Isolation, Identification, and Evaluation of Biocontrol Potentials of Rhizosphere Antagonists to Rhizoctonia solani (원예작물(園藝作物) 모잘록병(Rhizoctonia solani $K\"{u}hn$)의 발생(發生)에 관여하는 근권길항균(根圈拮抗菌)의 분리(分離), 동정(同定) 및 생물적(生物的) 방제(防除) 검토(檢討))

  • Antagonistic microorganisms from rhizosphere soil were isolated, identified, and applied successfully as the biocontrol agents of damping-off caused by Rhizoctonia spp. Rhizosphere antagonists isolated from rhizosphere soil were identified as Trichoderma viride, T. harzianum, T. hamatum, T. polysporum, Gliocladium sp., Pseudomonas fluorescence, P. stutzeri, P. cepacia, Enterobacter sp., Serratia sp. and Erwinia herbicola. Of these, the most promising ones in vitro were T. virdie, T. harzianum, Gliocladium sp., Serratia sp., P. stutzeri, and P. cepacia. These above six antagonists were efficient in reducing disease incidence to $40{\sim}70%$ when the reselected rhizosphere antagonists preparations were applied to the soil at $10^6$ propagules per gram. Among six antagonists, T. viride was the most promising biocontrol agents against R. solani isolates in soil. The suppressive effect was more evident in steam-sterilized soil than in non-sterilized field soil.

  • Medicinal plants containing wellness-fusion-complex compound are increasingly being pursued as suitable alternative sources of various biological properties. In this study, inhibitory effect of Quintinia acutifolia, which is a New Zealand plant, on P388 murine lymphocytic leukemia cells using MTT [3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl-tetrazolium bromide] assay. Based on $^1H-NMR$, $^{13}C-NMR$ spectral data and other spectral analysis, 2,3,2'',3''-tetrahydroochanaflavone (1) and 2'',3''-dihydroochana-flavone (3) inhibited the leukemia cells were purified from the plants. 2,3,2'',3''-tetrahydroochanaflavone (1) and 2'',3''-dihydroochana-flavone (3) are biflavonoids possessing two basic flavonoids and actively inhibited growth of P388 murine lymphocytic leukemia cells with a 50% inhibitory concentration ($IC_{50}$) of $8.2{\mu}g/mL$ and $3.1{\mu}g/mL$, respectively. Specially, 2'',3''-dihydroochana-flavone (3) possessed unconjugated flavonone system, which isn't consist of a pair with B ring of 2,3,2'',3''-tetrahydroochanaflavone (1). Therefore, the two compounds could be considered as a candidate for development of anticancer drugs and need to much studies in the future.

FINITE ELEMENT STRESS ANALYSIS OF MAXILLARY TWO IMPLANTS-RETAINED OVERDENTURE ACCORDING TO POSITION OF IMPLANT FIXTURES (상악피개의치를 위한 임플랜트의 위치에 따른 응력분포에 대한 유한요소분석)

  • Statement of problem: There have been a few studies about unsplinted implant retainted maxillary overdenture. Purpose: The purpose of this study was to examine the effect of different position of implant for 2 implants-retained maxillary overdenture. Materials and methods: Three-dimensional finite element models were used to reproduce an edentulous human maxilla with an implant-retained overdenture. Two implants in the canine tooth positions on both side and in the second premolar tooth positions on both side models were examined. Axial loads of 100 N were applied to the occlusal surface at the right first molar tooth positions. Maximum stress at the implant-bone interface and stress at the cortical bone surface just under the loading point were observed. Results and conclusion: Within the limits of this study, maximum stresses were concentrated around implant of canine position at loading side. The second premolar area was thought to be more favorable to distribution of stress on mucosa, alveolar bone and implants than canine area for maxillary overdenture.

Detection of periodontal disease related bacteria from the implant-abutment interface in oral cavity (구강내 임플랜트-지대주 연결부에서 치주질환관련 세균의 검출)

  • Statement of the problem: Implant systems result in gaps and cavities between implant and abutment that can act as a trap for bacteria and thus possibly cause inflammatory reactions in the peri-implant soft tissues. Purpose: Porphyromonas gingivalis, Prevotella intermedia, Tannerella forsythia, Treponema denticola, and Aggregatibacter actinomycetemcomitans, related to implant-abutment interface microleakage. Material and methods: Samples were taken from 27 subjects with sterilized paper points and were transported in $1{\times}PBS$. The detection of periodontopathogens were performed by polymerase chain reaction with species-specific primers based on 16S rDNA. Results: Our data showed that the detection rate of P. gingivalis and P. intermedia in implant fixture was 59% and 82% in patients respectively. Detection rate of P. gingivalis and P. intermedia in implant crevice was 44% and 82% in patients. Detection rate of P. gingivalis and P. intermedias in tongue was 82% and 82% in patients. Conclusion: Current implant systems cannot safely prevent microbial leakage and bacterial colonization of the inner part of the implant.

