본 연구의 목적은 백삼, 오미자, 맥문동, 길경, 감초 등으로 구성된 백삼복합물이 난알부민으로 유도된 천식 마우스에서의 천식개선에 대한 융복합 연구를 통한 효능을 확인하는 것입니다. 난알부민으로 천식을 유발한 마우스에 백삼복합물을 100 mg/kg과 300 mg/kg을 경구 투여한 후 혈청 내 Th2 사이토카인 IL-4, IL-5, IL-13을 루미넥스를 이용하여 측정하였다. 또한, 혈청과 혈액 내 난알부민 특이-IgE 수치와 호산구, BALF 내 총 세포수는 자동혈구계산기 및 현미경을 통해 측정하였다. 그 결과, 혈청 내 IL-4, IL-5, IL-13 및 난알부민 특이-IgE 수치가 대조군에 비해 각각 감소하였다. 또한, 혈액 내 호산구 및 BALF 내 총 세포수 역시 감소를 보였다. 이러한 결과는 백삼복합물이 Th2 사이토카인과 천식 요인에 대해 감소가 나타나 효과적인 항천식 치료 약물로서 사용될 수 있음을 시사하는 바이다.
한외여과를 통하여 난백으로부터 라이소자임을 분리, 정제하는 연구를 수행하였다. 교반식 한외여과장치에 분획 분자량(MWCO)이 각각 10 kDa, 30 kDa 및 100 kDa의 셀룰로즈 재질 분리막을 사용하여 알부민과 라이소자임의 투과실험 데이터를 수집하였다. 단백질 용액은 20 mM, pH 6, 7, 8의 인산염 완충용액에 난단백 함량을 $1\%,\;2\%,\;3\%$ 및 $10\%$ 첨가하여 제조하였으며, 막간압력차(TMP)는 $0.5\~3$ bar의 구간에서 실험을 하였다. 분리막의 분획분자량이 증가하면서 투과유속은 증가하였으며, 분리막에서의 막간압력차가 증가하면서 그리고 공급원액의 난백농도가 감소하면서 투과유속은 증가하였다. 또한, 분리막의 분획분자량이 감소하면서 알부민의 라이소자임에 대한 선택도는 증가하였으며, 모든 분리막에서 pH 와 공급원액의 난백농도 그리고 막간압력차가 증가하면서 라이소자임의 알부민에 대한 선택도는 증가하였다.
Objectives : In this study, we investigated the effect of Saposhnikoviae Radix on allergic responses in ovalbumin(OVA)-induced Allergic rhinitis(AR) mice. Methods : BALB/c mice were orally administrated with Saposhnikoviae Radix water extract (SRW, 50 mg/kg) or anti-histamine drug, Ketotifen (10 mg/kg) as a reference drug, followed by sensitization and challenge of OVA. Mice were measured clinical symptoms and the serum levels of histamine, IgE, IL-4, and IFN-${\gamma}$, and observed histopathological changes of nasal mucosa H&E staining. Results : SRW significantly decreased rubs and the serum levels of histamine, IgE, and IL-4, and then increased the serum levels of IFN-${\gamma}$ in OVA-induced AR mice, and inhibited histopathological changes of nasal mucosa with inflammation and the eosinophils infilteration. Conclusions : These data suggest that SRW has anti-allergic effect through the inhibitory property of Saposhnikoviae Radix against allergic responses in allergic rhinitis.
Objectives : In this study, we investigated the effect of KOB, a polyherbal medicine for allergic rhinitis and its main herb, Astragali Radix on allergic responses in ovalbumin (OVA)-induced Allergic rhinitis(AR) mice. Methods : Sprague Dawley (SD)-rats were orally administrated with KOB (500 mg/kg), Astragali Radix water extract (ARW, 100 mg/kg) or anti-histamine drug, dosodium cromoglycate (50 mg/kg) as a reference drug, and then intraperitoneally injected with compound 48/80 (8 mg/kg). Rats were measured the mortality and serum levels of histamine. BALB/c mice were orally administrated with KOB (500 mg/kg), ARW (100 mg/kg) or anti-histamine drug, Ketotifen (10 mg/kg) as a reference drug, followed by sensitization and challenge of OVA. Mice were measured the serum levels of histamine and IgE, and observed histopathological changes of nasa mucosa H&E staining. Results : KOB and ARW significantly decreased the mortality and the serum levels of histamine in compound 48/80-induced anaphylatic rats. KOB and ARW also decreased the serum levels of histamine and IgE in OVA-induced AR mice, and inhibited histopathological changes of nasal mucosa with inflammation and the eosinophils infilteration. Conclusions : These data suggest that KOB has a strong anti-allergic effect through the inhibitory property of Astragali Radix, the main component of KOB against allergic responses in allergic rhinitis.
