Effects of Cordyceps ochraceostromat, Silkworm Cocoon and Conjugated Linoleic Acid Addition on the Quality of Cremi Manufactured using Spent Layer Recovered Protein (동충하초 분말, 누에고치 분말 또는 Conjugated Linoleic Acid의 첨가가 산란 노계 회수단백질을 이용하여 제조한 Cremi의 품질에 미치는 영향)

  • To investigate the effects of Cordyceps ochraceostromat, silkworm cocoon and conjugated linoleic acid (CLA) addition on the quality of cremi during storage, this study was conducted. Cremis were prepared using a 20% spent layer breast recovered protein to replace Alaska pollack with the addition of 0.1% silkworm cocoon powder (T1), 0.05% silkworm cocoon powder + 0.05% Cordyceps ochraceostromat powder (T2), and 0.05% silkworm cocoon powder + 0.05% CLA. All manufactured cremis were vacuum-packaged, stored at $10{\pm}1^{\circ}C$ and finally evaluated for their quality on 0, 2, 4 and 6 weeks of storage. The pH, whiteness, gel strength and breaking force of cremis from T2 were higher than those of samples derived from either T1 or T3. However, cremis from T2 showed low shear force values when compared to other cremis (p<0.05). Therefore, the results indicate that cremis contained 0.05% silkworm cocoon powder and 0.05% Cordyceps ochraceostromat combination (T2) may be recommendable to induce customers to buy, thereby, it may be beneficial to both manufacturers and consumers in market.

Anti-obesity and LDL-cholesterol lowering effects of silkworm hemolymph in C57BL/6N mice fed high fat diet (고지방식이 비만 유도 mouse에서 누에체액의 항비만 및 LDL-cholesterol 저하효과)

  • Purpose: Obesity, a worldwide epidemic, is associated with insulin resistance, hyperlipidemia, hypertension, cardiovascular disease, and certain cancers. Many strategies, including natural alternative anti-obesity agents, are used widely to prevent obesity. This study examined the effects of silkworm hemolymph on the weight control of C57BL/6N mice fed with a high-fat diet. Methods: The mice were divided into five groups: normal group (N), high-fat diet group (HFC), high-fat diet and silkworm hemolymph (at dose of 1 mL/kg BW (HFS-1), 5 mL/kg BW (HFS-5) and 10 mL/kg (HFS-10) for 12 weeks. Results: After 12 weeks treatment, the administration of silkworm hemolymph decreased the final body weight significantly along with a decrease in the weights of epididymal fat and total fat. The plasma LDL-cholesterol concentration was significantly lower in the HFS-1, HFS-5, and HFS-10 groups than in the HFC group. In addition, the leptin level of the HFS groups was significantly lower than those of the HFC group without a change in the plasma insulin concentration. Conclusion: These findings suggest that the silkworm hemolymph may have the potential to prevent obesity.

Effects of matured silkworm hemolymph on suppressing melanin synthesis (누에 숙잠 혈림프의 멜라닌 생성 억제 효과)

  • In this study, matured silkworm hemolymph was studied for inhibitory effect on melanin formation, tyrosinase activities. A matured silkworm hemolymph mass purification method was developed and some of suppressing melanin formation effects were investigated, too. As a result, the advanced purification method was higher in yield rate than existing one by 53%. A free radical scavenging activity of hemolymph was higher than sericin on the market($IC_{50}$, $202.9{\pm}33.1$) and lower than vitamin C($0.57{\pm}0.02$) by $6.4{\pm}0.3{\mu}g/m{\ell}$. An inhibitory effect on melanin synthesis was higher in hemolymph compared to Morus bark and arbutin by 9.15% and 11.56%, respectively. An inhibitory effect on tyrosinase activity was higher in hemolymph than Morus bark and lower than arbutin. Effect of hemolymph $45{\mu}g/m{\ell}$ on suppressing of tyrosinase was almost 90%. Thus, these results suggest that hemolymph of matured silkworm may have beneficial properties as a material for cosmetics.

