Enhancement of Growth and Bioactivity of Pleurotus eryngii Mycelia by Spent Coffee Ground (커피박 첨가에 의한 새송이버섯 균사체의 생육 및 생리활성 증가)

  • Pleurotus eryngii. one of the most popular edible mushrooms, has been well known for its biological activities such as antioxidation, antitumor and immune modulation. Spent coffee ground(SCG) that is a waste product from the coffee industry has been continuously investigated for its reutilization. In this study, SCG was added to the fungal cultuvation medium and analyzed for its effect on the growth and physiological activity of P. eryngii mycelia. It was clearly demonstrated that SCG could accelarate mycelia growth. 1% SCG culture was very notable by showing 2.5-fold higher dry cell weight comapred to the control culture, which suggested SCG as an excellent activator for the growth of P. eryngii mycelia. By the addition of SCG, polyphenol content was increased by two fold but there was no change in polysaccharide content. In the analysis of DPPH scavenging activity, SCG was determined as a valuable source in order to significantly increase the antioxidative activity of the mycelium.

Control of Colletotrichum acutatum and Plant Growth Promotion of Pepper by Antagonistic Microorganisms (길항균주를 이용한 고추탄저병균(Colletotrichum acutatum) 방제 및 식물생장촉진효과)

  • Anthracnose caused by Collectotrichum acutatum is the most devastating disease of pepper plants in Korea. In this study, we evaluated the effect of selected antagonistic bacteria on control of anthracnose and plant growth promotion of pepper plants under field conditions. Four different bacterial isolates used in the current study were isolated from the pepper rhizosphere (GJ01, GJ11) and tidal flat (LB01, LB14) in previous studies. Four bacterial isolates, together with a control strain (EXTN-1), showed antifungal activity against C. acutatum in a dual culture assay. To test for plant growth promotion effect, seedling vigor index and growth parameters of pepper were measured under field condition. As a result, all four bacterial isolates were effective for improving plant growth promotion. The strain GJ01 was the most effective in improving the seedling vigor on pepper, but the strain GJ11 in increasing the pepper fruit yield. The incidence of anthracnose was inhibited in the range of 63.2~72.5% by treatment of four bacterial isolates. The current study indicated that the four bacterial isolates could be used as potential biological control agents of anthracnose disease of pepper.

Changes in Growth and Nutrient Contents of Populus ablba var. pyramidalis treated with Fertilizers and Absorbents in an Arid Region of China (중국 건조지역에서 시비와 보수제 처리에 의한 신강포플러의 생장 및 묘목의 양분 함량 변화)

  • To investigate soil management practices for increasing tree growth in arid regions of China, Xinjiang poplar (Populus alba var. pyramidalis) seedlings were planted in a nursery and windbreak and were treated with fertilizers (nitrogen and phosphorus) and absorbents (artificial; K-SAM and natural: Yuho (Artemisia ordosica)) at Dengkou in Inner Mongolia. Seedling height, diameter at breast height (DBH), biomass, and nitrogen (N) and phosphorus (P) concentrations were measured 1.5 years after the treatments. Soil texture was sandy loam for nursery and clay loam for windbreak, soil pH was 8.78 for nursery and 8.51 for windbreak, and total soil N concentration was 0.06% for nursey and 0.08% for windbreak. [n general, seedling height and DBH increased in the N fertilizer treatment compared to control or other treatments both in nursery and windbreak. Also seedling growth increased after the treatments of artificial and natural absorbents in windbreak. Total above- and belowground biomass increased after the N + P treatment. Foliage N and P concentrations following N and/or P treatments were same in the nursey, however, they significantly increased in the windbreak indicating N deficiency. It appeared that N was a limiting factor for early growth of Xinjiang poplar seedlings and artificial and natural absorbents (K-SAM and Yuho) would be useful to increase seedling growth in the arid region.

Rooting Promotion of 'New Guinea' Impatiens (Impatiens hawkeri hybrida) Cuttings by Treatment of Plant Growth Retardants and Triazole Fungicides (식물생장억제제와 Triazole계 살균제 처리에 의한 'New Guinea' 임파첸스(Impatiens hawkeri hybrida) 삽수의 발근촉진)

