The Effective for Non-Point pollution Reduction Facility installation project (비점오염저감시설 설치사업의 효과 평가)

  • 토지이용의 고도화에 따라 비점오염원 부하는 증가하는 추세이며 기후변화에 따른 강우강도 증가 등으로 지표면에 축적된 고농도의 비점오염물질이 하천으로 유출, 수질오염을 가중시키고 있어 비점오염원 관리가 필요하다. 환경부에서 효율적인 비점오염원 관리를 위하여 2004년부터 현재까지 3단계에 걸쳐 비점오염원 관리 종합대책을 수행하고 있으며, 2008년부터 비점국고보조사업을 추진하여 비점오염저감시설 설치를 통한 수질개선 및 수생태계 건강성 확보에 기여하고자 하였다. 이에 본 연구는 비점국고보고사업을 통해 구축된 비점오염저감시설을 대상으로 시설 설치 및 운영 현황과 강우시 비점오염저감 효과 분석을 통한 시설의 평가를 수행하고자 한다. 연구대상시설은 시범시설 및 국고보조시설 총 70개소로 2005~2017년에 준공되었으며, 2016년부터 현재까지 총 4년동안 진행된 모니터링을 바탕으로 연구를 수행하였다. 시설의 용량은 34~97,000㎥의 범위로 SA/CA 1.2~6.6%의 범위이다. 강우시 모니터링은 선행무강우일수 3일이상을 고려하여 수행하였으며 도시지역의 경우 5mm 이상, 농촌지역 10mm 이상시 모니터링을 진행하였다. 시설의 유입과 유출부에서 수질 및 유량 모니터링을 진행하였으며, 수질오염공정시험법에 준하여 BOD, COD, SS, T-N 및 T-P 항목에 대해 분석을 수행하였다. 모니터링 결과, 국내 비점오염저감시설의 평균 부하량은 SS 250.4 kg/day, BOD 89.2 kg/day, COD 136.2 kg/day, TN 51.4 kg/day, TP 7.1 kg/day 로 분석되었으며, 유출부 의 경우 SS 83.8 kg/day, BOD 37.2 kg/day, COD 51.0 kg/day, TN 15.4 kg/day, TP 2.0 kg/day로 나타났다. 또한, 은 오염물질 유입 및 유출 부하량의 상관관계 분석결과 SS, BOD, COD의 유입 및 유출 부하량의 상관성은 높게 나타났으며 특히 유기물질(BOD, COD)의 상관성은 0.8이상으로 분석되었다(p<0.005). 이는 비점오염저감시설에 적용된 식생, 미생물, 여재 등을 통하여 물리학적 및 생태학적 처리를 통해 저감되기 때문으로 판단된다. 하지만, TN은 인위적 요인과 자연적 요인이 복합적으로 작용으로 배출 특성으로 상관성은 매우 낮은 것으로 분석되었다.

Characterization and Distribution of Various Flavobacterium sp. in Lake Soyang (소양호에 존재하는 새로운 Flavobacterium의 분포와 특징)

  • In this study, samples were collected from Lake Soyang in Kangwon-do in order to isolate novel Flavobacterium sp. strains. Totally, 21 strains of Flavobacterium showing 97%~98% similarity in 16S rRNA were selected and thoroughly investigated individual characteristics and ecological differences. As results, we could categorize Flavobacterium isolated from Lake Soyang into four major node groups, where most of Flavobacterium belonged to single group. Next, fatty acid analyses were performed demonstrating similar pattern of the majority of fatty acids as either iso $C_{15:0}$ or summed feature 3 (comprised $C_{16:1}$ ${\omega}7c$ and/or $C_{16:1}$ ${\omega}6c$) of the other Flavobacterium. However, other phenotypic data were different from the other Flavobacterium sp. Group. Our data showed that genetically related species of Flavobacterium have been distributed in Lake Soyang. Those Flavobacterium strains were phenotypically different from previously reported genus of Flavobacterium species. Taken together, we speculated that isolated Flavobacterium strains from Lake Soyang might be ecologically important members to maintain ecosystem.

