Studies on the Low-temperature Storage of Strawberry Pulp and Red Pepper Paste by Cryoprotectants (동결방지제에 의한 딸기펄프와 붉은생고추 페이스트의 저온저장에 관한 연구)

  • With adding cryoprotectants to red pepper paste and strawberry pulp, the effects of freezing point depression and the energy requirement for storage at $-15^{\circ}C$ were studied. In case of red pepper paste, the freezing point was pressed to $-9.6^{\circ}C$ by adding NaCl(15% w/w) and citric acid after removing stem. And the components of combined cryoprotectants to keep strawberry pulp in the unforzen state at $-15^{\circ}C$ were dextrose(25% w/'w), fructose(17% w/w) sorbitol(8% w/w) and ascorbic acid(0.2%, w/w). Also, when compared with No-treatment, the storage time reduced about 50% and resulted in about $70{\sim}80%$ cut-down in the energy requirement for storage of food products at low-temperature.

Effects of Cryoprotectants and Diluents on Cryopreservation of the Red Seabream, Pagrus major Sperm (참돔, Pagrus major 정자의 동결보존에 미치는 희석제와 동해방지제의 효과)

  • A series of experiments were conducted to compare the effects of various diluents and cryoprotectants on the motility and survival rate in cryopreservation of the red seabream, Pagrus major sperm. Sperm was efficiently cryopreserved using 300 mM glucose as a diluent. Two cryoprotectant, dimethyl sulfoxide (DMSO) and glycerol, were added to 300 mM glucose to formulate the extenders at concentrations between 5% and 30% by volume for freezing. The highest post-thawed sperm motility and survival rate were obtained with 10% DMSO.

  • This study was conducted to investigate protocol standardization for spermatozoa cryopreservation of the Scapharca broughtonii (Schrenck). Among the freezing rates, freezing at a height of 2 cm above liquid nitrogen surface for 5 minute gave higher activity and survival rate. Among the various diluents, Ringer's solution was the best for S. broughtonii sperm cryopreservation. The suitability of cryoprotectants dimethylsulfoxide (DMSO), dimethylacetamide (DMA), glycerol and methanol were tested against three freezing rates. DMSO gave significantly higher activity and survival rates than others.

  • This study was estimated on performance of deicers, corrosion inhibitors and low corrodible deicer used for removal of snow or ice on the road and on influence on structure. The weight loss rate after freezing and thawing of low corrodible deicer is lower than one of deicer, corrosion inhibitors and these combination. Relative dynamic elastic modulus of all except water, low corrodible deicer and NaCl+JF-1004 was radically reduced after freezing and thawing 150 cycles. And concretes after freezing and thawing were showed severe surface damage. It was found that individual use of low corrodible deicer and corrosion inhibitors had a problem of field application because of lack of early ice melting effect and considerably low durability. Products combined with NaCl was showed rapid weight loss by metal corrosion. Therefore, It will need to circumspectly select combination of deicers having low effect on concrete pavement and bridge if possible.

  • An experiment was performed to obtain cryopreservation techniques of Pacific cod Gadus macrocephalus sperm. Milt were cryopreservation using five cryoprotectant demethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol, methanol and propylene glycol (PG) with marine fish ringer's solution (MFRS) as diluent. Milt were cryopreserved each experimental methods like cryoprotectants (10% and 20%), equilibration time (3, 5 and 10 min) and freezing protocols (liquid nitrogen vapor above 3, 8 and 12 cm). Post-thaw sperm survival rate revealed the highest in 10% PG with optimum methods of equilibration time (3 min) and freezing protocol (liquid nitrogen vapor above 8 cm) about 21.3±1.8%. Hatching rate of fertilization eggs using fresh and cryopreserved sperm were no significantly different.

  • The purpose of this research is to develop the simple freezing method for sperm cryopreservation in the Sevenband grouper and Kelp grouper. The motility and survival rate were checked to investigate the effects of diluent and cryoprotectant on sperm cryopreservation.In cryopreservation experiments, 300 mM glucose and 15% dimethyl sulfoxide (DMSO) were used as a diluent and a cryoprotectant, respectively. As a results of cryopreservation, the motility rate of Sevenband grouper was found to be more than 90%, and that of Kelp grouper was found to be more that 60% in sperm 5 months after cryopreservation.

