The cell cultures and crude extracts of Bacillus sphaericus 1593 K-5 and its mutant Spo-Dl216 were respectively bioassayed against Culex pipiens var. pollens mosquito larvae. The B. sphaeriucs 1593 K-5 showed toxic activity against the larvae. LC$_{50}$ values (cells/$m\ell$) was 2.6$\times$10$^2$. Also the LC$_{50}$ ($\mu\textrm{g}$ Protein/$m\ell$) of the crude extract was 10.26. However, B. sphaericus Spo-Dl216 didn't show toxic activity against the larvae. The soluble cytoplasmic toxin in broken B. sphaeriucs 1593k-5 cells was partially purified by gel permeation chromatography and ion exchange chromatography. Among the fractions of the gel permeation chromatography only a single fraction was found to be toxic. LC$_{50}$ values ($\mu\textrm{g}$ protein/$m\ell$) of the active fraction was 0.182. The active fraction of the gel permeation was subjected to ion exchange chromatography. Only a single fraction showed toxic activity and its LC$_{50}$ values ($\mu\textrm{g}$ protein/$m\ell$) was 0.02..02.
We investigated to know the growth patterns of Aedes albopictus larva at the four different temperature conditions. Each of 120 individuals was placed into a $20m{\ell}$ vial and 12 sets (a set of 10 vials) were separated into 12 water tanks ($17{\times}24{\times}18cm^3$). Each water tank was composed of 3 the $1^{st}$ instar, 3 the $2^{nd}$ instar, 2 the $3^{rd}$ instar, and 2 the $4^{th}$ instar. Three sets of water tanks were placed under the four different incubator temperatures ($17^{\circ}C$, $21^{\circ}C$, $24^{\circ}C$, $28^{\circ}C$). We found that the eclosion rates were $20.00{\pm}5.77%$ at $21^{\circ}C$ and $3.33{\pm}3.33%$ at other temperatures. For the mosquito larva mortality rate, $1^{st}$ instar was $19.24{\pm}3.65%$, $2^{nd}$ instar was $16.48{\pm}3.25%$, $3^{rd}$ instar was $23.54{\pm}5.06%$, and $4^{th}$ instar was $40.74{\pm}7.08%$. The lowest mortality rate in growth stages according to temperature was $13.33{\pm}6.67%$ at $17^{\circ}C$ in $1^{st}$ instar larva, $7.41{\pm}7.41%$ at $21^{\circ}C$ at $2^{nd}$ instar larva, $10.74{\pm}6.43%$ at $24^{\circ}C$ in $3^{rd}$ instar larva, and $20.37{\pm}5.46%$ at $28^{\circ}C$ in $4^{th}$ instar larva. The survival period of mosquitoes in underwater were $26.33{\pm}0.67days$ at $17^{\circ}C$, $23.33{\pm}1.33days$ at $21^{\circ}C$, $20.00{\pm}2.52days$ at $24^{\circ}C$, and $11.67{\pm}1.20days$ at $28^{\circ}C$. From our results the most effective temperature to the normal growth of mosquito larva was $21^{\circ}C$, and the highest mortality rate was shown at the $4^{th}$ instar stage of larva growth. Our results would provide the basic data for the mosquito larva's growth pattern.
Two mosquito species were collected in still-water near farming area in Andong, Korea. Based on morphological characters, these two mosquitoes were identified as Aedes koreicus and Culex vagans, respectively. DNA barcode analyses supported the identification. An entomopathogenic bacterium, Bacillus thuringiensis subsp. israelensis (BtI), exhibited insecticidal activities against the two mosquito species and its virulence was more potent than that of B. thuringiensis subsp. kurstaki. It has been known that the bacterial metabolites of Xenorhabdus spp. suppress insect immunity and enhance pathogenicity of B. thuringiensis. This study tested the effect of the bacterial culture broth of Xenorhabdus spp. on enhancing BtI pathogenicity. Among three Xenorhabdus spp., culture broth of X. ehlersii (Xe) was relatively effective to enhance BtI pathogenicity against both mosquito species. Indeed, organic extracts of Xe culture broth suppressed the hemocyte-spreading behavior, suggesting the presence of immunosuppressant in the culture broth. These results suggest a formulation of BtPlus by mixing BtI spore and Xe culture broth to be applied to control the two mosquito species.
