First Report of Phytophthora palmivora in Cheju Island as the Causal Pathogen of Phytophthora Crown Rot of Cymbidium (제주도에서 처음으로 발생한 Phytophthora palmivora에 의한 심비디움 역병)

  • Phytophthora crown rot of cymbidium was observed in Cheju island since June of 1996. The disease initiated at the basal portion of infected plant progressed upward to lower leaves. Soon after distinct water-soaking lesions appeared on lower leaves, the plant was wilted, blighted and died. Four orchid farms at Sogwipo out of 16 surveyed in the island were infected by the disease estimating 5~20% infection rates. The causal fungus was identified as P. palmivora based on following distinguishing characteristics. All isolates were heterothallic as A1 types and readily produced chlamydospores with cultural age. Sporangia were conspicuous papillate, ellipsoidal to ovoid, highly deciduous with short pedicels ca. 3~4 ${\mu}{\textrm}{m}$. Koch's rules were satisfied by a pathogenicity test and re-isolation of the fungus from inoculated plants. The pathogen has never been reported in Cheju island previously and its firstly recorded as the cause of Phytophthora crown rot of cymbidium in Korea.

Leaf Spot of Cymbidium hybrida Caused by Fusarium proliferatum (Fusarium proliferatum에 의한 심비디움 저무늬병)

  • Leaf spot of Cymbidium hybrida caused by Fusarium sp. was observed at major cultivating areas including Seosan and Cheonan of Korea from 1996 to 1998. The major symptoms of the disease were small brown to black spots, 1∼2 mm I diameter, with yellow halo. Based on the mycological characteristics, Fusarium sp. isolated from the lesions was identified as Fusarium proliferatum. Macroconidia were slender, falcate to almost straight, usually 3 to 5 septate and thin walled. Microconidia were formed in chains from polyphialides, clavate or oval, usually 1-celled with flattened base. Chlamydospores were absent. The fungus showed pathogenicity to Cymbidium hybrida.

Detection of Cymbidium Mosaic Virus and Odontoglosum Ringspot Virus by ELISA and RT-PCR from Cultivated Orchids in Korea (ELISA와 RT-PCR에 의한 국내재배난에서 심비디움 모자이크 바이러스와 오돈토글로섬 윤문 바이러스이 검정)

  • This study was carried out to detect cymbidium mosaic potexvirus (CymMV) and odontoglossum ringspot tobamovirus (ORSV) in cultivated orchid plants in Korea. The standard double antibody sandwich enzyme-linked immunosorbent assay (ELISA) and reverse transcription polymerase chain reaction (RT-PCR) were carried out for detection of the viruses in the collected orchid samples. ELISA was suitable for massive-scale diagnostic method for virus detection in orchids. RT-PCR was rapid, time-saving and reliable detective method, and detection limit data showed that RT-PCR was 103 times more sensitive than ELISA. Of the 321 individual orchids representing 5 orchids genera tested by the ELISA, CymMV and ORSV were detected in 15.6% and 22.4%, and mixed infection of the both viruses with 4.9%, respectively. Of the Cymbidium plants tested, cultivated plants showed 52.5% virus infection rate with either CymMV or ORSV and both viruses.

A Comparison of Functional Fragrant Components of Cymbidium (Oriental Orchid) Species (기능성 지표물질 확인을 위한 동양란 심비디움(Cymbidium) 향기 성분 비교 분석)

  • We analyzed the functional fragrant components of three species of Cymbidium oriental orchids using gas chromatography-mass spectrometry (GC/MS). For the comparative analysis, C. goeringii 'Minchunran', 'Jugeumhwa', C. forrestii 'Chwigae', 'Songmae', 'Yongja', and C. faberi 'Choemae', 'Namyangmae', 'Hwaja' were investigated. Major fragrant components detected by GC/MS were selected on the basis of more than 3% value according to the analysis of peak area (%). We found that ${\alpha}$-bergamotene, which has a cytotoxic effect on breast cancer, cervical cancer, and glioblastoma, and nerolidol, which induces apoptosis of human hepatoma cells (HepG2), inhibits the growth of Streptococcus mutans and babesiosis, and has antibacterial properties, are common substances produced by C. goeringii L. Nerolidol and ${\beta}$-bisabolene, which is cytotoxic and suppresses the growth of malignant melanoma cells (B16-F10), HepG2, and leukemia cells (HL-60, K562), are major substances in C. forrestii R. Furthermore, ${\alpha}$-pinene, which inhibits the growth of gliobastoma cells (SF-767) and inhibits the anti-inflammatory action of hepatoma cells (BEL-7402); 1,8-cineole, which is a potential therapeutic agent for the treatment of gastric ulcers; and 1,3,7-octatriene, which functions as a pheromone, are the most common substances in C. faveri R. Thus, substances identified as major fragrant components in oriental orchid species have multiple beneficial applications in human health. This research forms the basis for further studies of the roles of major fragrant components in oriental orchids.

