Searching for Insecticidal Metabolites Produced by Insect Pathogenic Fungi (곤충기생성 진균이 생산하는 살충성 생리활성물질의 탐색)

  • During the screening of biological control agents for insect pests in the greenhouse, 70 dead insect-related fungi were isolated and tested insecticidal effect of their culture filtrates was tested. From this studies, CNAB-63 isolate showed strong control effects against the cotton aphids and the two-spotted spider mite as 65.19% and 77.55%, respectively. The insecticidal active compound of CNAB-63 isolate was purified from culture filtrate by silica gel chromotography, thin layer chromatography and HPLC. Purified active compound (CNAB) showed control effects against the two-spotted spider mite with 63.45% at $100\;{\mu}g/ml$ concentration. However, it did not exhibit antimicrobial activity against fungi and bacteria.

The Characteristics of Soil Oribatid Mite(Acari: Oribatida) Communities as to Differences of Habitat Environment in Mt. Jumbong, Nature Reserve Area in Korea (점봉산 천연보호림에서 서식환경 차이에 따른 토양날개응애 군집특성)

  • This research was conducted every month from June 1994 until August 1996 with the aim to understand the ecosystem structure through the analysis of oribatid mite community structure in soil subsequent to environmental difference of its habitats located at northward & southward slopes adjacent to each other at an altitude of 1,000 meters of Mt.Jumbong, which is a natural reserved forest, remaining intact. There appeared a significant difference [t-test, p<0.06] in comparison of the number of the species and individuals of Oribatid mite species which were collected and identified at two survey areas. The mean density and the number of the species collected and identified at the northward slopes, and southward slopes were $99.2{\pm}17.6,\;234.2{\pm}62.6$ and $24.7{\pm}3.0,\;40.8{\pm}5.8$, respectively. Species diversity index(H') was higher at the southward slopes($3.09{\pm}0.11$) than at the northward slopes($2.71{\pm}0.13$). The population size of Oribatid mite species was found by the percentage of each species density as against the whole density and classified into dominant species, influent species, and recessive species according to the percentage; as a result, O. nova and Suctobelbella naginata was found to be a dominant species at both survey slopes while Trichogalumna nipponica was found to be a dominant species, at southward but it wasn't collected at the northward slopes at all. The feeding habit of the dominant species at two survey slopes was found to be microphytophagous- eating soil microbe. There appeared a conspicuous difference in compositions of the number of the species, individuals and dominant species at the southward/northward slopes adjoining each other at an attitude of 1,000 meters and less similarity between the two survey slopes. Conclusively, It was found that the heterogeneity of microhabitat has a great effect on Oribatid mite's community characteristics.

Bionomics of Tetranychus urticae Koch on Eggplants under Various Silicate Regimes in Controlled Environment (시설 재배 가지에서 규소 시비 수준에 따른 점박이응애의 생물적 특성)

  • Development of T. urticae was studied on the leaves of eggplant grown in hydroponics with silica contents of 0 mM, 1.7 mM, and 5.1 mM. As the levels of sllica increased, those of N, P, K, Ca, and Mg decreased and those of Si increased. While contents of calory, crude protein, carbohydrates, and vitamin C decreased, those of ash, fiber, and sugar increased. Biomass and content of cholorophyll decreased as the level of silica increased while leaves became thicker. Laboratory leaf disc tests provided with various silica levels revealed that feeding preference of T. urticae was the highest at 0 mM and the lowest at 1.7 mM. The oviposition preference decreased as the levels of silica increased, on the lower leaf appeared to hinder the feeding and the oviposition of T. urticae. The development slowed down both in female and male. Adult life span of female shortend as the silica level increased and that of male was the longest as 5.7 days at 0mM and the shortest at 1.7 mM. Oviposition period also shortened as the silica content increased. The number of eggs laid was the most as 86.3 at 1.7 mM while average oviposition per day was the least as 77 at 0 mM. No differences in hatchability was detected whereas the ratio of sex was the highest as 0.71 in favor of female at 0mM. $R_o$ and T decreased as the levels of silica increased. Rm and $\lambda$ was the lowest at 5.1 mM while Dt was the longest as 1.8271 at 5.1 mM and the shortest as 1.6991 days at 1.7 mM. Silica content in eggplant turned to affect the development of T. urticae, however, it deterred T. urticae from feeding and oviposition and the rate of increase tended to decrease due to lesser nutrients in the eggplant leaf.