Objectives : In this study, we investigated the effect of Cassia obtusifolia Linne (Cassiae Semen; CS) extract on ovalbumin (OVA)-induced allergic asthma in mice. Methods : CR was extracted with 70% ethanol. For in vitro study, HMC-1, human mast cells were treated with CS extract at 0.2 and $0.5mg/m{\ell}$ for 1 h, and then stimulated with compound (C) 48/80 for 30 min. Primary spleenocytes were isolated from the spleen of mice, treated with CS extract for 1 h, and then stimulated with ConA for 24 h. For in vivo study, mice were sensitized at day 0, 7 and 14 with 0.2% OVA and then airway challenged using neublizer at day 21, 23, 25, and 27 to induced allergic asthma. CS extract at doses of 100 and 300 mg/kg body weight was orally administered during OVA challenge once per a day. The levels of allergic mediators such as histamine, OVA-specific IgE, IL-4, and $IFN-{\gamma}$ were measured in the sera of mice or culture supernatants by EIA and ELISA, respectively. The expression of IL-4 and $IFN-{\gamma}$ gene was determined by RT-PCR. The histopathological change of lung tissues was observed with hematoxylin and eosin (H&E) and Periodic acid Schiff (PAS) staining. Results : The treatment of CS extract in HMC-1 cells significantly inhibited C48/80-induced degranulation, and histamine release. The treatment of CS extract in spleenocytes suppressed the expression of IL-4 and $IFN-{\gamma}$ mRNA. The administration of CS extract in OVA-induced asthmatic mice significantly decreased the levels of OVA-specific IgE, and IL-4 in a dose-dependent manner with OVA-control group. In addition, CS extract inhibited the infiltration of inflammatory cells and bronchiolar damage with epithelial thickening in lung tissues of OVA-induced asthma mice, and also mucin accumulation. Conclusions : These results indicate that CS extract prevents asthmatic damage through regulating the allergic immune response.
Objective : Allergic rhinitis (AR) is an allergic inflammation of the nasal airways. Socheongryong-Tang traditionally has been used in Korea for treatment of allergy diseases. In the present study, we investigated whether SCRT suppresses the progression of AR in vivo and in vitro. Method : In this study, we investigated the effect of oral administration of SCRT on the Ovalbumin(OVA)-induced allergic rhinitis mouse models. Additionally, to find a possible explanation for the anti-inflammation effects of SCRT, we evaluated the inflammatory cytokine levels in LPS-stimulated mouse inflammation cell (RAW264.7) and mouse splenocyte. Results : The finding of this study demonstrated that SCRT reduced mast cells and inflammation cells infiltration in OVA-induced nasal cavity. Additionally, SCRT inhibited the production of inflammatory cytokines in splenocyte. However SCRT don't inhibited the production of inflammatory cytokines in RAW264.7 cells. Conclusion : Taken together, our results showed that oral administration of SCRT beneficial effects in allergic rhinitis, suggesting that SCRT might be a useful candidate for the treatment of allergic rhinitis.
Objectives : The root of Codonopsis pilosula (Fr.) Nannf. (Codonopsis Pilosulae Radix) has been traditionally used as a oriental medicine with an anti-thrombotic, antidiabetic, anticancer, and anti-gastric ulcer effects and immunological adjuvant. In this study, we investigated the effect of 70% ethanol extract of Codonopsis Pilosulae Radix (CPR-E) on ovalbumin (OVA)-induced allergic responses in mice. Methods : Mice were sensitized (1, 8, and 15 days) with OVA and airway challenged(22, 24, 26, 28, and 30 days) to induced allergic responses. CPR-E extract at doses of 50 and 100 mg/kg/body weight was orally administered from days 21 to 30 consecutively. The levels of allergic mediators such as histamine, OVA-specific immunoglobulin (Ig) E, and Th1/Th2 cytokines such as IFN-${\gamma}$ and IL-4 were measured in the sera of mice by ELISA. The histological change of lung tissue was observed by hematoxylin and eosin (H&E) staining. Results : CPR-E extract significantly decreased the serum levels of histamine, OVA-specific IgE, and IL-4 compared with those of OVA control group, but significantly increased the serum level of IFN-${\gamma}$. Based on H&E staining, CPR-E extract inhibited the infiltration of inflammatory cells into lung tissues with histological changes. Conclusions : These results indicate that CPR-E extract has anti-inflammatory and anti-allergic responses through regulating the cytokine balance, suggesting that the extract may be useful for the treatment of allergic inflammatory diseases such as bronchial asthma and allergic rhinitis.
