Effects of Chongkukjang Intake on Lipid Metabolism and Liver Function in Alcoholic Fatty Liver Rats (청국장식이가 알코올성지방간 쥐의 지질대사 및 간 기능개선에 미치는 영향)

  • This study was performed to investigate the effect of chongkukjang intake on lipid metabolism and liver function in alcoholic fatty liver rats. Thirty-five 7-weeks old Spargue-Dawley male rats were used as experimental animals. After inducing alcoholic fatty liver, rats were divided into two groups and fed ethanol+casein diet (ECD) or ethanol+chongkukjang diet (EChD). At 10th, 20th and 30th days of the feeding experimental diet, rats were sacrificed to get blood and liver samples for analysis of blood lipids, lipid peroxides, antioxidative enzymes and biochemical indices of liver function. The mean food intake was not significantly different between ECD and EChD groups. Daily weight again of EChD group was significantly higher than that of ECD group at days 20 and 30. Serum total lipid, triglyceride and total cholesterol of ECD group were significantly higher than those of EChD group, while HDL-cholesterol was significantly higher in EChD group. Liver TBARS level of ECD group was significantly higher than that of EChD group. However, liver conjugated diene level was significantly higher in ECD group only at day 10. SOD, CAT and GPx activities of EChD group were significantly higher than those of ECD group at days 20 and 30. In the indices of liver function, GOT and GPT of ECD group were significantly higher than those of EChD group at day 10. LDH was significantly higher in ECD group. γ-GTP was significantly higher in ECD group only at day 20. Serum alcohol concentration of ECD group was significantly higher than that of EChD group at day 30. ADH and ALDH activities of EChD group were significantly higher than those of ECD group at day 30. Therefore, chongkukjang intake seems to give a beneficial effect on improving lipid metabolism and liver function by increasing HDL-cholesterol level, antioxidative enzyme activites, alcohol enzyme activities and decreasing serum lipids, liver TBARS and conjugated diene.

Effects of Kimchi Lactic Acid Bacteria Lactobacillus sp. OPK2-59 with High GABA Producing Capacity on Liver Function Improvement (GABA 생성능 우수 김치 젖산균 Lactobacillus sp. OPK2-59의 간 기능 개선 효과)

  • This study investigated the effect of improved liver function in rats administered with ethanol by kimchi lactic acid bacteria with high GABA producing capacity. Sprague-Dawley male rats were divided into four groups; normal diet control (NC), ethanol control (EC), ethanol+Lactobacillus sp. OPK2-59 normal powder (EL1), ethanol+Lactobacillus sp. OPK2-59 GABA powder (EL2) and fed for 6 weeks. Analysis showed that there were no significant differences in body weight and feed consumption among the groups during the experimental period. Also, there were no significant differences in organ weight among the groups. The test results showed total cholesterol and triglyceride in the blood concentration that were increased by ethanol administration were significantly lowered in EL2 group. Liver triglyceride was also significantly lowered in the EL2 group compared with the EC group. Serum GOT and GPT, and liver GOT levels were significantly lower in the EL2 group compared with the EC group. Serum ethanol concentration was lower in the EL1 and EL2 groups compared with the EC group. SOD activities in liver were significantly increased in the EL1 and EL2 groups compared with the EC group. These results suggest that Lactobacillus sp. OPK2-59 GABA powder improves lipid and enzyme profiles of rats administered with ethanol.

Association Between Liver Enzyme and Risk of All-Cause Mortality: Use of Korean Genome and Epidemiology Study (KoGES) Data (간 효소(AST, ALT)와 전체원인사망 위험의 관련성: 한국인유전체역학조사 자료 활용)

  • This study was conducted to investigate the association of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) with all-cause mortality among populations. The data used were from a Korean Genome and Epidemiology Study (KoGES) based on health examinations and questionnaires. The subjects consisted of 10,110 persons aged 40 and over. Hazard ratio was analyzed using Cox's proportional hazard model. The hazard ratio of AST (${\geq}50.0\;IU/L$) was 2.198 (95% CI: 1.217-3.971) after being adjusted for age, sex, education, regular exercise, smoking, drinking, WHR, and TG. In conclusion, AST was an independent significant risk factor of all-cause mortality, and ALT showed a tendency to increase. Overall, these findings indicate that AST and ALT may be useful tools for predicting mortality.

