Quality Characteristics and Antioxidant Effects of Peanut Sprout Soybean Yogurt (땅콩나물 분말 첨가가 대두요구르트의 품질 및 항산화능에 미치는 영향)

  • To evaluate peanut sprouts as a functional food material, the quality characteristics and antioxidant activities of soybean yoghurt with different percentages (0.5, 1.0, 2.0, and 3.0%) of peanut sprout powder (PSP) contents were investigated. The pH of soybean yogurt with PSP decreased in a time- and dose-dependent manner during fermentation, whereas the titratable acidity increased under the same conditions. The Hunter L value (lightness) decreased while the b value (yellowness) and a value (redness) increased as the PSP concentration in soybean yogurt increased. Prior to fermentation, the sugar content of the soybean yogurt increased as the PSP level increased, but the contents decreased as fermentation progressed. Meanwhile, the viscosity of the soybean yogurt increased during fermentation, and the lactic acid bacteria population was higher than that of the control. The sensory properties, including the flavor, color, taste, and overall acceptability, of the soybean yogurt containing 0.5 and 1% PSP, respectively, were higher than those of other tested yoghurts (with 0, 2.0, and 3.0% PSP contents). The antioxidant activity of the soybean yogurt significantly increased at the PSP concentrations of over 1.0%. These results suggest that the addition of PSP has beneficial effects on the quality and functionality of soybean yogurt, and that the addition of 1.0% PSP is the most suitable for manufacturing soybean yogurt.

Antioxidative Effects of Peanut Sprout Extracts (땅콩나물 추출물의 항산화 효과)

  • For the long-term goal of using peanut sprouts as a functional food material, the total polyphenol content and the antioxidant activity of methanol extracts were examined with use of 9 day-old peanut sprouts and peanuts collected in Chungbuk, Gyeongbuk and Jeonbuk provinces of Korea, China and Vietnam. The polyphenol levels in the sprout extracts were higher than those of the peanut extracts. The phenolic content of the Gyeongbuk peanut sprout extract (20.4 mg/g) was the highest of the tested samples. After 9 days of germination the peanut sprout extracts had higher activities than those of the peanut extracts. In particular, the activity of Gyeongbuk peanut sprout extract was the highest (37.67% at a concentration of $200\;{\mu}L/mL$), and its reducing power demonstrated a similar trend. The DPPH radical scavenging activities were measured for methanol extracts of cotyledon, root and stem of Gyeongbuk peanut sprouts; the highest (90.96% at a concentration of $200\;{\mu}L/mL$) was the activity of cotyledon. ABTS radical scavenging and $\beta$-carotene bleaching activities also were higher in the cotyledon extract than in those of the root or the stem. The resveratrol content was higher in the Gyeongbuk peanut sprout extract than in the Gyeongbuk peanut extract ($15.05\;{\mu}g/g $and $1.42\;{\mu}g/g$, respectively). These results suggest that peanut sprouts potentially could be used as a functional food material exhibiting antioxidant effects.

Resveratrol Content and Nutritional Components in Peanut Sprouts (땅콩나물의 레스베라트롤 함량 및 영양성분 분석)

  • To assess the potential of peanut sprouts as a functional food material, the germination rate, resveratrol content, and nutritional components of sprouts were analyzed. Of all samples tested, Gyeong-buk peanuts had the highest germination rate. The resveratrol content was higher in peanuts than in peanut sprouts. The resveratrol level in Gyeong-buk peanut sprout ($15.5{\mu}g/g$) was the greatest of all tested samples. The cotyledon of Gyeong-buk peanut sprout had the highest resveratrol content ($24.89{\mu}g/g$), followed by the roots ($12.66{\mu}g/g$), but resveratrol was not detected in the stems. The levels of moisture, crude protein, crude fat, ash, and carbohydrate in Gyeong-buk peanut sprout were 6.69%, 35.58%, 33.08%, 2.96%, and 21.96% (all w/w), respectively, in dried material. Compared with peanuts, peanut sprouts contained higher protein levels, and a lower content of crude fat, but showed a minimal difference in mineral content. The amino acid content of peanut sprouts (2,551.8 mg/100 g) was higher than that of peanuts (87.89 mg/100 g). Specifically, the asparagine content of peanut sprouts (834.54 mg/100 g) was the highest of all amino acids. Saturated fatty acids, including myristic and palmitic acids, were detected in peanuts, and the levels of unsaturated fatty acids such as oleic (31.19 g/100 g) and linoleic acids (39.24 g/100 g) in peanut sprouts were higher than those of other fatty acids.