  • This study was carried out to investigate the effect of plant growth retardants (daminozide, chlormequat, hexaconazole, uniconazole, paclobutrazole) and fungicides of triazole chemical (microbutanil, difenoconazole, terbuconazole, bitertanol) on the rooting of 'New Guinea' impatiens cuttings. In both 'Papete' and 'Anguilla', the formation of adventitious roots was promoted by all concentrations of chlormequat, hexaconazole, uniconazole, and paclobutrazole treated. More adventitious roots were formed with hexaconazole, uniconazole, and paclobutrazole than with chlormequat. Length of the longest root and rooting zone decreased at higher concentrations of chlormequat, hexaconazole, uniconazole, and paclobutrazole in 'Papete', while 'Anguilla' showed less responses. In contrast with plant growth retardants, the formation of adventitious roots was remarkably promoted by fungicides of triazole chemical such as microbutanil, difenoconazole, terbuconazole, and bitertanol in 'Pepete' and 'Martinique' impatiens, but was not in 'Pagopago'. However, length of the longest root and rooting zone showed more sensitive response in 'Martinique' and 'Pagopago' than in 'Pepete'. These results suggest that fungicides of triazole chemical can be effectively used for rooting promotion of impatiens cuttings.

Effects of Ethylsalicylic Acid on Growth and Rubisco/Rubisco Activase in Tobacco Plant Cultured under Cadmium Treatment in vitro (카드뮴(Cd) 하에서 기내 배양된 담배의 생장과 Rubisco/Rubisco Activase에 대한 Ethylsalicylic acid의 영향 분석)

  • Growth induced by cadmium (Cd) and ethylsalicylic acid (ESA) and the effect of ESA on rubisco/rubisco activase were studied in tobacco. The effect of denaturants on rubisco/rubisco activase was also investigated. In order to determine optimal concentration of ESA for growth of tobacco, tobacco was treated with $10^{-6}$-10 mM. It was found that its growth was the highest at $10^{-4}$ mM ESA. In the experiment using control, Cd treated group, ESA treated group, and Cd and ESA mixture group, ESA alone showed the highest growth and Cd showed the lowest growth. Cd treated group was the lowest in both rubisco/rubisco activase content and activity. ESA reduced the rubisco/rubisco activase content, but increased their activity. The activity of rubisco was inhibited by treating L-cysteine, urea, thiourea, ${\beta}$-mercaptoethanol, and EDTA other than guanidine-HCl in control group. L-cysteine, urea, thiourea, and guanidine-HCl treatments showed no change, but ${\beta}$-mercaptoethanol and EDTA increased rubisco activase activity. In conclusion, ESA inhibited the content of rubisco and promoted its activity, whereas promoted the content of rubisco activase and inhibited its activity. In addition, the content and activity of rubisco and rubisco activase inhibited by Cd were recovered by ESA. The activity of rubisco and rubisco activase by Cd and ESA was inhibited by the denaturant and the recovery of ESA inhibited by Cd was lost by the denaturant.

Herbicidal and Antifungal Activities of the aqueous extracts of Persicaria longiseta (개여뀌의 제초 및 항균활성 탐색)

  • The study researched germination of the plants and growth of Fungus according to concentration of aqueous extracts in order to provide basic data for developing natural agricultural resources by using Persicaria longiseta. The seed germination of Amaranthus spinosus was inhibited at 25% P. longiseta extract, while Agrostis stolonifera ssp. palustris was not affected at all concentrations tested. Especially, the seed germination rate and fresh weight of Trifolium incarnatum at 20% P. blumei extract were higher than those of control plot. The early growth of most receptor plant seedlings was promoted at 25% and 50% of P. blumei extracts, but the radicle growth of all receptor plants was significantly inhibited at > 25% of P. longiseta extract. The response of receptor plants to P. longiseta extract was different according to the plant species and the plant parts. The growth of plant pathogenic fungus in PDA medium showed an increasing inhibition tendency with increasing concentrations of P. longiseta extract. Especially, P. longiseta extract showed the greatest antimicrobial activity against Phytophthora infestans, Phythium graminicola, and Pythium venterpoolii. The content of total phenolic compound in P. longiseta was higher in leaves (1082.3 mg/L) but lower in roots (228.6 mg/L) and stems (207.8 mg/L), which is an allelopathic chemical. As these results are summarized, P. longiseta have competitive advantage because they release phenolic compounds with allelopathic effect and affect on germination, growth and fungi growth on underground flora compared to native plants and they have eligibility for natural herbicide and germicide.

