Physioecological Characteristics and Phylogenetic Relationships Based on Multivariate Analysis on Colocasia antiquorum Schott (토란의 생리.생태적 특성 및 다변량 해석법을 통한 유연관계 분석)

  • Twenty-four major ecological and physiolosical characters of the 54 accessions of taro(Colocasia antiquorum) were observed in the growing seasons of 1995 and 1996 in $Ch{\breve{o}}nju$, Korea. The average days to emergence of the 54 accessions were 21.7 and the accession which had the shortest days to emergence was Kurye #2. Petiole color of most accessions was green, except $Ich{\breve{o}}n$ #1, Iri #1, Puan #1 and Taiwan #1 which had purple petioles. All the accessions had cormels except Taiwan #1 which had long runners without cormels. Average yield of the accessions was 3,719 kg/10a and total carmel weight, average carmel number, average carmel and average corm weight were 787g/plant, 39.2/plant, 20.3g/cormel, and 263.6g/plant, respectively. Accessions $Ch{\breve{o}}ngju$ #1 and Wanju #1 showed superiority in the three important characters, total carmel weight/plant, average carmel weight, and corm weight. Multivariate analysis for the 24 characters indicated that petiole color and carmel type had relatively higher coefficient of variation, 56.8 and 44.4 %, respectively. Fifty four taro accessions could be classified into 11 groups with D(${\sqrt{D^{2}}$) value of 110 on the basis of phenotypical characteristics. Most accessions were inclued in Group II. The Group I in which Taiwan #1 was included was genetically the most distant from the other groups. Accessions with purple petioles were grouped into the three different subgroups in which green petiole accessions were not included, indicating high dissimilarity between the different petiole color accessions. All the accessions grouped into the Group VIII, $Ch{\breve{o}}ngju$ #1, Wanju #1, Wanju #3, and $Taech{\breve{o}}n$ #2, had higher bioogical yield. Some accessions collected from the same district showed a high dissimilarity by being classified into the different subgroups.

Changes in the Physiological Activities of Four Sweet Potato Varieties by Cooking Condition (고구마 네 가지 품종의 조리방법에 따른 생리활성 변화)

  • The present study was performed to investigate antioxidant, anticancer, and antimicrobial activities of four Korean sweet potato variaties and to identify the changes in these biological activities under different cooking conditions. Total polyphenol content was 3.8-73.6 mg/g in 80% ethanol extracts of sweet potatoes. The polyphenol content was highest Sinjami variety (p < 0.05). Radical scavenging activity against DPPH and $ABTS^{{\cdot}+}$ was high in Sinjami (p < 0.05) and the ethanol extract from Sinjami also showed effective superoxide dismutase (SOD)-like activity, which decreased significantly by steaming and roasting (p < 0.05). Ethanol extracts from the four sweet potato variaties did not inhibit cancer cell growth in MCF-7 or HepG2 cells at concentrations of 1, 10, and $100\;{\mu}g$/mL. Of the investigated sweet potato variaties, only Sinjami exhibited strong antimicrobial activity against Escherichia coli, Staphylococcus aureus, and Salmonella typhimurium. The antimicrobial activity of Sinjami against E. coli, St. aureus, and S. typhimurium decreased following steaming and roasting (p < 0.05). These results indicate that the Sinjami Korean sweet potato had higher polyphenol content, radical scavenging activity, SOD-like activity, and antimicrobial activity than those of the other variaties and consuming raw Sinjami might be beneficial for maintenance of biological activities.

  • Two individuals ($sp_1$, $sp_2$) of purple and one individual ($sd_1$) of red hearted flower were selected from 18 years old Hibiscus syriacus trees obtained from the seeds treated with colchicine, and their morphological and physiological characteristics were investigated and following results were obtained. 1. The somatic chromosome number of the selected individuals, $sp_1$, $sp_2$, and $sd_1$ were 2n=160, while that of the check tree was 2n=80, indicating that the selected individuals, $sp_1$, $sp_2$ and $sd_1$ were tetraploid. 2. Peroxidase isoenzyme bands of high activity in selected individuals, $sp_1$, $sd_1$ and check tree were mostly in cathode, fixed band was f and v bands, and frequency of each band and their activity were not different between selected individuals, $sp_1$ and $sd_1$ and check tree. 3. The flowers of $sp_1$ individual were large in size and more dark purple than check tree's. The flowers of $sp_2$ individual were not increased in size, but they were dark purple and red heart at the base of the petal was expanded to 2/3 of the petal length. The flower of $sd_1$ individual was also large and some of the red lines from the petal base were extended to 2/3 of the petal length, which was much longer than those of the check tree. 4. Thickess of leaves, length of guard cells, diameter of pollens, wood fiber lengths and woody fiber widths were all increased in $sp_1$, $sp_2$ and $sd_1$ as compared to those of the check tree. 5. Survival percentage of cuttings was 80% with $sp_1$ and 36% with $sd_1$, and their growth performance were inferior to control in their second growing season.

