내산소성 비피더스균의 분리와 생육 특성 1. 내산소성 비피더스균의 분리와 동정

  • 건강한 성인의 분변으로 부터 호기적인 평판배지에서의 생육이 가능한 비피더스균을 선별 분리하는 조건과 상기 분리주의 특성을 규명한 결과는 다음과 같다. 1.5%의 탈섬유소 말혈액을 첨가한 BLB Agar Medium에 인체 분변을 호기적으로 표면 배양하여 내산소성 비피더스균 변이주를 분리하였다. 상기 분리주는 BLB Agar Medium 평판배지에 호기적으로 배양 했을때 $1.1\;{\times}\;10^9$ cfu/ml 이상의 높은 균수를 유지하였다. 2. 상기의 내산소성 비피더스균 분리주를 BLB Agar Medium 에 호기적으로 배양했을때 0.5mm, 연유갈색, 원형, 융기상의 윤택한 집락을 형성하였으며 현미경 검경 결과 Y 자형 분지상과 V자형 만곡간균상, 팽윤상 등의 전형적인 비피더스균 형상으로 나타냈다. 3. 탄수화물 발표실험 결과 상기의 내산소성 비피더스균 분리주는 Bif. adolescentis 로 동정되었으며 F6PPK GKA효소 활성 함께 매우 높은 ${\alpha}$-Galactosidase, $\beta$ - Galactosidase, ${\alpha}$-Galactosidase, 효소 활성을 나타냈다. 한편 아무것도 첨가하지 않은 순수한 우유 중에서의 발효성 및 호기적인 생육능이 매우 양호하였다.

  • The aim of this study was to evaluated the efficacy of mixture of Lactic Acid Bacteria (LAB) and bifidobacteria supplement, which are contained with Lactobacillus acidophilus, Bifidobacterium longum SPM1205, and Pediococcus pentosaceus for the management of constipation in animal model and clinical trials. 5 ICR mice and 4 female constipation subjects were orally taken mixture of LAB and bifidobacteria for 2 weeks. We investigated the number of fecal LAB and harmful enzymes activities before and after mixture of LAB and bifidobacteria application. As a result, fecal LAB count was increased and harmful enzymes activities of intestinal microflora were generally decreased after mixture of LAB and bifidobacteria application. Also, 61 female subjects were randomly assigned to receive either mixture of LAB and bifidobacteria or lactose and were taken three times a day for 2 weeks. Then, we analyzed mixture of LAB and bifidobacteria effect through the questionnaires. Daily consumption of this mixture of LAB and bifidobacteria improved the constipation in constipation group (56.3%) compared with lactose application group (26.7%). Furthermore, after mixture of LAB and bifidobacteria treatment, frequency of hard stool decreased from 0.22 to 0.03. These results indicated that mixture of LAB and bifidobacteria application is effective to improve the constipation.

The Effect of Fermented Milk by Bifidobacterium bifidum on Serum Lipid Metabolism in Rats Treated High Fat Diet (비피더스균(Bifidobacterium bifidum )에 의해 발효된 우유가 고지방식을 섭취한 흰주와 혈청 지질대사에 미치는 효과에 관한 연구)

  • Selecting B. bifidum K-7 out of the bifidobacteria separated from healthy adults in the age of 20s which shows high degree of acid tolerance and bile tolerance, as the main bacterium, this study of find how the bifido-bacteria cause hypocholesterolemic effect in the high fat diet. In order to do this Sparague-Dawley male rats with the initial weight 200 g in average were assigned to four experimental group: 1) high fat diet & milk, 2) high fat diet & the milk added with bifidobacteria, 3) high fat diet & the milk added with microencapsulated bifidobacteria, 4) high fat diet & the fermented milk by bifidobacteria. The numbers of bifidobacteria ingested daily per rat through each type of the above mentioned milk are 10$^{9}$ CFU, 10$^{9}$ CFU respectively. Hypocholesterolemic effect and high level of serum phospholipid were observed in the group fed with fermented milk being compared with the group fed wih no bacterium but not in the groups fed with the milk with bifidobacteria and microencapsulated bifidobacteria. Thus, it was confirmed that the hypocholesterolemic effect is not due to the bifidobacterium itself but to the fermentation on milk.

  • The effects of diet on the composition of fecal microflora in mouse and the aclivilies of several enzymes in the leces were investigated. Vegetarian dietary groups were found to contain about ten lines higher numbers of Locmbocillus and B$&bacterizml than animal dietary groups. An~rnal dietmy groups were found to contain about 5 tolo times higher numbers of anaerobic Closhidia and Bocieriocles than the vegeterian detary groups. Fccal microbial $\beta$-glucosidase, $\beta$-glucm'onidase, ii-yptophanase and orease activilies in ihe animal dietary groups were shown lo be 30 to 50% hgher than those in h e vegetarian detary groups.

  • Several transglucosylated xylitols were synthesized using intermolecular transglucosylation reaction of cyclodextrin glucanotransferase (CGTase) and their bifidogenic effects were investigated. The CGTase from Thermoanaerobacter sp. showed the highest transglycosylation activity on xylitol compared to those obtained from other strains. Extruded starch was identified to be the most suitable glucosyl donor for transglucosylation reaction on xylitol molecule by CGTase. The optimum reaction conditions for transglucosylation were also studied using extruded starch as a glucosyl donor. The transglucosylated xylitols were purified by activated carbon column chromatography with ethanol gradient elution from 0 to 18%, and their chemical structures were analyzed by fast atom bombardment mass spectrometer, $\^$13/C-nuclear magnetic resonance spectrometer, and enzyme digestion method. Two transglucosylated xylitol, F-I and F-II, which had one or two glucose molecules attached to maternal xylitol by ${\alpha}$-1,4-linkage, were mainly obtained. F-II showed increased stimulation effect on the growth of Bifidobacterium breve compared to xylitol, indicating the possibility utilized as a new functional alternative sweetners having bifidogenic effects.