The Surface Characteristic Changes of Hydroxyapatite Coated Ti Disc When Immersed in NaCl Solution (NaCl 수용액에 담근 Hydroxyapatite 코팅된 타이타늄 시편의 표면 변화)

  • In previous studies, methods for enhancing cellular response on the Hydroxyapatite coated implant surface were described. In this study, the changes of surface characteristics such as surface roughness, contact angle, surface energy and surface morphology were observed when Hydroxyapatite coated Ti discs were immersed in NaCl solution for various time. Hydroxyapatite coated Ti discs were immersed in 0.9% NaCl solution for 7, 14 and 21 days at $37^{\circ}C$. The control group comprises dry identical discs not immersed in a solution. (n=3) All discs were dried in air completely and the surface roughness was measured using confocal laser scanning microscopy(CLSM). Static contact angle was recorded by video contact angle analyzer after dropping distilled water on the surface. The surface energy was calculated from contact angles of the three liquids. Surface was observed using a field emission-scanning electron microscope(FE-SEM). As a result, the surface roughness of immersed Hydroxyapatite coated Ti discs increased significantly and the contact angle decreased comparing with control group discs. The surface energy of immersed discs increased except for discs immersed for 14 days.

Analysis of Isoflavones for 66 Varieties of Sprout Beans and Bean Sprouts (66품종 나물콩과 콩나물의 isoflavone 분석)

  • Selection of high-quality varieties of sprout bean and bean sprout containing high levels of total isoflavones was performed by high performance liquid chromatography. The range and mean of total isoflavone contents of sixty six varieties of sprout beans were $247{\sim}2,256$ and $1209{\pm}470\;mg/kg$, respectively, with KLG10618, KLG11118, KLG10600, KLG10022, KLG1085 and sohokong containing the highest amounts of isoflavone among the samples. Medium-sized bean variety with green seed-coat color contained highest amount of total isoflavones among samples. The range and mean of total isoflavone contents of thirty varieties of bean sprouts were $768{\sim}3,343$ and $1,898{\pm}577\;mg/kg$, respectively, with soho bean sprout containing the highest total isoflavone content (3,343 mg/kg, dry basis). Total isoflavone contents of bean sprouts increased gradually during cultivation period, reaching maximum level on the 5th day of cultivation, and were the highest in the order of roots, cotyledon, and hypocotyl.

  • This experiment was conducted to select and rear the adaptable cultivar which isused for various purpose and to build a year-round cultivation using with characters of aerial part and analysis of chemical components in 28 local cultivar of Artemisia princeps Var. The growth of aerial part, which are stem length, stem diameter, no. of branch, node, leaf size and leaf pubescence, in north local cultivars was more vigorous than those of south local caltivars. Fresh weight of "Myeongcheonssuk " was highest in local caltivars, and in view of local area, local cultivars which was collected from Iri Si and Euiseong Gun was shown the highest fresh weight. Local cultivars as mentioned above had a characters with plenty of leaves, nodes and pubescence of leaf is long. In chemical components of leaves in morth cultivar, content of N, MgO, Zn, Cu, Mn was high, while $P_2O_5,\;K_2O$ in middle local cultivars, Fe in south local cultivars. The more content of carbohydrate in leaves was high, the content of $K_{2}O$ had a increasing tendency. In local cultivars which was collected form Iri Si and Euiseong Gun with the highest fresh weight, content of $N,\;P_{2}O_5,\;K_{2}O$ was high, on the other hand content of MgO, Zn, Ca, Mn, Fe, was low in leaves of Artemisia Princeps Var.