In order to isolate naturally occurring novel Bacillus thuringiensis strains, we investigated the distribution of B. thuringiensis from granary which exist in Kyong-gi province, Korea. A total of 146 strains of B. thuringiensis producing spore and crystal wre isolated. The toxicity of B. thuringiensis isolates was examined against lepidopteran larvae (Bombyx mori), dipteran larvae (Culex pipiens) and coleopteran larvae (Sitophilus oryzae.), respectively. The results showed that a large number of B. thuringiensis isolates from granary dusts were isolated and most isolates wer toxic to lepidopterous larvae. Also, non-toxic B. thuringiensis isolate was common. In order to isolate many B. thuringiensis, therefore, it suggested that granary is favorable locality.
For the expression of the cry11Aa gene highly toxic to dipteran insects, we constructed two cyanobacteria-Escherichia coli(E. coli) shuttle vectors, pCYASK5-l and pCYASK5-2. These vectors were transformed into E. coli and selected with kanamycin. The expression of the cry11Aa gene in E. coli was characterized by SDS-polyacrylamide gel electrophoresis and Western blot analysis. Two E. coli transformants harboring pCYASK5-1 and pCYASK5-2 expressed the cry11Aa gene in size of 72 kDa and 64 kDa, respectively and showed over 89% mortality against Culex pipiens larvae.
In order to determine the seasonal prevalence and population density of vector mosquitoes in Kyungpook Province, a survey based on average numbers of female mosquito per trap-night, were carried out during the period from May to November in 1984. Among the 34, 571 mosquitoes collected in Kyungsan county in animal shelters and human dwellings by light traps, approximately 45.0 per cent were Culex tritaeniorhynchus, 34.0 per cent Culex pipiens pallens, and 19.0 per cent Anopheles sinensis. By comparison, distribution of mosquito species in Ankang town, in which Japanese encephalitis had been reported in the past, were quite different and A. sinensis was the most abundant species, being constituted in 75.8 per cent, followed by C. tritaeniorhynchus(23.2 per cent), and C. pipiens pallens(O.6 per cent). In the general pattern of seasonal prevalence, C. tritaeniorhynchus first appeared in mid-June, and trapped in large numbers during the periods from mid-August to early September, showing a simple sharply pointed one-peaked curve, while C. pipiens pallens was found to be active through almost the entire season showing irregular curves with several peaks. The trend of seasonal distribution of A. sinensis was similar to that of C. tritaeniorhynchus, but the earliest dates of appearance and disappearance differed from that of C. tritaeniorhynchus. The results of hourly catches of mosquitoes by human baited traps have clearly shown the differences in the biting rhythm among the vector mosquitoes of this Province, such as the facts that C. tritaeniorhynchus and A. sinensis are rather constantly active all through a night with more or less inconspicuous peaks twice a night, while C. pipiens pallens exhibits an irregular curve, and the numbers of A. vexans nipponii, C. vagans and Armigeres subalbatus are very small and are not sufficient for estimating the biting rhythm. Four genera and fourteen species of larval mosquitoes collected from eighteen kinds of habitats which classified according to the categories followed by Bates (1949). The main breeding sites contributing to peak C. tritaeniorhynchus adult densities in this surveyed areas were evidently ricefields, ground pools, puddles and swamps and marshes, etc. Summarizing the results, this study indicated that the highest population density of C. tritaeniorhynchus was encountered in late August and early September, showing a simple sharply pointed one-peaked curve.