  • We investigated the effect of peanut sprout extract (PSE) on andropause symptoms. PSE was evaluated for impotency and benign prostatic hyperplasia via phosphodiesterase (PDE) or 5-alpha reductase II inhibition assay. Inhibition of PDE and 5-alpha reductase II activities in PSE was significantly higher than that of the non-treated control. To investigate the effects of testosterone levels by PSE, we performed cell media test using TM3 cells. Production of testosterone in TM3 cells was elevated by PSE. These results indicate that PSE was able to alleviate andropause symptoms.

Cultivar Selection for Peanut Sprouts and Investigation on the Growth Stage for the High Level of Resveratrol (땅콩나물용 품종선발과 고함량 레스베라트롤 생육단계 구명)

  • This study was conducted to find out optimum cultivars, and growth temperature and stage in peanut sprout for resveratrol production. Morphological characteristic, days to 50% of the final germination percentage($T_{50}$) and early growth vigor were measured in 8 different peanut varieties. In result 'Jopyeong' appeared to be the most appropriate cultivar for peanut production because of its lower contamination rate, lesser lateral root formation, and thicker hypocotyl length. Optimum temperature for growing peanut sprouts was determined 27. Content of resveratrol was examined by high performance liquid chromatography(HPLC) to investigate appropriate growth stage of peanut sprouts for resveratrol production. Resveratrol was higher than $17.0{\mu}g/g$ in peanuts sprouts 9 days after plating. Considering peanut sprout's shapes and content of resveratrol into account, it was most appropriate to harvest in 9-day after germination.

A study on Suppression Condition of Lateral Root Development for Healthy Production of Peanut Sprouts (땅콩나물 생산에서 세근발생 억제 조건에 관한 연구)

  • The present study was conducted to establish the suppression condition of lateral root development for producing high-quality peanut sprouts. The result of Chamsiru or 6BA treatment on peanut sprouts 'Jopyeong' and 'Vietnam' showed that the higher concentration of the solutions showed the better effect for suppressing lateral root development, regardless of the kinds of lateral root inhibitor. Compared to Chamsiru, 6BA showed better effect for suppressing lateral root development, and the effect was more obvious for 'Jopyeong' than 'Vietnam'. The alternant irrigation of Chamsiru and 6BA solution could produce high-quality peanut sprouts by inhibiting bacteria activity in solution and promoting of hypocotyl and root growth, while suppressing the lateral root development. The treatment of mixture solution of $GA_3$ and Chamsiru improved the overall growth of peanut sprouts, and the effect was the highest in the treatment of '$GA_3\;50mg{\cdot}L^{-1}$ + Chamsiru $8.40mg{\cdot}L^{-1}$'.

Establish of Optimum Cultivation Temperature for the Production of Peanut Sprouts (고품질 땅콩나물 생산을 위한 최적 재배수온 조건 확립)

  • The present study was conducted to development mass production methods for peanut sprouts that is considered as a field of blue ocean among the agricultural products. 'Jopyeong' was the best as a major cultivar for peanut sprouts production. The manual for the production of high-quality peanut sprouts is as following. Germination temperature appropriate for production of high-quality peanut sprouts was $27^{\circ}C$. Peanut sprouts at the growth stage of 8th day, and older plants with advanced growth showed deteriorated merchantable and eating quality. Resveratrol compound was not found in the seeds, but its highest amount was detected from 9-day old sprouts. The best water temperature applicable to high quality peanut sprout production was $25^{\circ}C$. The growth of peanut sprout was inhibited by the high temperatures above $35^{\circ}C$ and low temperatures below $15^{\circ}C$.