Biological Treatment of Processed-Leachate from Landfills by Reed (Phragmites australis)-Bed in a Continuous Flow System (갈대-상(床)을 이용한 쓰레기 매립지 침출수의 생물학적 연속흐름 처리)

  • We investigate the biological treatment of processed-leachate from SUDOKWON landfill site in Korea by the reed (Phragmites australis)-bed with a continuous flow system. The reed individuals showing superior growth in processed-leachate experiment were selected among the reeds of thirteen natural habitats by means of the comparisons between the removal rates of nutrient salts, eco-physiological responses and growth. The reed-beds (combination of the reed individuals showing superior growth with helper microorganisms) were continuously supplied with processed-leachate. We monitored the effluents that passing through the reed-beds during the experiment period. After five weeks, analysis results of effluent from each reed-bed were as follows; chromaticity, total nitrogen, total phosphorus, biological oxygen demand, chemical oxygen demand, total dissolved solid and salinity decreased $29.5{\sim}36.9\%,\;49.4{\sim}67.2\%,\;42.1{\sim}94.6\%,\;74.5{\sim}88.8\%,\;15.6{\sim}20.8\%,\;17.5{\sim}35.4\%\;and\;15.3{\sim}34.7\%$, respectively. These results represented the substantial improvement of water-quality after passing through reed-bed in a continuous flow system.

Changes in Soil Biota Affected by the Application of Organic Materials in Reclaimed Upland and Paddy-converted Soils Cultivated with Korea Ginseng (개간 및 답전윤환 인삼재배지에서 유기물처리에 따른 토양생물상 변화)

  • Cultivation of Korea ginseng in newly reclaimed and paddy-converted fields has been increasing, and evaluation of organic amendment effectiveness is needed in the two soil types. Soil organisms influence organic matter decomposition, and their responses to applications of organic matter were studied. De-oiled cake and compost were applied at $20Mg\;ha^{-1}$ and $40Mg\;ha^{-1}$ in both soil types. Changes in microflora were assessed by analyzing phospholipid fatty acid (PLFA). The abundance of nematodes and microarthropods was measured. Microbial PLFA indicators for microorganisms and microarthropod abundance were greater in reclaimed upland than in paddy-converted soil. There were few differences in the microflora and fauna of reclaimed uplands, regardless of treatment. In paddy-converted soil, the abundance of Oribatida was increased by the application of compost at $20Mg\;ha^{-1}$ and was correlated with PLFA indicators of fungi. The results suggested a minimal influence of organic amendments in reclaimed upland, because the organic matter content and abundance of soil organisms are low in mineral soils. In paddy-converted soil, the effects of organic amendment differ among different soil organisms, and soil properties are important mediators of the effect.

Rapid Detection of Cochlodinium polykrikoides by Sandwich Hybridization Integrated with Invertase Assay (Invertase 분석이 통합된 sandwich hybridization에 의한 Cochlodinium polykrikoides의 신속 검출)

  • Rapid and accurate identification of marine microalgae causing harmful algal blooms (HABs) is a crucial tool for predicting and managing HABs. We previously developed a nuclease protection assay sandwich hybridization (NPA-SH) method for the in situ detection of blooming microalgae Cochlodinium polykrikoides. In this study, we improved the applicability of the NPA-SH method for the detection of C. polykrikoides by simplifying the reaction step. For this purpose, invertase (INV) was conjugated to the signal probe instead of using fluorescence, and sucrose was used as a reactant to induce a color reaction. The INV-signal probe conjugation was confirmed by SDS-PAGE and epifluoromicroscopy. The treatment time and appropriate amounts of the probe and sucrose that optimized the reaction were determined. As a result, the developed INV-SH reduced the treatment time in the field compared with NPA-SH, and also enabled the use of a relatively small volume and low-priced personal glucose meter, as well as an absorbance meter. INV-SH is the first C. polykrikoides species identification technology to which INV has been applied and could be an improved field technique.

