Antimicrobial activities of Lindera obtusiloba Blume and Zanthoxylum piperitum DC extracts (생강나무(Lindera obtusiloba Blume)와 초피나무(Zanthoxylum piperitum DC) 추출물의 항균활성)

  • Ethanol and hot water extracts were prepared from Lindera obtusiloba Blume (LO) and Zanthoxylum piperitum DC (ZP) and used to evaluate their antimicrobial activities and thermal stability against six foodborne pathogens (3 gram-positive and 3 gram-negative bacteria). The antimicrobial activities were assessed using the agar diffusion method, and the thermal stabilities of extracts were examined after heat treatment at 60, 70, 80, and $100^{\circ}C$ for 10 min. The zones of inhibition by the LO extract or the ZP extract of the tested microorganisms were in the range of 21-30 mm and 19-25 mm, respectively, at 100 mg/mL concentrations. The 60% ethanol extract and the hot water extracts from LO showed the strongest antimicrobial effects against MRSA and Staphylococcus aurues, respectively. For the extract from ZP, the strongest antimicrobial effect was shown against S. aurues by 60% ethanol, and the weakest antimicrobial effect was shown against E. coli by the hot water extracts. The ZP extracts showed that the gram-positive bacteria were more sensitive than gram-negative bacteria. For the thermal stability of the extracts, the antimicrobial effects stabilized after heat treatment. Overall, the data suggest that the extracts have a potential for application in various food products for which a natural antimicrobial additive is desired.

Quality Characteristics of Seasoned Pork with Water Extracts of Allium hookeri Root during Storage (Allium hookeri 뿌리 열수 추출물을 첨가한 양념돈육의 저장 중 품질 특성)

  • This study examined the antioxidant and antimicrobial activities of 70% ethanol extracts and water extracts of Allium hookeri root. We evaluated the effects of water extracts of A. hookeri root on storage of seasoned pork added with water extracts at $4^{\circ}C$ for 21 days in order to evaluate its potential as a functional food material. A. hookeri root water extracts displayed antioxidant activities (total polyphenol content and DPPH and ABTS radical scavenging activities) that were superior to those of 70% ethanol extracts. The 70% ethanol extracts and water extracts of A. hookeri root showed antimicrobial activities against food-borne Staphylococcus aureus bacteria that were about 1/400 times greater than that of vancomycin. Chemical composition analysis was conducted on pork seasoned with sauce containing 5%, 10%, and 15% water extracts. Moisture and crude ash contents significantly decreased as the amount of water extracts increased (P<0.05), and the highest crude protein content was in the 10% group. Acidity of seasoned pork increased proportionally in the early stages of storage, whereas it significantly decreased as the amount of water extracts increased after day 12 (P<0.05). Although the total number of bacteria in seasoned pork continuously increased during storage, growth of bacteria was significantly restricted as level of A. hookeri root water extracts increased (P<0.05). In the sensory evaluation, pork seasoned with 10% A. hookeri root water extracts showed the highest scores for taste, texture, and overall acceptance (P<0.05). In summary, A. hookeri water extracts display antioxidant and antimicrobial activities that can improve quality characteristics of seasoned pork and have potential as natural preservatives to restrict bacteria growth. Regarding the amount of extracts, 10% was determined to be the most appropriate level to minimize changes in seasoned pork during storage and improve sensory quality.

  • The anticancer activity of Schizandra chinensis Baillon was investigated for the development of functional food resources. The antiproliferative activity of hot water extracts of Schizandra chinensis Baillon in human colon cancer cell line (HT-29) were identified using cell viability, morphology study, cell cycle and RT-PCR analyses. HT-29 cells were cultured in several concentrations (0, 1.0, 2.0, 4.0 mg/mL) of water extracts of Schizandra chinensis Baillon. In our study, colon cancer cell growth could be inhibited by hot water extracts of Schizandra chinensis Baillon in a dose-dependent manners. It was associated with morphological changes and apoptotic cell death with cell shrinking, chromatin condensation, apoptotic bodies and cell cycle analysis. These results suggest that Schizandra chinensis Baillon may inhibit the growth of human colon cancer cells by various apoptosis-aiding activities as well as apoptosis itself.