  • Effects of some artificial diets containing pulverized silkworm pupae On the oviposition of a green lacewing, Chrysopa formosa, were studied. The pulverized silkworm pupae with honey seemed to be a good diet for the egg-laying activity of field-collected adults in the beginning of rearing, but their fecundity progressively decreased. Newly emerged adults. did. not lay eggs when fed the above diet, but they were able to lay eggs when the silkworm pupae were defatted and provided with honey. An adult female laid an average of 166.8, 110.2, 96.1, and 66.1 eggs after spending 5, 9, 9, and 11 days for the initiation of oviposition when supplied with various diets i.e. the cabbage aphid, defatted silkworm pupae with additive constituents, mixture of intact and defatted silkworm pupae with additive constituents, and defatted silkworm pupae with honey, respectively.

Studies on the Applicability of Furazolidone to the Silkworm Rearing Industry as a Useful Remedy for Certain Silkworm Diseases (Report I) (푸라졸리돈의 누에병치료약으로서의 응용가능성에 관한 연구 (제1보))

  • The author studied the applicability of Furazolidone to the silkworm rearing industry as a useful remedy for certain silkworm diseases, at the silkworm rearing house of the college of agriculture, Seoul national university, during both the spring and the autumn silkworm rearing season of 1972. Discovering the fact that Furazolidone, when put on the mulberry leaves in a powdered form, is eaten along with the leaves by silkworms and thus the systematic administration of Furazolidone to silkworms is possible, the experimenter carried on a series of experiments (1. determining the in vitro antibacterial activity of Furazolidone to four pathogens of silkworm diseases-Bacillus thuringiensis, Aspergillus oryzae, Aspergillus flavus, and Isaria farinosa, 2. observing the prophylactic and therapeutic effect of Furazolidone against the experimental flacherie caused by inoculation of B. thuringiensis, and 3. examining the toxicity of Furazolidone to silkworm larvae). As the results of the experiments the investigator found out the fundamental fact that Furazolidone exerts a good prophylactic and therapeutic effect against flacherie which is the most common and important silkworm disease: Furazolidone, in in vitro test, inhibited completely the growth of B. thuringiensis, the pathogen of bacterial flacherie, at the concentration of 1 ${\mu}$g/mι. with the tube method and at the concentration of 5 ${\mu}$g/mι. with the plate method, and the drug showed an excellent prophylactic effect and a considerably good therapeutic effect, depending on the time of administration, on the 5th instar silkworms inoculated B. thuringiensis, at the tentative dose of 150mg. per 10 silkworms administered once a day for 2 days. For the practical administration of Furazolidone against flacherie, the dose, the time and duration of administration, and the form of preparation, will be investigated more closely.

Utilization of Grass Powder as an Ingredient of Artificial Diet fro Silkworm, Bombyx mori L. (누에 인공사료 소재로서 목초분말의 이용)

  • The cost of artificial diets for silkworm is very expensive, so that it is essential to lower it in order to extend the period of silkworm rearing on artificial diets. In the present paper, the utilization of several kinds of grass powders was tried to use as an ingredient of artificial diet to reduce its production cost. The value of grass powder as an ingredient of artificial diet was assessed on the base of ingestability and growth of silkworms. As a result it was found it was found that they varied with the kinds and harvesting season of grasses. But it is appeared to be possible to use a grass powder as an ingredient for silkworm diet, when it contains 10~20% of grasses. Moreover, to cut the cost of diet, it is recommanded that the diet containing grass powder could be a applied to the polyphagous silkworm variety.

