본 연구는 치주낭에 biofilm형태로 부착되어 질환을 유발시키고 항생제 빚 항균제에 저항을 일으키는 세균 독성요소를 규명하기 위해 시행된 기초연구이다. 치주질환의 주 병원균의 하나인 Porphyromonas gingivalis 381 biofilm의 세포외막에 특이하게 발현되는 단백질을 규명하기 위한 기초적인 자료를 얻기 위해 시행하였다. Porphyromonas gingivalis 381을 통상적인 세균 배양용 broth를 사용하여 혐기성 세균 배양기로 24시간 배양한 것을 대조군으로 하고, tissue culture plate를 이용하여 혐기성 배양조건 하에서24시간동안 biofilm을 형성하여 실험군으로 설정하였다. 세균을 수획하여 세포외막을 분리하고 isotonic isoelectric focusing을 시행한 결과 주로 약 20-30 kilodaltons에 해당하는 수종의 세균세포막 단백질이biofilm으로 배양한 세균에서 더 상승적으로 발현됨이 관찰되었고, 상이한 수종의 단백질도 planktonic culture broth로 배양한 세균에서 다 상승적으로 발현됨을 관찰할 수 있었다. 이것은 세균의 배양조건과 환경에 따라 그 외막 단백질이 서로 다르게 발현됨을 입증하는 기초적인 자료로서 향후 단백질의 동정과 성격을 규명하는 근간 실험으로 추진할 계획이다.
Two groups of entomopathogenic bacteria, Xenorhabdus and Photorhabdus, are known to suppress insect immune responses by inhibiting eicosanoid biosynthesis. This study used these bacterial culture broths to develop novel biochemical insecticides against the diamondback moth, Plutella xylostella. Though the bacterial culture broths alone showed little insecticidal activity, they significantly enhanced pathogenicity of Bacillus thuringiensis against the fourth instar larvae of P. xylostella. Sterilization of the bacterial culture broth by autoclaving or $0.2\;{\mu}m$ membrane filtering did not influence the synergistic effect on the pathogenicity of B. thuringiensis. Three metablites identified in the culture broth of X. nematophila also showed similar synergistic effects. In field test, both entomopathogenic bacterial culture broth also enhanced the control efficacy of B. thuringiensis against P. xylostella.
Entomopathogenic bacteria (Xenorhabdus nematophila, X. sp. and Photorhabdus temperata subsp. temperata) isolated from entomopathogenic nematodes express potent insecticidal activity in insect hemocoel. They are also known to suppress insect immune mediation by inhibiting phospholipase $A_2$, leading to host immunosuppression. This study analyzed effects of their cultured broths on inhibiting insect immunosuppression. For this, we removed all bacterial cells using $0.2\;{\mu}m$ pore sized membrane from the bacteria-cultured broth. All three sterilized cultured media, in dose-dependent manners, significantly inhibited hemocyte-spreading behavior of 5th instar larvae of Spodoptera exigua. However, they showed differential inhibitory activities among different bacterial species, in which X. nematophila showed the most potent inhibitory activity. This immunosuppressive effect was applied to increase the pathogenicity of Bacillus thuringiensis (Bt). All three bacterial cultured broths including bacterial cells significantly potentiated Bt pathogenicity against young S. exigua larvae when each of them was orally administered in a mixture of low dose of Bt. Finally, we tested the effect of oral administration of the cultured media containing the immunosuppressive compound(s) secreted by the bacteria. The membrane-sterilized cultured broths were mixed with the low dose of Bt and then orally administered to the young S. exigua. Only the cultured medium of X. nematophila showed increase of Bt pathogenicity. These results indicated that the; cultured media of the three bacteria possessed immunosuppressive factor(s), which may act to potentiate Bt toxicity to young S. exigua larvae.