Cellular Protective Effects of Peanut Sprout Root Extracts (땅콩나물 뿌리 추출물의 세포 보호 효과)

  • In this study, the cellular protective effect and antioxidative property of peanut sprout root extracts were investigated. Cellular protective effects of peanut sprout root extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction of extracts exhibited a cellular protective effect in a concentration dependent manner. Particularly, the aglycone fraction of extracts showed prominent cellular protective effects in a concentration range (5~50 ${\mu}g/mL$). They are more effective than that of (+)-${\alpha}$-tocopherol, known as a lipid peroxidation chain blocker. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of peanut sprout root extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of extracts ($OSC_{50}$; 1.59 ${\mu}g/mL$) showed a similar ROS scavenging activity compare with that of L-ascorbic acid (1.50 ${\mu}g/mL$), known as a strong antioxidant. On the other hand, the order of free radical (1,1-diphenyl-2-picrylhydraxyl, DPPH) scavenging activity ($FSC_{50}$) was (+)-${\alpha}$-tocopherol > 80% MeOH extract > aglycone fraction > ethyl acetate fraction. These results indicate that peanut sprout root extracts can function as an antioxidant in biological systems, particularly skin exposed to solar UV radiation by scavenging $^1O_2$ and other ROS, and to protect cellular membranes against ROS.

Neuroprotective Effects of Methanolic Extracts from Peanut Sprouts (땅콩나물 추출물의 신경세포 보호 효과)

  • The neuroprotective effects of extracts from various parts of peanut sprouts on glutamate-induced neurotoxicity in N18-RE-105 cells were investigated. This study was performed to evaluate the neuroprotective activity of methanolic extracts from the whole (WME), heads (HME), and stems (SME) of peanut sprouts. The neuroprotective effects of these extracts were measured by MTT reduction assay, LDH release assay, phase-contrast microscopy, and flow cytometric analysis on the N18-RE-105 cells. Among these extracts, the HME showed the greatest neuroprotective effects, and was further fractionated with hexane, diethyl ether, ethyl acetate, and water, according to degree of polarity. Out of the fractionated extracts, the diethyl ether layer showed the highest activity on glutamate-induced cytotoxicity in N18-RE-105 cells. The sub-G1 DNA contents of the glutamate-induced severely apoptotic N18-RE-105s were measured by flow cytometric analysis to confirm the HME's anti-apoptotic activity. Interestingly, after incubation with 100 mg/ml of the HME, the proportion of sub-G1 cells of the glutamate-stressed N18-RE-105s had been greatly reduced, from 58.5% to 9.1%. These results imply that HME may have strong potential as a chemotherapeutic agent against neuronal diseases.

Evaluation of Characteristics of Peanut Sprout Using Korean Cultivars (땅콩 품종을 이용한 싹나물 특성 평가)

  • This experiment was conducted to select suitable cultivars and evaluate growth characteristics to get basic information for sprouting peanut. On sprouting peanut, it showed a rapid increase in trans-resveratrol content that has effects on anti-cancer, anti-inflammatory, blood-sugar-lowering and other beneficial cardiovascular in mouse. For this experiment, characteristics of peanut sprouts were tested in 37 cultivars grown for 7 days at $26^{\circ}C$ temperature. There were a lots of variations in the growth characteristics among cultivars as followers : The range of 100 grain weight was 56 to 142 g, hypocotyl length was 4.3 cm to 5.8 cm, diameter of hypocotyl was 5.0 to 8.0 mm, epicotyl length was 0.8 cm to 4.6 cm, seedling ratio per seed number was 84% to 100%, weight per seedling was 4.9 g to 8.4 g, the rate of hypocotyl cleavage was 0% to 46%, the content of trans-resveratrol was $22.5\;{\mu}g/g$ to $88.2\;{\mu}g/g$ and sprout yield was 360% to 820%. The selection points considered were high sprout yield, high seedling rate, high resveratrol content, low brownish cotyledon, no hypocotyl cleavage, and fat hypocotyl etc. The best cultivar selected was 'Jokwang' that showed 7.8 mm diameter, clean cotyledon color, 100% seedling rate, 0% hypocotyl cleavage, $63.3\;{\mu}g/g$ resveratrol, and 820% sprouting yield. This cultivar was expected to be of use as a new food and nutraceutical material. Relationship between growth characteristics showed that root length had significant positive correlations with epicotyl length, resveratrol content and sprouting yield but negative correlations with hypocotyl diameter and cleavage. Hundred grain weight showed negative correlations with resveratrol content, seedling rate and sprouting yield but positively correlated with curved hypocotyl rate and hypocotyl cleavage positively. This result showed small grain seed will be more appropriate for sprouting peanut.
