Melanocytes, derived from neural crest, make melanin and protect skin from the hazardous ultraviolet light. Melanocytes with dendritic process has similar morphology with neurogenic cells and share growth factor receptors such as neurotrophin receptors. Melanogenesis can be regulated by ultraviolet light and inflammation of the skin. In addition, several factors such as hormone, cytokines, arachidonic acid can affect the proliferation and melanogenesis of melanocytes. For melanogenesis, melanocytes need expression of various genes including tyrosinase, TRP-1, TRP-2. In addition, melanin need to be transferred from melanocytes to surrounding keratinocytes. The biology of melanocytes is complex and mechanism of melanocytes proliferation and melanogenesis is still under the investigation.
We induced the activation of melanocytes in the epidermis of C57BL/6 mice by ultraviolet B (UVB) irradiation and observed the effect of Bu-Zhong-Yi-Qi-Tang (BZYQT) on the formation, and decrease of UVB-induced epidermal melanocytes. C57BL/6 mice were irradiated by UVB $80\;mJ/cm^2$ (0.5 mW/sec) daily for 7 days, and BZYQT was intraperitoneally or topically applied pre- or post-irradiation. For the estimation of change of epidermal melanocytes, light microscopic observation with dihydroxyphenylalanine (DOPA) stain was performed. Split epidermal sheets prepared from the ear of untreated mice exhibited 11-16 melanocytes/$mm^2$, and one week after UV irradiation, the applied areas show an increased number of strongly DOPA-positive melanocytes with stout dendrites. But intraperitoneal or topical treatment with BZYQT before each irradiation interrupted UVB-induced pigmentation and resulted in a marked reduction in the number of epidermal melanocytes as compared to radiation control skin. The number and size of DOPA-positive epidermal melanocytes were also significantly decreased in intraperitoneally injected or topically applicated group after irradiation with BZYQT at 3rd and 6th weeks after irradiation. The present study suggests the BZYQT as inhibitor of UVB-induced pigmentation and depigmenting agent.

This study investigated antioxidation activity through the content of total polyphenol, that of flavonoid and DPPH radical scavenging activity and measured the cytotoxicity against B16F10 melanoma and inhibiting function of melanin biosynthesis to evaluate antioxidation activity and melanocyte effect of pueraria lobata root extract. As the results of study, it was recognized that the toxicity did not show against B16F10 melanoma and the increase of generating melanin was inhibited as the results of measuring the inhibition function of melanin biosynthesis after inducing the generation of melanin by ${\alpha}$-MSH against B16F10 melanoma cell. The high contents of polyphenol and flavonoid was found as the contents of pueraria lobata root extract increases and DPPH radical scavenging activity as the results of antioxidation activity. Through this study, it was recognized that pueraria lobata root extract has the feasibility that can be used as the material of cosmetics as it has the excellent effect of antioxidation activity and inhibiting the generation of melanin against melanocyte, low toxicity against skin cell and its safety against melanocyte of skin was found.

Pueraria thomsonii Benth. as a medicinal ingredient, has been traditionally used in Chinese medicine to treat fever, acute dysentery, diarrhea, diabetes, and cardiovascular disease. The effects of P. thomsonii flower on skin have not been reported yet. In this study, the whitening effect of P. thomsonii flower was verified using B16F1 melanoma cells and HS68 fibroblasts. P. thomsonii flower extract reduced melanin contents of B16F1 cells in a dose-dependent manner. To identify its active components, we analyzed P. thomsonii flower extract using high performance liquid chromatography (HPLC). As a result, we identified three major isoflavones of tectorigenin, tectoridin, and tectorigenin 7-O-xylosylglucoside. At a non-cytotoxic concentration, the three components also reduced melanin contents of B16F1 cells in a dose-dependent manner. The depigmentation effects were attributed to the reduced gene expression of tyrosinase and microphthalmia-associated transcription factor (MITF). In order to elucidate another depigmentation mechanism, their effects on DKK-1, a fibroblast-derived depigmentation factor, was determined in HS68 cells. As a result, P. thomsonii flower extracts, tectoridin and tectorigenin 7-O-xylosylglucoside, reduced DKK-1 gene expression, while tectorigenin increased DKK-1 gene expression in a dose-dependent manner. These results suggest that tectorigenin can be used as an effective whitening agent that inhibit melanin synthesis in melanocytes and promote the secretion of depigmentation factor from fibroblasts.

Melanin is synthesized by the melanocytes in basal layer of epidermis and distributed over all the layers of skin including corneocytes in stratum corneum, outermost layer of the epidermis. Melanin of corneocytes was stained using Fontana-Masson method and quantified by image analysis. The correlation between melanin contents and skin color value was estimated in the skin of 71 Korean women. Melanin covering area (MCA) showed good correlation with $L^*$ value and $ITA^{\circ}$ (Individual Typology Angle) (r = 0.6049, 0.6651, respectively). MCA can be used as new parameter for skin color study and has potential application for evaluating the efficacy of the skin whitening cosmetics.