Antioxidant Activities of Hot Water Extract from Cornus walteri Wanger against Oxidative Stress Induced by tert-Butyl Hydroperoxide in HepG2 Cells (tert-Butyl Hydroperoxide로 산화 스트레스가 유도된 HepG2 세포에서 말채나무 열수추출물의 항산화 활성)

  • The objective of this study was to investigate the effect of hot water extract from Cornus walteri Wanger (CWE) on tert-butyl hydroperoxide (TBHP)-induced oxidative stress in HepG2 cells. Generation of reactive oxygen species (ROS), concentrations of cellular lipid peroxidation products and reduced glutathione, and antioxidant enzyme activity were used as biomakers of cellular oxidative status. Cells pretreated with CWE (25~200 ${\mu}g/mL$) showed an increased resistance to oxidative stress in a dose-dependent manner, as revealed by a higher percentage of surviving cells compared to control cells. ROS generation induced by TBHP was significantly reduced when cells were pretreated with 200 ${\mu}g/mL$ CWE for 4 h. Pretreatment with CWE (5~50 ${\mu}g/mL$) prevented the decrease in reduced glutathione and the increase in malondialdehyde and ROS evoked by TBHP in HepG2 cells. Finally, CWE pretreatments prevented the significant increase of glutathione peroxidase, catalase, glutathione reductase, and superoxide dismutase activities induced by TBHP. These results show that CWE has significant protective ability against a TBHP-induced oxidative insult and that the modulation of antioxidant enzymes by CWE may have an important antioxidant effect on TBHP-induced oxidative stress in HepG2 cells.

Antioxidant and Nitrite Scavening Activity and α-Glucosidase Inhibitory Effect of Water Extract from Schizandra chinensis Baillon (오미자 열수 추출물의 항산화 활성과 아질산 소거능 및 α-Glucosidase 저해 효과)

  • The nutraceutical role of omija (Schizandra chinensis Baillon) water extract (OWE) was determined through the analysis of antioxidant activity, nitrite scavening activity, and xanthine oxdiase and $\alpha$-glucosidase inhibitory effects. Antioxidant activity of OWE was measured by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) free radical scavenging activity and superoxide dismutase-like activity (SODA). DPPH radical scavenging activity and SODA increased in a dose-dependent manner, and was about 49.0% at 2.5 mg/mL and 69.2% at 5 mg/mL, respectively. The xanthine oxidase and $\alpha$-glucosidase inhibitory activities of OWE were about 88.8% and 86.2% at 1 mg/mL, respectively. Nitrite scavenging activity of OWE was about 54.9%, 42.4%, and 34.2% on pH 1.2, 3.0, and 6.0 at 1 mg/mL, respectively. These results suggest that OWE has a strong antioxidant activity, and xanthine oxidase and $\alpha$-glucosidase inhibitory effects.

Protective Effect of Semisulcospira libertina Extract on Induced Hepatitis in Rats (다슬기 열수 추출물이 간독성이 유도된 흰쥐에 미치는 보호 효과)

  • This study aimed to investigate the protective effect of Semisulcospira libertina extract on liver injury induced by D-galactosamine in rats. After the administration of S. libertina extract, the local fat degeneration and infiltration of inflammatory cells in liver tissues were significantly decreased and peripheral hemorrhages around portal triads and central necrosis around central veins were found to be protective. The elevated levels of plasma ALT, AST, and LDH, the ALP activation lipid peroxidation, and the lipid contents of a damaged liver were recovered in experimental rats administrated with S. libertina extract, suggesting its role in blood enzyme activation and lipid content restoration within damaged rat liver tissues. Moreover, the expression rate of TNF-α, which accelerates inflammation and induces tissue damage and necrosis, was significantly decreased. In addition, the activities of antioxidant enzymes were more effectively upregulated compared to those of the control group induced hepatotoxicity. All data showed that S. libertina extract has a preventive role against liver damages, such as inflammation and tissue necrosis, as instigated with D-galactosamine by improving the activities of blood enzymes and antioxidant enzymes and modulating the expression of inflammation factor, suggest S. libertina extract is an effective medicinal resource for the restoration of hepatotoxicity.