  • Our review begins with the maize hybrid for grain, called 'Seakso 1,' which was developed in 2008 by the Gangwon Agricultural Research and Extension Services in Korea, and subsequently registered in 2011. In this study, we aimed to investigate the lipid metabolic enzyme activity and inhibitory effect on the adipocyte differentiation, in 3T3-L1 cells of the identified Seakso 1 corn husk and cob extracts (EHCS). We investigated the pancreatic lipase inhibitory effect and anti-adipogenic effect of EHCS.The lipid accumulation and adipocyte differentiation were measured by the procedure of Oil Red O staining, Real-time PCR and the Western blot analysis. The pancreatic lipase inhibitory activity of EHCS was measured at higher levels than those of the positive control (orlistat) at 100, 500, and $1,000{\mu}g/mL$. In particular, EHCS was noted as being significantly inhibited and including a measured adipocyte differentiation and lipid accumulation, when treated during the adipocyte differentiation process in 3T3-L1 cells. Based on the Oil Red O staining, EHCS inhibited lipid accumulation at 19.19%, 33.30% at $1000{\mu}g/mL$, $2000{\mu}g/mL$, respectively. The real-time PCR and Western blot analysis showed that EHCS significantly decreased in the mRNA expression and protein level of obesity-related factors, such as peroxisome-proliferatorsactivated-receptor-${\gamma}$ ($PPAR{\gamma}$) and CCAAT enhancer-binding-proteins ${\alpha}$ ($C/EBP{\alpha}$). This study potentially suggests that the Saekso 1 corn husk and cob extracts may improve lipid metabolism and reduce lipid accumulation.

Change of neutrophil count after treatment of intravenous immunoglobulin in children with idiopathic thrombocytopenic purpura (소아 특발혈소판감소자색반병에서 면역글로불린 투여 후 호중구수치의 변화)

  • Purpose : The aim of this study was to investigate the incidence and course of neutropenia following intravenous immunoglobulin (IVIG) therapy in children with idiopathic thrombocytopenic purpura (ITP). Methods : From January 2001 to June 2006, fifty-four patients with ITP were enrolled in this study. Forty-two of 54 patients were treated with IVIG, while the other 12 were treated with anti-D immunoglobulin (Anti-D Ig). Post-treatment absolute neutrophil counts (ANC) were compared between patients who received IVIG and those who received Anti-D Ig. Comparison of post-treatment ANC between patients who treated with two different IVIG regimens (400 mg/kg/day for 5 days and 1 g/kg/day for 2 days) was also performed. Results : Pretreatment ANC were not significantly different between the two treatment groups. After treatment with IVIG, 32 out of 42 patients (76.2%) showed more than 50% decrease of ANC from the baseline. On the other hand, only 2 out of 12 patients (16.7%) showed more than 50% decrease of ANC from the baseline after treatment Anti-D Ig. No significant difference was observed in the decline of ANC between the first IVIG treatment (42 patients) and repeated IVIG treatment groups (7 patients). There was no statistical difference in post-treatment ANC between patients who treated with two different IVIG regimens. The neutropenia induced by IVIG had resolved spontaneously in 38 out of 39 patients (97%) after several days. Conclusion : Neutropenia following IVIG administration may not be an uncommon finding in children with ITP. It seems to be transient and self limited.

Antioxidant Activities of Extracts Prepared from Sweet Potatoes with Different Flesh Colors (육질색 종류별 고구마 추출물의 항산화 활성)

  • Antioxidant activities of ethanol extracts from three types of sweet potatoes with distinctive flesh color (white, orange, and purple) were investigated. Total phenolic content of purple-fleshed sweet potato (PF) was always 2- to 5-fold higher than those of white-fleshed (WF) and orange-fleshed sweet potato (OF). 2,2-diphenyl-1-picryl-hydrazyl free radical scavenging activity of 70% ethanol extracts was highest ($EC_{50}$ = 0.179 mg/mL) for PF and lowest ($EC_{50}$ = 2.127 mg/mL) for OF. And then, reducing power was also highest ($EC_{50}$ = 0.236 mg/mL) for PF and lowest ($EC_{50}$ = 1.587 mg/mL) for WF. Antioxidant activities of 70% ethanol extracts were closely related to the amount of total phenolics. These results imply that antioxidant activity of PF is 6.7 to 11.9 times higher than those of WF and OF.

