The Molecular Phylogenetic Study of Filipendula (Rosaceae) (터리풀속(Filipendula)의 분자계통학적연구)

  • 터리풀속(Filipendula)은 장미과(Rosaceae), 장미아과(Rosoideae)에 속하는 다년생 초본이며, 북반구 온대지역의 산지지역에 서식하며 15-20여 종이 보고되어 있고, 이 중 10여종이 한국, 중국, 일본, 타이완 등의 동아시아 지역에 분포한다. 본 연구의 목적은 DNA 염기서열 자료를 이용하여 터리풀속(Filipendula)내 종들간의 계통관계를 규명하기 위하여 본 연구를 수행하였다. 이를 위해서 11종 29개체의 터리풀속(Filipendula)샘플과 외군인 산딸기나무속(Rubus)에 속하는 3종 5개체의 샘플을 이용하였다. 추가로 Genbank에서 3속 10종 18개의 염기서열을 다운받아 비교분석에 이용하였다. 계통연구를 위하여 엽록체에 존재하는 atpF-atpH, psbK-psbI, psbA-trnH, matK, rbcL, 5개 마커와 핵에 존재하는 ITS, 총 6개 마커의 염기서열을 생산하였다. 총 52개의 샘플에 대하여 엽록체유전체 5개 마커지역은 염기서열 길이가 3,485bp였고 핵 ITS지역은 631bp였으며, 이들을 합한 염기서열 길이는 4,116bp였다. 계통분석결과, 터리풀속(Filipendula)은 단계통군을 이루었다. F. occidentalis와 F. vulgaris가 기저분류군을 이루었고 이들은 각각의 아속에 해당한다. 그리고 나머지 종들은 모두 하나의 단계통군을 이루었다. 위의 결과들은 1961년 시미즈가 본 속을 Hypogyna아속, Filipendula아속, Ulmaria아속으로 나눈 분류시스템과 일치한다. 나아가 분자계통수에서 Ulmaria아속은 크게 4개의 subclade로 구분되었다. 먼저 subclade I에는 F. vestita, F. kiraishiensis, F. tsuguwoi, F. multijiuga, F. purpurea 등 5개 종으로 구성되었다. Subclade II는 F. ulmaria 한 종으로만 구성되었다. Subclade III에는 F. glaberrima, F. koreana, F. formosa, F.camtschatica 로 구성되었으며 subclade III에는 한국에 서식하는 3종이 포함되었다. Subclade IV에는 F. rubra, F. angustiloba, F. palmata, F. intermedia 4종으로 구성되었다. 이번연구에서는 Ulmaria아속내에 4개의 subclade가 존재함이 처음으로 확인되었다.

Taxonomic Review of the Umbelliferous genus Sium L. in Korea: Inferences based on Molecular Data (분자생물학적 자료에 의한 한국산 개발나물속의 분류학적 고찰)

  • The taxonomy of umbel genus Sium L., Apiaceae in Korea was reviewed on the basis of molecular phylogenies derived from sequences of nuclear ribosomal DNA internal transcribed spacer (ITS) regions. The ITS sequence-derived phylogeny indicates that S. heterophyllum, endemic to Korea, is identical to S. tenue, which is known as endemic to Ussuri regions. Comparisons of sequence pairs across both spacer regions gave divergence values and revealed the identity between S. tenue and S. heterophyllum on Mt. Moonsoo. On the other hand, the ITS sequences support species delimitation of S. ternifolium, as reported recently as a new species that differs from other Sium species. The ITS sequence divergence values of 1.4 and 1.6% support species delimitation between S. serra and S. ternifolium.

