Verification of the Antioxidant Effects and Whitening Activity of fermented Ambrosia trifida L. Extracts in B16F10 Cells (단풍잎돼지풀 발효 추출물의 항산화 효과 및 B16F10 세포에서의 미백 활성 검증)

  • The purpose of this study was to verify the antioxidant and whitening effects of fermented Ambrosia trifida L. extract (ATFE) and to verify its usefulness as a cosmetic material. The antioxidant effects were measured by assessing the electron-donating capacity and 2,2-azino-bis(3-ethyl-benthiazoline-6-sulfonic acid) (ABTS) radical scavenging ability of these extracts. ATFE was shown to have an electron-donation capacity of 68.4% at a concentration of 1000 ㎍/ml. While its ABTS+ radical scavenging ability was shown to be 58.7% at the same concentration. The ATFE tyrosinase inhibitory effect, which is related to skin-whitening, was shown to be 32.35% at a concentration of 1000 ㎍/ml and a cell viability assay using melanoma cells showed a 14.8% reduction in cell viability at a concentration of 100 ㎍/ml. Surviving cells were then used in western blot analyses to evaluate the protein inhibitory effects of ATFE at 25, 50, 100 ㎍/ml where β-actin was used as a positive control. The whitening effects of these extracts were also evaluated by western blot and show that the expression of microphthalmia-associated transcription factors, Tyrosinase-related proteins (TRP)-1, TRP-2 and Tyrosinase were all inhibited, 51.14%, 55.4%, 38.6%, 83.77% respectively, at 100 ㎍/ml ATFE. The efficacy of the whitening effects was verified and the suitability of ATFE as a cosmetic material was assured.

Whitening Effect of Abelmoschus esculentus on Melanoma Cells (B16F10) (B16F10 세포에서의 오크라 추출물의 미백 활성 검증)

  • In this study, the whitening effect of Abelmoschus esculentus extract was investigated to confirm its applicability in cosmetics. To determine the whitening effect, the tyrosinase-inhibitory activity of Abelmoschus esculentus hot water extract (AEWE) and 70% ethanol extract (AEEE) was measured. At the final concentration of 1000 ㎍/ml, AEWE showed an inhibitory activity of 22.2% and AEEE of 32.8%. To determine the whitening effect at the cellular level, the viability of melanoma cells treated with AEWE and AEEE was evaluated using the MTT assay. At concentrations of 100 ㎍/ml or less, both AEWE- and AEEE-treated groups showed cell survival rates of >95%. Furthermore, in both AEWE- and AEEE-treated melanoma cells, the melanin content decreased in a concentration-dependent manner. The inhibitory effects of AEWE and AEEE used at 5, 10, 50, and 100 ㎍/ml on protein expression were measured by western blot, with β-actin as the positive control. At a concentration of 100 ㎍/ml, AEWE showed an inhibitory effect of 88.1%, 24.8%, 62.2%, and 42.9% on microphthalmia-associated transcription factors (MITF), tyrosinase, tyrosinase-related proteins (TRP)-1, and TRP-2 factors, respectively. At the same concentration, AEEE showed inhibitory effect of 65.3%, 58.3%, 66.2%, and 65.3% against MITF, tyrosinase, TRP-1, and TRP-2 factors, respectively. In conclusion, the whitening effects of AEWE and AEEE were verified, and their applicability as a natural ingredient in cosmetics was confirmed.

Whitening Effects of Solvent Fractions Isolated from Vitex rotundifolia (만형자 용매 분획물의 미백 개선 효과)

  • Recently many effort focused to understand the mechanical insights of melanogenesis to develop the agent for hyper-pigmentation. So this study was performed to investigate the depigmentation of Vitex rotundifolia. With B16F10 mouse melanoma cell, we have seen inhibition of the tyrosinase, MITF, TRP-1, TRP-2, and melanin synthesis, which eventually were dose dependently decreased by Vitex rotundifolia. Specially, Vitex rotundifolia decreased the protein levels of tyrosinase and TRP-1. In conclusion, Vitex rotundifolia showed the whitening activity in all the experiments mentioned above and we expect that it can be used for preventing melanin synthesis.

Studies on Antioxidant, Anti-inflammation and Tyrosinase Inhibitory Activities of Melissa officinalis Extracts and Their Fractions (레몬밤 추출물과 분획물의 항산화, 항염 및 티로시나제 저해활성 연구)

  • This study was carried out to evaluate the antioxidant, anti-inflammation, and tyrosinase inhibitory activity of Melissa officinalis extracts and their fractions. The total polyphenol contents of the extracts were 33.02-302.76 mg GAE/g and total flavonoid contents were 9.98-325.07 mg CE/g. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, DPPH radical scavenging activity was observed in the extracts and water fractions of M. officinalis and similar to that of ascorbic acid ($30{\mu}M$). In addition, the treatment of chloroform fraction significantly inhibited the production of nitric oxide (NO) in RAW 264.7 cells, indicating that they have anti-inflammatory activity. Tyrosinase inhibitory activity of $200{\mu}g/mL$ of 100% ethanol reflux extract showed better inhibitory activity than arbutin treatment at statistically significant level. As a result, it is considered that M. officinalis can be used as an effective cosmetic ingredient having antioxidant, anti-inflammation, and whitening activity.