Mineral Absorption by Cymbidium Jungfrau in the Solution Culture (서양 심비디움 양액재배에서의 무기물 흡수)

  • $N(^{15}N)$ and $P(^{32}P)$ absorption by 2 year-old Cymbidium Jungfrau in solution culture were investigated. Growth, photosynthesis rate, chlorophyll content and mineral composition of Cymbidium in the solution culture with bark or granular rockwool were compared with these parameters in the conventional pot culture. Nitrogen absorption by Cymbidium was higher in full sunlight than in 60% of sunlight while P absorption was higher in 60% of sunlight. Sixty seven % of N absorbed in plant was redistributed to the bulb(39%) and leaves(28%) while 46% of P absorbed was fund in the bulb (36.2%) and leaves (10.2%). Accumulation of P in leaves was 3-fold lower than that of N. N and P absorption in 0.5 or 1 year- old daughter plant Bowing vigorously were greater than in immature daughter or mother plant. The absorption rate of phosphorus in Cymbidium was 350-fold lower than that of barley. Greater shoot length and bulb diameter, and higher fresh weight, photosynthesis rate and chloroployll content were observed in the solution culture than in the conventional pot culture. Solution culture had-also more content of N, P, K and Mg in leaves, bulb and root than conventional pot culture but did not that of Ca. A large part of the nutrient absorption was occurred during vegetative growth. Also, There was no difference between bark and rockwool in the solution culture due to the improvement of poor dispersion of nutrient solution in bark.

A New Cymbidium Cultivar 'Orange Bowl' with Orange Colored Flower and Medium Sized Plant (오렌지색계 중형 심비디움 'Orange Bowl' 육성)

  • Cymbidium 'Orange Bowl' (Lucky Rainbow 'Randevous' ${\times}$ 'Eastern Star') was developed from a cross between hybrids at the National Institute of Horticultural & Herbal Science, Rural Development Administration in 2006. A cross was made between the pink colored flower C. Lucky Rainbow 'Randevous' as maternal line and pure yellow colored flower, C. 'Eastern Star' as paternal line in 1995. The seed germination, cultivation, selection, and characteristic trials were conducted from 1996 to 2006. The line was named as Wongyo F1-18 and phenotype was characterized in 2006 as a new 'Orange Bowl'. The 'Orange Bowl' has having light yellow basal color (RHS, YO21D) and orange line (RHS, OR30B) on both of sepal and petal with red lip (RHS, OR30B). 'Orange Bowl' has about 10.9 flowers per flower stalk and flower size of 7.4 cm. General appearance of the petals and sepals is slightly incurved shape. The plant size is intermediate having erect peduncle. Blooming is started from the late of January (mid winter) under optimal culture condition. Leaf attitude and twisting is half- erect and very weak respectively. This hybrid has attractive floral arrangement, long flower stalk (71.8 cm) and vigorous growth. We expect that 'Orange bowl' has a great potential for exporting to Chinese market.

Effects of Plant Growth Regulators on in vitro Propagation of Cymbidium kanran and Cymbidium hybrida (한란 및 심비디움의 기내 증식에 미치는 생장조절물질의 효과)

  • This study was carried out to determine the effects of plant growth regulators on organogenesis from Cymbidium kanran and Cymbidium hybrida. Optimal rhizome formation from Cymbidium kanran was obtained on MS medium with 10 ppm kinetin+2 ppm NAA. and optimal protocorm formation from Cymbidium hybrida was obtained on MS medium with 10 ppm kinetin+0.05 ppm NAA. However, in this study the optimal media for the callus induction from both explants was not identified. Optimal shoot induction from rhizome of Cymbidium kanran was obtained on MS medium with 10 ppm BA+2 ppm NAA and 5 ppm BA+2 ppm NAA. Optimal shoot induction from protocorm of Cymbidium hybrida was obtained on MS medium with 10 ppm kinetin+2 ppm NAA.

