The study was carried out to establish in vitro microtuber production from leaf and petiole explant cultures of Pinellia ternata. Culture conditions were optimized by investigating the effect of plant growth regulators, different media, and gelling agents on the efficiency of microtuber indurtion. Among the different combinations of plant growth regulators tested, the combination of 0.1 mg/l 2,4-Dichlorophenoxyacetic (2,4-D) and 0.5 mg/l 6-benzylaminopurine (BAP) showed the highest yield fer microtuber production from leaf (3.9) and petiole (4.7) explant culture on MS medium far 6 weeks. SH(Schenk and Hildebrandt) medium was the most effective medium for microtuber induction and the half strength SH medium was better than SH medium for microtuber production from both leaf and petiole culture. Gelrite was better than agar in the formation of microtubers and 4% Gelrite showed the highest number of microtubers per explant frome leaf (5.9) and petiole (7.8) culture. Germination rate of microtubers after cold storaged for one months long was 86% from in vitro culture and 43% from autoclaved soil.
The influence of growth regulators (NAA and BA) and sucrose concentrations (0, 3, 5, 7, 9%) on in vitro rapid-propagation of virus-free sweet potato [Ipomoea batatas (L.) Lam.] was investigated with single-node or shoot-tip culture of two cultivars ('Matnami' and 'Shinhwangmi'). The survival rate and growth of shoot-tip explant was also investigated under the presence or absence of light (blue and red LED = 7:3, 150±5 μmol·m-2·s-1 PPFD) during minimal-growth in vitro conservation at 15℃. Vine length, vine diameter, fresh weight and dry weight were enhanced without callusing of explant in the MS medium supplemented with 0.2-0.5 mg·L-1 BA. The growth of single-node and shoot-tip explants were significantly enhanced with the increase of vine length, number of leaf, number of root, fresh weight, and dry weight in the solid medium containing 5% sucrose and 0.2 mg·L-1 BA. Vine elongation of shoot-tip explants were highest in the liquid medium containing 3% sucrose than the solid medium. The survival rate of minimal-growth in vitro conservation was 100% in 5 months under the presence of light (LED, 150±5 μmol·m-2·s-1 PPFD) at 15℃, but the explants in dark condition died in 3 months. The light was absolutely necessary for the in vitro conservation under minimal-growth conditions of virus-free sweet potato plantlets at 15℃, and the high density of explants (10 plantlets per Petri Dish) was increased the efficiency of mass conservation.
Shoot formation was investigated from in vitro cultivation of exotic hybrid poplar (Populus koreana ${\times}$ P. trichocarpa) with a specific stomatal character occurring both upper and lower surface of leaves. Two different explants (stem and leaf segment) of Peace poplar were cultured on half strength Murashige and Skoog (MS) basal medium supplemented with the various concentrations of thidiazuron as a plant growth regulator. Most adventitious shoots were produced from excised ends of stem or mid-veins of leaf segments. The highest average numbers of shoots were 7.1 and 5.3 with the treatments of 0.02mg/L TDZ in both explants of stem and leaf segment. The highest shooting rates were achieved to 83.3% and 47.6% with the concentrations of 0.01mg/L and 0.02mg/L TDZ by axillary bud and leaf cultures, respectively.
The twin-scale segments of nerine (Nerine bowdenii) were cultured to investigate the influence of NAA, BA and sucrose concentrations on in vitro bulblet formation. The formation of bulblets from twin-scale segments showed a good response both the percentage of bulblet formation and the number of bulblets per explant on MS medium supplemented with 1mg/L NAA and 2 mg/L BA. Formation of bulblet showed the highest efficiency on medium containing 30g/L, and the formation of bulblets was strongly inhibited on medium containing over 90g/L. When the twin-scale segments formed bulblets were subcultured three times to the same medium by 60 day subculture interval, the number of bulblets per explant was 6.5, 7.3 and 8.2 in order of first, second and third. The bulblets over 3mm in diameter were hypertrophied and rooted after transferring to the hormone-free MS medium. The plantlets over 50mm in height were successfully acclimatized in the soil mixed with the same volume of vermiculite and perlite, and the survival rate was over 95%.