Characteristic Comparison of the Composition Classification on Potash Glass Beads Excavated in Korea (한국 포타쉬 유리구슬의 조성 분류에 따른 특성 비교)

  • In this study, the potash glass beads of 281 samples in 30 sites analyzed until now were classified according to the chemical composition. And the color, size, manufacturing technique and distribution period were compared. Korea potash glass beads are divided into 3 types depending on the stabilizer content. I, II type is the CaO and $Al_2O_3$ content of less than 5%. Relatively, I type has a high CaO and II type has a high $Al_2O_3$. In contrast, III type comprises more than CaO 5%. I, II type is the saltpetre, III type is estimated using plant ash as row material of flux. A review of the properties by type, I type is cobalt blue, copper blue and purple beads. The outer diameter is sized to the range 1.4~7.4mm. Also it was produced by the drawing technique. It was used continuously from BC 1C until around AD 6C. On the other hand, II type is outer diameter of 1.9~3.6mm and a copper blue beads. manufacturing technique is the same as the I type. This seems to have been in use since around AD 1C to 4C. Finally, III type is brown, colorless, amber beads and an outer diameter of about 10mm. It was formed by winding technique and appeared in the tombs of Goryeo and Joseon Dynasty. As a result, 3 types of potash glass beads distributed in Korea is likely to flowing through the various trade routes from different provenances.

A New Stable Bolting, High Quality and High Yielding Variety ‘Anpung’ of Angelica gigas Nakai (저추대 고품질 다수성 참당귀 ‘안풍’)

  • Korean angelica(Angelica gigas Nakai), a oriental medicinal plants is important and in great demand in Korea. This crop is a biennial which is to die after flower, so as could not harvest the root. A new variety 'Anpung' was developed from the selection of the local collecting population by medicinal crop team at NCES in 2001. This showed stable bolting during its cropping year and have high quality and yield potential. The color is greenish purple in stem and red purple in flower. It was longer in plant height and root length, thicker in root diameter, more leave and latheral roots comparing with the check variety 'Manchu'. The dry root was 72 g/plant showing heavier than check variety of 65 g/plant. It showed such low bolting rate as 0.5%, on the other hand 49.3% in landrace when it cultured by transplanting with heating nursery seedlings, and $0{\sim}15%$ with field nursery seedlings. The dry root yield of the variety was averaged 3,630 kg/ha out yielding the check variety by 11% at the three regional yield test from 1999 to 2001. Finally, 'Anpung' showed higher content of extracts up to 46.3% in 'Anpung' and 45% in check variety, decursin and decursinol contents was 3.48% and 2.79%, showing slightly higher than check variety.

New Variety 'Johyun' of Coix lacryma-jobi var. mayuen Stapf with Early Maturity and Short Plant Height (율무 조숙 단간 다수성 신품종 '조현')

  • ‘Johyun’ was a new adlay variety which was developed from three way crosses of (Suwon-6xOkayama)x UCN300-25 at the Northern Agricultural Research Station, Gyeonggi Province ARES. This variety had purple sheath and green leaf blade in primitive growth stage and green leaf and culm in heading stage. Colors of pistil and seed coat were milky-white and dark-brown, respectively. Plant height of Johyun was 179 cm and shortened 22 cm less than Yulmoo-1 which was tested by check variety. Johyun shortened 8 days of heading date and 15 days of ripening date compared with Yulmoo-1. Yield of Johyun was 320 kg/10a of hulled grains in three regional yield trials and 18% higher than Yulmoo-1. This variety was adaptable to single cropping in central and northern region of Korea.

A New Cultivar Hibiscus syriacus 'Red Bohanjae' with Small Violet-Pink Flowers for a Pot Plant (분화용 무궁화 자색 단심 'Red Bohanjae' 육성)

  • A new cultivar of Hibiscus 'Red Bohanjae' (Hibiscus syriacus 'Red Bohanjae') was developed after crossing between Hibiscus syriacus 'Andong' (♀) with white flowers and Hibiscus syriacus 'Samchully' (♂) with pink flowers to improve flower quality and growth habit. The Hibiscus breeding program was initiated in 2002, and H. syriacus 'Red Bohanjae' was preliminarily selected as '02-RV-17' in 2003 due to its stable flower quality with rich pink flowers and small tree type. The selected line was further evaluated for growth characteristics, leaf shape, leaf size, flowering characteristics, and tested for distinctness, homogeneity, and stability during 2004-2007. The characteristics preserved by grafting. 'Red Bohanjae' showed a small growth habit with slightly upright branches. The tree had potential use as a potted plant or bonsai because of its dwarf form. The new cultivar 'Red Bohanjae' had violet-pink color flowers (RHS N78C) with red eye spots, small size (9.2 cm) and spatulate (I-type) petals. Leaves were 4.2 cm long and 2.3 cm wide. After the evaluation of plant characteristics for 3 years (2005-2007), it was registered as cultivar 'Red Bohanjae' (4118, plant variety protection rights number) in 2012. 'Red Bohanjae' having violet-pink flowers with red eye and dwarf tree form seems to be suitable for pot or bonsai cultivation in both indoor and outdoor conditions.