Effect of Styrene on Hepatic Activities of Antioxidant Enzymes in Rats (스티렌이 흰쥐의 간 조직 중 항산화계 효소 활성에 미치는 영향)

  • Styrene is a commercially important chemical used mainly in the production of raw materials and plastics. To determine the effect of styrene on hepatic activities of antioxidant enzymes, styrene was treated to Sprague-Dawley rats at 50 mg/kg, 200 mg/kg and 400 mg/kg (i.p) twice a day for 4 days. There were determined the significantly increased activities of serum AST (aspartate aminotransferase), ALT (alanine aminotransferse), and the increased content of MDA (malondialdehyde) at the dose of 400 mg/kg compared to the control. The hepatic activities of XO (xanthine oxidase) and CYPdAH (cytochrome P450 dependant aniline oxidase) in the dose of 400 mg/kg compared to the dose of 200 mg/kg were more increased, which means the excessive ROS (reactive oxygen species)s were produced during Phase I. In addition, significantly decreased were rates of the hepatic activities of GPx (glutathione peroxidase), CAT (catalase), SOD (superoxide dismutase) and GST (glutathione S-transferase) at the dose of 400 mg/kg compared to the control. And, the group at the dose of 400 mg/kg showed more significantly decreased GSH (glutathione) level than the group at the dose of 200 mg/kg. The decrease in GSH could ascribe to the toxic metabolites of styrene, such as styrene oxide. In conclusion, these results indicate that the excessive ROSs and the toxic metabolites of styrene may result in the hepatotoxicity, and be related to their imbalanced activities for antioxidant enzymes.

Protective Effect of Eriobotrya japonica Lindl. on Hepatotoxicity by Carbon Tetrachloride (비파식초의 사염화탄소에 의한 간독성 보호 효과)

  • This study was carried out to investigate the effect of natural Eriobotrya japonica Lindl. vinegar on the liver protective effect of animals exposed to carbon tetrachloride. Eriobotrya japonica Lindl. vinegar (200 mg/kg) was administered at the same time for 28 days, and hepatotoxicity was induced by intraperitoneal injection of carbon tetrachloride on the $29^{th}$ day. The aspartate aminotransferase and alanine aminotransferase levels were significantly decreased (p < 0.001) and the superoxide dismutase and catalase activities were significantly increased (p < 0.001) in the Eriobotrya japonica Lindl. vinegar group compared to the control group. Histopathological observations showed that the Eriobotrya japonica Lindl. vinegar showed hepatic cell structure similar to normal group, and these results showed that it had an effect of suppressing and protecting the damage of liver cell. Therefore, Eriobotrya japonical Lindl. vinegar is considered to be a healthy functional food of the liver.

Purification and Characterization of Glutathione Peroxidase Isolated from Rat Erythrocyte and Histochemical Study of its Localization in Liver of White Rat (흰쥐 적혈구에 있는 Glutathione Peroxidase의 순화 및 성질과 간에서의 용작부위에 대한 조직화학적 연구)

  • A glutathione peroxidase from white rat (Wistar strain)erythrocytes was partially purified and characterized. In addition, localization of this enzyme in the liver was studied by histochemical method. A glutathione peroxidase was purified approximately 33.5-folds by ammonium sulfate precipitation, Sephadex filtration column and DEAE-Sephadex column chromatography. The optimum temperature of the crude glutathione peroxidase was $40^\\circC$, and the optimum pH was 7.5. This crude glutathione peroxidase was most stable at $30^\\circC$ and the values of Km and Vmax were calculated to be 8.5mM and 15.6 $\\mu$moles/min for glutathione, and 40 $\\mu$M and 10.5 $\\mu$moles/min for hydrogen peroxide, respectively. The molecular weight of this enzyme was estimated by Sephadex G-200 gel filtration to be approximately 90, 000. By electron microscopic examination, histochemical reaction products were microbodies that were prominent in the peripheral parts of the lobule. The reaction products exhibited round shapes, the diameter of which varied $0.2\\sim0.7 \\muM$ and their boundary membranes were not distint.