An entomopathogenic bacterium, Photorhabdus temperata ssp. temperata (Ptt), suppresses insect immune responses and facilitates its symbiotic nematode development in target insects. The immunosuppressive activity of Ptt enhances pathogenicity of various microbial pesticides including Bacillus thuringiensis (Bt). This study was performed to select a cheap and efficient bacterial culture medium for large scale culturing of the bacteria. Relatively cheap industrial bacterial culture media (MY and M2) were compared to two research media, Luria-Bertani (LB) and tryptic soy broth (TSB). In all tested media, a constant initial population of Ptt multiplied and reached a stationary phase at 48 h. However, more bacterial colony densities were detected in LB and TSB at the stationary phase compared to two industrial media. All bacterial culture broth gave significant synergism to Bt pathogenicity against third instars of the diamondback moth, Plutella xylostella. Production of bacterial metabolites extracted by either hexane or ethyl acetate did not show any significant difference in total mass among four culture media. Reverse phase HPLC separated the four bacterial metabolites, which were not much different in quantities among four bacterial culture broths. This study suggests that two industrial bacterial culture media can be used to economically culture Ptt in a large scale.
We evaluated cultivation methods to obtain pure cultures of previously uncultivated bacteria from soil. Soil bacteria (suspensions) were inoculated onto various oligotrophic media with one of the following additives: 1) soil extract; 2) anthraquinone disulfonate (humic acid analogue); 3) acyl homoserine lactones (quorum-signaling compounds); 4) catalase (for the protection of bacteria from exogenous peroxides). After the relatively long period (60 days) of incubation with elevated concentrations of $CO_2$ (5%, v/v), the media containing catalase showed the highest colony count. We purified 147 randomly selected colonies from the media and the isolates were subjected to the phylogenetic analyses of their 16S rRNA gene sequences. Phylogenetic analysis revealed that approximately 30% of the isolates might belong to novel species or novel family, suggesting that the media and incubation conditions used could be useful for the cultivation of as-yet-uncultured bacteria. Especially, bacteria belonging to the phylum Acidobacteria, ubiquitous bacterial taxon known as an uncultured bacterial group (at least difficult to culture from environmental samples), were successfully cultured in this study.

Perchlorate ($ClO_4^-$) is an emerging contaminant detected in soil, groundwater, and surface water. Previous study revealed bacterial community in the enrichment culture tdegraded perchlorate using elemental sulfur as an electron donor. Quantitative and qualitative molecular methods were employed in this study to investigate archaeal community in the enrichment culture. Real-time qPCR showed that archaeal 16S rRNA gene copy number in the culture was about 1.5% of bacterial 16S rRNA gene copy number. This suggested that less archaea were adapted to the environment of the enrichment culture and bacteria were dominant. DGGE banding pattern revealed that archaeal community profile of the enrichment culture was different from that of the activated sludge used as an inoculum for the enrichment culture. The most dominant DGGE band of the enrichment culture was affiliated with Methanococci. Further research is necessary to investigate metabolic role of the dominant archaeal population to better understand microbial community in the perchlorate-reducing enrichment culture.
Bacteriolytic myxobacteria have been known to secrete various antifungal metabolites against several soilborne phytopathogens including Phytophthora. Among the three isolates of Myxococcus spp., KYC 1126 and KYC 1136 perfectly inhibited the mycelial growth of Phytophtora capsici in vitro. In order to show the biocontrol activity on Phytophthora blight of hot pepper, we tried to find the best way of application of a myxobacterial isolate. Although KYC 1126 fruiting body was easily grown on the colony of Escherichia coli as a nutrient source, it did not control the disease when it was pre-applied in soil. Before the bioassay of a liquid culture filtrate of KYC 1126 was conducted, its antifungal activity was confirmed on the seedlings applying with the mixture of the pathogen's zoospore suspension and KYC 1126 filtrate. On greenhouse experiments with five and four replications, the control value of KYC 1126 on phyllosphere and rhizosphere was 88% and 36%, respectively. Whereas, the control value of dimetnomorph+propineb on phyllosphere was 100% and that of propamorcarb on rhizosphere was 44%. There was a phytotoxicity of the myxobacterial filtrate when seedlings were washed and soaked for 24 hours. Gummy materials were covered with roots. And stem and petiole were constricted, then a whole seedling was eventually blighted.