Road-map for Environmental Friendly Integrated Pest Management(IPM) of Insect Pests on the Strawberry Vinyl-houses of Farmer's Field (딸기재배 하우스에서 발생하는 해충의 환경친화적 종합적방제를 위한 로드맵)

  • Road-map for the environmental friendly integrated pest management(IPM) of insect pests was drawn up on the strawberry vinyl-houses of farmer's field. Major insect pests were occurred Tetranychus urticae and Aphis gossypii during the strawberry plant seeding in the vinyl house and open field. Also, same insect pests were occurred in the vinyl house during harvesting season of strawberry. For the control of T. urticae and A. gossypii, Phytoseiulus persimilis and Aphidius colemani as natural enemies were input to the vinyl house, respectively. However, because these natural enemies could not control insect pest populations, acaricide and insecticide were sprayed. Then natural enemies were input again in the vinyl house. Natural enemies could not endure the intense cold and differences of temperature and relative humidity between day and night during strawberry harvesting season. So, their behavior and control activity of pests were more decrease than pests. Firstly, natural enemies are input in the vinyl house during the early breeding season of strawberry, secondly, acaricides and insecticide are sprayed for the control of mites and aphids, respectively, during the middle breeding season in the hard winter. Finally, natural enemies are re-input in the vinyl house during the middle and late breeding season.

Bionomics of Tetranychus urticae Koch on Eggplants under Various Potassium Regimes in Controlled Environment (시설 재배 가지에서 칼리 시비 수준에 따른 점박이응애의 생물적 특성)

  • Development of T. urticae was studied on the leaves of eggplant grown in hydroponics with potash contents of 0 mM, 2 mM, 6 mM, and 12 mM. As the levels of potash increased, that of nitrogen decreased and that of P, K, Mg increased in the plant. While contents of crude protein and fiber decreased, those of ash and sugar increased. Carbohydrate content was the highest at 2 mM. Water contents increased as those of potash increased with the exception at 0 mM. Biomass was the heaviest as 552.7 g at 6 mM and the lightest at 0 mM. Leaf thickness and the content of chlorophyll increased as the content of potash increased. Laboratory leaf disc tests provided with various potash levels revealed that feeding and oviposition preferences of T. urticae were high at 6 mM and 12 mM, respectively. Ratio of damaged leaf by naturally occurring T. urticae on eggplants of 99 days post-transplant in the greenhouse was the highest at 6 mM. Development of immature stages of T. urticae shortened as the levels of potash increased with a less tendancy in male than in female. No differences were detected in adult longevity and oviposition period but the number of eggs laid was the most as 84.7 at 6 mM and the least as 40.6 at 0 mM. There were no differences in the rate of egg hatch and the ratio of sex. $R_o,\;r_m,\;and\;{\lambda}$ were the highest at 6 mM and the lowest at 0 mM. T and Dt were the lowest at 6 mM and the highest at 0 mM. There was a descending trand of T. urticae developmet when levels of potash either gets high or low in the hydroponics.

