To investigate the efficacy of enzymatic extract of Ecklonia cava and its polyphenol extract (AG-DK) as cosmetic ingredients, the anti-oxidative effect, anti-glycation effect, anti-melanogenic effect, and anti-inflammatory effect of the extracts were evaluated in vitro. The enzymatic extract of E. cava ($SC_{50}$ 42.9 ppm) and AG-DK ($SC_{50}$ 6.4 ppm) showed a strong DPPH free radical scavenging activity. The anti-glycation ability of the enzymatic extract of E. cava and AG-DK was tested using bovine serum albumin (BSA), which inhibited the formation of advanced glycation end-products (AGEs) in the BSA/glucose system. The enzymatic extract of E. cava ($IC_{50}$ 97.2 ppm) and AG-DK ($IC_{50}$ 7 ppm) had inhibitory effects on tyrosinase activity. Moreover, the enzymatic extract of E. cava and AG-DK had an anti-inflammatory effect through the inhibition of nitricoxide (NO) and prostaglandin E2 ($PGE_2$). These findings suggest that the enzymatic extract of E. cava and AG-DK can be applied to skin-care products as cosmetic ingredients.

Dendropanax morbifera Leveille (Araliaceae) is an endemic species growing in the south-western part of South Korea and has been used in folk medicine. However, the effects of Dendropanax morbifera Lev. on skin biology remain to be elucidated. In this study, we isolated 1-tetradecanol and ${\beta}$-sitosterol from the n-hexane fraction of Dendropanax mobifera Lev. and To investigate the whitening effect of the fraction, we tested the inhibition of tyrosinase activity of 1-tetradecanol. The results show that the inhibitory effect of the 1-tetradecanol was higher than water extract and n-hexane fraction. And 1-tetradecanol significantly reduced melanin contents of B16F10 cells compared to more than water extract and n-haxane fraction dose-dependantly without cell cytotoxicitiy (below $100{\mu}g/mL$). We also investigated the skin moisturizing effect using HR-1 hairless mice. The transepidermal water loss (TEWL) in the 1-tetradecanol treated group was significantly smaller than that in the other groups. To investigate the effect of the preventing hair loss by ${\beta}$-sitosterol, we observed HR-1 hairless mice through periodic growth feature. The results suggest that hair loss of mice by ${\beta}$-sitosterol was delayed and it's hair density showed the highest. These data provide evidence that Dendropanax morbifera Lev. may be a potent candidate for the improvement of both skin whitening, moisturizing and alopecia from the point of cosmetic industry view.
This study was designed to determine the biological activities of Chionanthus retusus flower extracts. Water and 90% ethanol extracts of C. retusus flower were prepared. The inhibitory activities of water and ethanol extracts with a phenolic content of $200{\mu}g/mL$ against xanthine oxidase were 25.60% and 15.92%, respectively. Further, the water extract did not show any inhibitory activity against ${\alpha}$-glucosidase whereas the ethanol extract showed 100.00% inhibition of ${\alpha}$-glucosidase. The inhibitory activities of the extracts against tyrosinase were 17.27% (water extract) and 36.13% (ethanol extract), which suggest that the extracts may have a whitening effect. The water extract did not inhibit elastase activity but showed a collagenase-inhibitory activity of 20.21%. On the contrary, the ethanol extract showed 96.26% and 35.93% inhibition of collagenase and elastase, respectively. These findings suggest that the extracts may have an anti-wrinkle effect. Lastly, the extracts showed a hyaluronidase inhibitory activity of 36.96% (water extract) and 88.70% (ethanol extract), suggesting that they may have an anti-inflammatory effect. The results indicate that C. retusus flower extracts containing phenolic compounds can be used as functional resources because they have anti-gout, carbohydrate degradation-inhibitory, whitening, anti-wrinkle, and anti-inflammatory effects.
This study was conducted to evaluate various beneficial functions of soaked and germinated Robusta coffee beans. Vietnam Robusta coffee beans were soaked in purified water or Salicornia extract for 12 h. The total polyphenol content of non-germinated coffee (NGC), Salicornia extract-germinated coffee (SGC), and water-germinated coffee (WGC) was found to be 16.71-20.17%. First, NGC, SGC, and WGC at concentrations of 25-100 ㎍/mL showed significant anti-oxidant effects on DPPH radical scavenging and xanthine oxidase activity. NGC, SGC, and WGC also inhibited tyrosinase activity and melanin formation in α-MSH-induced B16F10 cells. In addition, the anti-obesity property of germinated coffees was tested by the Oil Red O staining method. NGC, SGC, and WGC inhibited adipogenesis in 3T3-L1 cells without cytotoxicity. Taken together, germinated Robusta coffees with enhanced flavor showed beneficial multifunctional effects, such as anti-oxidant, anti-melanogenesis, and anti-adipogenesis effects.