지구온난화에 따라 농업부문 신재생에너지의 중요성이 증대되고 있으며, 화본과 식물은 바이오에너지작물의 중요한 소재를 제공하고 있다. 화본과 식물의 기내대량증식연구의 일환으로 홍띠식물의 기내 재생 식물체의 유전적 안정성에 대한 기초자료를 제공할 목적으로 기내배양으로 재분화시킨 홍띠(Imperata cylindrica 'Rubra') 재분화 식물체 중 녹색체 재생식물체를 대상으로 ISSR 표지를 사용하여 유전적 안정성을 조사하였다. 재분화식물체는 MS (Murashige and Skoog, 1962)배지에 생장조절제를 첨가한 배지에서 배양하였다. 생장점 부위를 적출하여 캘러스를 유도하고(0.1 mg/L 2,4-D와 2 mg/L BA), 캘러스 증식(0.1 mg/L 2,4-D와 0.05 mg/L BA), 신초 재분화( 0.01 mg/L NAA와 2 mg/L BA) 후 MS배지에서 식물체를 양성하고 순화시켰다. 배양은 26±2℃, 25 µmol/m2/s, 14h/10h (day/night) 광조건 하에서 실시하였다. 재분화식물체는 홍띠 및 녹색 재분화식물체 2 종류로 나타났는데, 이는 생장점에서는 홍띠가 분화되었음에도 불구하고 생장점 주변조직에서 유래한 녹새체가 분화된 후 우세하게 자라서 녹색재생체가 우점하는 것으로 추정된다. ISSR 분석은 대조구로 모식물체 홍띠를(8개체), 재분화식물체는 녹색체 중, 1년간 노지포장에서 재배중인 녹색체(10개체)와 실험실내 화분에서 재배중인 시료를(10개체) 사용하였다. ISSR 밴드패턴을 비교한 결과, 재분화체는 실내포트 재배식물체 10.3%, 노지1년 재배식물체 8.3%로 대조구의 4.1%보다 유전적 다형성 비율이 2배 이상 높게 나타났다. 또한 재분화식물체들의 유전적 유사도를 평가하고 군집분석을 실시하였다.
The 2,4-D + BA combination in MS medium showed high regenerating ability and induced compact callus from leaf and petiole segments of three strawberry cultivars, 'Bokyo', 'Suhong', whereas the NAA + BA combination resulted in friable callus. Band of RAPD products obtained with primer 212 from the callus was different from the band of mother plant when callus induced from leaf segment of 'Suhong' cultivar was maintained in MS medium containing NAA or BA for 8 months. The RAPD bands obtained from mother plants of 'Bokyo' and 'Yeobong' were different from that of callus maintained in the presence of MS liquid medium containing 2,4-D(0.2 mg/$\ell$) subcultured every two weeks for 6 months(12 subcultures).
This study was aimed to mutant induction in Gentiana scabra BUNGE through the tissue culture techniques and comparison of gentiopicroside content in each mutant plants derived from tissue culture. In order to induce the mutants of Gentiam scabra, seed, apical and lateral bud were soaked in $NaN_3$, EMS and MNU solutions and cultured on MS basal medium containing 0.5mg/l NAA, BA $NaN_3$, MNU-treated buds survived about 50% but not survived in EMS treatment. Seed germination rate was extremely low in EMS and MNU treatments but various types of mutant plantlets were obtained by those treatments. 63% of elongated type and 37% of dwarf type were obtained from tissue culture treated by various mutagen. Gentiopicroside contents of regenerated plantlets was analyzed. Root of dwarf type contained more gentiopicroside(1.383%) than that of elongation type (0.083%).

Series of in vitro experiments in Lilium callosum were conducted to investigate efficient multiplication through finding the optimal cultural environment, and organogenic capability of cultural explants, and then to determine the progressive method for enhancing bulblet growth in Lilium callosum scale culture. Twenty-four hr photoperiod was most effective for the growth of bulblet and the formation of other organs. Optimum light intensity for bulblet growth was 2,500~5,000 Lux. When bulbets were subcultured, growth of bulblets were enhanced by removing excessive leaf blade. Number of bulblets per scale increased as mother scale size increased, whereas diameter of bulblet from the small size mother scale increased. Bulblet formation and development was induced when explants were placed above the medium to be exposed to more light.
To measure the time required for ginseng explants to become determined to form flower buds, we cultured zygotic embryos, seedlings, and cotyledonary nodes on MS medium supplemented with BA and GA$_3$of 5 ${\mu}$M each (flower inducing medium, FIM) for various periods and transferred to the basal medium. The explants required a minimum of 10 days on FIM to be determined. Histological observations revealed that the axillary meristem to be fated to develop into flower bud remained in a state of shoot meristem during the first 10 days of culture and differentiated into flower bud after 15 days of culture. We suggest that the in-vitro flowering system described in this study is useful in investigating (a) regulatory element(s) to cause the phase change from the vegetative to reproductive state by comparing predetermined explants with determined ones at the molecular level.
The regeneration and growth of bulblets from the bulb scale segments of Hyacinthus orientalis L. cv. Pink Pearl were more efficient in IBA than IAA at the same concentrations (1.0 and 3.0 mg/l). The normal (base-down) orientation of explants was more effective for bulblet regeneration and root growth than the inverted (base-up) orientation. The growth of bulblets and roots was increased higher in the perlite than the agar medium. These results suggested that the alternate culture system, first cultured in the agar medium for bulblet regeneration, and then in the perlite medium for bulblet growth, may be more useful for efficient in vitro culture of hyacinth (H. orientalils) cv. Pink Pearl.
