Control of Sperm Penetration In Vitro by Cumulus Cells in Porcine Oocytes (돼지의 체외수정시 난구세포에 의한 정자침입의 조절)

  • The functional role of cumulus cells on the penetration and polyspermy during in vitro fertilization in porcine was examined. The penetration rate was significantly higher (P<0.01) in oocytes with (61%) than without (25%) cumulus cells, but significant differences in polysper-my rates were not observed. When hyaluronidase was added to the fertilization medium with different concentrations, penetration rates in oocytes with cumulus cells were higher than in oocytes without cumulus cells at 0 (61% vs 34% ; P<0.05), 0.01 (56% vs 35% ; P<0.05), 0.1 (66% vs 30% ; P<0.05) and 1.0mg/$m\ell$ (39% vs 27%). The polyspermy rates were lower in oocytes without than with cumulus cells, and had a tendency to decrease with high concentrations of hyaluronidase. In another experiment, the penetration and polyspermy rates had a tendency to increase as time of sperm-oocyte culture was prolonged. At 16 and 20 h after insemination, the penetration rates were significantly higher (P<0.05) in oocytes with (48 and 62% for 16 and 20 h) than without (25 and 31% for 16 and 20 h) cumulus cells in medium containing hyaluronidase. Polyspermy rates were significantly (P<0.05) lower in oocytes without (13% and 16%) than with (37% and 48%) cumulus cells at 16 and 20 h after insemination. In cumulus-free oocytes inseminated in medium containing different concentrations of cumulus cells, the penetration rates were significantly (P<0.05) higher in medium with than without hyaluronidase. The proportion of polyspermy was lower in medium without than with hyaluronidase at 0 (10% vs 0%), 10$^2$(25% vs 0%), 10$^4$(24% vs 14%) and 10$^{6}$ (29% vs 10% ; P<0.05) cumulus cells/$m\ell$. These results suggest that cumulus cells can have a positive influence on sperm penetration, its action on polyspermy control does appear to function primarily on zona pellucida by co-culture of cumulus cells and oocytes in medium without hyaluronidase.

Effect of Sperm Preincubation Medium with Ascorbic Acid and/or Ferrous Sulfate on Porcine In-Vitro Fertilization (돼지의 체외수정시 Ascorbic Acid와 Ferrous Sulfate의 첨가하에서 정자 전배양의 영향)

  • The aim of this work was to study the effects of ascorbic acid (Asc) and/or ferrous sulfate (Fe$^{2+}$) and spernatozoa preincubation on the in vitro fertilization in porcine. Porcine follicular oocytes matured in culture were inseminated with frozen-thawed boar spermatozoa preincubated for 0, 1, 2, 3, 4 and 5h. The penetration rates (37~51%) were not significantly different between durations of spermatozoa preincubation in medium with 0.1 mM Asc. The addition of 1.0 mM Fe$^{2+}$ during spermatozoa preincubation were not significantly affecting the penetration rates (41~56%). When spermatozoa were preincubated with Asc and Fe$^{2+}$, the penetration rates had a tendency to increase with time of spermatozoa preincubation, and were significantly (P<0.05) higher in spermatozoa preincubated with that than without Asc and Fe$^{2+}$ for 5 h. On the other hand, when spermatozoa were preincubated in fertilization medium without Asc and/or Fe$^{2+}$, the penetration rates were significantly (P<0.05) higher in medium with Fe$^{2+}$ than with Asc or Asc and Fe$^{2+}$ for in vitro fertilization. The rate of polyspermy in penetrated oocytes in medium with Asc and Fe$^{2+}$ decreased with the period of spermatozoa preincubation. Despite different culture conditions for spermatozoa preincubation, no differences were observed in polyspermy rates in the presence of Asc and/or Fe$^{2+}$ These results indicate the advantage of preincubating spermatozoa with Asc and Fe$^{2+}$ and an addition of Fe$^{2+}$ during in vitro fertilization with spermatozoa preincubated maintain penetration potential without increased polyspermy rates on in vitro fertilization in porcine oocytes.on in porcine oocytes.

Biodiversity and Characteristic Communities Structure of Freshwater Ecosystems in the Western Area of DMZ, Korea (민통선이북지역(DMZ) 서부평야 일대의 수생태계 생물다양성 및 군집 특성)

