Ability of Lipophilic Extract Obtained from Plants to Inhibit Tyrosinase Activity in Reverse Micelles (역미셀계를 이용한 지용성 식물체 추출물의 tyrosinase 저해효과 분석)

  • The abilities of petroleum ether-extracts prepared from 75 plants to inhibit tyrosinase activity were evaluated in reverse micelles composed of isooctane/AOT(100 mM)/phosphate buffer(20 mM, pH 8.0) containing tyrosinase(105.3 units/mL) and 3,4-dihydroxyphenylalanine(0.18 mM). Compared with control which has no plant extracts, garlic could completely inhibit in vitro melanogenesis by tyrosinase, and Chinese quince, sweet potato, onion, radish bud and apple did more than 60%. Lipophilic extracts of medicinal plants and herbs such as rosemary, coriander, cinnamomi ramulus, crataegii fructus, ramulus biotae folium, mume fructus, menthae herba, eucommiae cortex and clove also inhibited tyrosinase activity more than 60%. When the extraction yield of lipophilic materials was considered together with their inhibition effect on tyrosinase, it was possible to select plants of which tyrosinase inhibitors could be produced in high quantity from unit weight. Using reverse micelles, the analysis of the capacity of lipophilic materials to inhibit tyrosinase activity which was difficult up to present could be possible.

Inhibitory Effect of a Decoction of Eucommiae ulmides OLIVER and Dipsacus asperoides C. Y. Cheng et T.M.Ai on Collagen II-induced Arthritis Mice (Collagen II-induced Arthritis 생쥐에 대한 두충(杜仲).속단(續斷) 배합약물의 관절염 억제 효과)

  • Objectives : The object of this study was to verify the inhibitory effect of a decoction of Eucommiae ulmides OLIVER (EU) and Dipsacus asperoides C. Y. Cheng et T.M.Ai (DA) on Collagen II-induced Arthritis Mice (CIA mice). Methods : DBA/1OlaHsd mice were immunized with bovine type II collagen. Boostnig same collagen 21 days later, arthritis was induced and then administrated orally the extract of EU+DA (200 or 50 mg/kg) once a day for 4 weeks and compared with that of methotrexate (MTX, 0.3 mg/kg) as a positive control. Results : Administration of EU+DA suppressed the inflammatory progression of CIA mice and the results were 1. Arthritis index of CIA mice was decreased. 2. EU+DA decreased the production of TNF-$\alpha$, IL-6, IL-$1{\beta}$ in the serum of CIA mice. 3. EU+DA decreased the level of IgM. 4. EU+DAincreasaed $CD3^+$, $CD4^+$, $CD4^+$/CD25 but decreased $CD19^+$, $CD3^+/CD49b^+$(NKT), $CD3^-/CD49b^+$(NK), $B220^+/CD23^+$ in PBMC of CIA mice. 5. EU+DA decreased $CD3^+$, $CD4^+$, $CD3^+/CD69^+$ of paw joint in CIA mice. 6. EU+DA decreased subsynovial inflammation. Conclusions : This results demonstrated that extract of EU+DA suppressed the inflammatory progression of CIA mice and supported further studies are required to clarify a mechanism of therapeutic role.

In Vitro Anti-inflammatory Activities of Herbal Extracts with Eucommia ulimoides and Ulmus davidiana (두충과 느릅 혼합추출물에 의한 염증 억제 효과)

  • This study confirmed the cytotoxicity and anti-inflammatory activities of natural herbal extracts (HE) including Eucommia ulimoides and Ulmus davidiana against human mast cell (HMC-1). HE was extracted with distilled water (at $75^{\circ}C$) and then freeze-dried for 5 days. Finally, the HE was sterilized by gamma radiation with $^{60}Co$ ${\gamma}$ source at room temperature. Cytotoxicity of the HE against HMC-1 cell was measured using cell counting kit-8 (CCK-8) assay. In addition, inflammatory cytokines such as $TNF-{\alpha}$, IL-6 and IL-8 were evaluated by ELISA kit on the HMC-1 cells with calcimycin A23187 and phorbol 12-myristate 13-acetate (PMA). The results showed that HE had no toxicity and reduced $TNF-{\alpha}$, IL-6, IL-8 response on HMC-1 cells.

Effects of Eucomiae Cortex on the Depression of Male Aged Mice (두충(杜沖)이 수컷 노화쥐의 우울증에 미치는 효과)

  • In order to investigate the effects of Eucomiae Cortex extracts on the depression caused by aging, histochemistry and immunohistochemistry were performed on the hippocampus of aged rats and the following results were obtained. Experimental animals were divided into three groups as follows: 8 week old ICR male mice, Aging-elicited group (AE group) and Eucomiae Cortex treatment group (EC group) 50 week old male ICR mice were used. The control group and AE group did not take any treatment and did not restrict diets and negatives. In the EC group, 0.51g/kg of Eucomiae Cortex extract was dissolved in distilled water once a day for 6 months. The Eucomiae Cortex extract reduced pyramidal neuronal damage in C3 hippocampus and dentate gyrus, increased DJ-1, SHH positive responses in aged mouse hippocampus, and 8-OHdG positivity was reduced, ${\beta}$-endorphin positivity was reduced in aged mouse substantia nigra. Therefore, based on the above results, Eucomiae Cortex extract reduces damage of pyramidal neurons in the hippocampus caused by aging, inhibits neuronal cell death, induces proliferation and differentiation of stem cells in the hippocampus, reduces DNA damage-induced oxidative stress, so improves the reduction of hippocampus volume. It is also thought to improves depression due to aging through ${\beta}$-endorphin which enhances mood through the inhibition of pain.

