Changes of Bacterial Diversity Depend on the Spoilage of Fresh Vegetables (신선 채소류의 부패에 따른 세균의 다양성 변화 및 세균에 의한 채소 부패 조사)

  • Almost 10~30% of vegetables were discarded by the spoilage from farms to tables. After harvest, vegetables are often spoiled by a wide variety of microorganisms including many bacterial and fungal species. This investigation was conducted to extent the knowledge of relationship the spoilage of vegetables and the diversity of microbes. The total aerobic bacterial numbers in fresh lettuce, perilla leaf, and chicory were $2.6{\sim}2.7{\times}10^6$, $4.6{\times}10^5$, $1.2{\times}10^6\;CFU/g$ of fresh weight, respectively. The most common bacterial species were Pseudomonas spp., Alysiella spp., and Burkholderia spp., and other 18 more genera were involved in. After one week of incubation of those vegetables at $28^{\circ}C$, the microbial diversity had been changed. The total aerobic bacterial numbers increased to $1.1{\sim}4.6{\times}10^8$, $4.9{\times}10^7$, and $7.6{\times}10^8\;CFU/g$ of fresh weight for lettuce, perilla leaf, and chicory that is about $10^2$ times increased bacterial numbers than that before spoilage. However, the diversity of microbes isolated had been simplified and fewer bacterial species had been isolated. The most bacterial population (~48%) was taken up by Pseudomonas spp., and followed by Arthrobacter spp. and Bacillus spp. The spoilage activity of individual bacterial isolates had been tested using axenic lettuce plants. Among tested isolates, Pseudomonas fluorescence and Pantoea agglomerans caused severe spoilage on lettuce.

Distribution of Foodborne Pathogens from Garlic Chives and Its Production Environments in the Southern Part of Korea (남부지방 부추와 재배환경의 식품매개병원균의 분포)

  • Recently, foodborne illness outbreaks linked to fresh produce are being increasingly reported in the United States, the EU, and Korea as well. Some of this increase may be due to improved surveillance, increase in consumption, change in consumers' habits, and complex distribution systems. Garlic chive is a green, fresh-cut vegetable consumed year-round as a nutrition-rich herb in Korea. It is also prone to contamination with foodborne pathogens during pre-harvest, as amendment with high amounts of livestock manure or compost to soil is required in its cultivation. Our aim in this study was to evaluate microbial contamination of garlic chives, garlic chives cultivation soil, compost, and irrigation water in the southern part of Korea. Samples were collected in A, B, and C regions in 2019 and 2020, and 69, 72, 27, and 40 of garlic chives, soil, compost, and irrigated water, respectively, were analyzed for the presence of sanitary indicator bacteria (total aerobic bacteria, coliforms and Escherichia coli), Bacillus cereus, Staphylococcus aureus, pathogenic E. coli, E. coli O157:H7, Listeria monocytogenes, and Salmonella spp. In A, B, and C regions, levels of total aerobic bacteria, coliform, B. cereus, and S. aureus on all samples were between 1.14 and 8.83 log CFU/g, 0.43 and 5.01 log CFU/g, 0.41 and 5.55 log CFU/g, and 1.81 and 6.27 log CFU/g, respectively. B. cereus isolated from garlic chives and environmental samples showed β-hemolysis activity. Incidence of S. aureus in garlic chive and its production environments in 2020 was different from 2019. In this study, B. cereus and S. aureus were the only pathogenic microorganisms detected in all samples. As a result, this work suggests that continuous monitoring in the production and pre-harvest environment is required to improve hthe hygiene and safety of garlic chive.

Growth Characteristics and Nutrient Uptake of Kalanchoe Plants (Kalanchoe blossfeldiana 'Marlene') at Different Light Intensities and Nutrient Strengths in Ebb and Flow Subirrigation Systems (Ebb and Flow 저면관수 시스템에서 광강도와 양액농도에 따른 칼랑코에(Kalanchoe blossfeldiana 'Marlene') 생육 및 양분흡수 특성)