Ecological studies of Aedes togoi, the vector of malayan filariasis, were ci,wind out at Tolsando, Yosu and Sokcho area in 1991. The adult population of Aedes togoi was continuously appeared from the first week of April to the end of November showing the highest density in .truly. The larvae of Aedes togoi were found at rock pools from March to December in Sokcho area and the density was highest in July and August, whereas in the southern coastal area (Yosu), the larvae were found throughout the year and the density was the highest in Tune. The rate of larvae inhabited below 0.5% salinity was 45.7% in Sokcho and 51.7% in Yosu. The feeding activity of Aedes togoi was nocturnal, with the peak period of 01 :00-03:00 hours. Indoor feeding activities were slightly higher than outdoors showing the biting ratio of 1 :0.8 (indoor: outdoor). The average number of Aedes togoi attracted to CO2 gas was 8.5 whereas 117 Anophelei sinensis was attracted. The result indicates that CO2 is not an effective attractant for host seeking of Aedes togoi compared to Anopheles sineteis. The most common place was bedroom with 54.5% of total collections and next to stock place (18.2%), floor(9.1%) and kitchen (9.1%).
To describe characteristics of a hemolysin in mosquitocidal Bacillus thuringiensis subsp. gyangiensis strain 21-2, Escherichia coli HB101 was transformed with a gene encoding hemolysin in the strain 21-2. Transformant 47 con-tained 2.5 kb DNA was selected by ELISA, immunoblot and DNA electrophoresis. Transformant 47-5 was recon-structed after digestion of the 2.5 kb DNA with Hind m. Transformant 47-5 contained 1.8 kb DNA and expressed 23 kDa Protein which had mosquitocidal activity to Aedes aegypti. The 23 kDa Protein itself in vitro didn't show hemolytic activity on human erythrocytes, but the protein had the activity after proteinase K treatment.
A combined field release of indigenous larvivoroug fish (Aplocheilus latipes) and hybrid herbivore (Tilapia mossambicus miloticus) was conducted to determine the effectiveness of biological control, in particular, against malaria vector (anopheles sinensis), breeding in weedy habitat of rice fields at the University rice paddies at Suwon during the period of June through September in 1988. A combined fish introduction at the release rate of 2.0 fish per $M^{2}$ for Aplocheilus and 1-pair $10M^{2}$ water surface for Tilapia resuited in 70.8% Anopheles larval reducton in a week period, the rates of rontrol increased to 73. 5% and 80.2% in 4th and 5th week respectively after the fish introdurtion, maintaining mosquito suppression in range of 80~82% control after the 5th week against Anopheles and Culex spp. combined. In a single fish treatment with Aplocheilus at 2.0 $fish/M^{2}$ release rate, Anopheles larval reduction ranged on the average 51.4~56.5%in 4 wrek period which was later integratpd with B.t.i. (Rl53.78) treatment at 1.0 kg/ha dosage rate to suppress vector mosquito population down and the the results was compared with that of combined fish introdurtion.
A confined field assessment on the integrated control of vector mosquitoes (Anopheles sinen- sis and Culex tritaent'orhynchus) was conducted by timely treatment of Bacillus thuringiensis(H-14) formulation in the presence of larvivorous fish (Aplocheilus lalipes) in natural rice fields of $30,000M^2$ at Yongam, South Korea Cholla Province from July through October in 1988. In presence of larvivorous fish (Aplocheilus) at lower density of 0.6 fish per $M^2$ water surface, mosquito vector control rates ranged 55. 0~57. 6% from July through August, and when the mosquio density started to increase with over 10 larvae on the average per sample, B.t.(H-14) formulation treatment at the rate of 1. 0 Kg/ha made 100% vectors reduc¬tion in 24 hours, the control sustained above 98% until the test was terminated on October 11th. In the fish introduced rice paddies with Aphyocypris chinensis at the density of above 1. 5 fish per $M^2$, a satisfactory degree of vector control was obtained by 88. 2~96. 7% in 2 week period until September 21st. In the absence of larvivorous fishes, B.t. (H-14) treatment made 100% control in 24 hours, however, vector population rebound appeared in day 7, required to make additional treatment to suppress population down.