Antioxidative, Anti-inflammatory, and DNA Damage Protective Effect in Cortex Extracts of Eucommia ulmoides by Roasting (추출방법에 따른 두충 껍질 추출물의 항산화, 항염증 활성 비교)

  • Background: Eucommia ulmoides has long been used as an herbal medicine for the treatment of diabetes, hypertension and other diseases in many Asian countries. Methods and Results: This study aimed at evaluating the antioxidant and anti-inflammatory properties of its water (EU-DW, and REU-DW) and ethanol (EU-EtOH, and REU-EtOH) extracts, as well as those of non-roasted E. ulmoides (EU) and roasted EU (REU) cortex. The following were assessed in each extract: total phenolic and flavonoid contents, 1,1-diphenyl-2-picryl-hydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and nitrite radical scavenging activities, reducing power, DNA damage prevention activity, and nitric oxide (NO) inhibition activity. Both EU and REU extracts showed high phenolic and flavonoid content, dose-dependent DPPH radical scavenging capacity, high reducing activity, and considerable DNA damage prevention activity. EU extracts showed remarkable ABTS free radicals scavenging capacity. REU extracts showed a higher radical scavenging capacity and played an important role in inhibiting NO production. Conclusions: The results of this study suggested that aqueous and ethanol extracts of EU and REU possess antioxidant capacities, and prevent oxidative damage to DNA, probably owing to their phenolic and flavonoid content. Therefore, EU and REU could be candidates antioxidant supplements.

  • Periodontitis is a chronic inflammatory disease and gums pull away from the teeth and form spaces. The main goal of treatment is to control the infection and anti-inflammation. Eucommia ulmoides Oliv. Bark (EU) is a traditional Korean herbal used for the treatment of arthritis and hypertension. In the present study, we investigated the anti-inflammatory properties of EU in Porphyromonas gingivalis-LPS (LPS-PG) induced YD-38 epithelial cells. We observed significant inhibition of nitric oxide (NO) production and the expression of interleukin-8 (IL-8), interleukin-6 (IL-6), tumor necrosis factor alpha ($TNF-{\alpha}$), cyclooxygenase-2 (COX-2) in LPS-PG-induced YD-38 cells. These results support that EU has an anti-inflammatory effects for the treatment of periodontitis.

The Growth Promoting Effect of Enterobacteria Bifidobacterium infantis KCTC 3270 by Combination of Natural Products Bearing hntioxidative Capacity (장내 유용세균 Bifidobacterium infantis KCTC 3270의 성장 촉진을 위한 항산화 천연산물의 조합구성)

  • The growth of enterobacteria, Bifidobacterium infantis KCTC 3270, was promoted by natural products bearing antioxidative capacity and mixed combinations of two, three and four kinds of them. B. infantis was expressed a good growth by Aurantii nobilis pericarpium majus L., Astragali radix, Epimedii herba, alone, and two mixed combinations of Epimedii herba and Theae folium, Epimedii herba and Crataegi fructus, Epimedii herba and Eucommiae cortex, and three mixed combinations of Theae folium, Epimedii herba and Chelidonium majus L., Theae folium, Epimedii herba and Aurantii nobilis pericarpium, Theae folium, Chelidonium majus L. and Auantii nobilis pericar pium, Theae folium, Chelidonium majus L. and Astragali radix, Theae folium, Aurantii nobilis pericarpium and puerariae radix, Epimedii herba, Chelidonium majus L. and Aurantii nobilis pericarpium, and four mixed com-binations of Theae folium, Chelidonium majus L., Astragali radix and Epimedii herba, and Theae folium, Chelidonium majus L., Astragali radix and Puerariae readix, and Theae folium, Epimedii herba, Aurantii nobilis pericarpium and Puerariae radix, and Theae folium, Chelidonium majus L., Aurantii nobilis pericarpium and Puerariae radix. The best four mixed combination ofr the growth of B. infantis was Theae folium, Chelidonium majus L. Astragali radix and Epimedii herba, which promoted 2.1 times than that of control, and its antioxidative capacity was also 5 times higher, and the ratio of elimination of hydroxyl radical was more than 80% in each dilution rate. As these combinations of natural products might activate some parts of body, they could be applied to pharmaceutical applications, functional foods, antiaging tea, also expected to promote useful bacterial growth for fermentative beverage bearing multifunction.