  • This study surveyed the diversity and community characteristics of benthic macroinvertebrates and freshwater fish, which are the main animal classifications in a hydro ecosystem, from May to September 2017 in the western plains of the Demilitarized Zone (DMZ). The results showed a total of 125 species of benthic macroinvertebrates belonging to 66 families, 19 orders, and four phyla in the streams and wetlands. Among benthic macroinvertebrates, Coleoptera (27 spp.: 21.60%) was the largest group in terms of species richness followed by Odonata (26 spp.: 20.80%), non-Insecta (22 spp.: 17.60%), and Hemiptera (11 spp.: 8.80%) occupying in the lentic area. Of the feeding function groups (FFGs), predators (51 spp.: 56.67%) showed a relatively larger presence, indicating the dominance of hygrophilous invertebrates that usually inhabit the freshwater wetlands or ponds. Of the habitat oriented groups (HOGs), climbers (33 spp.: 24.44%) and burrowers (17 spp.: 12.59%) were the dominant groups. This observation is typical in a slow flowing habitat and can lead to the disturbance of the ecosystem due to cannibalism among predators. Cannibalism can be caused by stress induced by various population and environmental factors. For the ecosystem services benchmark (ESB) value, site 13 appeared to be the highest with 82 marks while other sites showed relatively lower rates and indices (III water quality class with ${\alpha}$-mesosaprobic). The analysis result of stability factors showed that almost all sites were evaluated to be the I characteristic group with high resilience and resistance or the III characteristic group that was sensitive to environmental disturbance and formed uneven and unstable communities. Of the freshwater fishes, 46 species (3,405 individuals) belonging to 39 families and 18 orders were identified in all the investigated sites. Among them, Cyprinidae (30 spp.: 65.2%) was the largest group, and Zacco koreanus was identified as the dominant species (728 individuals, 21.4%). The survey of freshwater fish communities found both stable communities (sites 7 and 13) with low dominant index (0.39) and high diversity index (2.29) and unstable communities (sites 2, 3, 8, and 10) in opposite tendency. This survey found five Korean endemic species, 17 species belonging to the export controlled species, two endangered species level II (Lethocerus deyrollei and Cybister chinensis), and rare species (Dytiscus marginalis czerskii) among benthic macroinvertebrates. The survey also found an invasive species, Ampullarius insularus, which was distributed throughout the whole area and thus can continuously disturb the ecosystem in the western plain area in the DMZ. Of freshwater fish, one natural monument (Hemibarbus mylodon) and three endangered species level II (Acheilognathus signifer, Gobiobotia macrocephalus, and G. brevibarba) were observed. The survey also found four introduced species (Pomacea canaliculate, Carassius cuvieri, Lepomis macrochirus, Micropterus salmoides) in the western DMZ area, indicating the need for the protection and conservation measures.

Analysis of Genetic Diversity and Structural Changes in Hanwoo Proven Bulls Population (한우 보증씨수소 집단의 유전적 다양성 및 구조 변화 분석)

  • In this study, 844 Hanwoo proven bulls in South Korea (called KPN) were classified into 8 groups based on their birth year. Microsatellite (MS) marker information for paternity identification of each individual is provided at the homepage of the National Agricultural Cooperation Federation, Korea (NACF) and is mainly for the analysis of genetic diversity and structural changes. The polymorphism analysis of KPN whole groups revealed the average number of alleles in each marker (number of alleles), the expected heterozygosity ($H_{ex}$), the observed heterozygosity ($H_{ob}$), the polymorphism information content (PIC) and the $F_{is}$ mean as 10.54, 0.764, 0.773, 0.727 and -0.014, respectively. For group D, with the birth year 2004-2005, the $H_{ex}$ and $H_{ob}$ were 0.777 and 0.792 respectively and the PIC was 0.740. The $H_{ex}$ of group C and D, with birth years 2003-2004 and 2007-2008, respectively, were greater than $H_{ob}$. In all the other groups, $H_{ob}$ was greater than $H_{ex}$. Genetic composition and structure were analyzed using STRUCTURE software. According to the analyzed results, the generation of Hanwoo groups showed changes in specific genetic components according to the flow. It was confirmed that the continuous improvement in the Hanwoo affects the genetic structure of the proven bulls group. The results of this study are expected to be used for enhancing the efficiency of Hanwoo improvement project.

Comparison of soil arthropod community characteristics in organic and conventional orchards in Gangwon Province (강원지역 유기 및 관행 과수원에서의 절지동물 군집 특성 비교)

  • This study conducted an investigation into the effects of fruit type and cultivation practices (organic and conventional) on soil characteristics and soil arthropod communities within apple, blueberry, grape, peach, and pear orchards. The collection of soil arthropod communities was achieved through the utilization of pitfall traps, with concurrent measurements taken for soil moisture content, electrical conductivity, and temperature. The findings of this study unveiled substantial impacts attributed to fruit type and cultivation practices on soil characteristics. Specifically, within organic apple orchards, discernibly higher levels of soil moisture content, electrical conductivity, and temperature were observed when compared to their conventional counterparts. The investigation into soil arthropod communities yielded a total of 1,527 individuals, classified in to five phyla and 15 orders. The range of abundance, species richness, and diversity indices varied across conventional and organic orchards. Cultivation practices were found not to exert a significant influence on soil arthropod community characteristics. However, Non-metric Multidimensional Scaling (NMDS) analysis indicated a significant differentiation in soil arthropod community structure based on cultivation practices. This study underscores the importance of considering vegetation structure and environmental characteristics that may influence soil arthropod communities comprehensively when assessing the impact of cultivation practices on soil arthropods. Furthermore, it emphasizes the need to account for both the characteristics and structure of soil arthropod communities in understanding the implications of cultivation practices on these organisms.

