Basic Physiological Activities of Bifidobacterium infantis Maeil-K9 and Lactobacillus plantarum KCTC3099 Selected by Anticarcinogenic Activities. (항암 활성능이 우수한 Bifidobacterium infantis Mneil-K9과 Lactobacillus plantarum KCTC3099의 기초 생리활성)

  • This study was conducted to confirm the availability of lactic acid bacteria as probiotics haying inhibitory effects to cancer cell line. Five lactic acid bacteria showing anti-cancer activity were compared by acid tolerance, bile tolerance, antibiotics resistance, milk fermentation, stability, and cell adherence activity to colon epithelial cell. The results obtained are as follows : In acid tolerance, all strains did not have a resistance below pH 3.0 and 3.5 except Lactobacillus plantarum KCTC3099. In antibiotics resistance, Lactococcus lactis and L. plantarum KCTC3099 were resistant to cotrimoxazol (128 mg/1), and Bifidobacterium adolescentis Maeil-K8 and B. infantis Maeil-K9 were resistant to doxycylin and gentamycin (4 mg/1). In case of cell adherence ability to Caco-2 cell, B. infantis Maeil-K9 was found to be superior to others as 3.1%, while the others were less than 0.5%. When the strains were cultured to milk base, viable counts of the strains tested increased more 1 log cycle than inoculation, but acid production was very low except L. plantarum KCTC3099. Also, L. plantarum KCTC3099, B. adolescentis Maeil-K8, and B. infantis Maeil-K9 were stable in fermented milk base during storage. In conclusion, L. plantarum KCTC3099 and B. infantis Maeil-K9 were confirmed to be superior for the availability as probiotics.

Probiotic Properties of Lactic Acid Bacteria isolated from Feces and Kimchi (베트남인 분변 및 김치로부터 분리된 유산균의 프로바이오틱스 기능성 연구)

  • The purpose of this study was to investigate the probiotic properties and antioxidant capacity of lactic acid bacteria isolated from Vietnamese feces and the Korean traditional food kimchi. Six isolated strains were identified as Lactobacillus sp. by 16S rRNA sequencing. All strains showed good resistance to low pH (1.5, 2.0, and 3.0) and 0.3% oxgall bile acids. Culture filtrates from the six strains showed various antioxidant effects, including DPPH, ABTS, reducing power, and metal chelating ($Fe^{2+}$) activities. Two of the six Lactobacillus strains showed potential probiotic activity. Heat resistance and adhesion assays were conducted by mixing the selected strains, Lactobacillus acidophilus V4, Lactobacillus plantarum V7, and Lactobacillus paracasei DK121 isolated from kimchi. The results showed that the heat resistance of these strains was similar to that of a commercial strain, L. plantarum LP. In addition, a mucin attachment assay using the mixture of selected strains (V4, V7, and DK121) showed high binding activity to the mucous layer. In conclusion, a mixture of V4, V7, and DK121 shows promising probiotic activity and may be useful for the development of health-related products.

Probiotic Properties of Lactic Acid Bacteria Isolated Traditional Fermented Foods (전통발효식품 유래 유산균의 프로바이오틱스 특성 연구)

  • This study performed to investigate the probiotic properties of lactic acid bacteria 200 strains isolated from traditional fermented foods. Based on being higher tolerance to bile salts and showing higher acid resistance, 4 LAB Strains were selected in the screening experiment; Lactobacillus plantarum SRCM 102224, Lb. plantarum SRCM102227, Lb. paracasei SRCM102329, Lb. paracasei SRCM102343. Antibacterial activity against various pathogens, acid and bile salt tolerance, hemolytic phenomenon, cell surface hydrophobicity, and antibiotic resistance were examined. Among the tested strains, SRCM 102343 (95.9%) was highly observed hydrophobicity compared to Lb. rhmanosus GG (13.4%) as control. In this study, the in vitro adhesion properties of 4 strains of LAB was investigated using human intestinal caco-2 cell cultures. SRCM102329 and SRCM102343showed higher adherence to caco-2 cells than Lb. rhamnosus GG. The antibacterial activities of 4 strains LAB were investigated. the 3 strains showing strongly antimicrobial activity against Escherichia coli ATCC10798, Staphylococcus aureus KCCM11593, Listeria invanovii KCTC3444, Bacillus cereus ATCC11778 and S. enterica serovar. Typhi KCTC1926. These results suggest that selected strains have good probiotic potential for application in functional foods.