Effects of Eucommiae Cortex on Myofiber Type Transition and MyoD Expression in Hind Limb Muscle Atrophy of Rats (두충(杜沖) 이 근육위축 흰쥐의 후지 근섬유형 및 MyoD 발현에 미치는 영향)

  • Objectives : Eucommiae cortex is reported that it helps bone and skeletal muscle stronger. In case of bone, many report is presented, but reports related to skeletal muscle are rarely existed. So in order to investigate effects of Eucommiae cortex on the skeletal muscle atrophy following stroke, cerebral infarct was induced by the middle cerebral artery occlusion (MCAO) in the rats. Methods : In order to induce MCAO rats, nylon suture was advanced and then blocked middle cerebral artery(MCA). Water extract of Eucommiae cortex was treated for 15 days, once a day orally, after the MCAO. Effects were evaluated with muscle weights, muscle fiber type composition, cross-sectioned area of muscle fibers in soleus and gastrocnemius of the unaffected and affected hind limbs. And MyoD protein expression in gastrocnemius was demonstrated with immunohistochemistry and western blotting. Results : In the affected hind limb of the MCAO rats, muscle weight loss of gastrocnemius and tibialis anterior muscles were attenuated by Eucommiae cortex treatment. In soleus muscle of the affected hind limb of the MCAO rats, increase of type-I fibers and decrease of type-II fibers were induced by Eucommiae cortex treatment. In soleus muscle of the affected hind limb of the MCAO rats, decrease of cross-sectioned areas of type-I fibers was attenuated by Eucommiae cortex treatment. In gastrocnemius muscle of the affected hind limb of the MCAO rats, increase of type-I fibers and decrease of type-II fibers were induced by Eucommiae cortex treatment. In gastrocnemius muscle of the affected hind limb of the MCAO rats, decreases of cross-sectioned areas of type-I and type-II fibers were attenuated by Eucommiae cortex treatment. In gastrocnemius muscle of the affected and unaffected hind limb of the MCAO rats, MyoD expressions were increased by Eucommiae cortex treatment. Conclusions : These results suggest that Eucommiae cortex has a protective effect against muscle atrophy, through the inhibition of the muscle cell apoptosis, following the central nervous system demage.

Effect of Eucommiae Cortex on Hind Limb Muscle Atrophy of Sciatic Nerve Transectioned Rats (두충이 좌골신경손상 흰쥐의 후지 근육위축에 미치는 영향)

  • In oriental medicine, it is known that Eucommiae Cortex (EC) has strengthening and rehabilitative effects on the bone-muscle dysfunction. This study aimed to evaluate the effect of EC on the skeletal muscle atrophy. The muscle atrophy was induced by unilateral transection of the sciatic nerve in Sprague-Dawley rats. EC (water-extract, 170mg/100 g body weight) was treated once a day for 12 days. In this study, the effect of EC examined the muscle weight of hind limb, cross section areas of muscle fibers, fiber type compositions, apoptosis related factors (Bax and Bcl-2). EC reduced muscle atrophy in soleus (SOL), medial gastrocnemius (MGT), extensor digitorum longus, and tibialis posterior significantly in the damaged hind limb. EC increased type-I muscle fibers and decreased type-II muscle fibers significantly in SOL of the damaged hind limb. EC enlarged cross section areas of type-I and type-II muscle fibers significantly in SOL. EC enlarged cross section areas of type-I and type-II muscle fibers significantly in. EC reduced apoptotic nuclei and atrophic muscle fibers in SOL and MGT. EC reduced Bax positive muscle nuclei in SOL and MGT. EC up-regulated Bcl-2 positive muscle fibers in SOL and MGT. These results suggest that EC has an anti-atrophic effect and anti-apoptotic effect against myonuclear apoptosis induced by the peripheral nerve damage.

Screening of Inhibitory activities on Angiotensin Converting Enzyme from Medicinal plants (약용식물의 Angiotensin Converting Enzyme 저해활성 탐색)

  • Angiotensin converting enzyme(ACE) belongs to the class of zinc protease and plays an important role in the regulation of blood pressure. In this experiment, we investigated the inhibitory activities of medicinal plant extracts on ACE. Fifty medicinal plants were selected and the extracts were prepared by refluxing with 70% methanol. Among the extracts, eleven medicinal plant extracts such as Sedum sarmentosum Bunge, Petasites japonicus(s.et z.) Max, Rubus coreanus, Morus bombycis Koidz, Acorus calamus var. angustatus, Glycyrhiza glabra, Equisetum hyemale, Portulaca oleracea L., Prunella vulgaris var. lilacina Nakai, Sorbus commixta Hedl, Allium thunbergii showed more than 50% inhibitory activities, and Paeonia suffruticosa Andr., lnula helenium, Acanthopanax senticosus Harms, Dendrobium moniliforme, Juglans mandshurica, Zizyphus jujuba, Leonurus artemisia, Aster scaber Thunb, Vitex rotundifolia, Platycodon grandiflorum, Prunus persica, Ligularia fischeri showed $40{\sim}49%$ inhibitory activities. Therefore these extracts which contain high ACE inhibitory activities may be useful as antihypertension agents and to the treatment of hypertension.