Changes of the Intestinal Microflora and Fecal Properties by Intake of Yoghurt Added Capsulated or Uncapsulated Bifidobacteria (캡슐및 비캡슐 비피더스균 첨가 요구르트의 섭취에 따른 장내균총 및 분변성상의 변화)

  • Fourteen healthy volunteers ranged in ages from 20 to 30 were served to administrate two types of yoghurt (2 bottles/day) such as one added uncapsulated-Bifidobacteria (Y-UCB) and the other added capsulated-Bifdobacteria (Y-CB) for 4 weeks, and the changes of intestinal microflora and fecal properties were studied. After administration of Y-UCB, the viable cell counts of fecal Bifidobacteria (p<0.01) and Lactobacilli (p<0.05) were significantly increased, however, fecal pH, moisture content and tile viable cell counts of coliform bacteria in feces were not changed when they were compared to those before administration (control). After administration of Y-CB, the viable cell counts of Bifidobacteria were significantly increased (p<0.01) and viable cell counts of coliform bacteria were significantly decreased (p<0.05), however, fecal pH, moisture content, and viable cell counts of Lactobacilli were not changed when they were compared to those before administration. High level of fecal Bifidobacteria and low pH were maintained after 2 weeks from ceasing the administration of both types of yoghurt when they were compared to those before administration. In conclusion, there were not significant differences between two types, Y-CB and Y-UCB in the changes of fecal pH, moisture content, and the viable cell counts of Bifidobacteria, Lactobacilli, coliform bacteria after the administration.

Potential prebiotic effects of yacon extract, a source of fructooligosaccharides, on Bifidobacterium strains (프락토올리고당을 함유한 야콘 추출물의 비피더스균에 대한 잠재적인 프리바이오틱 효과)

  • This study was conducted to investigate the potential prebiotic activity of an extract from yacon tuberous root on the growth of probiotic strains Bifidobacterium and Lactobacillus. Results showed that the amount of fructooligosaccharides per 100 g fresh root was the highest (7.60 g), followed by sucrose (0.72 g), fructose (0.34 g), and glucose (0.26 g). The in vitro culture test of intestinal beneficial bacteria, including Bifidobacterium longum BORI, B. bifidum BGN4, and B. lactis AD011 showed effective growth on the MRS-Yacon medium containing yacon extract, whereas the growth of Lactobacillus acidophilus AD031, L. plantarum BH02, and L. fermentum BH03 did not differ from that of the control groups. In particular, B. longum BORI showed better growth than the control group after 10 h of incubation. These results indicate that yacon can be a natural prebiotic source of fructooligosaccharides, which can exert a prebiotic effect on intestinal microflora by selectively enriching Bifidobacterium.

Growth Characteristics of Bifidobacteria and Quality Characteristics of Soy Yogurt Prepared with Different Proteolytic Enzymes and Starter Culture (단백분해효소와 배양방법의 종류에 따른 비피더스균의 생육특성 및 soy yogurt의 품질특성)

  • The quality characteristics of soy yogurt prepared with different proteolytic enzymes and starter culture were evaluated. In order to facilitate the growth of lactic acid bacteria and subsequent production of lactic acid, soy protein isolate(SPI) was hydrolyzed using three kinds of proteases; one extracted from Aspergillus oryzae, bromelain and ${\alpha}-chymotrypsin$. The cultural systems employed thereafter for lactic fermentations were: 1) Bifidobacterium bifidum, 2) B. bifidum and Lactobacillus acidophilus, 3) B. bifidum and Lactobacillus bulgaricus. In soy yogurt, pH was more decreased by mixed culture method than single culture method with the accumulation of lactic acid. Viable cells of lactic acid bacteria in soy yogurts were measured $10^8$ CFU/g by the single culture method while $10^9$ CFU/g by the mixed culture method except ${\alpha}-chymotrypsin$ treatment. The amount of free amino acids in soy yogurts were substaintially increased by enzyme treatment. Viscosity was decreased by enzyme treatment, resulting in higher viscosity by ${\alpha}-chymotrypsin$ treatment. Water holding capacity was found to be higher in the single culture method in case of enzyme treatment. Among the various volatile flavor components isolated and identified after enzyme hydrolysis, acetaldehyde, ethanol, diacetyl, butyl alcohol contents tended to increase by lactic fermentation.

Characterization of Functional Kimchi Using Bifidobacterium lactis (Bifidobacterium lactis를 이용한 기능성 김치의 특성)

  • This study was conducted to investigate the application of bifidobacteria on kimchi. Among several Bifidobacterium species, we selected Bifidobacterium lactis (DSM 10140), which is resistant to oxygen, acid and salt. Bifidobacterium lactis was cultured in a supplemented deMan, Rogosa and Sharpe (SMRS) medium under aerobic conditions. Its acid-tolerance and salt-tolerance were pH 3.0 and 3.5% (NaCl), respectively. The viability of Bifidobacterium lactis added to kimchi was confirmed by PCR, using specific primers on Bifidobacterium lactis. In sensory evaluation, kimchi containing Bifidobacterium lactis showed similar scores in overall acceptability with the control kimchi. Consequently, these results showed that it would be possible to prepare functional kimchi using Bifidobacterium.
