Chestnut leaf (CL) exhibits numerous pharmacological effect including anti-allergy and anti-microbial properties. However, the anti-wrinkle effect of CL is not completely understood. In this study, to find a possible explanation for the anti-wrinkle effects of CL, we evaluated the effects of CL on radical scavenging activity, elastase inhibition activity and collagenase inhibition activity. CL was extracted with various solvents including water (CLW), 70% ethanol (CLE) and 70% acetone (CLA). The results showed that CLW, CLE, and CLA have the radical scavenging activity. In addition, we showed that CLW, CLE, and CLA have the elastase inhibition activity and collagenase inhibition activity. Consequently, the CL has a potent anti-wrinkle effect and it could be a useful cosmeceutical product for anti-aging purposes.

  • In this study, we conducted research to find anti-wrinkle skin care ingredients from natural sources via the measurement of procollagen type 1 levels in the medium of normal human dermal fibroblast (NHDF) and found that a Nelumbo nucifera leave extract (NLE) was as the best effective ingredient. By the high performance liquid chromatography analysis, we confirmed isoquercitrin was one of the main compounds of NLE. Moreover, it also increased the procollagen type 1 production without cytotoxicity of NHDF. NLE and isoquercitrin exerted free radical scavenging activity. Especially, isoquercitrin exhibited strong intracellular antioxidant capacity in human skin-derived cells, HaCaT and NHDF. Finally, we performed clinical test for the inhibitory effect of NLE on skin wrinkle formation. A randomized study was conducted in 22 healthy female volunteers, aged between 30 and 65 yrs, with moderate to moderately severe facial wrinkles. Volunteer applied a 1.7% NLE cream (isoquercitrin 51 ppm) and a control cream at each facial side (left/right) twice daily for 8 weeks. The 1.7% NLE cream improved skin roughness through reducing the wrinkle of the craw's feet significantly without any skin side effect. Our results demonstrate that NLE and isoquercitrin can increase the collagen production and exert antioxidant activity. Therefore, we expect that the new non-toxic herbal extract, isoquercitrin-containing NLE will be interesting natural ingredient of cosmetics with anti-wrinkle efficacy.

  • In this study, Camellia sinensis Leaf cultivated in Boseong was extracted by vacuum distillation and hydrothermal extraction methods, and we performed in vitro experiments such as antioxidant, anti-wrinkle, whitening, and antibacterial activity test and HPLC analysis. Wrinkle improvement (elastase inhibition assay) and whitening effect (mushroom tyrosinase inhibition assay) of Boseong vacuum distilled extract have excellent results and IC50 values of vacuum distillation extract was measured at a lower concentration than that of the hydrothermal extract. The antioxidant activity of the Boseong hydrothermal extract was excellently measured in DPPH radical and ABTS+ radical scavenging activity, SOD-like activity, and total polyphenol content. In the antibacterial experiment, the Boseong hydrothermal extract formed a clear zone for the Staphylococcus aureus and Cutibacterium acnes strain. As a result of HPLC analysis, EGCG and caffeine in the hydrothermal extract were 40.29 mg/g, 9.94 mg/g respectively, and caffeine in the vacuum distilled extract was 3.40 mg/g. Therefore, we examined that Boseong Camellia sinensis vacuum distilled extract has excellent anti-wrinkle and whitening effect, and its hydrothermal extract has good antioxidant and antibacterial efficacy.

  • Ginseng berry (GB) contains Ginsenoside Re and has anti-inflammatory and anti-wrinkle properties. In this study, TLC fractions 1, 2, and 4 of the ginseng berry amino acid complex were identified and analyzed by HPLC. And identified a peptide (AP-1) by LC/MASS analysis of fraction 1. The anti-inflammatory activity was confirmed by investigating the inhibitory effect of AP-1 on NO production. In addition, collagen synthesis using procollagen type I C-peptide (PIP) ELISA kit was 50% higher effective than that of the control group. From these results, the peptide isolated from ginseng berry amino acid complex is considered to have anti-inflammatory and anti-wrinkle effect, and may be useful as an anti-inflammatory and anti-aging cosmetic raw material.

  • Chronic exposure of solar ultraviolet (UV) light to human skin results in photoaging with wrinkle formation. This study was performed to investigate anti-wrinkle effects of Lindera obtusiloba extract (LO) on UVB-induced wrinkle formation. We first measured cell proliferation and type I pN collagen synthesis activities in normal human dermal fibroblasts. Cell proliferation and type I pN collagen synthesis were increased by 33.8 % and 91.8 %, respectively, compared with no treatment control. SKH-1 hairless mice were topically applied 5 % LO solution for 10 weeks with UVB irradiation three times a week. After 10 weeks, a visual assessment and replica assay were performed on each mouse. According to visual assessment of close-up photos and skin replica, application of 5 % LO solution inhibited UV-induced wrinkle formation in mouse skin as compared to the vehicle-applied control mice. These results indicated that LO could protect skin wrinkle formation caused by chronic photo-irradiation in hairless mice.

  • The study studied dermal protection and crease improvement from the sunlight of the lipid extract and narinjin. Sunlight was investigated in HR-1 (motherless mice) to identify changes in epithelial thickness and changes in collagen fibers, which account for around 90% of dermis, as the inhibitory efficacy of collagenase dissolving collagen also plays an important role in wrinkles. The experiment was validated using narinjin and jisil extract. First: The components of jisil and narinjin were analyzed. Second, antioxidant capabilities were confirmed with DPPH. Third: The inhibitory activity of collagen was measured. Studies have shown that the skin's upper skin thickness suppression of dermal extracts and narinzine has increased and that collagen thicknesses and wrinkles have decreased significantly compared to controls.

  • In this study, the skin-improving effect was investigated through the oral intake of DuOligo (51.67% lactulose and 15.8% galactooligosaccharides). Thirty seven healthy women (40~60 in ages) were divided into placebo group (dextrin) and treatment group (DuOligo group), and each group was given a sample for oral intake for 8 weeks. After that, corneometer value, transepidermal water loss (TEWL), melanin index, erythema index, and wrinkle index were measured. As a result, moisture content of DuOligo group increased 38.22% than control group after 8 weeks significantly (p < 0.01). The TEWL of control group decreased by $3.39g/h/m^2$ after 8 weeks but the TEWL of DuOligo group decreased by $5.32g/h/m^2$ after 8 weeks. The melanin index of the control group was significantly increased with times, but the melanin index of the DuOligo group did not show any significant difference with times. The total wrinkles, length of total wrinkles, number of wrinkles, and depth of wrinkles significantly decreased (p < 0.05) compared to the control group. These results suggested that the applicability of DuOligo as a skin improving functional material was confirmed.