Distribution of Microflora and Mesofauna in the Forest Soils of Gwangneung Experimental Forest (광릉시험림 산림토양의 미생물상 및 중형동물상 분포)

  • The aim of this study was to investigate the distribution of soil biota across forest types and soil depths in Gwangneung Experimental Forest. We selected 5 sites, which are 3 deciduous- and 2 coniferous forests. The abundance of microflora in these areas was analyzed by the phospholipid fatty acid (PLFA) indicators, and the density of mesofauna was measured by the abundance of nematodes and microarthropods. The abundance of soil microflora was higher in the A layer than in the B layer based on the PLFA indicators, but there were no differences between deciduous and coniferous forests. Average density of nematodes was higher in the A horizon than in the B horizon. The density of fungivorous nematodes was higher in the deciduous forests. Mean densities of Collembola and Oribatida were 44% and 42% of microarthropods in soil samples, respectively. The results of microbial PLFA indicators were significantly correlated with the density of their consumers, which reflected that the food web in fungi- and bacteria-based soil developed in this forests. This study suggested that the distribution of soil organisms largely separated by soil depths and was slightly affected by the forest type in the Gwangneung Experimental Forest.

Vertical Profiles of Alkaline Phosphatase Activity in Dam Reservoirs and its Relation with Microbial Parameters (댐 저수지에서 alkaline phosphatase 활성의 수직변화와 미생물 요인들과의 상관관계)

  • The alkaline phosphatase activity (APA) of two dam reservoirs and inflowing streams were measured monthly in 2000. During summer months in 2001, the vertical profiles of APA and related parameters were also examined in one of the reservoirs. The APA was relatively high during the summer season in the epilimnion while it was almost invariable in the hypolimnion. A small increase in APA was observed at just above the bottom. The APA fluctuation was independent of the concentration of soluble reactive phosphorus. It was assumed that APA is not indicative of the phosphorus availability status. An examination of size-fractionated samples suggested that APA in reservoirs was attached to particles larger than $0.4{\mu}m$, whereas in streams it existed in a dissolved form. There was a positive significant correlation between chlorophyll a concentration and APA in the photic zone. In the aphotic zone, APA correlated positively with the colony count of heterotrophic bacteria, but not with microscopic total bacterial counts.

Quality Control to Improve Reliability of Automatic Water Quality Data (수질자동측정망 자료의 신뢰성 제고를 위한 정도관리)

  • The automatic water quality monitoring system (AWQMS) have been installed to immediately response to any pollution incident. It also make it possible to conduct the task efficiently regarding water quality control. The purpose of this study is to enhance reliability by securing accuracy of automatic water quality data through quality assessment (QA) for temperature, pH, dissolved oxygen (DO), electric conductivity (EC), total organic carbon (TOC). The result of comparison between manual and automatic data, relative accuracy of general items (temperature, pH, EC, DO) and TOC were mostly satisfied with guideline (i.e. less than 20%). On the other hand, relative accuracy of DO between sampling site and housing site was somewhat against the guideline. The contamination by attaching algae and microorganism in the pipeline is considered as main cause. After backwashing the pipeline, DO concentration was increased up to 53%. Therefore, pipeline management is recognizable as important thing to secure reliability of water quality data.

Community Changes of Ectomycorrhizal Fungi by Thinning in a Forest of Korea (간벌처리에 따른 외생균근균의 군집 변화)

  • This study was conducted to investigate the effect of thinning on ectomycorrhizal fungal communities in a forest. Ectomycorrhizal root tips were collected from forest soils in thinning and non-thinning sites and identified using morphological characteristics and molecular analysis of ITS rDNA sequences. As a result, species richness of ectomycorrhizal fungi was significantly increased and ectomycorrhizal fungal community composition was changed by thinning. These results suggest that forest management such as thinning, could be an important factor affecting mutualistic relationships and belowground microorganisms in forest ecosystems.

Bacterial Abundances and Enzymatic Activities under Artificial Vegetation Island in Lake Paldang (팔당호에 설치된 인공식물섬에서의 세균 수와 체외효소 활성도의 변화)

  • For analyzing function of a microbial ecosystem which was created under the artificial vegetation island (AVI) installed at Lake Paldang, zooplankton and bacterial numbers and exoenzyme activities (${\beta}$-glucosidase and phosphatase) were measured biweekly from 3 November 2()()1 to 20 April 2002 at AVI site and control site. Under the AVI, the water quality was worse than control site in term of comparing the environmental parameters. But, zooplankton number of AVI site was 25 times higher than that of control site. Respiratory active bacterial numbers were 3-8 times higher at AVI site. In addition, enzymatic activities were higher at AVI site than those of control site. These results suggest that the zooplankton-phytoplankton-bacteria relationships are closely coupled with each other and organic materials are eliminated by respiration of zooplankton and bacterial activities.
