Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products Formation (VII) (한국약용식물의 최종당화산물 생성저해활성 검색(VII))

  • In this study, 49 Korean herbal medicines have been investigated with an in vitro evaluation system using glycation end products (AGEs) formation inhibitory activity. Of these, 18 herbal medicines ($IC_{50}$ < $50{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Of these, five herbal medicines ($IC_{50}$ < $50{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Particularly, Mallotus japonicus (twigs and leaves), Rhus javanica (twigs and leaves), Boehmeria nivea (whole plants), Quercus acuta (stems), and Eurya japonica (stems) showed more potent inhibitory activity (approximately 9-37 fold) than the positive control aminoguanidine ($IC_{50}=76.47{\mu}g/ml$).

  • Advanced glycation end products (AGEs) has been shown to play an important role in the development of the diabetic complications. The AGEs inhibitors or cross-link breakers attenuate various functional and structural manifestations of diabetic complications. In this study, 69 China herbal medicines have been investigated with an in vitro evaluation system using AGEs inhibitory activity. Of these, 28 herbal medicines $IC_{50}$=<50 ${\mu}g/ml$) were found to have stronger AGEs inhibitory activity compared with aminoguanidine ($IC_{50}$=59.77 ${\mu}g/ml$). Particularly, 5 herbal medicines, Camptotheca acuminata (stem, leaf), Eurya groffii (stem, leaf), Cornus Capitata (leaf), Mucuna birdwoodiana (root), Nelumbo nucifera (fruit, seed) showed more potent inhibitory activity (approximately 6-27 fold) than the positive control aminoguanidine.

  • In this study, 64 Korean herbal medicines have been investigated with an in vitro evaluation systems using glycation end products (AGEs) formation inhibitory activity. Of these, 30 herbal medicines ($IC_{50}&lt;50{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Of these, four herbal medicines ($IC_{50}&lt;5{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Particularly, Cornus controversa (branches and leaves), Acer ginnala (stems and leaves), Platycarya strobilacea (flowers) and Picrasma quassioides (stems), showed more potent inhibitory activity (approximately 17-27 fold) than the positive control aminoguanidine ($IC_{50}=77.04{\mu}g/ml$).

  • Advanced glycation end products (AGEs) have been postulated to play a central role in the development of diabetic complications. A variety of different agents that inhibit AGEs have been under investigation. In this study, 54 herbal medicines from China have been investigated with an in vitro evaluation system using AGEs formation inhibitory activity. Of these, 6 herbal medicines ($IC_{50}&lt;5{\mu}g/ml$) were found to have significant AGEs formation inhibitory activity. Particularly, herbal medicines Punica granatum (peels), Terminalia chebula (fruits), Rheum palmatum (roots), Oxyria digyna (stems and leaves), Anisodus luridus (roots) and Quercus schottkyana(stems and leaves) showed more potent inhibitory activity (approximately 9-43 fold) than the positive control aminoguanidine ($IC_{50}=77.04{\mu}g/ml$).

  • The present study was conducted to investigate the effect of whole wheat bread with added Lindera obtusiloba (LO) and Capsosiphon fulvescens (CF) ethanol extracts on serum glucose and lipid levels in Sprague Dawley rats. Rats were divided into five groups depending on the diet administered: normal bread (NC), whole wheat bread (W), whole wheat bread with LO leaves extract (WL), whole wheat bread with CF extract (WC), and whole wheat bread with freeze-drying CF (WDC). After 4 weeks of consuming the experimental diet, the blood glucose level and hemoglobin A1c contents were found to be significantly lower in the W, WL, WC, and WDC groups than in the NC group. The high-density lipoprotein-cholesterol levels increased in the WL group when compared to those in the NC group and triglycerides levels decreased in all wheat groups compared to those in the NC group. These results suggest that wheat breads containing LO and CF extracts are effective for preventing hypercholesterolemia and obesity.

Hydrothermal Pretreatment of Ulva pertusa Kjellman Using Microwave Irradiation for Enhanced Enzymatic Hydrolysis (구멍갈파래의 효소 가수분해 증진을 위한 마이크로파 이용 열수 전처리)

  • The green algae have cellulose as a main structural component of their cell wall and the cellulose content in green algae is much higher than other marine algae such as brown algae and red algae. Furthermore, green algae do not contain lignin in their cell wall and store starch as food in their plastids. Thus, it was investigated that the effect of hydrothermal pretreatment process utilizing microwave irradiation for Ulva pertusa Kjellman, a division of green algae, which is expected to be utilized for bioenergy production, on the enzymatic hydrolysis. The hydrothermal temperature have an effect on the pretreatment of Ulva pertusa Kjellman, but the effect of power of microwave irradiation is negligible. The rate of enzymatic hydrolysis was increased as the hydrothermal temperature increased until $140^{\circ}C$. The enzymatic hydrolysis of pretreated Ulva pertusa Kjellman under the optimum pretreatment conditions (50 W of microwave irradiation power and $150^{\circ}C$ of hydrothermal temperature) with cellulase, ${\alpha}$-amylase, and Novozyme 188 having ${\beta}$-glucosidase acitivity resulted in the saccharification of 96 wt% of total carbohydrate in Ulva pertusa Kjellman during 3 hrs, while it took 24 hrs for the enzymatic hydrolysis of untreated Ulva pertusa Kjellman. It confirmed that the hydrothermal pretreatment was effective on Ulva pertusa Kjellman for the enzymatic hydrolysis.

