Anti-wrinkle Activity of Lindera obtusiloba Extract (생강나무 추출물의 광노화에 의한 주름형성 억제 효과)

  • Chronic exposure of solar ultraviolet (UV) light to human skin results in photoaging with wrinkle formation. This study was performed to investigate anti-wrinkle effects of Lindera obtusiloba extract (LO) on UVB-induced wrinkle formation. We first measured cell proliferation and type I pN collagen synthesis activities in normal human dermal fibroblasts. Cell proliferation and type I pN collagen synthesis were increased by 33.8 % and 91.8 %, respectively, compared with no treatment control. SKH-1 hairless mice were topically applied 5 % LO solution for 10 weeks with UVB irradiation three times a week. After 10 weeks, a visual assessment and replica assay were performed on each mouse. According to visual assessment of close-up photos and skin replica, application of 5 % LO solution inhibited UV-induced wrinkle formation in mouse skin as compared to the vehicle-applied control mice. These results indicated that LO could protect skin wrinkle formation caused by chronic photo-irradiation in hairless mice.

Effect of Lindera obtusiloba extract on cancer metastasis (생강나무 추출물의 암전이 억제효과)

  • Objectives : In the present study, anti-metastatic properties of the methanol extract of L. obtusiloba (MLO) were evaluated. Methods : To determine the effect of MLO on cancer metastasis, we checked matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) activities and expressions in B16F10 melanoma cells. In addition, we performed cell migration assay as well as invasion assay using Matrigel. Finally, we used an in vivo lung metastasis model to confirm the anti-metastatic activity of MLO. Results : 1. MLO showed potent inhibitory effects on MMP-2 and MMP-9 activities and expressions via down-regulation of activation of NF-${\kappa}B$ in B16F10 melanoma cells. 2. Melanoma cell migration and invasion were down-regulated by MLO treatment. 3. Not only in vitro model, but MLO also significantly suppressed lung metastasis in vivo. Conclusions : The present results indicate that MLO has strong inhibitory effect on cancer metastasis. Therefore, L. obtusiloba could be a valuable anti-metastatic agent.

Isolation and Structure Elucidation of Flavonoid Glycosides from Lindera obtusiloba BL. (생강나무에서 분리한 Flavonoid 배당체의 화학구조)

  • 생강나무 (Libdera obtusiloba BL.)의 페놀성 화합물들을 분리하기 위하여 잎고 줄기의 MeOH 엑스로부터 계통 분획한 EtOAc 분획물은 silica gel column chro-matography로 분리하여 2종의 화합물을 얻었다. IR, UV,$^1H-NMR$, $^13C-NMR$ 등의 분석을 행하여 화학구조를 밝힌 결과 이들 화합물의 화학구조는 잎에서 $quer-cetin-3-O-\alpha-L-rhamnopyranoside(quercitrin),$ 줄기에서는 quercetin $3-O-\beta-D-galactoyranoside(hyperoside)로$ 각각 결정하였다. 이 화합물들은 생강나무에서 처음으로 분리하였다.

Diversity of Foliar Endophytic Fungi Isolated from Lindera obtusiloba in Korea (생강나무(Lindera obtusiloba)의 잎에서 분리한 내생균의 다양성)

  • Leaves of Lindera obtusiloba were collected from four sites in Gangwon Province, Korea. Endophytic fungi were isolated from the leaves and identified using ITS sequences of rDNA. Total twelve species belonging of endophytic fungi were identified; Alternaria alternata, Annulohypoxylon annulatum, Creosphaeria sassafras, Diaporthe eres, Discosia sp., Epicoccum nigrum, Glomerella acutata, Glomerella cingulata, Paraconiothyrium brasiliense, Pestalotiopsis neglecta, Phomopsis amygdali, Xylaria sp. The endophytic fungus, Phomopsis amygdali, was the most dominant species isolated from L. obtusiloba in this study and the fungal diversities varied in the different sites.

Essential Oils from Leaves and Twigs of Lindera obtusiloba (생강나무 잎과 가지의 정유성분)

  • Essential oils which were isolated from leaves and twigs of Lindera obtusiloba Blume by steam distillation method were analyzed by the NIST and Wiley 6 Library method, using GC-MS. 24 components from the experiment were identified, and sesquiterpenoids were major of the components. The main constituents in the leaves were germacrene B (17.78%), ${\beta}-caryophyllene$ (17.50%), phytol isomaer (13.36%) and $(-)-{\beta}-elemene$ (11.07%), and the main in the twigs were ${\beta}-elemol$ (24.47%), ${\alpha}-cadinol$ (13.73%), camphor (9.83%) and ${\beta}-citronellol$ (9.54%). Acyclic diterpene such as phytone, phytol and phytol isomer were isolated for the first time in the leaves of L. obtusiloba.

  • This study was analyzed to the characteristics of the Lindera obtusiloba eaf teas by various manufacture methods. Moisture content was the highest in the tea processed by air dry method, and the lowest in the tea roasted after steaming. The contents of soluble solid was higher in the fermented tea than in the others, and the lowest in the roasted tea. The content of tannin was the higher in the roasted after steaming tea than in the others, and the lowest in the fermented tea. The content of vitamin C was the higher in the roasted tea than in the others, and the lower in the steamed tea. The mineral contents of leaf teas was orderly K, Na, Mn, Ca, Fe, Mg, and Zn. The roasted after steaming tea was estimated to the most excellent in the sensory evaluation of leaf teas.