Antitumor Effects of Cistanchis Herba Aqueous Extracts on MCF-7 Human Breast Cancer Cell-Xenograft Athymic Nude Mice, through Potent Immunomodulatory Activities (유방암 세포(MCF-7) 이식 누드 마우스에서 육종용 열수 추출물의 항암 효과 평가)

  • 목 적: 본 연구의 목적은 유방암 세포(MCF-7) 이식 누드 마우스 모델을 이용하여 육종용 열수 추출물의 면역활성 효과를 통한 항암 활성을 체계적으로 평가하는 것이다. 육종용 열수 추출물은 유방암 치료제로 자주 사용되는 대표적인 경구용 항암제인 tamoxifen 경구 투여군과 비교하여 분석 연구하였다. 방 법: 총 110마리의 6주령 암컷 누드 마우스를 준비하여, 7일간 적응 후 체중이 일정한 마우스를 선정하여 우측 둔부 피하부위에 MCF-7 세포를 이식하였다. 종양세포 이식을 한 지 20일 후, 종양 크기 및 체중을 기준으로 그룹 당 8마리씩 본 실험에 사용하였다. MCF-7 이종 이식 21일 후부터 매일 1회씩 35일간 육종용 열수 추출물을 10 ml/kg의 용량(400, 200 및 100 mg/kg)으로 경구 투여하였으며, tamoxifen 역시 10 ml/kg의 용량(20 mg/kg)으로 경구 투여하였고, 정상 및 종양 이식 매체 대조군에서는 멸균증류수만 종양 이식 21일 후부터 동일한 방법으로 35일간 경구 투여 하였다. 결 과: 본 실험의 결과, MCF-7 세포 이식을 함으로써 현저한 비장 및 하악하 림프절 무게, 혈중 IFN-γ의 함량, IL-1β 및 IL-10의 함량, 비장내 TNF-α, 비장세포 및 복강 대식구의 활성의 감소가 관찰되었고, 비장 및 하악하 림프절의 림프구 감소에 의한 조직병리학적 위축 또한 관찰되었다. 그리고 체중 및 증체량의 감소 역시 관찰되었으며, 혈중 IL-6 함량의 증가, 난소 주위의 지방 무게의 감소 및 조직병리학적으로 난소 주위의 축적 지방 조직 위축 현상이 인정되어, 종양 이식 후에 전형적인 종양과 관련된 면역억제와 악액질 현상이 유발된 것으로 판단되었다. 한편 육종용 열수 추출물 400, 200 및 100 mg/kg 경구 투여군에서는 종양 이식 대조군에 비해 유의성 있는 현저한 항암활성이 투여 용량 의존적으로 관찰되었다. 또한 tamoxifen 20 mg/kg 경구 투여군에서는 종양 관련 악액질 소견이 오히려 악화되는 반면, 육종용 열수 추출물 경구 투여군에서는 면역활성 및 악액질 억제 효과가 관찰되었다. 결 론: 본 연구 결과, 육종용 열수 추출물의 적절한 경구 투여는 심각한 부작용 없이, 종양 관련 악액질 소견을 포함하여, 효과적인 유방암 치료 수단을 제공할 수 있을 것으로 기대된다.

