Antioxidant and anticancer activities of Adenophora triphylla leaf and root extracts (새싹 잔대 잎과 뿌리의 항산화 및 항암 효과)

  • The root of Adenophora triphylla is a highly valued medicinal resource that is used to prevent human obesity, cancer, and inflammation, whereas young leaves or sprouts of A. triphylla are used as food ingredients. In this study, we compared the antioxidant and anticancer activities of 70% ethanol extracts of A. triphylla roots and leaves. The leaf extract exhibited stronger 2,2-diphenyl-1-picrylhydrazyl (DPPH)-radical scavenging activity, reducing power, and oxygen radical absorbance capacity (ORAC) than the root extract. Furthermore, the leaf extract was observed to be a potent source of anticancer compounds that were effective against A549 (lung cancer), LNcaP (prostate cancer), SKOV3 (ovarian cancer), and Caco-2 (colorectal cancer) cells. These results indicate that not only the roots but also the leaves of A. triphylla can serve as valuable sources of functional materials in the pharmaceutical industry.

Antifungal Activity of Zanthoxylium schinifolium Against Fusarium graminearum, a Barley Powdery Mildew Fungus. (보리 흰가루병 곰팡이 Fusarium graminearum에 대한 산초 추출물의 항진균 활성)

  • The powdery mildew, a fungal plant disease found in varieties of plant cultures, is occurred by attack with Fusarium sp., Sphaerotheca sp., Leveilluna sp., and Eryshipe sp.. In this study we investigated the control of Fusarium graminearum, a barley powdery mildew fungus, by natural plant extracts. Among the 900 plant extracts tested, Zanthoxylum schinifolium, Ligusticum acutilobum, Bidens frondosa L., Dictamnus dasycarpus, Evodia officinalis, Disporum sessile, Scopolia japonica Max., Styrax japonica S. et Z., Dictamnus dasycarpus Turcz., Sinomenium acutum Rehder et Wils., Eugenia aromaticum, Rubus parvifolius L., Reynoutria elliptica, Coptis chinensis, Paeonia lactiflora Pall., Rheum undalatum, Paeonia suffruticosa, Oenothera odorata Jacq., Euphorbia pekinensis Rupr., and Nepeta cataria were selected based on spore germination inhibition assay. Further mycelial growth inhibition assay with economical and safety considerations led us to finally select Z. schinifolium (sancho) for control of F. graminearum. To produce antifungal sancho extract, methanol was suitable for extraction and subsequent fractionations of the extract showed that the water residue mainly had antifungal activity. The sancho extract and its fractions showed minor antibacterial activity against different pathogenic or food spoilage bacteria, but they did not show any harmful effects against young tomato plant by treatment of $1,000\;{\mu}g/ml$ in green chamber test. These results suggested that the extract of sancho has high potentials on control of a powdery mildew fungus, F. graminearum.

Effect of Plant Extract [FGF271] on Estrogen Replacement (식물유래 추출물(FGF271)의 여성호르몬 대체 효과)

  • A certain group of phytochemicals such as isoflavone have been proven to act as a phytoestrogen. After thorough the study of different bibliographic herbs excluding soybeans, dates, pomegranates, and other publicized plants, three different edible herbs by Korean Food Regulation were extracted for the animal study on the effect of estrogen replacement. The herbal extract(FGF271) has been orally administered into 51 weeks old partial ovariectomized rats for 5 weeks with the different dosages of 100 and 1,000 mg/kg, respectively. It was observed that 1) serum estrogen level was increased in both 100 and 1,000 mg/kg group, 2) the distension of uterus was made dose dependently and significantly different in 1,000 mg/kg group (p.0.05) from control in the gross findings, 3) the weight of uterus was increased in 1,000 mg/kg group, and 4) the action on reproductive tissues was clear in the microscopic findings in terms of hyperplasia of endometrial epithelial cell, cystic change of submucosa, dilatation of uterus (significantly increased in 1,000 kg/mg), and follicular cystic changes in ovary. As a result, FGF271 seemed to act as a phytoestrogen by inducing the change in ovary and uterus and by increasing the serum estrogen concentration.

Study on Antioxidative Activity of Plant Extracts in Fish Oil (식물 추출물이 어유의 산화 안정성에 미치는 영향)

  • Extract from forty four species of natural plants in Korea were obtained. Each extinct (1,000 ppm) was added into fish oil and their antioxidative activities were screened by rancimat, From the results, seven species including Brassica juncea, Solanum tuberosum, Capsella bursa, Ulums davidiana, Broussonetia kazinoki, Oenanthe javanica, and Rhus chinensis were selected due to their relatively prolonged induction periods. When the concentration (100 - 1,000 ppm) of the selected extract was considered, the most prolonged induction period was observed from Solanum tuberosum throughout the studied concentrations, suggesting that its extinct showed the strongest antioxidative activity against fish oil. In addition, peroxide value (POV) and thiobarbituric acid reactive substance (TBARS) value were considered at $60^{\circ}C$ for 5 days. Among the selected extract, extract from Solanum tuberosum showed the lout POV while the extract from Brassica juncea var showed the least TBARS values after 5 days. Also, the free radical scavenging effect against DPPH radical were studied with the selected extract, showing that the extract from Rhus chinensis was found to be the most effective.