Spatial and Temporal Distribution and Characteristics of Zooplankton Communities in the Southern Coast of Korea from Spring to Summer Period (봄과 여름철의 남해안 동물플랑크톤 시·공간적 분포와 군집 특성)

  • The zooplankton composition, abundance, community structure, and species diversity in the major commercial fishery species spawning grounds in the southern coast of Korea were investigated in this study. A total of 80 taxa were sampled, with the mean abundance range of 5,612-11,720 ind. m-3 and the mean biomass range of 41.6-1,086.8 mg m-3. The dominant species were Paracalanus copepodites, Paracalanus parvus s. l., Oithona copepodites, Paracalanus nauplii, Noctiluca scintillans, Oithona similis, and Ditrichocorycaeus affinis. The species diversity indices were highest in August, suggesting that diversity is influenced by neritic and oceanic warm-water species. A cluster analysis with non-metric multidimensional scaling (nMDS) revealed three groups of zooplankton communities. The April and May samples clustered into Group A, having the highest mean total zooplankton abundance and lowest species diversity, consisting mainly of temperate species located in the middle region of the southern coast of Korea. Cluster Group B was from the early summer season (June) and contained the highest species diversity with some oceanic and neritic zooplankton species. Cluster Group C from the summer season (July and August) mainly comprised P. parvus s. l. and O. similis. The redundancy analysis (RDA) indicated that abundance is positively correlated with salinity, and chlorophyll-a concentrations.

Effects of Environmental Factors on the Seasonal Variations of Zooplankton Communities in the Semi-enclosed Yeoja Bay, Korea (반폐쇄적 여자만 동물플랑크톤 군집의 계절변화에 따른 환경요인의 영향)

  • Effect of environmental factors on the seasonal variations of zooplankton communities was investigated in the semi-closed Yeoja Bay, Korea from February, April to August, and November 2021. Out of a total 49 species of zooplankton were collected with a predominant of neritic copepods (mainly Paracalanus orientalis, Acartia omorii, Acartia ohtsukai, Centropages abdominalis, Ditrichocorycaeus affinis, and Oithona sp.), accounting for 58.9% of the total abundance of zooplankton. The diversity indices indicated a relatively highest in July, suggesting that diversity is influenced by seasonal temperature, N. scintillans, and neritic copepods species. A cluster analysis with non-metric multidimensional scaling revealed four groups of zooplankton communities. The February sample clustered into Group A, having the lowest mean total abundance and species diversity of zooplankton, consisting mainly of N. scintillans located the whole region. Cluster Group B from the spring season (April to May) and contained the species diversity with some neritic copepods. Cluster Group C from the summer season (June to August) mainly comprised P. orientalis, A. ohtsukai, Oithona sp., and hydromedusae. Cluster Group D from the autumn season (November) mainly comprised P. orientalis, Temora discaudata. Redundancy analysis indicated that abundance is positively correlated with temperature, salinity, and pico chlorophyll-a concentrations. This study showed that planktonic larvae (such as branchyura larvae) and some copepods (including A. omorii, A. ohtsukai, C. sinicus, and C. abdominalis) were significantly vulnerable to zooplankton community of temperature, salinity, and pico chlorophyll-a concentrations.

The Effect of Feeding with Sasa quelpaertensis Nakai Extract on Change in Economic Traits of the Pig (제주조릿대 (Sasa quelpaertensis Nakai) 추출물 급여가 돼지의 경제형질 특성 변화에 미치는 영향)

  • This experiment investigated the effects of feed additives of Sasa quelpaertensis Nakai (SQN) extract on Landrace pigs on economic traits such as the quality, physiological characteristics, and productivity. Sixteen pigs with an average age of 154 days were selected as experimental subjects. The experiment was conducted by dividing the group into eight pigs for the supplementation group, feeding with SQN extract, and another eight for the control group feeding without SQN extract. Water was fed ad libitum. On the 30th day, there was no significant difference between meat quality and productivity. However, the glucose and thyroxine were statistically lower with the supplementation group than with the control group (p<0.05). Also, the levels of creatinine difference between 1.18 ± 0.12 mg/㎗ with the supplementation group and 0.70 ± 0.06 mg/㎗ with the control group (p<0.05). However, all serum biochemistry values were within a normal range, with no health problems. The present study will help solve the problem of reducing the diversity of plant species in Halla Mountain by increasing the availability of the SQN as a pig feed additive.