  • The objective of this study was to determine the effects of light intensity and electrical conductivity (EC) of nutrient solution on the growth and nutrient uptake of potted kalanchoe plants (Kalanchoe blossfeldiana 'Marlene') with growth stage in ebb and flow subirrigation systems. The plants were grown at four ECs of 0.5, 1.0, 1.5, and 2.0 $dS{\cdot}m^{-1}$ for seedling stage and four ECs of 1.0, 1.5, 2.0, and 3.0 $dS{\cdot}m^{-1}$ for short day stage under three daily photosynthetic photon flux (PPF) of 6.5, 10.3, 18.2 $mol{\cdot}m^{-2}{\cdot}d^{-1}$. At seedling stage, plant height was the longest under the lowest light intensity, and particularly dry weights and leaf areas were the highest at PPF 10.3 $mol{\cdot}m^{-2}{\cdot}d^{-1}$. Dry weights and leaf areas were the highest at EC 1.5 $dS{\cdot}m^{-1}$ regardless of light intensity. At short day exposure, plant height was the longest under the lowest light intensity. Dry weights, leaf areas, and number of pedicels of the plants significantly increased as light intensity increased. Under all light intensity conditions, dry weights, leaf areas, and number of pedicles increased until EC becomes to 1.0 - 2.0 $dS{\cdot}m^{-1}$. And after reached the highest at EC 2.0 $dS{\cdot}m^{-1}$, they decreased at EC 3.0 $dS{\cdot}m^{-1}$. By comparing the ion uptakes at EC 1.5 $dS{\cdot}m^{-1}$ of seedling stage and EC 2.0 $dS{\cdot}m^{-1}$ of short day stage in which the plants grew better, we confirmed that ion balance of nutrient solution among $NO_3{^-}$-N, $H_2PO_4{^-}$, $K^+$, $Ca^{2+}$, and $Mg^{2+}$ were significantly changed at short day stage compared to seedling stage. For better growth of the plants, both ion balance and EC of nutrient solution should be considered under different light intensities at short day stage while control of EC is enough at seedling stage.

Evaluation on Microbial Contamination in Red Pepper and Red Pepper Cultivated Soil in Korea (고추와 고추 재배 토양의 미생물 오염도 조사)

  • Red pepper is widely used as a spicy flavor ingredient in the food industry and many households. The objective of this study was to assess the total aerobic bacteria count, coliforms count and incidence of Escherichiacoli, Salmonella spp., Escherichiacoli O157:H7, Listeria monocytogenes, and Bacillus cereus in red pepper and red pepper cultivated soil. The total aerobic bacteria number in red pepper and soil were in the range of 2.97 to 8.13 and 5.91 to 7.65 log CFU/g, respectively. The coliforms in red pepper and soil were in the range of 1.87 to 6.71 and 0.67 to 6.16 log CFU/g, respectively. E. coli was detected in 3 of 54 soil samples. In 3 out 63 red pepper and 53 of 54 soil samples, B. cereus was detected, while Salmonella spp., E.coli O157:H7, and L.monocytogenes were not detected. The results from this study provide an important basic information associated with the microbiological safety of fresh vegetables. Continuous caution is needed to prevent the contamination of pathogenic microorganisms during its farming.

Analysis of the Volatile Flavor Components in Plum ($Prunus$ $salicina$) Irradiated with an Electron Beam (전자선 조사한 자두의 휘발성 유기성분 변화)

  • The changes in the volatile organic compounds in plum after its electron beam irradiation and storage were determined using the simultaneous distillation extraction method and gas chromatograph-mass spectrometry. There were 44, 46, 45, 47, and 38 volatile compounds in the 0-, 0.25-, 0.5-, 0.75-, and 1 kGy irradiated samples, respectively. Also, the volatile flavor components of the plum that was stored for 30 days were identified as 48, 40, 40, 39, and 40 components. The compositions of the volatile compounds of the control and irradiated samples showed a similarity after the storage. Especially, the more important volatile flavor of the plum was identified as hexanal of the C6compounds, (E)-2-hexenal and (Z)-3-hexenal. In particular, hexanal, (E)-2-hexenal, and (Z)-3-hexen-1-ol increased in all the doses, where as hexanol and (E)-2-hexen-1-ol decreased. Among the lactone compounds, ${\gamma}$-hexalactone, ${\gamma}$-octalactone, and ${\gamma}$-decalactone were identified during the storage period in the raw samples. Hexanonic acid and 2-hexenoic acid were not identified during the storage of the samples, and 2-methylprrole was detected only when the storage samples were irradiated at a dose higher than 0.5kGy. Therefore, it was shown that there was no effect on the variation of the volatile organic component suntil 1 kGy in the plum was irradiated with an electron beam.