Antioxidant Effects of Ethanol Extracts from Flower Species of Compositae Plant (국화과 식물 중 꽃 에탄올추출물의 항산화효과)

  • The present experiments were carried out to develop new antioxidants from EtOH extract of flower resources of Compositae plant concerning total polyphenol and flavonoid content, scavenging activities on DPPH and ABTS radicals, ferrous ion chelating effects, and inhibition activity on lipid peroxidation of linoleic acid. EtOH extracts from Matricaria recutica, Cosmos bipinnatus, Synurus deltoides, and Aster pilosus showed higher level of total polyphenol content, and the flavonoid content was the highest in C. bipinnatus. However, scavenging activity on DPPH radicals was the highest in EtOH extract of Hieracium pilosella-1.1 times higher than BHT control. The activity on ABTS radical scavenging was the highest in EtOH extracts of M. recutica and S. deltoides-2.0 and 1.2 times higher than ascorbic acid, and 2.2 and 1.3 times higher than BHT, respectively. Ferrous ion chelating effects was also the highest in M. recutica, but the level was much lower than EDTA. Inhibition activity on lipid peroxidation of linoleic acid, measured at every 4 days for 32 days, was superior and longer lasting with M. recutica and Achillea alpina, compared to that of BHT. In conclusion, antioxidant activity was different depending on species, so selection of proper plant species for the development of potential antioxidant is very important.

The Study on Antimicrobial and Antifungal Activity of the Wild Seaweeds of Jeju Island (제주 자생 해양식물들의 항균 및 항산화 활성 연구)

  • In this study, we investigated the antimicrobial and antifungal activity from the wild seaweeds of Jeju island. The active ingredients of the seaweeds were prepared by 80% methanol extraction. Antimicrobial and antifungal activity of seaweed extracts was examined. We found that 6 plant extracts among 45 plants, namely, Codium contractum, Undaria pinnatifida, Ishige sinicola, Ishige okamurai, Ishige okamuriai, Ecklonia cava, Hizikia fusiformis, Ulva fasciata, Ulva pertusa, Sargassum siliquastrum, Ecklonia kurome, Gracilaria textorii, significantly inhibited growth of harmful microorganisms. Additionally, according to DPPH assay. 2 plant extracts were found to have antioxidant activities. Taken together, these results suggest the possibility that 11 plant extracts can be utilized as an antimicrobial agent.

Screening of Plant Resources of anti-viral activity (항바이러스 활성 식물자원 탐색)

  • Aqueous extracts from eleven species of Korean medicinal plants were screened for antiviral activity against transmissble gastroenteritis virus (TGEV) and porcine epidemic diarrhae virus (PEDV). Seven of eleven plant extracts were found to have cytotoxicity for ST cells at the concentration of 1mg/ml, and ten extracts except Ephedra sinica were found to have non-cytotoxicity for Vero cells at the same concentration, Extracts of Zanthoxylum piperium, Cudrina tricuspidata, Clerodendron trichomum, Sophora flavescens, effectively inhibited TGEV, respectively. There were the antiviral activity against TGEV in the extracts of Glesdistsia sinensis, Euonymus alatus, Ficus carica, Solanum nigrum, Ephedra sinica, Xanthium strumarium, Acanthopanax seeliflorus in spite of their cytotoxicities. Nine extracts except Xanthium strumarium and Acanthopanax seeliflorus were found to have the antiviral activitity against PEDV. Extracts of Zanthoxylum piperium, Sophora flavescens was shown to have antiviral activities against both TGEV and PEDV.

Physicochemical Properties of Dextran Produced by Leuconostoc mesenteroides SM according to Concentration of Yeast Extract and its Modulation of Rheological Properties (효모 추출물 농도에 따른 Leuconostoc mesenteroides SM에 의해 생산된 dextran의 물리화학적 특성 및 물성개량)

  • Dextran was produced by $Leuconostoc$ $mesenteroides$ SM with various contents of yeast extract and its rheological properties were modulated, via an addition of hydroxypropyl methylcellulose (HPMC) of different viscosities. The conversion yield reached 90% after adding 3% yeast extract, which remained constant, thereafter. The acidity of the cultures was approximately 1.4 and 0.9% after fermentation for 24 h at 25 and $30^{\circ}C$, respectively. The total dextran content (107.3 g/kg) was the highest in the presence of the 3% yeast extract. Under the same conditions, the consistency, viscous modulus (G"), and elastic modulus (G') of the cultures were $37.6\;Pa{\cdot}s^n$, 38 Pa, and 50 Pa, respectively. The rheological properties of the culture were changed drastically by the fortification with HPMC of higher concentration and viscosity. The addition of 10% HPMC (4,000 cp) resulted in a significant increase in G" to 1,950 Pa. Furthermore, adding HPMC to a viscous culture resulted in a remarkable increase in both hardness and firmness.

