichia stipitis CBS 5776 also produced 40.2g/l ethanol utilizing about 96% of 100g/l xylose after 72hr fermentation. At this point, the overall volumetric ethanol productivity was 0.56g/1-hr and the ethanol yield was 0.42 g-ethanol/g-xylose consumed, which corresponds to 82% of the theoretical yield.