Comparison of Nutrient Replenishing Effect under Different Mixing Methods in a Closed-loop Soilless Culture using Solar Radiation-based Irrigation (적산 일사 제어법으로 관수하는 순환식 수경재배에서 배액 혼합 방식에 의한 재사용 양액 내 양분 조정효과 비교)

  • Electrical conductivity, drainage, and irrigation amount of nutrient solution are important factors for determination of the mixing ratio of fresh and reused nutrient solutions in closed-loop soilless culture. Generally a fixed mixing ratio is applied in commercial scale greenhouses using solar radiation-based irrigation system. Although it ensures continuous supply of fresh nutrient solution in the mixing process, occasional discharge of the drainage is inevitably required. This study was conducted to compare the nutrient replenishing effect under different mixing processes and to investigate appropriate mixing process. For this experiment, a fixed mixing ratio (FR), modifiable mixing ratio (MR), and open-loop (OP) as control were applied. Mixing ratio was determined by a set value of EC for dilution of collected drainage in FR and the set values of 1.0 and $2.0dS{\cdot}m^{-1}$ were used as treatments (FR 1.0 and FR 2.0), respectively. In MR, mixing ratio was determined based on EC and volume of drainage within irrigation volume per event. The volume of drainage stored in the drainage tank tended to increase in FR 1.0. Although such trend was not observed in FR 2.0 and MR, the volume of drainage stored in MR was lower than that in FR 2.0. The ion balance of $Mg^{2+}:K^+:Ca^{2+}$ or $SO^{2-}_4:NO^-_3:PO^{3-}_4$ in the drainage and reused nutrient solution changed within a narrow range regardless of treatment.

  • The purpose of this thesis is to analyze the error happening in the process of solving the simultaneous inequality with two unknown qualities and to propose the correct teaching method. We first introduce a problem about the simultaneous inequality with two unknown qualities. And we will see the solution which a student offers. Finally we propose the correct teaching method by analyzing the error happening in the process of solving the simultaneous inequality with two unknown qualities. The cause of the error are a wrong conception which started with the process of solving the simultaneous equality with two unknown qualities and an insufficient curriculum in connection with the simultaneous inequality with two unknown qualities. Especially we can find out the problem that the students don't look the interrelation between two valuables when they solve the simultaneous inequality with two unknown qualities. Therefore we insist that we must teach students looking the interrelation between two valuables when they solve the simultaneous inequality with two unknown qualities.

Detection of Hydrocarbons Induced by Electron Beam Irradiation of Almond (Prunus amygosalus L.) and Peanut (Arachis hypogaea) (전자선 조사한 아몬드(Prunus amygosalus L.)와 땅콩(Arachis hypogaea)에서 유래한 지방분해산물 분석)

  • Food irradiation has recently become one of the most successful techniques to preserve food with increased shelf life. This study aims to analyze hydrocarbons in almonds (Prunus amygosalus L.) and peanuts (Arachis hypogaea) induced by electron beam irradiation. The samples were irradiated at 0, 1, 3, 5 and 10 kGy by e-beam and using florisil column chromatography fat, and content was extracted. The induced hydrocarbons were identified using gas chromatography-mass spectrometry (GC/MS). The major hydrocarbons in both irradiated samples were 1,7-hexadecadiene ($C_{16:2}$) and 8-heptadecene ($C_{17:1}$) from oleic acid, 1,7,10-hexadecatriene ($C_{16:3}$) and 6,9-heptadecadiene ($C_{17:2}$) from linoleic acid and 1-tetradecene ($C_{14:1}$) and pentadecane ($C_{15:0}$) from palmitic acid. Concentrations of the hydrocarbons produced by e-beam were found to be depended upon the composition of fatty acid in both almonds and peanuts. The $C_{n-2}$ compound was found to be higher than $C_{n-1}$ compound in oleic acid and palmitic acid, while in case of linoleic acid, $C_{n-1}$compound was higher than $C_{n-2}$ compound. The radiation induced hydrocarbons were detected only in irradiated samples, with 1 kGy or above, and not in the non-irradiated ones. The production of 1,7-hexadecadiene ($C_{16:2}$), 8-heptadecene ($C_{17:1}$), 1,7,10-hexadecatriene ($C_{16:3}$) and 6,9-heptadecadiene ($C_{17:2}$), in high concentration gave enough information to suggest that these may be the possible marker compounds of electron beam irradiation in almonds and peanuts.