  • 최근 디지털 프린팅 기술의 핵심기술로 떠오르고 있는 잉크젯 프린팅 기술은 최근 기존의 문서인쇄 뿐 아니라, 직물 인쇄, 태양전지 등의 다양한 반도체 소자 제조에 널리 활용되고 있으며, 점차 그 응용 분야를 넓혀가고 있다. 특히 thermal 방식의 잉크젯 피린팅 기술은 etching, thin film process, lithography등의 반도체 공정 기술을 이용하여 제작할 수 있기 때문에, 현재 잉크젯 프린팅 기술은 대부분 thermal 방식을 체택하고 있다. 이러한 thermal 잉크젯 프린팅 방법에서는 잉크를 토출시키기 위하여, 전기적 에너지를 열에너지로 전환하는 전자저항막층이 필수적으로 필요하게 되는데, 이러한 전자저항막층은 수백도가 넘는 고온 및 잉크와 접촉으로 인한 부식 및 산화 문제가 발생할 수 있는 열악한 환경에서 사용되므로, Ta, SiN과 같은 보호층을 필수적으로 필요로 한다. 그러나 최근 잉크젯 프린터의 고해상도 고속화, 대면적 인쇄성 등과 같은 다양한 요구 증가에 따라, 잉크젯 프린터의 저전력 구동이 이슈로 떠올라 열효율에 방해가 되는 보호층을 제거할 필요성이 제기되고 있다. 지금까지는 Poly-Si, $HfB_2$, TiN, TaAl, TaN 0.8 등의 물질들이 잉크젯 프린터용 전자저항막 물질로 연구되거나 실제로 사용되어져 왔으나, 이러한 물질들을 보호층을 제거하는 경우 쉽게 산화되거나, 부식되는 문제점을 가지고 있다. 따라서, 기존 전자저항막의 기능을 만족시키면서, 산화나 부식에 대한 강한 내성을 가져 보호층을 제거하더라도 안정적으로 구동이 가능한 하이브리드 기능성(히터 + 보호층)을 가지는 잉크젯 프린터용 전자저항막 물질의 개발이 시급한 실정이다. 본 연구에서는 자기조립특성을 가져 정밀제어가 가능한 원자층증착법(Atomic Layer Deposition)을 이용하여 원자/나노 단위의 미세 구조 컨트롤을 통해 내열 내산화 내부식성 저온도저항계수를 동시에 가지는 다기능성 전자저항막을 설계 및 개발하고자 하였다. 전자저항막 개발을 위하여 우수한 내부식 내산화성을 가지고 결정립 크기에 따른 온도저항계수 조절이 가능한 platinum group metal들과 전기 저항 및 내열성 향상을 위한 물질의 복합구조막을 원자증증착법으로 증착하였다. 또한, 전자저항막 증착시 미세구조와 공정 변수가 내부식성, 내산화성, 그리고 온도저항계수에 미치는 영향을 체계적으로 연구하여, proto-type의 inkjet printhead를 구현하였다.

Selection of Acid-tolerant and Hetero-fermentative Lactic Acid Bacteria Producing Non-proteinaceous Anti-bacterial Substances for Kimchi Fermentation (비단백질성 항균물질을 생산하는 김치발효용 내산성 Hetero 발효형 유산균주 선발)

  • Twenty-three strains of Leuconostoc species and 45 strains of Weissella species inhibiting the growth of Lactobacillus sakei, one of the most populous lactic acid bacteria in over-ripened kimchi, were isolated from kimchi in our previous study. Among these hetero-fermentative 68 strains, Leuconostoc mesenteroides CK0128, Weissella cibaria CK0633, and W. cibaria KK0797 exhibited a relatively high survival rate in MRS medium, which was adjusted to pH 4.3 using an acid mixture consisting of acetic and lactic acids, and produced a large amount of exopolysaccharides. The culture supernatants of 3 strains were fractionated by a molecular weight cutter and lyophilized. The fractions with a molecular weight smaller than 3,000 Da showed antagonistic activity against Staphylococcus aureus and Lb. sakei. The anti-bacterial substances were very stable to heat treatments ($121^{\circ}C$, 15 min) and active at acidic conditions below pH 5. ${\alpha}$-Amylase, lipase, and proteolytic enzymes (proteinase K and pepsin) did not affect their activities. These non-proteinaceous anti-bacterial substances inhibited the growth of several food pathogens.

Preparation of Yogurt Added with Potato and its Characteristics (감자를 첨가한 요구르트의 제조와 특성)

  • The curd yogurt (total solid content: 14%) was prepared from milk added with skim milk powder and potato puree. Potato puree comprised 9.5. 13,8 and 17.9% (dry basis) of the milk-potato mixture, and the effect of potato on the quality of yogurt was investigated. Addition of potato remarkably stimulated acid production and propagation of lactic acid bacteria, and viable cells reached above $3.9{\times}10^{10}$ CFU/ml after 12 hours. As potato content increased, the ratio of lactic acid content to total acidity decreased, while citric acid increased. The major organic acids of yogurt were lactic acid, citric acid, and acetic acid. Viscosity of yogurt was increased in proportion to the increment of the potato content. After 24 hours of incubation, the sensory score of yogurt containing 13.8% (dry basis) potato showed better sensory acceptability. When curd yogurt added with potato was kept at $5^{\circ}C$ for 15 day, its keeping quality was relatively good. Viable cells of lactic acid bacteria and ${\beta}-galactosidase$ activity decreased rapidly at pH 1.5, and 2.5, but the group added with potato was more stable than control.