Antimicrobial activities of Lindera obtusiloba Blume and Zanthoxylum piperitum DC extracts (생강나무(Lindera obtusiloba Blume)와 초피나무(Zanthoxylum piperitum DC) 추출물의 항균활성)

  • Ethanol and hot water extracts were prepared from Lindera obtusiloba Blume (LO) and Zanthoxylum piperitum DC (ZP) and used to evaluate their antimicrobial activities and thermal stability against six foodborne pathogens (3 gram-positive and 3 gram-negative bacteria). The antimicrobial activities were assessed using the agar diffusion method, and the thermal stabilities of extracts were examined after heat treatment at 60, 70, 80, and $100^{\circ}C$ for 10 min. The zones of inhibition by the LO extract or the ZP extract of the tested microorganisms were in the range of 21-30 mm and 19-25 mm, respectively, at 100 mg/mL concentrations. The 60% ethanol extract and the hot water extracts from LO showed the strongest antimicrobial effects against MRSA and Staphylococcus aurues, respectively. For the extract from ZP, the strongest antimicrobial effect was shown against S. aurues by 60% ethanol, and the weakest antimicrobial effect was shown against E. coli by the hot water extracts. The ZP extracts showed that the gram-positive bacteria were more sensitive than gram-negative bacteria. For the thermal stability of the extracts, the antimicrobial effects stabilized after heat treatment. Overall, the data suggest that the extracts have a potential for application in various food products for which a natural antimicrobial additive is desired.

Study on Classification of Quercus mongolica Forests in Kangwon-do by Phytosociological Method and TWINSPAN (식물사회학적(植物社會學的) 방법(方法)과 TWINSPAN에 의한 강원도 신갈나무림(林)과 분류(分類)에 관(關)한 연구(硏究))

  • Employing the relev$\acute{e}$ method of Muller-Dombois & Ellenberg and Braun-Blanquet, 100 plots were sampled in the Quercus mongolica community of Mt. Odae, Mt. Chumbong and Mt. Jungwang in Kangwon-do. To analyze the data, phytosociological method and TWINSPAN were used. 1. Quercus mongolica forests were recognized as Quercus mongolica-Acer pseudosieboldianum community group, and were classified into Quercus mongolica-Carpinus cordata community, Quercus mongolica-Acer pseudosieboldianum typical community, Quercus mongolica-Lespedeza maximocoiczii community and Quercus mongolica-Abies nephrolepis community by phytosociological method. Quercus mongolica-Carpinus cordata community was subdivided into Acer mandshuricum subcommunity and typical subcommunity. 2. According to the classification of TWINSPAN, it was categorized into five groups, such as Quercus mongolica-Acer mandshuricum community, Quercus mongolica-Carpinus cordata community, Quercus mongolica-Acer pseudosieboldianum community, Quercus mongolica-Lindera obtusiloba community, and Quercus mongolica-Abies nephrolepis community. 3. Since the results of phytosociological community classification, which identify five groups of the community, were consistent with that of TWINSPAN, it proved that these two methods could be complement when one do a community classification.

  • 본 연구에서는 산불발생 시 삼림 내 가연물의 화재강도 및 산불위험성을 예측하기 위하여 영동지역에서 자생하는 관목류 가운데 주요 분포수종으로 생강나무와 초피나무를 대상으로 생엽과 가지부위를 채취하여 착화특성, 화재전파특성, 피난특성을 고찰하였다. 발화온도 범위는 400$^{\circ}C$${\sim}$440$^{\circ}C$로 확인되었으며 생강나무의 생엽은 착화가 가장 빠르게 개시되었으며 가지부위는 생엽보다 착화는 늦으나 착화 후 화염유지시간이 길고 비교적 빠른시간에 많은 열량을 방출하는 것으로 나타났으며 초피나무의 가지부위는 연기방출량과 CO 및 $CO_2$의 방출량이 많은 것을 확인할 수 있었다.

Antioxidant and Anti-inflammatory activity of Saccharomycopsis fibuligera fermented Lindera obtusiloba flower extract (Saccharomycopsis fibuligera 발효 생강나무 꽃 추출물의 항산화 및 항염효과)

  • In this study, the antioxidant activity and anti-inflammatory activity of Lindera obtusiloba flower fermentation extracts were identified. To evulate antioxidant activity and antioxidant concentration, polyphenol concentration measurements, flavonoid concentrations measurements, DPPH experiments, and ABTS experiments were conducted. In Flavonoids, DPPH and ABTS assay, antioxidant activity were increased after fermentation. At this time, the flavonoid concentration was 5.0%, the DPPH experiment showed 33.27% and the ABTS experiment showed 29.82% antioxidant increase. In the anti-inflammatory experiment, we conducted a cytotoxicity experiment and an anti-inflammatory experiment for LPS-induced inflammation. Cytotoxicity showed low cytotoxicity in both control and fermentation groups, but lower cytotoxicity in fermentation groups. In the case of NO production inhibition, fermented Lindera obtusiloba flowers showed an increase in anti-inflammatory activity by more than 50% compared to the control group, showing that they can be used as a cosmetic ingredient with anti-inflammatory function.