Review on breeding, tissue culture and genetic transformation systems in Cymbidium (심비디움 육종, 조직배양 및 형질전환 연구동향에 관한 고찰)

  • Cymbidium is horticulturally important and has been one of the most commercially successful orchid plants as well as cut flowers around the world including Korea. Up to now, a huge number of elite Cymbidium cultivars have been released on the commercial market via cross-hybridization, mutation and polyploidization breeding techniques. To investigate on breeding system in Cymbidium, we inquired the brief history and techniques of breeding and the current status on Cymbidium breeding in Korea. Also, the general propagation process of elite Cymbidium lines via tissue culture should be presented. However, the slow process of conventional breeding and the lack of useful genes in Cymbidium species delays the introduction of new cultivars to the commercial market. To solve these limitations, efficient regeneration and genetic transformation systems should be established in the improvement of Cymbidium breeding program. During the last several decades, some progress has been made in tissue culture and genetic transformation in Cymbidium species. We review the recent status of tissue culture and genetic transformation systems in Cymbidium plants.

Effects of BA on Flowering of Cymbidium encifolium 'Tekkotsusosin' (BA처리가 심비디움 '철골소심'의 개화에 미치는 영향)

  • This study was conducted to investigate the optimum concentration and time of BA treatments for the improvement of flowering in Cymbidium encifolium 'Tekkotsusosin'. The flowering rate was increased by the higher concentration of BA treatment, and reached 100% by $400mg{\cdot}L^{-1}$ treatment. But high concentration induced the flower malformation. Therefore, the optimum concentration of BA for increased of flowering rate was about $300mg{\cdot}L^{-1}$. In the time of BA treatment, plants which were treated in June to August by BA $300mg{\cdot}L^{-1}$ were flowered, especially up to 100% in August. However, those treated in April, May, and September did not.

  • 식물조직 배양공정의 기계화 기술개발을 위한 기초자료를 얻고자 식물조직 배양기술의 개발 또는 식물묘를 대량생산하는 연구지도기관 및 농원 등의 배양실 10개소를 대상으로 배지의 분주, 배양용기의 밀봉 및 세척, 배양체의 접종공정 등에 대한 작업실태를 조사분석한 결과를 요약하면 다음과 같다. 가. 배양기술의 개발 또는 식물묘 대량생산은 대부분이 나리, 호접란, 풍란, 심비디움 등 화훼류였으며, 배지는 모두 한천이 혼합된 고체배지, 배양용기는 형상 및 용량 등이 다른 플라스크, 배양병, pE용기 등이 사용되었다. 나. 배지의 분주작업은 조사대상 10개배양실중 6개소는 작업자가 비이커를 사용하여 분주하였고 4개소는 자동 또는 반자동분주기를 사용하였으며, 인력 및 반자동분주기의 사용시 배양용기간의 분주량은 기준량의 $\pm$25%범위에서 차이가 있었다. 다. 액체배지의 분주시 온도는 36.8~88.$0^{\circ}C$, 점도는 3.7~11.8cP 범위였고 점도는 배지에 혼합하는 한천, 사과 등 유기물의 량에 따라 차이가 있으므로 배지의 분주작업을 기계화하기 위해서는 배지의 온도와 점도, 배양용기와 분주량 등을 종합적으로 고려하여 배지의 점도에 따라 분주량을 조절할 수 있도록 개발할 필요성이 있다고 생각된다. 라. 배양용기의 밀봉소재는 플라스크의 경우 호일 또는 고무마개와 호일, 배양병은 모두 나사산이 있는 캡, PE용기는 뚜껑을 사용하였으며, 밀봉작업은 모든 배양실에서 인력에 의존하였고 작업능률은 호일만으로 밀봉하는 경우 시간당 180~240병, 캡은 시간당470~600개 정도였다. 마. 배양체의 접종작업은 모든 배양실이 인력에 의존하였으며, 배양체를 배지와 분리하여 불필요한 부분을 제거하고 배양작물에 따라 생육정도를 2~3등급으로 구분하여 배양용기의 배지 위에 치상하는 과정으로 수행되었으며, 작업능률은 호접란의 경우 배양병에 25본을 접종하는데 시간당 6병, 심비디움은 원형 플라스크에 25본을 접종하는데 시간당 10병 정도였다. 바. 식물체의 대량증식에 사용되는 플라스크, 배양병, PE용기 등 배양용기의 세척작업은 농원의 1개배양실에서 간이식 세척기, 이 외의 9개배양실은 모두 물에 담겨 두었다가 세제와 브러쉬 등을 사용하여 인력으로 세척하고 있어 생력화 기술개발이 요구되었다.