The Selection and Sensitivity to Environmental Mutagen of Silkworm Reared Artificial Diet in a Screening System Using Specific Locus Mutation of the Silkworm, Bombyx mori (누에의 가시 돌연변이형질을 이용한 인공사료육 누에의 적품종 및 변이원 감수성 조사)

  • We investigated the selection and sensitivity to environmental mutagen of silkworm reared artificial diet to develop all-year-round operation system using a specific locus mutation of Bombyx mori. In the system, mutagenicity could be detected by the egg colour manifested by the pe and/or re genes, which is a kind of recessive visible mutation of the insect. Among, hi, Backokjam, C5, and N12, varieties of silkworm, AT was higher than other varieties in eclosion rate of female, and C5 and N12 were higher in fertility of male. Bakokjam was higher in eclosion rate of female, rate of moth to lay eggs normally in female and male, no. of eggs layed in female and fertility of female. As above results, Bakokjam was finally selected as the most fitness one among varieties of silkworm reared artificial diet. But the sensitivity to mutagen was lower than other varieties. In the sensitivity to mutagen, AT was the most sensitivity to mutagen in tested varieties of silkworm. To use AT variety in this system, AT was improved major characteristics, eclosion rate, fertility, rate of moth to lay eggs normally, and so on, by crossing of other varieties including Bakokjam.

Selection of Mutant Silkworm with Oxidation-deficient Haemolymph for Insect Cell Culture (곤충세포 배지 개발을 위한 체액산화지연 돌연변이 누에계통 선발)

  • Insect cell culture system has been demonstrated the effective means of producing medical and agricultural products. Furthermore, Fetal bovine serum (FBS) is in wide use in insect cell culture. Silkworm hemolymph was tested to develop as a substitute for FBS and was effective in insect cell growth. Hemolymph is oxidized and darkens visibly during the collection from silkworms due to the activity of tyrosinase in it. Toxic quinones are produced by the oxidation and consequently inhibit the cell growth. Heat treatment can be used to prevent the oxidation; however, the oxidation may occur during the collection of hemolymph before it is heat-treated. Hemolymphs collected from 257 different strains of silkworms were examined to select the slowly oxidized hemolymphs. Hemolymphs collected from mutant strains such as $Y_4$, TBO and $wE^b$ showed relatively slow color changes. Oxidation rates of the hemolymphs were measured by the absorbance change using a spectrophotometer. The absorbance of mutant hemolymph reached the saturation value at $20^{\circ}C$ in each 330 min ($Y_4$), 360 min (TBO) and 450 min ($wE^b$) min, whereas the total oxidation time of the wild-type (Baekokjam) hemolymph at the same temperature was 120 min. The cell growth in the medium supplemented with mutant species hemolmph was more effective that in the medium supplemented with Baekokjam species hemolymph.

Production of the yellow fluorescent silk using the fibroin heavy chain protein expression system in transgenic silkworm (피브로인 H-chain 재조합 단백질 발현시스템을 이용한 황색형광실크의 제작)

  • We constructed the fibroin H-chain expression system to produce enhanced yellow fluorescent proteins (EYFP) in the silk of transgenic silkworm. Fluorescent silk could be made by fusing EYFP cDNA to the heavy chain gene and injecting it into a silkworm. The EYFP fusion protein, each with N- and C-terminal sequences of the fibroin H-chain, was designed to be secreted into the lumen of the posterior silk glands. The expression of the EYFP/H-chain fusion gene was regulated by the fibroin H-chain promoter. The yellow fluorescence proving that the fusion protein was present in the silk. Accordingly, we suggest that the EYFP fluorescence silk will enable the production of novel biomaterial based on the transgenic silk.

Sex-Hormone Replacement Effect of Silkworm Pupa and Mixture with Herbs (미성숙 흰쥐에서 누에 번데기 및 한약재 혼합물의 여성호르몬 대체효과)

  • In this study we isolate substances to serve as dietary resources in order to replace the female hormone. Silkworm (Bombyx mori) is one of the most attractive hosts for large-scale production of eukaryotic proteins which have been proven safe as a dietary resource. We report on the estrogenicity of a mixture of silkworm pupa and herbs (Ginseng,Ulkeum, and Hasuo) using the immature rat uterotrophic assay in vivo. Silkworm pupa aqueousextract (KW) and silkworm oil extract (KO) induced effects on the immature rat uterotrophic assay. KO showed neither positive uterotrophic response nor inhibition on E2 induced effect, while KW and MK (mixture of KW and herbs) showed both of the effects. It is concluded that ethanol extracts from silkworm might be a good, therapeutic, natural product for hormone-deficient diseases.