목적 : 월견초는 다량의 불포화 지방산인 리놀렌산과 감마 리놀렌산을 함유하고 있으며, 천식성 기침이나 아토피피부염에 효능이 있는 것으로 알려져 있다. 본 연구는 월견초의 전초와 종자 추출물의 피부 멜라닌 합성에 대한 억제 효과를 조사하였다. 방법 : B16F10 멜라닌세포주를 이용하여 멜라닌, tyrosinase 활성 및 세포생존율을 측정하였다. 또한 멜라닌 합성- 관련효소인 tyrosinase, TRP-1, TRP-2의 단백질발현과 $\alpha$-MSH를 처리하여 색소침착을 유도 한 뒤 단백질 발현을 조사하였다. 결과 : 월견자는 B16F10 세포의 멜라닌 합성을 $5\;{\mu}g/ml$와 $10\;{\mu}g/ml$ 농도에서 각각 대조군의 81.3%, 68.3%로 억제하였고 tyrosinase의 활성도 이와 유사하게 억제하였다. 멜라닌 합성-관련효소들의 단백질발현을 관찰한 결과 월견초와 월견자는 tyrosinase 발현을 억제하였으며 TRP-1과 TRP-2의 발현에는 영향을 주지 않았다. 특히 $\alpha$-MSH에 의한 과색소 유도 시 tyrosinase 발현이 현저하게 감소되었으며, 월견자의 멜라닌 합성 억제 효과가 월견초 보다 높게 나타났다. 결론 : 이상의 연구 결과 월견자는 멜라닌세포의 tyrosinase 단백질 발현과 tyrosinase 활성을 억제하여 멜라닌 생성을 감소시키는 것으로 사료된다.

C57BL/6 mice were irradiated by UV-B $80\;mJ{\cdot}cm^{-2}$ ($0.5\;mW{\cdot}sec^{-1}$) daily for 7 days, and HH was intraperitoneally, orally or topically applied pre- or post-irradiation. For the estimation of change of epidermal melanocytes, light microscopic observation with dihydroxyphenylalanine (DOPA) stain was performed. Split epidermal sheets prepared from the ear of untreated mice exhibited 13~15 melanocytes${\cdot}mm^{-2}$, and one week after UV irradiation, the applied areas showed an increased number of strongly DOPA-positive melanocytes with stout dendrites. But intraperitoneal, oral or topical treatment with HH before each irradiation interrupted UV-B-induced pigmentation and resulted in a marked reduction in the number of epidermal melanocytes as compared to the number found in UV-B-irradiated, untreated control skin. The number and size of DOPA-positive epidermal melanocytes were also significantly decreased in intraperitoneally injected or topically applicated group after irradiation with HH at 3rd and 6th weeks after irradiation. The present study suggests the HH as inhibitor of UV-B-induced pigmentation, and depigmenting agent.

본 연구에서는 80% 식물성 알코올을 추출 용매로 사용해 오디를 빛을 차단 후 실온에서 3일 간 추출하였다. 3회 여과한 후 최소 온도($40{\sim}60^{\circ}C$)에서 농축한 뒤 동결 건조하여 파우더 형태로 사용하였다. 오디(Morus alba)의 에탄올 추출물은 B16/F10 세포의 항산화 및 멜라닌 합성 억제 효과를 나타내었다. 멜라닌 함량과 세포 내 tyrosinase 활성을 Western blotting으로 측정 하였다. Tyrosinase와 tyrosinase-related protein (TRP) -1은 tyrosinase-related protein (TRP) -2보다 강력하게 억제되었으며, 이들 결과는 tyrosinase와 TRP-1은 흑갈색을 띠는 eumelanin의 생합성의 억제와 강한 상관관계가 있음을 보여 주었다. ${\alpha}$-melanocyte-stimulating hormone (${\alpha}$-MSH) 처리 한 B16/F10 흑색 종 세포에서 M. alba 에탄올 추출물은 멜라닌 생성 연관 단백질의 발현 및 멜라닌 생성이 용량 의존적으로 억제 하였다. 멜라닌 함량과 세포 내 tyrosinase 활성을 Western blotting으로 측정 하였다. 또한 DPPH와 SOD를 사용하여 항산화 활성을 분석하였고 총 폴리 페놀과 총 플라보노이드 함량을 측정 하였다. MTT assay 분석을 사용하여 M. alba 에탄올 추출물의 세포 독성을 측정 하였다. B16/F10 멜라닌 생성 세포의 tyrosinase 저해 활성 및 사멸 효과가 일반적으로 효과적이었다. 따라서 M. alba 에탄올 추출물은 항산화 및 미백 효과를 나타내며, 기능성 화장품의 천연 성분으로서 우수한 것으로 여겨진다.
Giant congenital melanocytic nevi are very rare, with an estimated incidence of 1 in 20,000 live births. They have a high risk of malignant melanoma transformation and neurological deficits such as neurocutaneous melanocytosis and epilepsy. Early evaluation, surgical intervention and careful long term follow up are recommended to monitor for malignant transformation. We report one case of giant congenital melanocytic nevi diagnosed at birth with the related literatures.