This review describes the characteristics of various unculturable soil bacteria, successfully-cultivating examples of those bacteria, and the diverse factors to be considered for successful cultivation. Most importantly, the selection of proper media is very important because unculturable bacteria demand different types of nutrients at various concentrations of substrates, nitrogens and phosphorus. To develop a new medium to successfully culture unculturable bacteria from soil, molecular ecological studies should be combined together. The inoculum size on a plate is also important: less than 50 bacterial cells are recommended to be plated on a single culture plate. The environmental factors such as pH and salt concentration of the medium need to be adjusted as similar as possible to mimic the original soil environments, and the trial of the various temperatures and extended period of cultivation are better. Since one cannot simply tell about which one was unculturable among a great number of colonies grown on a newly developed medium, some suitable detection methods and fast identification methods are required. Many soil bacteria live with cooperation one another in their communities, so that enrichment such as coculture of using other bacterial metabolites and subsequent pure cultures can also guarantee successful cultivation of the previously uncultured bacteria in soil. Here, this review will discuss for the future perspectives to culture the unculturable soil bacteria.

헤민과 메나디온 제한에 의한 Porphyromonas gingivalis(P. gingivalis) W50의 병독력의 변화를 검색하고자, 실험관내 병독력을 NIH 3T3 세포의 세포활성 변화로 관찰하였고, 생체내 병독력은 배양상청액을 ICR mouse 피하조직에 주사한 후의 염증반응을 관찰하였다. 헤민 존재 하에 배양한 P. gingivalis W50 배양상청액에 의한 mouse 3T3 세포의 세포활성은 헤민과 메나디온 없이 배양한 세포의 활성보다 낮았다. 헤민과 메나디온을 첨가하지 않고 배양한 세균의 생체내 병독력은 중등도의 염증세포 침윤과 울혈에 의한 출혈, 미약한 세포간질의 부종과 근육 파괴를 보였다. 메나디온 존재 하에서 배양한 세균은 미약한 염증세포의 침윤, 울혈에 의한 출혈 및 근육의 파괴가 관찰되었다. 헤민 존재하에서 중등도의 울혈에 의한 출혈, 미약한 세포간질의 부종, 염증세포의 침윤 및 근육파괴가 관찰되었다. 헤민과 메나디온 존재 하에서 배양한 세균은 심한 염증세포의 침윤과 중등도의 세포간질의 부종 및 울혈에 의한 출혈을 보였다. 이상의 연구 결과 P. gingivalis W50 배양 상층액의 병독력은 헤민에 의하여 영향을 받는 것으로 생각된다.
Clumping of cells is one of the major obstacles to culture wild myxobacterial strains in liquid media. In an effort to solve this problem, we tried a method isolating spontaneous mutants that grow dispersed in liquid media from a wild myxobacterial strain. Myxococcus stipitatus KYC1001, a newly isolated strain from Gyearyongsan National Park in Korea, clumps and sticks to the surface of culture vessels as other wild myxobacteria behave in liquid media. Taking an advantage of the characteristics that dispersed mutant cells would grow dispersed while most other wild type cells would clump and stick to the surface of culture vessels, spontaneous dispersed mutants were enriched by repeated subculturing of culture supernatant. A resultant mutant, KYC2001, did not form any clumps nor stick to the surface of culture flasks, but grew completely dispersed in liquid. Meanwhile, three other spontaneous mutants, KYC2002, KYC2003, and KYC2004, shelved partially dispersed phenotype. A major portion of the cells grew dispersed in liquid but they still formed some clumps.