Molecular Phylogeny and Distribution of Far Eastern Oryzias latipes Based on Mitochondrial Cytochrome b Gene Sequence (미토콘드리아 cytochrome b 유전자 염기서열 분석에 의한 극동지역 송사리의 계통과 지리적 분포의 상관관계)

  • To examine the relationship of geological distribution and phylogenetic tree of O. latipes in the Far East, we analyzed cytochrome b (cyt b) gene in the mitochondrial genome. In this study we employed the entire sequence of cyt b of 53 samples collected from nine Korean locations and 117 cyt b data retrieved from the GenBank. From 170 Oryzias latipes cyt b sequence data, 142 different haplotypes were identified and phylogenetic relationship was reconstructed based on the dataset. According to the phylogeny, haplotypes were divided into three major haplogroups A, B and C, and their relationships were well correlated to their distributional patterns. Haplogroup A which is widely distribute in the southern part of Korea is separated in the geographical distribution from the haplogroup B which is found from China to the western part of Korea. Haplogroup C is only found in Japan.

Morphological Characteristics of the Blue Trevally, Carangoides ferdau (Perciformes: Carangidae) and its Phylogenetic Relationships among Korean Relatives (흑전갱이, Carangoides ferdau의 형태적 특징 및 분자계통분류학적 위치)

  • As Carangoides ferdau was previously reported based on its underwater photograph, morphological descriptions have been incomplete up to the presence in Korea. On the base of two samples collected at the coast of Jeju island, morphological characters of C. ferdau are described in detail. This species is characterized by having the forepart of second dorsal fin much prolonged, 7~8 transverse dark bands on body, and snout length almost equal to eye diameter. It is morphologically very similar to C. orthogroammus, but is easily distinguished in having transverse dark bands instead of yellow spot on the body of C. orthogroammus. Phylogenetic relationships based on the mitochondrial cytochrome b (1,141 base pairs) sequences shows that C. ferdau is closely related to C. orthogroammus, and C. dinema also has a sister group relationship with C. ablongus. Both genetic distances (p-distances) are 8.2%, respectively.

Identification of Sphaerulina azaleae on Korean Azalea in Korea Based on Morphological Characteristics and Multilocus Sequence Typing (형태적 특징 및 다좌위 염기서열 분석에 의한 산철쭉 모무늬병균 Sphaerulina azaleae 동정)

  • From 2008 to 2017, Korean azalea (Rhododendron yedoense f. poukhanense) showing angular, necrotic leaf spots were found in Jeju and Hongcheon, Korea. The lesions occurred frequently, detracting from the beauty of the glossy green leaves of the plant and causing premature defoliation. Therefore, to identify the fungus associated with the lesions, morphological characterization and molecular phylogenetic analysis of actin (Act), translation elongation factor 1-alpha (EF), internal transcribed spacer (ITS), 28S nrDNA (LSU), and RNA polymerase II encoding the second largest subunit (RPB2) of the two representative isolates were performed. The phylogenetic tree inferred from the neighbor-joining method showed the isolates clustering in the Sphaerulina azaleae group. Therefore, the fungus associated with the angular leaf spots on the Korean azalea was identified as Sphaerulina azaleae.

Antibacterial Activity against Salmonella enteritidis JK-15 and LPS Changes Caused by Rose Flower Extracts (장미꽃 추출물에 의한 식중독 세균 Salmonella enteritidis JK-15에 대한 살균활성 및 그에 따른 LPS 변화)

  • The aim of this work was to investigate the antibacterial effect of the food-poisoning bacterium, Salmonella enteritidis JK-15 exposed to rose extracts. Initially, the isolate S. enteritidis JK-15 was enriched and isolated from stale food. BIOLOG and 16S rRNA analyses revealed that strain S. enteritidis JK-15 was 98% similar to the S. enteritidis species cluster; therefore we have designated this strain as S. enteritidis JK-15. Bactericidal effects of S. enteritidis JK-15 exposed to rose extracts ranging from 5 mg/ml to 100 mg/ml were monitored, and complete bactericidal effects were achieved within 6 h at 100 mg/ml and 12 h at 50 mg/ml, respectively. SDSPAGE with silver staining revealed that the amount of lipopolysaccharides increased or decreased in the strain S. enteritidis JK-15 treated to different concentrations and exposing periods of rose extracts in exponentially growing cultures. Scanning electron microscopic analysis, demonstrated the presence of irregular rod shapes with umbilicated surfaces for cells treated with rose extracts.