Isolation and Identification of Probiotic Lactobacillus Isolates for Calf Meal Supplements (사료 첨가용 생균제를 위한 Probiotics 유산간균의 분리 및 동정)

  • Fifty four acid-resistant and bile-resistant isolates of lactic acid bacteria were initially isolated from the faces of Korea native cattle and Holstein using MRS agar and LAPT agar, and ten strains with superior activity against bile salt were finally selected LS1, LS15, and LL6 isolates showed survival of 66.5%, 82.6% and 80.7% against the simulated stomach liquid(pH 2.5), respectively. LL6 and LL7 isolates had the highest inhibitory activities against the pathogenic bacteria such as Salmonella typhimurium, Staphylococcus aureus, and Clostridium perfringens. By using API 50 CHL kit LS1, LS2 and LM1 isolates were identified as a L. fermentum. LL6 and LL7 isolates as a L. acidophilus, and LS3 isolate as a L. plantarum. The other four isolates belong to genus Lactobacillus. All the isolates tested were sensitive to some antibiotics such as ampicillin, amoxicillin and erythromycin, but resistant to colistin and ciprofloxacin. LB1, LL6 and LL7 isolates were resistant to gentamicin and neomycin. Especially, the LL6 isolate showed the highest resistance to both of the simulated stomach liquid and bile salt, in addition to the highest inhibitory activities against Sal. typhimurium, Staph. aureus and Cl. perfringens.

Probiotic Properties of the Candida kefyr Isolated from Kefir (Kefir에서 분리한 Candida kefyr의 생균제를 위한 특성)

  • To search direct fed microbials, we isolated a Candida sp. from kefir grain. The isolated Candida sp. strain showed 99.8% of identity to the species of Candida kefyr by API 20C kit. Enzyme activity of Candida kefyr was higher in amylase (0.33±1.12μmol/min/mg) than that in phytase (0.052±0.98μmol/ min/mg) cellulase(0.051±μmol/min/mg) and xylanase (0.011±0.98mol/min/mg). The maximum numbers of Candida kefyr in growth curve were reached at 30 h fermentation. Candida kefyr showed high resistances to acidic environment, which was not perfectly extincted even at pH 2.0. And it showed high tolerance to bile salt which had almost 97.2% of survival in the presence of 1.0% bile salt.Especially, Candida kefyr showed high heat stability which remained 10% of initial microorganisms at 60℃. Candida kefyr was not generally inhibited by most of 11 antibiotic agent which contained tetracycline groups. These results suggest that the isolated Candida kefyr has a useful properties as probiotics.

Rapid in vivo Colonization Screening of Probiotic Bacteria Isolated from Human Infants using Caenorhabditis elegans Surrogate Host (Caenorhabditis elegans 생체대체모델을 이용한 한국 영유아분변 유래 프로바이오틱스 균주의 in vivo 장 우점능 검토)

  • The ability of probiotics to adhere to the intestinal epithelium likely plays an important role in their colonization of the gastrointestinal tract. Here, we performed high-throughput screening (HTS) for suitable characteristics of potential probiotic bacteria using attachment and colonization ability through a C. elegans surrogate in vivo model. A total of 100 strains of lactic acid bacteria (LAB) isolated from infant feces were subjected to the colonization assay using C. elegans intestine. Based on colonization ability, we showed that nine isolates have a high attachment ability during whole experimental periods (up to 168 h), compared to Lactobacillus rhamnosus strain GG as a control. Also, through the use of an in vitro cell attachment model, nine isolates revealed highly binding activity to the mucus layer. Next, the selected 9 isolates were assayed for their survival ability when exposed to acidic and bile conditions as well as cholesterol reduction and the utilization of prebiotic substrates. As a result, the isolated nine strains were determined to be highly resistant to acid and bile conditions. In addition, they have significant activity for the reduction of cholesterol and utilization of several prebiotic substrates as a carbon source. Finally, the selected nine strains were identified by either L. rhamnosus or L. plantarum (4 strains for L. rhamnosus and 5 strains for L. plantarum, respectively). Taken together, we propose that the direct colonization of probiotics using C. elegans may be applicable to the rapid screening of valuable probiotic strains in vivo.

A Fundamental Study on Sulfate Resistance of Inorganic Binder with High Acid Resistance (고내산성 무기바인더의 내황산성에 관한 기초적 특성 연구)

  • This study analyzed sulfate resistance of strongly acid-resistant inorganic binder based on industrial byproducts. According to the study experiment, compared to OPC mixture, the mixture of high acid-resistant inorganic binder had excellent chemical resistance against 10% H2SO4 solution. In the case of ordinary portland cement, its sample with 28 days of immersion had severe corrosion on its mortar erosion part, and thus external appearance was damaged greatly, and compression strength decreased by around 57% and more.