Indole Acetic Acid Production of Rice Paddy Soils (논토양의 Indole Acetic Acid 생성능)

  • This study was conducted to evaluate the indole acetic acid (IAA) formation in soils as a biological indicator for the health of rice paddy soils with control, nitrogen sole, chemical fertilizer (NPK), and chemical fertilizer plus compost (CNPK) plots. There was a positive relationship between colorimetric method and high performance liquid chromatography for IAA in soils determined, and the values were similar between two methods, as $0.83{\sim}1.23{\mu}g\;IAA\;g^{-1}h^{-1}$ in colorimetric method, $0.80{\sim}1.29{\mu}g\;IAA\;g^{-1}h^{-1}$ in HPLC method. Numbers of dehydrogenase-producing bacteria and the IAA production in soils were high in NPK and CNPK plots comparing with control and nitrogen sole plots. Also there was high correlation between numbers of dehydrogenase-producing bacteria and IAA production in soils.

Drosophila robusta species group 2종 (D.lacertosa 와 D.sordidula)의 mtDNA 변이에 의한 종분화정도

  • Drosophila lacertosa and D. sordidula are members of the robusta species group in virilis section of Drosophila. The mtDNA of both species was analyzed, using 10 restriction endonucleases. The mtDNA genome size of D. lacertosa and D. sordidula was 15.7 kbp, altogether, and the numbers of mtDNA fragment were 26 and 32, respectively. Restriction cleavage map of mtDNA in these species was constructed. The patterns of cleavage map were very different between two species and it means that speciation was taken for a long time ago.

Effects of Rice Embryo and Embryo Jelly with Black Rice Bran Pigment on Lipid Metabolism and Antioxidant Enzyme Activity in High Cholesterol-Fed Rats (쌀 배아와 흑미 미강 색소 첨가 배아젤리가 고콜레스테를 식이 흰쥐의 지질대사와 항산화효소 활성에 미치는 영향)

  • We investigated the effect of rice embryo and embryo jelly with black rice bran pigment on lipid metabolism and antioxidant activity. Thirty 4-week-old male Sprague-Dawley rats were fed high cholesterol diets supplemented with 15% rice embryo and 25% embryo jelly added black rice bran pigment, respectively, for 6 weeks. Plasma and hepatic lipid profile, lipid peroxidation, and the activity of antioxidant scavenger enzymes in liver were examined. Supplementation with rice embryo and embryo jelly had no effect on food intakes in high cholesterol-fed rats. The plasma triglyceride concentration was not significantly different among the groups. Supplementation with rice embryo and embryo jelly resulted in lower plasma and hepatic total cholesterol (TC) concentration and high-density lipoprotein-cholesterol (HDL-C)/TC ratio and atherogenic index compared to the control group, while the plasma HDL-C concentration tended to elevated. Rice embryo and embryo jelly tended to lower plasma and hepatic levels of thiobarbituric acid reactive substances than the control group. Moreover, hepatic antioxidant enzyme activities, including superoxide dismutase and glutathione peroxidase, were significantly higher in the rice embryo and embryo jelly groups. In conclusion, rice embryo and embryo jelly was very effective in improving the lipid metabolism and reducing oxidative stress by up-regulating the hepatic antioxidant enzymes in high cholesterol-fed rats.

뱀장어속 어류 2종의 동위효소 및 mtDNA 분석

  • 뱀장어속(Anguilla)의 뱀장어(A. ioponica)와 무태장어(A. marmoruta) 및 꾀붕장어(Anago anogo)의 유전적 특징과 종간 유연관계를 밝히기 위하여 isozyme 분석과 mtDNA 분석을 실시하였다. Isozyme 분석결과 20개의 효소 및 비효소단백질에서 총 39개의 유전자를 검출하였고 각종 특유의 genetic marker를 확인하였다. 3종의 평균 유전적 변이는 뱀장어가 HD=0.057. HG=0.065, 무태장어는 HD=0.067 HG=0.053, 체불장어가 HD=0.018. HG=0.020으로 각각 나타났다. 뱀장어와 무태장어의 평균 종간유전적 근인관계는 S=0.420 (D=0.869)로 멀게 나타났다. 6 base를 인지하는 10종류의 제한효소를 처리한 결과 mtDNA 절편양상은 각 종내에서 동일하게 나타났으나 각종간에 차이를 보였고, 종간 평균염기 치 환율은 뱀장어와 무태장어가 p=3.4%, 뱀장어속 2종과 꾀붕장어는 p=9.6%로 나타났다.