  • Studies were made on the morphology and life cycle of the mite, Pyemotes tritici $Lagr{\acute{e}}z$-Fossot & $Montagn{\acute{e}}$ (Trombidiformes; Pyemotidea) ,parasitic on larvae of cigarette beetle, Lasioderma serricorne F., which is one of the most serious pests of stored tobacco, cigar, and cigarette in Korea. One generation time was $20.9{\pm}0.7$ days out of which $9.5{\pm}0.3$ days were spent for feeding and $10.3{\pm}0.8$ days for reproduction of progenies. A female of this insect-parasitic mite produced $56.7{\pm}6.9$ progenies during her reproduction period. The body size of a newly-laid male or female was $280{\mu}m$ long and $85{\mu}m$ wide. As female of this mite sucked on, their abdomen grew larger and larger to reach $825{\mu}m$ in width and $0.346mm^3$ in volume after $9{\sim}10$ days. By sucking the humor of a host, the abdomen of a female mite became almost a global shape in two days. The increase rate of abdominal width was the biggest on the second or the third day of feeding while the volume of abdomen reached to the largest on $6{\sim}8$ days after feeding. The largest number of the daily young produced on 4-th day after a female began to reproduce.

A Study on Distribution of Soil Microarthropods in Pinus rigida Plantations following Strip-Cutting (대상벌채(帶狀伐採)한 리기다소나무 조림지(造林地)의 토양미소절지동물(土壤微小節肢動物) 분포(分布)에 관(關)한 연구(硏究))

  • This study was carried out to investigate composition and distribution of the soil microarthropods in Pinus rigida plantations following strip-cutting from May 1998 to April 1999. The results of this study were as follows; 1. The individuals of soil microarthropods found during the research periods were 181, 904 and were identified into 21 orders in 7 classes. The classes were Arachnida, Insecta, Malacostra, Pauropoda, Diplopoda, Chilopoda and Symphyla. 2. The population density of the soil microarthropods was 82,962 individuals at the control area, 62,688 individuals at the reserve area and 36,254 individuals at the cutting area. The microarthropods in the control area were 2 times higher than those in the cutting area. 3. Although the major dominant taxa, Acari and Collembola, decreased in densities at the cutting area, the density reduction in Acari was higher than that in Collembola. This resulted in the increase of Collembola in terms of the relative abundance at the cutting area. 4. Among the total soil microarthropods, Arachnida was 59.74%, followed by Insecta of 39.82%. Such two groups comprise 99.56% of soil microarthropods. 5. According to the relative population density, Acari was 99.18% in Arachnida and 59.25% in the total and Collembola 93.99% in Insecta and 37.42% in the total. Therefore, individuals of Acari and Collembola was 96.67% of the total individuals. And the next abundant groups were Hymenoptera (0.95%), Diptera (0.64%) and Pseudoscorpiones (0.39%). 6. The population density of the soil microarthropods fluctuated seasonally, showing the bimodal pattern, being high in fall and spring. The highest density occurred in November and the lowest in July.

Method Comparison of Chemical-Resistance Level Determination and Field Resistance of Two-Spotted Spider Mite, Tetranychus urticae Koch to Benzomate, Cyhexatin, and Dicofol (점박이응애의 약제저항성(藥劑抵抗性) 수준결정방법(水準決定方法) 비교(比較)와 Benzomate, Cyhexatin 및 Dicofol 저항성(抵抗性) 조사(調査) 연구(硏究))

  • This study was conducted to compare the methods of slide dip and leaf dip techniques in the resistant level determination of spider mites to acaricides used; .and to investigate resistant levels of two-spotted spider mite, Tetranychus urticae Koch to benzomate, dicofol and cyhexatin in Honam region of Korea. Comparisons of the resistant strains and susceptible strain based on $LC_{50}$ values were summarized. Using the slide dip and leaf dip techniques with benzomate acaricide, the Keumchun strain of T. urticae showed 75-and 69-fold resistant levels, respectively, as compared with the Kwangju susceptible strain, but the Hackyo strain was 38-and 35-fold resistant levels, respectively. There was no significant difference (P>0.05) between the slide dip and leaf dip methods. With dicofol acaricide, resistant levels of the Keumchun strain of T. urticae were 71 times by using the slide dip and 74 times by using the leaf dip as resistant as the Kwangju susceptible strain while the Hackyo strain was 24 times by the former method and 23 times by the later method. There was also no significant difference (P>0.05) between both the methods. The slide dip technique is more recommendable than the leaf dip technique for determination of degree of chemical resistance to the spider mites with accuracy and less variation of the results. Using the slide dip method with cyhexatin, the Bia, Keumchun and Hackyo strains of T. urticae were 19-, 18- and 9-fold resistant levels, respectively, over the Kwangju susceptible strain.