Antifungal Activity and Plant Growth Promotion by Rhizobacteria Inhibiting Growth of Plant Pathogenic Fungi (식물병원성 진균을 억제하는 근권세균의 항진균능과 식물생장촉진능)

  • Since many pesticides cause various health and environmental problems, alternative measures to replace them are needed, and the bacteria producing the antifungal substances can be one of them. In this study, several rhizobacteria were isolated and their antifungal activities against some important plant pathogenic fungi were examined. Pseudomonas otitidis TK1 and Paenibacillus peoriae RhAn32 inhibited the growth of Fusarium oxysporum f. sp. niveum and F. oxysporum f. sp. lycopersici by 49.8% and 45.6%, and 45.1% and 48.3%, respectively compared to those of the control. P. peoriae RhAn32 also decreased the growth of F. oxysporum f. sp. raphani by 37.5%. This growth inhibition might be due to the production of antifungal substances, such as siderophore, hydrogen cyanide and chitinase, which were produced by these rhizobacteria. P. otitidis TK1 also produced plant growth hormones indole acetic acid and indole butyric acid at $293.41{\mu}g/mg$ protein and $418.53{\mu}g/mg$ protein, respectively. When P. otitidis TK1 and B. cereus TK2 were inoculated together with F. oxysporum f. sp. lycopersici to the 4 weeks grown tomato seedlings and incubated additional 8 weeks, the stem lengths of tomato increased up to 45.7% and 55.3% and root lengths were raised to 64.9% and 60.8%, respectively than those of the control group. The wet weights increased by 118% and 182%, respectively compared to the control group.

Effects of Growth Regulators on Shoot Differentiation and Bulblet Formation in Shoot-Tip and Bulb-Scale Cultures of Lilium longiflorum (백합 경단 및 인편배양으로부터 유식물체 분화 및 자구형성에 미치는 생장조절제의 영향)

  • Regulation of organ differentiation by growth regulators was investigated through the shoot-tip and bulb-scale cultures of Lilium longiflorum (cv Georgia). When shoot tips were placed on MS medium supplemented with 0.1 mg/L NAA alone or 0.1 mg/L NAA and 0.1 mg/L BA, axillary shoots were proliferated. Root diffentiation and growth were stimulated on the basal medium. Although growth regulation did not seem to be necessary when bulb scales were used as explants for shoot differentiation, its differentiation was promoted vigorously by 0.2 mg/L NAA, but suppressed by BA. Bulblets were formed from bulb-scale-derived plantlets cultured on MS medium supplemented with 0.1 mg/L IBA. And more bulblets were formed from the plantlet in MS medium supplying 0.2 mg/L NAA with 6% than3% sucrose

Effects of Cutting Condition on Rooting and Early Growth of a Rare Endemic Juniperus chinensis var. sargentii HENRY (삽목조건이 희귀수종 눈향나무의 발근과 초기생장에 미치는 영향)

  • This study was conducted to elucidate the best cutting condition in propagation of Juniperus chinensis var. sargentii in respect of survival rate, rooting rate, and early growth after cutting via process of cutting time, cutting media, cutting type and growth regulators. As for cutting time, rooting rate was 68.3% on April, which was higher than 39.5% on May and 32.5% on June. The best cutting media was stream sand of rooting rate 72.7%. Those of mixed soil and decomposition were 40.1% and 27.5% each. The effect of growth regulators was different according to the kind of cutting media. There was no effect on stream sand media which showed the highest rooting rate, while it was very effective on the media of decomposition of granite, which rooting rate showed the lowest. Hardwood as cutting materials showed somewhat higher survival and rooting rate compared to semi-hardwood. The cutting with hardwood condition to stream sand on April, it is regarded to be the most effective condition for Juniperus chinensis var. sargentii to increase survival and rooting rate. Therefore in this experiment it is considered that the best season cutting is April, and the most useful media and plant materials stream sand, and hardwood respectively.

Growth Promotion of Lettuce by Biofertilizer, BIOACTIVE, Prepared from Bacillus subtilus HR-1019 and N-acetyl-thioproline (Bacillus subtilus HR-1019와 N-Acetyl-thioproline으로 제조한 미생물처리제, BIOACTIVE에 의한 상추의 생장 촉진)

  • A biofertilizer, BIOACTIVE, was manufactured by N-acetyl-thioproline (ATCA) and mineral phosphate solubilizing bacteria. The growth promoting effect of the biofertilizer on lettuce was evaluated under three different pot conditions, and its stability was assessed in the field. According to the results of the pot experiments, plant growth was improved compared with that of control: 128%, 122%, and 153% for the leaf number, leaf length, and leaf mass, respectively. Applying the manufactured biofertilizer increased the concentration of phosphate: 118% and 132% in the cultivation soil and plant cells, respectively. These show that BIOACTIVE may have potential as an effective biofertilizer in agriculture.