Effects of Phytoestrogens on Glucose Metabolism in C57BL/KsOlaHsd-db/db Mice (주요 Phytoestrogen들이 제2형 당뇨 마우스의 당질대사에 미치는 효과)

  • This study was conducted to evaluate the antihyperglycemic effects of three phytoestrogens, genistein, coumestrol, and enterolactone, in type 2 diabetic animals. Forty male C57BL/KsOlaHsd-db/db mice were used as a diabetic animal model. The animals were divided into four groups and fed a phytoestrogen-free AIN-76 diet (control), or one of three phytoestrogen-supplemented (3.75 mg/100 g diet) AIN-76 diets for six weeks. During the experimental period, fasting blood glucose levels were measured on week 0, 2, 5, and 6 of the experiment, and oral glucose tolerance tests were performed on the 5th week. After the experimental period, blood concentrations of HbA1c, insulin, and glucagon were measured, and hepatic glycogen content and glucose regulating enzyme activities were analyzed. Fasting blood glucose, HbA1c level, and the area under the blood glucose curve in the oral glucose tolerance test were significantly lower in all of the phytoestrogen-supplemented groups compared to the control group. Plasma glucagon levels were also significantly lower in all of the phytoestrogen-supplemented groups compared to the control group. Hepatic glycogen level was significantly higher in the coumestrol-supplemented group compared to the other groups. However, there were no significant differences in the activities of glucokinase and glucose-6-phosphatase between the groups. These results suggest that all of the three major phytoestrogens tested in the present study were effective in lowering blood glucose levels in type 2 diabetic animals. However, further studies need to be conducted to elucidate the exact mechanism for the hypoglycemic effects of phytoestrogens.

Anti-aging Effects of Prescription Extracts Containing Forsythia viridissima L. (연교를 함유한 처방단 추출물의 항노화 효과)

  • Forsythia fructus has been shown to have antioxidative, anti-inflammatory, antibacterial, anti-aging and whitening effects. This work was carried out to investigate the anti-aging effects of Forsythia viridissima-prescription extracts (Yeongyoseungma-tang, Gamiyeongyoseungma-tang, Hoechunyangkyeok-san) on skin. Skin anti-aging effect of Forsythia viridissima-prescription extracts (Yeongyoseungma-tang, Gamiyeongyoseungma-tang, Hoechunyangkyeok-san) was evaluated by using antioxidant assay, anti-glycation activity, inhibitory effect of tyrosinase activity, expression of type I procollagen and UVA-induced matrix metalloproteinase-1 on HS68 cells, and reduction of ${\alpha}$-MSH-induced melanin on B16F1 cells. Forsythia viridissima-prescription extracts showed anti-oxidative and anti-glycation activity. The pectinex hydrolysed extract from Yeongyoseungma-tang 75% EtOH extract and Gamiyeongyoseungma-tang 75% EtOH extract increased the type I procollagen synthesis, and decreased the UVA-induced MMP-1 expression on HS68 cells. The pectinex hydrolysed extracts of Hoechunyangkyeok-san 75% EtOH extract had the inhibitory effect of melanin synthesis on B16F1 cells. Based on these results, we suggest that Forsythia viridissima-prescription extracts may be useful as a potential source of functional anti-aging cosmetics.

Effects of Hwanggeum-tang Water Extract on the Expression of Pro-inflammatory Responses Elicited by Advanced Glycation End Products in THP-1 Cells (황금탕(黃芩湯) 추출물이 THP-1 세포에서 당화종말산물에 의한 염증반응에 미치는 효과)

  • Hwanggeum-tang (HGT) was recorded in Dongeuibogam as being able to treat Sogal whose concept had been applied to Diabetes Mellitus (DM). Advanced glycation end products (AGEs) play important roles in the development of diabetic complications such as atherosclerosis by eliciting inflammatory responses. In this study, we examined the suppressive effects of HGT against inflammation elicited by AGEs. AGEs treatment increased the expression of pro-inflammatory cytokine gene TNF-${\alpha}$; chemokines MCP-1, IP-10; pro-inflammatory cyclooxygenase COX-2 on the THP-1 cells. HGT had suppressed the expression of pro-inflammatory genes and protein levels in AGE-treated THP-1 cells. HGT had also decreased intracellular ROS production stimulated by AGEs. These results suggest that HGT has beneficial effects for the improvement diabetic vascular complication through suppressing inflammatory responses elicited by AGEs.

  • In this study, the inhibitory effect on advanced glycation end products (AGEs) formation of 43 Korean herbal medicines has been evaluated. Among them, 16 Korean herbal medicines were showed to have significant effect ($IC_{50}$; <50 ${\mu}g/ml$) compared to positive reference, aminoguandine ($IC_{50}$: $76.47{\pm}4.81{\mu}g/ml$). Especially, five herbal medicines, Rubus coreanus (leaves, $IC_{50}$: $4.49{\pm}0.03{\mu}g/ml$), Rubus coreanus (twigs, $IC_{50}$: $3.80{\pm}0.34{\mu}g/ml$), Ampleopsis brevipedunculata (stems, $IC_{50}$: $7.43{\pm}0.09{\mu}g/ml$), Lindera erythrocarpa (leave, $IC_{50}$: $8.14{\pm}0.20{\mu}g/ml$), and Lindera erythrocarpa (stems, $IC_{50}$: $3.69{\pm}0.14{\mu}g/ml$) showed more potent inhibitory activity (approximately 9-20 fold) than the positive control aminoguanidine.