In this study, the evaluation of antioxidative activity and componential analysis of C. obtusa leaf extracts was carried out. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of C. obtusa leaf extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction ($OSC_{50}$; 0.22 ${\mu}g/mL$) and aglycone fraction of C. obtusa leaf extracts (0.20 ${\mu}g/mL$) showed about 7 times more prominent ROS scavenging activity than L-ascorbic acid (1.50 ${\mu}g/mL$). The cellular protective effects of fractions obtained from C. obtusa leaf extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction and aglycone fraction of C. obtusa leaf extracts showed the cellular protective effects in a concentration dependent manner (5~25 ${\mu}g/mL$). The inhibitory effect ($IC_{50}$) of ethyl acetate fraction and aglycone fraction on tyrosinase exhibited 74.43 and 53.80 ${\mu}g/mL$, repectively. The aglycone fraction showed four times higher tyrosinase inhibitory effect than arbutin (226.88 ${\mu}g/mL$), known as a whitening agent. The aglycone fraction of C. obtusa leaf extracts showed three bands in TLC chromatogram and three peaks in HPLC chromatogram (360 nm). Three compounds were identified as taxifolin, quercetin and kaempferol. These results indicate that the fractions of C. obtusa leaf extracts can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against reactive oxygen species. The fractions of C. obtusa leaf extracts can be applicable to new functional cosmetics for antioxidan and whitening effects.

To develop a new natural whitening agent for cosmetics, we investigated the inhibitory effects of Pogostemon cablin Bentham extracts (PCE) and its active component on melanogenesis. PCE showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $24.2{\pm}2.85{\mu}g/mL$ and $IC_{50}=118{\pm}0.43{\mu}g/mL$, respectively. PCE reduced melanin contents of B16 melanoma cells in a dose-dependant manner and decreased to about 23 % at a concentration of $20{\mu}g/mL$ without cell cytotoxicity (below $100{\mu}g/mL$). And the PCE reduced intracellular tyrosinase activity about 18 % at concentration of $50{\mu}g/mL$. We purified one active compound from PCE and identified its structure. It was identified as patchouli alcohol, sesquiterpene family, by 1H-NMR, $13_C$-NMR, and Mass analysis. Patchouli alcohol also inhibited ROS scavenging activities in DPPH radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.14{\pm}0.12{\mu}g/mL$ and $49{\pm}3.24{\mu}g/mL$, respectively. Patchouli alcohol inhibited melanin synthesis in a dose dependent manner ($IC_{50}=3.9{\mu}g/mL$). And the patchouli alcohol reduced intracellular tyrosinase activity about 40 % at concentration of $10{\mu}g/mL$. Patchouli alcohol inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that PCE and patchouli alcohol reduced melanin formation by the inhibited of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that PCE could be used as a useful whitening agent.
A 70% ethanol extract of Erigeron annuus (L.) Pers. was investigated for its whitening activity for application as a functional ingredient in cosmetic products. At the E. annuus extract concentration of $100{\mu}g/ml$, the electron-donating ability was found to be 67.83%, the tyrosinase inhibitory effect (related to skin-whitening) was 69%, the elastase inhibitory effect (related to skin-wrinkling) was 69%, and the astringent effect was 80%. The $ABTS^+$ radical-scavenging ability was 87% at the $500{\mu}g/ml$ concentration. In the cell viability test measured on melanoma cells, 96% of the cells treated with $100{\mu}g/ml$ of the extract were viable. According to the western blot results, the protein expression of the microphthalmia-associated transcription factor (MITF), tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 was decreased by 60.22%, 47.83%, 54.79%, and 67.88%, respectively, at the extract concentration of $100{\mu}g/ml$. The protein expression of phosphorylated extracellular signal regulated kinase (p-ERK) and phosphorylated cAMP response element-binding protein (p-CREB) was decreased with increasing concentrations of the extract. Reverse transcription-polymerase chain reaction of the extract showed that the mRNA expression of MITF, tyrosinase, TRP-1, and TRP-2 was decreased by 86.51%, 85.22%, 74.26%, and 66.66%, respectively, at $100{\mu}g/ml$ extract concentration. The findings suggest that the 70% ethanol extract from E. annuus (L.) Pers. has potential as a cosmeceutical ingredient with whitening effect.

