  • We constructed the fibroin H-chain expression system to produce Discosoma sp. red fluorescent protein variant2 (DsRed2) in transgenic silkworm cocoon. Fluorescent cocoon could be made by fusing DsRed2 cDNA to the heavy chain gene and injecting it into a silkworm. The DsRed2 fusion protein, each with N- and C-terminal sequences of the fibroin H-chain, was designed to be secreted into the lumen of the posterior silk glands. The expression of the DsRed2/H-chain fusion gene was regulated by the fibroin H-chain promoter. The use of the 3xP3-driven EGFP cDNA as a marker allowed us to rapidly distinguish transgenic silkworms. The EGFP fluorescence became visible in the ocelli and in the central and peripheral nervous system on the seventh day of embryonic development. A mixture of the donor and helper vector was micro-injected into 1,020 Kumokjam, bivoltin silkworm eggs. We obtained 6 broods. The cocoon was displayed strong red fluorescence, proving that the fusion protein was present in the cocoon. Accordingly, we suggest that the DsRed2 fluorescence silk will enable the production of novel biomaterial based on the transgenic silk.

  • Silk protein has been explored to be used for biomedical applications for several decades. However, it has not been used in this field cause to their irreversible crystallization after dissolving in water. The existing methods of silk protein hydrolysis using silkworm cocoon were used with harmful solvents and through a very complicated process. Therefore, we have developed novel methods for the production of water-soluble hydrolysate using silkworm gland. We manufactured two types of silkworm gland-derived hydrolysate (water-soluble SGH, SSGH; total SGH, TSGH) and compared the characteristics with commercial cocoon-derived sericin hydrolysate (CSH). The molecular weight of SGH ranged from 7 to 50 kDa (SSGH) and 5 to 15 kDa (TSGH) within glycine, alanine, and aspartic acid as a main amino acid composition. In contrast, CSH ranged from 15 to 50 kDa within serine and aspartic acid. The results of FTIR implied that SGH was more soluble form than CSH, as shown by the decrease in the ${\beta}$-sheet structure at $1630cm^{-1}$ on amide I peak. In comparison with 10% fetal bovine serum, 0.1% (1 mg/ml) SSGH had equivalent effect on the proliferation of human dermal fibroblasts and mesenchymal stem cells. All results of the SSGH made by novel manufacturing process indicate the SSGH is more preferable as a culture medium supplement than cocoon-derived sericin.

  • This study was aimed for identification of a useful genetic resources from the entomopathogenic bacteria infected-midgut of the silkworm, Bombyx mori L. We analyzed the appropriately midgut-immunizing condition of $4^{th}$ instar larvae by a feeding infection using several entomopathogenic bacteria. Xenorhabdus nematophila was selected as a suitable bacteria for midgut immunization of Jam 123, B. mori. We constructed a subtraction cDNA library from the mRNA of the immunized midgut, respectively. A total of 1,000 clones were randomly selected from the subtracted cDNA library, and then performed a differential display hybridization analysis with forward and reverse probes. In conclusion, nine clones were identified as differential expressed genes, which presumed that these genes were involved in gut immunity of silkworm. The total number of clones analyzed in this work is not enough to have a brief overview of a understanding on the midgut immunity factors of silkworm. Therefore, further defined studies on these molecules biological roles will give us well-fined information about the innate immune mechanism of silkworm.

  • Freeze-dried silkworm powder has been approved as a functional ingredients for blood glucose control since 2009. Proximate and nutrient compositions of silkworm powder was summerized as bellow. Silkworm powder contained 4.7% moisture, 57.7% crude protein, 9.5% crude fat, 5.4% crude fiber, and 9.3% ash. The larval protein contained 18 known amino acid including glutamic acid and aspartic acid. Fat is composed of 3:7 ratio of saturated fatty acid to unsaturated fatty acid including various essential fatty acids. It contained 34.7 linolenic acid, 24.4% oleic acid, 20.2% palmitic acid, 9.2% stearic acid, and 7.7% linoleic acid. Five minerals, K, P, Ca, Mg, S were abundant in the silkworm powder with other minerals such as Cu, Mn, Fe, and Zn.

  • The just molted larvae of the fifth instar were fed on with cadmium trated mulberry leaf and on the third day of the fifth instar the daily change of the total protein, activities of GOT and GPT in larvae haemoymph were sexually analyzed. The result obtained are summarized as follows : 1. The total haemolymph was decreased by cadmium treatment. A decreasing ratio of the total haemolymph protein was higher at the later development stage of the male fifth instar than the female fifth instar. 2. There seemed a slight difference in albumin content of the female blood along with the fifth larvae development between control and cadmium treatment where 7% decrease of blood albumin took place with cadmium treatment comparing to that of cotrol. Contrarily a decrease of blood globulin was made on both sexes. 3. The activities of GOT and GPT were inhibited by cadmium treatment.

  • Purification method of unsaturated fatty acid from silkworm pupa has been set up. Over 70% of total fatty acid was unsaturated form. Unsaturated, fatty acid from silkworm pupa improved hyperlipemia in the induced hyperlipemial mice, increased the production of collagen, inhibited the activity of collagenase, and cured the dryness-induced skin very quickly.

  • To isolate naturally occurring novel Bacillus thuringiensis strains for useful pest con시 agents, A total of 64 isolates of B. thuringiensis producing spore and crystal were obtained from 583 soil samples in Korea. Toxicity of isolates was assayed against Lepidoptera (Bombyx ori), Diptera (Culex pipens) and Coleoptera (Sitophilus oryzae), respectively. The results showed that B. thuringiensis isolates toxic Lepidoptera are 42.2% , both Lepidoptera and Diptera are 31.3% and Diptera are 20.3%, demonstrating that spectrum of insecticidal activity of B. thuringiensis isolates is various. But no B. thuringiensis isolates have toxicity against Coleoptera.

  • In the light of the present situation of fertilizing presently apllied in farmer's mulberry fields, it was considered to be value enough to carry out the experiment on whether its degree of fertilizing have no affect on silkworm growth and their cocoon harvest as well, so that the results thus obtained from the experiment are as following; The amount of fertilizing, N 44.4kg, P$_2$O$\sub$5/ 18.4kg, and K$_2$O 34.4kg, respectively were minimum for the . east affect on the silkworm growth and cocoon yields.

  • To investigate the effect of hemolymph of silkworm larvae on the multiplication of Bombyx mori nuclear polyhedrosis virus (BmNPV) in BmN-4 cells, BmN-4 cells were infected with BmNPV, which were sequentially Heated with the hemolymph exracted from B. mori larvae. When the culture media TC-100 containing 3% fetal bovine serum was mixed with 10% hemolymph heated at 65$^{\circ}C$ for 30 minutes, the released polyhedra by multiplication of BmNPV in BmN-4 cells were increased more than those of non-treated. However, multiplication of BmNPV in BmN-4 cells treated with non-heated hemolymph was not effective, since non-heated hemolymph was toxic for the cell growth. The result of plaque assay showed that plaque forming units in BmN-4 cells treated with heated hemolymph are significantly increased, suggesting that efficiency of multiplication of BmNPV in BmN-4 cells is due to increase not of cell growth but of infectivity of BmNPV.