Objectives : Allergy has been described as an inflammatory with hypersensitivity resulting from seasonal or perennial responses to specific allergens. The root of Platycodon grandiflorum (Jacq.) A. DC.(Platycodi Radix; Campanulaceae) has been traditionally used to treat chronic diseases such as bronchitis, asthma, pulmonary tuberculosis, inflammation and hyperlipidemia. In this study, we examined the effect of 70% ethanol extract of Platycodi Radix (PR-E) on ovalbumin (OVA)-induced airway inflammation in mice. Methods : Mice were sensitized and challenged by OVA inhalation to induced chronic airway inflammation, and then were intragastrically administered PR-E extract at doses of 50 and 200 mg/kg/day from days 21 to 30 consecutively. The levels of allergic mediators such as histamine, OVA-specific immunoglobulin (Ig) E, and cytokines such as IL-4 and IFN-${\gamma}$ were measured in the sera of mice by ELISA. The histological change of lung tissue was observed by hematoxylin and eosin (H&E) staining. Results : PR-E extract significantly decreased the serum levels of histamine, OVA-specific IgE, and Th2 cytokine, IL-4 compared with those in the OVA-induced group. PE-E extract significantly increased the serum level of Th1 cytokine, IFN-${\gamma}$. Based on lung histopathological studies, inflammatory cell infiltration and mucus hypersecretion were inhibited by PE-E extract administration compared to that in the OVA-induced group. Conclusions : These findings indicate that PE-E extract may be useful as an adjuvant therapy for the treatment of bronchial asthma.
Objectives In the past, studies have been shown that Forsythiae Fructus (FF), Ulmi Cortex (UC) and JwaGwiEum (JGE) are effective in treating allergic reactions. However, no report shows the difference in effects of FF, UC and JGE in allergic rhinitis (AR). Therefore, the purpose of this study was to differentiate the effects of FF, UC and JGE in AR induced by ovalbumin (OVA) in mice. Methods From this experiment, the effects of FF, UC and JGE were several - changes in body weight, hematologic changes such as WBC, RBC, Hb and PLT counts, immunological changes such as levels of histamine, IgE, IL-4 and expressions of iNOS & COX-2 mRNA. Moreover, histological change of nasal mucosa was also investigated. Results FF administration group and JGE administration group significantly inhibited level of IgE compared to the control group. Also, FF administration group, UC administration group and JGE administration group inhibited IL-4 and expressions of iNOS & COX-2 mRNA. In histological assessment, FF administration group, UC administration group and JGE administration group showed reduced amount of subepithelial edema, desquamated mucosa and hyperplasia of basement membrane from OVA.
Objectives : In this study, we investigated the effects of Agastachis Herba water (AH-W) extract on compound 48/80-induced mast cell degranulation and histamine release in human mast cells and also anti-asthmatic effect of AH-W extract on ovalbumin (OVA)-induced asthma in mice. Methods : Human mast cells, HMC-1 were treated with AH-W extract in the presence or absence of compound 48/80 (C48/80). Mast cell degranulation was observed by microscope, and the histamine release was measured in culture medium by ELISA. For preparation of asthmatic in vivo model, mice were sensitized (0, 7, and 14 days) with OVA and airway challenged (21, 23, 25, 27, and 29 days). AH-W extract at doses of 100 and 300 mg/kg/body weight was orally administered during OVA challenge once per a day. The levels of immunoglobulin (Ig) E, and Th1/Th2 cytokines, IFN-$\gamma$ and IL-4 were measured in the sera of mice by ELISA. The histopathological change of lung tissues was observed by hematoxylin and eosin (H&E) and Periodic Acid Schiff (PAS) staining. Results : The treatment of AH-W extract significantly decreased the mast cell degranulation and histamine release in C48/80-stimulated HMC-1 cells. In addition, The administration of AH-W extract at does of 100 and 300 mg/kg significantly decreased the serum levels of OVA-specific IgE compared with those of OVA control group. In H&E and PAS staining, AH-W extract inhibited OVA-induced airway inflammation, and inflammatory cells infiltration, and also histopathological damages on lung tissues such as bronchiole epithelial desquamation, goblet cells hyperplasia, and mucin releasing. Conclusions : These results indicate that AH-W extract may improve asthmatic symptoms through mast cell stabilization and inhibiting the lung inflammation in bronchial asthma.