Splenocyte-mediated immune enhancing activity of Sargassum horneri extracts (괭생이 모자반 추출물의 비장세포 면역활성 증강 효과)

  • Purpose: This study examined the immunological activity and optimized the mixture conditions of Sargassum horneri (S. horneri) extracts in vitro and in vivo models. Methods: S. horneri was extracted using three different methods: hot water extraction (HWE), 50% ethanol extraction (EE), and supercritical fluid extraction (SFE). Splenocyte proliferation and cytokine production (Interleukin-2 and Interferon-γ) were measured using a WST-1 assay and enzyme-linked immunosorbent assay, respectively. The levels of nitric oxide and T cell activation production were measured using a Griess assay and flow cytometry, respectively. The natural killer (NK) cell activity was determined using an EZ-LDH kit. Results: Among the three different types of extracts, HWE showed the highest levels of splenocyte proliferation and cytokine production in vitro. In the animal model, three different types of extracts were administrated for 14 days (once/day) at 50 and 100 mg/kg body weight. HWE and SFE showed a high level of splenocyte proliferation and cytokine production in the with and without mitogen-treated groups, whereas EE administration did not induce the splenocyte activation. When RAW264.7 macrophage cells were treated with different mixtures (HWE with 5, 10, 15, 20% of SFE) to determine the optimal mixture ratio of HWE and SFE, the levels of nitric oxide and cytokine production increased strongly in the HWE with 5% and 10% of SFE containing group. In the animal model, HWE with 5% and 10% of SFE mixture administration increased the levels of splenocyte proliferation, cytokine production, and activated CD4+ cell population significantly, with the highest level observed in the HWE with 5% of SFE group. Moreover, the NK cell activity was increased significantly in the HWE with 5% of SFE mixture-treated group compared to the control group. Conclusion: The optimal mixture condition of S. horneri with immune-enhancing activity is the HWE with 5% of SFE mixture. These results confirmed that the extracts of S. horneri and its mixtures are potential candidate materials for immune enhancement.

Cryopreservation of Umbilical Cord as a Source of Mesenchymal Stromal Cells and Growth Factors (간엽줄기세포와 성장인자의 공급원으로서 제대 조직의 동결 보관)

  • Background: Umbilical cord (UC) is a promising source of mesenchymal stromal cells (MSCs). We compared the characteristics of MSCs from cryopreserved UC with those from fresh tissues, and demonstrated the possibility of UC cryopreservation for acquisition of MSCs from cryopreserved UC. Methods: Each UC was sliced into two types ($1{\sim}2mm^3$ vs. 0.5 cm), and cryopreserved in liquid nitrogen using different media (autologous cord blood plasma, aCBP vs. RPMI 1640). A fresh aliquot of $1{\sim}2mm^3$-sized UC was used as control tissue. After one week, the cryopreserved tissues were thawed and cultured. For the 0.5 cm UC, a slicing step into $1{\sim}2mm^3$ was needed. Cell count, viability, proliferative activity, and surface antigens were determined from harvested MSCs. Several growth factors (EGF, IGF-1, PDGF, TGF-${\beta}$, bFGF, and VEGF), were measured from the culture supernatant. Results: Eleven UC were enrolled in the study. Efficiencies of obtaining MSCs were higher in cryopreserved UC using RPMI 1640, compared with use of aCBP; the same result was observed for 0.5 cm sized UC, compared with $1{\sim}2mm^3$ sized UC. No difference in proliferative activity was observed between MSCs from fresh and cryopreserved UC. The amount of growth factors in culture supernatant using RPMI 1640 was larger than that of fresh tissues. Conclusion: We obtained growth factors from the supernatant as well as MSCs from cryopreserved UC. As with a cord blood bank, in the future, cryopreservation of UC for acquisition of both MSCs and growth factors would be possible in a time of need.