Distribution of Sexually Transmitted Viral Diseases in Busan (부산지역 바이러스성 성병 감염양상 연구)

  • This study was performed to evaluate the prevalence of high risk Human papilloma virus (HPV), Herpes simplex virus type 1, 2 (HSV-1,2), Hepatitis B virus (HBV) and Human Immuno deficiency virus (HIV) infection with sexual transmitted viral diseases in Busan during 2004 to 2005. six hundred seventy four samples of cervical swabs were tested for sexually transmitted viral diseases. Among the isolated viruses, 23 (3.4%) samples were HPV and 3 (0.4%) and 9 (1.3%) samples were HSV 1 and 2, respectively. Among the 586 serum samples tested for viruses, HSV IgM 121 (3.6%), HSV-1 IgG 487 (83.1%), HSV-2 IgG 135 (23.0%), HBsAg 26 (4.4%), HBeAg 7 (1.2%), and HIV (0%) types were found. HPV genotypes were detected in 16 patients, of which 13 cases were high risk type HPV, 3 cases were low risk type HPV, and multi infection were detected in 7 cases. In the age distribution of the patients, 7.2% of infection tested from cervical swabs occurred in under the age of 20, while 100% of infection was found to occur in those who were 40 years old or older in the serum samples. The outbreak pattern in their occupations was found to be the highest at the health organization (amusement quarter) for the cervical swabs, and at infirmary (commercial sex worker) for the serum samples, respectively.

Characterization and Outbreak Pattern of Isolated Influenzavirus in Busan, 2000-2002 Years. (최근 3년 부산지역에서 분리한 인플루엔자바이러스의 특성 및 발생 양상)

  • Influenzavirus were isolated from patients with acute respiratory infections in Busan during 2000-2002 and characterized their antigenic properties. In 2000, 39 viruses were isolated and they were identified as Influenzavirus. Among the isolated influenzavirus, 23 were A type influenzavirus and 16 were B type influenzavirus. As a result of antigenic characterization, the influenzavirus were determined to A/Sydney/05/97 (H3N2)-like, A/Beijing/262/95 (H1N1)-like, and B/Harbin/07/94-like virus. In 2001, 56 viruses were isolated and all of the viruses were identified as influenzavirus. They were A/Panama/2007/99 (H3N2)-like and A/Newcaledonia/20/99 (H1N1)-like virus In 2002, 114 Influenzaviruses were isolated. Among the isolated influenzavirus, 83 were A type influenzaviruses, and 31 were B type influenzaviruses. They were A/Panama/2007/99 (H3N2)-like, A/Newcaledonia/20/99 (H1N1)-like and B/Beijing/243/97, B/Honkong/22/2001, B/Sichuam/379/99-like virus when determined their antigenic properties. These results may be useful for the establishment of reliable epidemic data.

Effects of Dandelion on Oxygen Free Radical Generating and Scavenging System of Brain in Streptozotocin-Induced Diabetic Rats (서양민들레가 Streptozotocin으로 유발한 당뇨 횐쥐의 뇌조직 중 유해 활성산소 생성 및 제거 효소계에 미치는 영향)

  • Many studies have shown that hyperglycemia leads to an increase of lipid peroxidation in diabetic patients and animals, reflecting a rise reactive oxygen species production. It is increasingly recognized that brain is another site of diabetic organ damage. Accordingly, this study was to investigate the effect of dandelion on oxygen free radical generating and scavenging system of brain in streptozotocin (STZ)-induced diabetic rats. Male Wistar rats were divided into diabetic (control) and diabetic-dandelion supplemented groups. Dandelion was supplemented for 4 weeks with dandelion leaf and root powder (DLP, DRP) or dandelion leaf and root water extract (DLW, DRW) based on 11.4 g of raw dandelion/kg diet. Diabetes was induced by single injection STZ (55 mg/kg B.W., i.p.)in a citrate buffer. Oxygen free radical generating enzymes, cytochrome P-450, amino-pyrine N-demethylase, aniline hydroxylase and xanthine oxidase, were lowered in dandelion supplemented-groups compared to the control group. Superoxide dismutase, catalase and gluthathione peroxidase activities of brain were also lower in dandelion leaf and root supplemented-group than in the control group, whereas glutathione S-transferase activity and gluthathione content were increased in dandelion supplemented-groups compared to the control group. With regard to the lipid peroxidation products, the malondialdehyde content of brain was lower in dandelion supplemented groups. Therefore, it could be suggested that powder and water extract of dandelion leaf or root are beneficial in preventing diabetic complication from lipid peroxidation and free radical in brain of diabetic rat brain.