Screening of Antagonistic Bacteria having Antifungal Activity against Various Phytopathogens (다양한 식물병원성 곰팡이에 항진균 활성을 갖는 길항미생물의 탐색)

  • The aim of this study was to isolate a potential multifunctional biocontrol agent from bacteria for control of multiple plant diseases as an alternative to fungicides. A total of 201 strains were isolated from soil undamaged by repeated cultivation in Sunchang and their ability to produce antibiotics, siderophores and extracellular enzymes such as protease, cellulase and amylase was investigated. Selected strain SCS3 produced cellulose, protease and amylase. This strain also produced siderophores and showed excellent antifungal activity against various phytopathogens. SCS3 was identified as Bacillus subtilis using 16S rRNA sequencing, and named Bacillus subtilis SCS3. Finally, physiological and biochemical characteristics of B. subtilis SCS3 were examined. From the results, B. subtilis SCS3 was found to be a useful multifunctional biocontrol agent against various phytopathogens.

Phylogenetic Relationships of Morus Species on the Basis of RAPD (RAPD를 이용한 뽕나무속 식물의 유전적 유연관계 분석)

  • Phylogenetic relationships among mulberry varieties (Morus species) were analyzed on the basis of RAPD in order to identify the possibility of classification for the species. Polymorphisms under RAPD method were compared among 41 mulberry varieties. From the results of RAPD for 41 mulberry varieties by use of 30 different primers, 151 polymorphic bands were formed out of 201 ones. Under the dendrogram based on cluster analysis with the polymorphic bands, the varieties were classified into 7 groups including two large and five small ones on 0.747 value of genetic similarity coefficient. In the large groups 19 and 16 varieties were belong to group I and III, respectively. On the other hand, relatively high genetic similarity was shown among the varieties belonging to the group I, II and III. While, relatively low similarity were done between them in the group IV-Ⅵ and the other groups, and Mohusang in the group Ⅶ showed the lowest phylogenetic relationship with the other varieties.

  • The movement of vesicles from the neuronal cell body to specific destinations requires molecular motors. The squid giant axon represents a powerful model for studies of the axonal transport mechanism because the axoplasm can readily be separated from the sheath by simple extrusion. In a previous study, vesicular movements in the axoplasm of the squid giant axon were inhibited by the kinesin antibody. In the present study, we cloned and sequenced the cDNAs for squid brain KIFs. Amplification of the conserved nucleotide sequences of the motor domain by polymerase chain reaction (PCR) using first-strand cDNAs of the squid optic lobe identified six new KIF proteins. Motif analysis of the motor domains revealed that the squid KIFs are homologous to the consensus sequences of the mouse KIFs. The phylogenetic tree generated by using the maximum parsimony (MP) method, the neighbor-joining (NJ) method, the minimum evolution (ME) method, and the maximum likelihood (ML) method showed that squid KIFs are closest to mouse KIFs. These data prove the phylogenetic relationships between squid KIFs and mouse ones.

Characterization of Pyrenophora avenae Isolated from Discolored Black Oat Seeds in Korea (귀리 흑변 종자에서 분리된 Pyrenophora avenae의 특성)

  • In January 2017, discolored black oat seeds were found in the storage depot of a farmhouse in Jeongeup. Pyrenophora sp. was detected in 45% of the oat seeds surveyed. All Pyrenophora isolates obtained from the seeds were identified as Pyrenophora avenae based on the sequences of internal transcribed spacer (ITS) rDNA regions and glyceraldehyde 3-phosphate dehydrogenase (GPDH) gene and validated by morphological and cultural characterization. A phylogenetic tree constructed using the ITS and GPDH sequences showed that the Korean isolates of P. avenae comprise of four genetically distinct groups. Pathogenicity test validated that the fungus is an infectious agent responsible for discolored black seeds and leaf blotch in oat plants. This is the first study report that P. avenae causes leaf blotch disease of oat in Korea.