Inheritance and Cross Resistance of Acequinocyl Resistance in Twospotted Spider Mite, Tetranychus urticae (Acequinocyl 저항성 점박이응애의 유전과 교차저항성)

  • The development of resistance to acequinocyl was found in the population of the twospotted spider mite, Tetranychus urticae, collected from rose greenhouses in Gimhae, Gyeongnam province in January 2001. This pest is reared on 5 years treated with acequinocyl (over 200 times), and increased 87.8 folds in resistance as compared to susceptible strain (S). Inheritance of acequinocyl resistant strain (R) and cross resistance of this strain to 8 acaricides against T. urticae adults and eggs was investigated. There were differences of susceptibility in the acequinocyl concentration-mortality relationships in $F_1$ progenies obtained from reciprocal cross with the S and R strain ($S(female){\times}R(male)$, $R(female){\times}S(male)$). Degrees of dominance were -0.75, -0.57 in $F_1$ progenies of adult and egg of $S(female){\times}R(male)$. Inheritance in $F_1$ progenies of $S(female){\times}R(male)$ was incomplete recessive. Degree of dominance were 0.81, 0.45 in $F_1$ progenies of adult and egg of $R(female){\times}S(male)$, respectively. These results suggest that inheritance of acequinocyl resistance is controlled by a complete dominance. The R strain exhibited cross resistance of 1.1 and 0.9 fold to amitraz, bifenazate, and negatively correlated cross resistance of 0.08 fold to emamectin benzoate in adult females. The R strain showed cross resistance of 37.7, 14.0, and 26.2 fold to amitraz, milbemectin and spriodiclofen in eggs, respectively. Particularly it showed high levels of cross-resistance to pyridaben with 6538.3 fold. These chemicals showed negatively correlated cross-resistance exhibited 0.4, 0.3, and 0.2 fold to ahamectin, bifenazate, and emamectin benzoate in eggs.

Toxicity Evaluation of 'Bt-Plus' on Parasitoid and Predatory Natural Enemies (기생성 및 포식성 천적에 대한 작물보호제 '비티플러스'의 독성 평가)

  • Effect of a new crop protectant 'Bt-Plus' on natural enemies was analyzed in this study. Tested natural enemies included two parasitic species of $Aphidius$ $colemani$ and $Eretmocerus$ $eremicus$, and four predatory species of $Harmonia$ $axyridis$, $Orius$ $laevigatus$, $Amblyseius$ $swirskii$, and $Phytoseiulus$ $persimilis$. 'Bt-Plus' was formulated by combination of three entomopathogenic bacteria ($Xenorhabdus$ $nematophila$ (Xn), $Photorhabdus$ $temperata$ subsp. $temperata$ (Ptt), $Bacillus$ $thuringiensis$ (Bt)) and bacterial metabolite (BM). All three types of 'Bt-Plus' showed significantly higher toxicities against fourth instar $Plutella$ $xylostella$ larvae than Bt single treatment. Two types of bacterial mixtures ('Xn+Bt' and 'Ptt+Bt') showed little toxicity to all natural enemies in both contact and oral feeding assays. However, 'BM+Bt' showed significant toxicities especially to two predatory mites of $A.$ $swirskii$ and $P.$ $persimilis$. The acaricidal effects of different bacterial metabolites were evaluated against two spotted spider mite, $Tetranychus$ $urticae$. All six BM chemicals showed significant acaricidal effects. The BM mixture used to prepare 'Bt-Plus' showed a high acaricidal activity with a median lethal concentration at 218.7 ppm (95% confidence interval: 163.2 - 262.3). These toxic effects of bacterial metabolites were also proved by cytotoxicity test against Sf9 cells. Especially, benzylideneacetone, which was used as a main ingredient of 'BM+Bt', showed high cytotoxicity at its low micromolar concentration.