The objectives of this study were to determine inhibitory effect on tyrosinase activity and antioxidant activity of the methanolic extract from grape seeds and to investigate relationships between tyrosinase inhibitory activity and antioxidant activity of the extract. The 80% methanol extracts of grape seeds were fractionated subsequently with hexane, chloroform, ethyl acetate and water. The methanolic extract and fractions from grape seeds inhibited tyrosinase activity in a concentration dependent manner. The methanolic extracts showed the highest inhibitory effects on tyrosinase activity. The inhibitory effects of the ethyl acetate fraction from the methanolic extract on tyrosinase activity was higher than other fractions. The ethyl acetate fraction from methanolic extracts showed higher antioxidant activity and contained higher polyphenolic and flavonoid contents compared to other fractions. The correlation coefficients among the polyphenoilc content of methanolic extracts, ABTS radical cation scavenging activity and inhibitory effect of tyrosinase were relatively high. These results suggest that grape seeds may have potential as a depigmentation agent for cosmetics and functional food products.

In this study, the antioxidative and inhibitory effects on tyrosinase and elastase of Hippophae rhamnoides (H. rhamnoides) leaf extracts were investigated. The ethyl acetate fraction of H. rhamnoides extracts showed more effective free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$ = 4.68 ${\mu}g$/mL). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of the aglycone fraction in the luminol-dependent $Fe^{3+}$-EDTA/$H_2O_2$ system was 0.19 ${\mu}g$/mL. The aglycone fraction exhibited more prominent cellular protective effects (${\tau}_{50}$, 133.3 min at 10 ${\mu}g$/mL) in the $^1O_2$-induced photohemolysis of human erythrocytes. The inhibitory effect ($IC_{50}$) of the aglycone fraction on tyrosinase was 54.86 ${\mu}g$/mL, and more effective than arbutin known as whitening agent. These results indicate that fractions of Hippophae rhamnoides extract can be used as antioxidants in biological system, particulaly skin exposed to UV radiation by quenching and/or scavenging $^1O_2$ and other ROS, and protecting cellular membranes against ROS.

This study was to improve cosmetical activity of thiamine di-lauryl sulfate (TDS) by encapsulation of nanoparticle with lecithin. Results showed that most of the nanoparticles containing the TDS were well formed in round shape with below 150 ~ 200 nm diameter as well as they were fairly stable in various pH ranges by measuring zeta potentials. The nanoparticles of TDS resulted in 85% cell viability of human normal fibroblast cells (CCD-986sk) when added at the highest concentration (1.0 mg/mL). The nanoparticles of Acer mono sap showed highest free radical scavengering effect as 88.1% in adding sample (1.0 mg/mL), compared to TDS solution of non-encapsulation (81.6%). The nanoparticles of TDS reduced the expression of MMP-1 on UV-irradiated CCD-986sk cells down to as 41.4%. The TDS solution and nanoparticles showed significant anti-microbial activities agaionst the salmonella typhimurium and listeria monocytogenes at 5 and 6 days as compared with control. Anti-microbial activities of TDS nanoparticles were similar to positive control. These results indicated that TDS nanoparticles may be a source for functional cosmetic agents capable of improving cosmetical activity such as antioxidant, whitening, and anti-wrinkling effects and can be further developed as natural preservative in cosmetics.