Antioxidant Activity of Asteraceae Plant Seed Extracts (14종 국화과(Asteraceae) 식물 종자 추출물의 항산화 활성)

  • Approximately 10% of all angiosperms belong to the Asteraceae family. Plant species belonging to this family have traditionally been used as medicinal plants in the Korean Peninsula. We investigated the antioxidant activity of seed extracts from 14 species belonging to the Asteraceae family. Seeds with ≥ 90% percentage of filled seed and ≥ 50% final germination were used. The total phenolic content was the highest in Dendranthema zawadskii var. tenuisectum (13.5 mg of gallic acid equivalents (GAEs)/g seeds), followed by Dendranthema zawadskii var. latilobum (11.8 mg of GAEs/g seeds), and Callistephus chinensis (11.0 mg of GAEs/g seeds). The total flavonoid content was highest in C. chinensis (9.8 mg of quercetin equivalents (QEs)/g seeds), followed by D. zawadskii var. tenuisectum (7.2 mg of QEs/g seeds) and Taraxacum officinale (6.3 mg of QEs/g seeds). Our results showed that 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) scavenging activity was highest in D. zawadskii var. tenuisectum (57.4 ㎍/ml), followed by T. officinale (59.1 ㎍/ml) and D. zawadskii var. latilobum (65.0 ㎍/ml), with a half maximal inhibitory concentration (IC50) of DPPH scavenging activity. Furthermore, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity was highest in C. chinensis (26.2 ㎍/ml), followed by D. zawadskii var. tenuisectum (38.4 ㎍/ml), T. officinale (40.2 ㎍/ml), with a half maximal inhibitory concentration (IC50) of ABTS scavenging activity. Based on a cluster analysis according to the antioxidant activity, the 14 species were classified into five groups, with group 4 having the highest antioxidant activity and group 0 having the lowest antioxidant activity. D. zawadskii var. latilobum, D. zawadskii var. tenuisectum, T. officinale, and C. chinensis belonging to groups 3 and 4, exhibited high phenolic content and antioxidant activity and can be considered potent plant-derived natural antioxidants.

Industrialization of Jeju Plant Resources (제주 자원식물의 산업소재화)

  • 과학기술의 발달로 인해 국민의 생활수준 향상과 건강에 대한 관심 증대 등으로 인해 화장품을 비롯한 향장품산업, 기능성식품 및 천연물 신약 등에 대한 자원식물 유래 천연물질 수요가 증가되고 있다. 자원식물을 활용한 기능성 제품의 개발은 국민의 건강증진 뿐만 아니라 국제경쟁력 강화나 윤택한 삶의 추구에도 절대적 역할을 할 것으로 기대되고 있으며, 이러한 수요에 따라 제주의 특산 또는 우수 자원식물을 활용한 생리활성 물질에 대한 연구들이 활발하게 진행되고 있다. 우리나라에 자생하는 관속식물은 총 4,500여 종으로 국가생물종목록에 작성되어 있으며 제주의 생물자원 중 관속식물은 해발 1,950m 한라산의 영향으로 2,200여 종이 분포한다고 최근 기록되고 있다. 이 중 식용 및 약용으로 활용 가능성을 갖고 있는 식물자원들도 약 800여 종이 분포한다고 보고되고 있다. 이들 자원은 대한약전, 생약규격집, 중약본초, 중약대사전 등 문헌에 근거한 자원으로 공정서에 수록된 약용식물 약 500여 종 중 제주 분포 약용식물도 217종으로 조사되고 있다. 이러한 제주의 약용식물 자원은 다양한 한의약재로 활용되고 있으며 민간약으로 활용되는 약용식물도 170여 종이 포함되어 있다. 특히, 최근에는 나고야의정서 발효에 따른 자원전쟁이 시작되어 자원식물을 포함한 생물자원 관리의 중요성이 그 어느 때보다 부각되고 있어 각 지역마다 자생 생물자원에 대한 주권을 확보하기 위한 연구들이 많이 추진되고 있다. 이와 더불어 지자체연구소인 생물종다양성연구소에서도 제주에 분포하는 자원식물들에 대한 주권 확립을 위한 생태사진 900여 종, 표본 1,400여 종, 유전자 정보 800여 종 등을 확보하고 있으며 추출물 2,000점을 확보하여 산업소재화 연구를 추진하고 있다. 저자는 여기에 제주의 자원식물을 활용하여 화장품 원료등재, 건강기능식품개별인증 소재 및 천연의약품 소재 등 산업소재화 연구를 수행했었던 생물종다양성연구소의 연구 결과를 소개함으로써 유사한 목적으로 연구를 수행하는 대학, 연구소 및 기업으로의 정보를 제공하여 제주의 자원식물을 활용한 산업소재화 분야에 작은 도움이나마 제공하고자 한다.