여러 종류의 동해방지제를 이용한 소 체외수정란의 동결 및 일단계 융해후의 체외배양 생존율 및 수태율

  • 치밀 난구세포로 둘러싸인 소 난자를 $38^{circ}C$. 5% $CO_2$ 배양기에시 5% superovulated cow serum(SCS)이 첨가된 m-TCM 199 medium 으로 $20{\sim}22$ 시간 배양하였으며, 수정능이 획득된 정자와 체외수정하였다. 7일${\sim}$8일경의 수정란을 1.3M methyl cellosolve(MC), 1.1M diethylene glycol(DEG), 1.8M ethylene glycol(EG), 1.6M propylene glycol(PG) 및 1.1M 1,3-butylene glycol(BG) 용액에서 10분간 평형시킨 후 0.25 ml 스트로내에 장전하였다. 스트로를 $0^{\circ}C$의 alcohol bath freezer에 넣고 $-6^{\circ}C$까지 $-1^{\circ}C$/분 속도로 냉각, 식빙 후 10분간 정체시켰으며, $-0.3^{\circ}C$/분 또는 $-0.5^{\circ}C$/분으로 $-30^{\circ}C$까지 냉각 후 스트로를 액체질소에 침지하여 보관하였다. 수정란이 들어있는 스트로를 $30^{\circ}C$ 온수에서 융해하였으며, 수정란을 TCM 199 medium 으로 옮긴 후 5% SCS가 첨가된 TCM 199 medium 에서 48시간 배양하였다. 수정란이 양호한 형태를 유지하며 나중의 발육단계로 진행된 것을 생존한 것으로 간주하였다. 각 종류의 동해방지제에서 동결된 수정란의 일부는 융해 후 동해방지제를 제거하지 않고 직접 비외과적으로 이식하였다. 동결-융해 후 동해방지제의 종류에 따른 탈출배반포 발달율은 EG 50.0%, MC 53.6%, DEG 56.9%, PG 58.0% 그리고 BG 11.5%였다. $-0.3^{\circ}C$/분 또는 $-0.5^{\circ}C$/분 으로 냉각한 수정란의 생존율은 두 그룹간에 유의적인 차이가 없었으나 (P<0.05), 탈출배반포 발달율은 -0.5분 $^{\circ}C$/분(22.6%, 12/53)보다 $-0.3^{\circ}C$/분(64.6%, 31/48) 냉각시에 유의적으로 높았다(P<0.01). 동해방지제의 종류에 따른 수정란의 수태율은 MC 48%(10/21). DEG 30%(3/10), EG 74%(20/27) 및 PG 40%(4/10) 였다. 이러한 결과로 보아 MC, DEG, EG 그리고 PG는 소의 체외수정란의 동결을 위한 동해방지제로서 이용될 수 있음을 보여주었다.

Use of Response Surface Methods to Optimize the Formula of Non-frozen Pound Cake (반응표면분석법에 의한 비동결 파운드 케이크의 최적 배합비 설정)

  • The optimal formula for non-frozen pound cake was sought using a central composite design with a quadratic model, by response surface methodology (RSM). Behavior on supercooling, freezing time, theological properties, and sensory attributes of pound cake were studied by using various concentrations of sugar, sorbitol, and glycerol. The freezing temperature of standard pound cake was $-16.16^{\circ}C$. The hardness, freezing point temperature, and sensory properties were shown as a quadratic relationship whereas moisture content was analyzed by a linear model. Optimized formula for non-frozen cake were suggested to include (sugar:sorbitol:glycerol) 77.6:0.0:20.4, 60.0:32.8:16.4, and 70.8:11.2:19.6 (all % of total sugar) by RSM. The freezing tines of optimized non-frozen pound cake were reduced by $27{\sim}45%$. The freezing Points of optimized pound cake were depressed below $-20^{\circ}C$. Sorbitol and glycerol may be used as cryoprotectants and preserve the desired sensory attributes of pound cake at low temperatures.