  • In this study, the skin whitening and anti-wrinkle activity of Abelmoschus manihot (A. manihot) flowers were evaluated as a cosmetic material. A. manihot flowers were extracted using water and 70% ethanol. Tyrosinase inhibitory activity was evaluated to increase concentration-dependent inhibitory activity in both water (AMW) and 70% ethanol extracts (AME), and was found to inhibit melanin biosynthesis at concentrations that did not affect survival in B16F10 cell lines at 10, 25, 50, 75, and 100 ㎍/mL. As a result of measuring the anti-wrinkle effectiveness of A. manihot flowers, 70% ethanol extract (AME) showed higher anti-wrinkle activity than water extract (AMW). Through the results of this study, it is considered that the A. manihot flower extract can be used as a functional cosmetic material with whitening and anti-wrinkle activity.

Depigmenting Effects of Esculetin and Esculin Isolated from Fraxinus rhynchophyllaHance (물푸레나무로부터 분리된 Esculetin와 Esculin의 미백 효능)

  • Stem bark extracts of Fraxinus rhynchophylla Hance were found to contain two major bioactive components, esculetin and esculin. Esculetin substantially inhibited melanogenesis in B16F10 melanoma cells, with an $IC_{50}$ value of $2.8{\mu}M$, and reduced melanin synthesis in Melan-A cells. Moreover, esculetin suppressed melanin biosynthesis by inhibiting mushroom tyrosinase activity, with an $IC_{50}$ value of $40{\mu}M$. Taken together, these results suggest that esculetin could serve as an effective skin-lightening agent that inhibits melanin production by regulating the activity of melanogenic enzymes.

Whitening Effect of Green Tea Seed Shell Ethanol Extracts (녹차씨 껍질 에탄올 추출물의 미백 활성)

  • The objective of this study was to evaluate the whitening effect of green tea seed shell as an industrial byproduct. Green tea seed shell extract (GTSE) was obtained by ethanol extraction, and the yield was 1.24%. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging and tyrosinase inhibitory activity of GTSE increased dose-dependently. To estimate inhibition of melanin synthesis, viability was tested in B16BL6 melanoma cells. GTSE treatment induced cytotoxicity at a concentration higher than $125{\mu}g/mL$ but did not induce cytoxicity lower than $62.5{\mu}g/mL$. Thus, we fixed the optimal concentration at $62.5{\mu}g/mL$. Using this optimal concentration, melanin synthesis inhibition was measured, and GTSE treatment significantly reduced melanin synthesis induced by ${\alpha}$-melanin stimulating hormone. Therefore, the results indicate that green tea seed shell extracts may have potential melanin synthesis inhibitory activity and may be useful for development of whitening material as a natural ingredient.

The Antioxidant and Skin Whitening Effect of Artemisia iwayomogi Extracts (더위지기 추출물의 항산화 및 미백 효과)

  • The purpose of this study was to investigate the antioxidant and skin whitening effects of Artemisia iwayomogi extract. Artemisia iwayomogi was extracted with 100% ethanol and water. The antioxidative and skin whitening effects of these extracts were determined with in vitro assays by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) method assessing the inhibitory effects on tyrosinase activity and melanogenesis in B16 melanoma cells. Radical scavenging activity of the extracts was tested by DPPH assay which showed a high DPPH radical scavenging activity ($SC_{50}$; 17.1 ppm in EtOH, 198.4 ppm in water). In term of tyrosinase inhibitory activity, Artemisia iwayomogi ethanol extract showed high inhibition activity ($IC_{50}$: 481.8 ppm). In B16 mouse melanoma cells, the ethanol extract significantly inhibited melanin synthesis by 36.8% at a concentration of 50 ppm. These results suggest that Artemisia iwayomogi ethanol extract has significant antioxidant activity and whitening activity.

In vitro activities of Grape Pruning Stems for Application of Cosmetic Ingredients (포도 전정가지의 화장품 소재로서의 응용)

  • Grape pruning stems constitute a scarcely investigated class of byproducts with limited reports on their bioactive polyphenol content and/or industrial applications. Herein we present the outcome of our investigation on grape pruning stems extracts, concerning the assessment of their total polyphenolic content and the detailed evaluation of their antioxidant properties. Results obtained indicate that grape pruning stems are particularly rich in flavonoids and trans-resveratrol. The antioxidant activities was analyzed and expressed as electron donating ability, ABTS cation radical decolorization, hydrogen peroxide scavenging activity and superoxide anion radical scavenging activity, the antioxidant activity of Vitis labrusca L. pruning stems extracted (GPSE) was higher than that of BHA and L-ascrobic acid. The whitening and anti-wrinkle activities display an capability. Results herein grape pruning stems used as a valuable resource for the extraction of resveratrol, which would be added to functional cosmetics and food materials.

Antioxidant and Skin Whitening Effects of Rhamnus yoshinoi Extracts (짝자래나무 추출물의 항산화 및 미백 효과)

  • The purpose of this study was to investigate the antioxidant and skin whitening effects of Rhamnus yoshinoi extracts. Rhamnus yoshinoi was extracted with 100% ethanol and water. The antioxidative and skin whitening effects of extracts were determined by in vitro assays using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and inhibitory effects against tyrosinase activity and melanogenesis in B16F1 melanoma cells. The radical scavenging activities of the Rhamnus yoshinoi extracts were tested by DPPH assay and showed high DPPH radical scavenging activities (SC50; 21.6 ppm in EtOH, 40.5 ppm in water). As for tyrosinase inhibitory activity, the Rhamnus yoshinoi ethanol extract had the highest inhibition activity ($IC_{50}$; 256.3 ppm). In B16F1 mouse melanoma cells, the Rhamnus yoshinoi ethanol extract significantly inhibited melanin synthesis by 53.36% at the concentration of 50 ppm. These results suggest that Rhamnus yoshinoi ethanol extract has significant antioxidant activity and whitening activity.