Antioxidant and anti-inflammatory activities of hot water extracts of Ligularia fischeri (곰취 열수 추출물의 항산화 및 항염증 활성)

  • This study was carried out to investigate the anti-oxidative and anti-inflammatory effects of hot water extracts of Ligularia fischeri cultivated in Youngyanggun. We obtained hot water extract (HWE) and cold water extract (CWE) from L. fischeri. The anti-oxidative activities of L. fischeri extracts were measured by 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity. The anti-inflammatory effects of L. fischeri were evaluated in human mast cell line-1 (HMC-1) cells stimulated with phorbol-12-myristate-13-acetate plus A23187 (PMACI). The solid yields of HWE was 150% higher than CWE solid yield. Total polyphenol contents of HWE were $198.07{\pm}0.24mg/g$. The value of anti-oxidative activities of HWE were shown $IC_{50}$ $28.2{\pm}0.04ug/mL$. We showed that HWE significantly reduced the PMACI-induced the production of IL-6 (0.01-1 mg/mL), IL-8 (0.1-1 mg/mL), and $TNF-{\alpha}$ (0.01-1 mg/mL). These results indicate that the HWE of L. fischeri can be used as a functional material due to its antioxidant and anti-inflammatory activities.

Antioxidant activity and inhibition activity against α-amylase and α-glucosidase of Juniperus rigida Sieb extracts (노간주나무(Juniperus rigida Sieb) 추출물의 항산화 활성 및 α-amylase와 α-glucosidase에 대한 저해활성)

  • In this study, the antioxidant effect of water and ethanol extracts from Juniperus rigida Sieb were investigated. The activities of each of the extracts were measured based on their total phenolic and flavonoid contents and using antioxidant test such as of 2,2-azinobis (3-ethyl benzothiazoline-6-sulfonic acid (ABTs) radical scavenging activities, ferric reducing antioxidant power (FRAP), angiotensin I converting enzyme (ACE) inhibition activity, antioxidant protection fator (PF), thiobarbituric acid reactive substances (TBARs) content, and ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibition activity assay. The result of the examination to measure the polyphenol content by investigating the antioxidativity of the J. rigida Sieb. extract showed 71.3 mg/g polyphenol content in the water extract, and 116.0 mg/g in the ethanol extract and a 17.7 mg/g flavonoid content in the water extract and in 76.4 mg/g in the ethanol extract. The ABTS radical cation decolorization showed 76.4% and 79.3% scavenging activities of the $500{\mu}g/mL$ water extract and ethanol extract, respectively. The FRAP showed 1.83 mM efficacy in the water extract and a lower 1.77 mM in ethanol extract. Both the water extract and the ethanol extract showed reduced ACE activities of 75.39% and 71.25% at $500{\mu}g/mL$, respectively. The antioxidant protection factor of the water and 70% ethanol extracts of J. rigida Sieb were 1.5 PF and 2.1 PF, respectively. In the TBARS inhibitory activity, the extracts showed 55.78% and 71.48% antioxidant activities at the $500{\mu}g/mL$ concentration. The results of the measurrement of the ${\alpha}$-amylase inhibitory activity indicated more than 90% of activity inhibition in the $500{\mu}g/mL$ concentration of the ethanol extract. For the ${\alpha}$-glucosidase inhibitory activity, the ethanol extract showed 70% activity inhibition at the $500{\mu}g/mL$ concentration.

Biological activities of Brassica rapa (Turnip) callus extracts by plant cell culture technology (식물세포배양기술을 이용한 순무 캘러스 추출물의 생리활성)

  • The purpose of this study was to investigate biological activities of Brassica rapa (Turnip) plant callus extracts of Ganghwa-gun of Incheon city using water, ultrasonic wave and ethanol extractions to develop functional materials. DPPH radical scavenging activities of the callus extracts were increased in a concentration-dependent manner, as compared with control. The astringent effects of the ethanol extracts were higher, as compared to water and ultrasonic extracts. In the collagen synthesis assay, the ethanol extract showed significant anti-wrinkle effects of 59% and 78% at a concentration of 5 ppm and 10 ppm, respectively. These results suggested that water, ultrasonic wave and ethanol extracts of turnip plant calluses are natural antioxidant sources. Especially, the ethanol extract can be regarded as a functional, natural cosmetic material with astringent and anti-wrinkle effects.
