The purpose of this study was to compare the cytotoxicity by MTT test and genotoxicity by Ames test of new calcium phosphate-based root canal sealers (CAPSEAL I, CAPSEAL II) with commercially available resin-based sealers (AH 26, AH Plus) , zinc oxide eugenol-based sealers (Tubliseal EWT, Pulp Canal Sealer EWT), calcium hydroxide-based sealer (Sealapex), and tricalcium phosphate based sealers (Sankin Apatite Root Canal Sealer I, II, III). According to this study, the results were as follows : 1. The extracts of freshly mixed group showed higher toxicity than those of 24 h set group in MTT assay (p<0.001). 2. CAPSEAL I and CAPSEAL II were less cytotoxic than AH 26, AH Plus, Tubliseal EWT, Pulp Canal Sealer EWT Sealapex and SARCS II in freshly mixed group (p<0.01). 3. AH 26 in freshly mixed group showed mutagenicity to TA98 and TA100 with and without S9 mix and AH Plus extracts also were mutagenic to TA100 with and without S9 mix. 4. Tubliseal EWT, Pulp Canal Sealer EWT and Sealapex in freshly mixed group were mutagenic to TA100 with S9 mix. 5. Among those of 24 h set groups the extracts of SARCS II were mutagenic to TA98 with and without S9 mix and AH 26 showed mutagenic effects to TA98 with S9 mix. 6. No mutagenic effect of CAPSEAL I and CAPSEAL II was detected. 7. There is no statistically significant difference between CAPSEAL I and CAPSEAL II at MTT assay and Ames test in both freshly mixed group and 24 h set group.
This study was conducted to examine the stages showing the antimutagenic effects on the microbial mutation by addition of the juice extracted from small water dropwort. It was not able to find out the signal showing the genic derepression or change of gene repair system by addition of the juice. And it was hardly possible to expect the conversion of 2-AF to inactive form by the juice. however the longer 2-AF and S-9 mix were contacted before addition of the juice, the stronger the microbial mutagenisity of 2-AF was, and after addition of the juice, the mutagenicity was decreased rapidly. It seems that some components in the juice act as inhibitor of a enzyme in S-9 mix, and block the conversion of 2-AF to the ultimate mutagen.
Objectives : This study was to assessment the toxicity of Sipjeondaebo-tang(Shiquan dabu-decoction) by Chromosomalanomaly test. Methods : Sipjeondaebo-tang(Shiquan dabu-decoction) water-extract in vivo Chromosomalanomaly test was performed using chiness hamster lung cell line. Results : Sipjeondaebo-tang water extract was negative in Chromosomalanomaly test at the doses of 0, 625, 1250 and $2500{\mu}g/m\ell$ at 6h and 24h.(S9- fraction). Chromosomalanomaly test(S+fraction) was also negative at the doses of 0, 1250, 2500 and $5000{\mu}g/m\ell$. Conclusions : It was concluded that Sipjeondaebo-tang extract did not induce Chromosomalanomaly in the chiness hamster lung cell.

Sophorae radix extract (SRE) has been registered as an environment-friendly organic material that is widely used in the cultivation of crops in Korea. Matrine, the active ingredient in SRE, was reported as a toxic substance in the nervous system in mice. However, no information is available on its toxic effects in other organisms. Therefore, antimutagenicity and two kinds of genotoxicity tests (bacterial reverse mutation and chromosome aberration test) of two samples of SRE were investigated in this study. Antimutagenicity test was experimented by using bacterial reverse mutation test. In the reverse mutation test, Salmonella Typhimurim TA98, TA1535 and TA1537 were used to evaluate the mutagenic potential of SRE. Bacterial reverse mutation test was also performed on positive and negative control groups in the presence of the metabolic activation system (with S-9 mix) and metabolic non-activation system (without S-9 mix). In the chromosome aberration test, Chinese hamster lung cells were exposed to SRE for 6 or 24 hours without S-9 mix, or for 6 hours with S-9 mix. Negative and positive control groups were experimented for chromosome aberration test. As a result, the number of mutated colonies induced by 4-NQO were reduced by SRE treatment in all strains, indicating that SRE may have antimutagenic effects. Reverse mutation was not shown at all concentrations of SRE, regardless of application of the metabolic activation system. In the chromosomal aberration test, one of the SRE sample gave a suspicious positive result at 250 ${\mu}g/ml$ in the presence of S-9 mix. For the more adequate evaluation of the genotoxic potential of SRE samples, other in vivo genotoxicity study is needed.
Gardenia yellow pigment produced by Gardenia jasminoides Ellis was tested for reverse mutagenic test in Salmonella typhimurium stains TA1535, TA1537, TA98 and TA100 at concentrations raging form 6.25 to 200$\mu\textrm{g}$/$m\ell$ per plate. No significant reverse mutagenic activity was observed in any of the S. typhimurium strains, in either presence or absence of S9 mix. There was no toxicity to the bacteria. These result indicate that yellow pigment doesn't have mutagenicity.
The purpose of this study was to call attention to the mental, physical and occupational hazards of the anticancer-drug-handling nurses by examining the possible urinary mutagenicity and measuring physical symptoms and stress level of the nurses exposed to anticancer drugs. The experimental group of the urinary mutagenicity assay was 14 nurses handling anticancer drugs at the medical wards of a hospital located in J city ; the control group was 12 psychiatric nurses of the same hospital. The test material was the nurses' 24hrs urine, which was concentrated by XAD-2 column chromatography. Tester strains were TA98(±S9 mix), TA100(±S9 mix), TA1535(±S9 mix) and TA1537(±S9 mix) ; Salmonella mammalian-microsomal test(Ames test) was employed for the urinary mutagenicity assay. The physical symptoms of which the nurses experienced were investigated through self-reports on open-questionnaires. The stress levels of the experimental group were measured by a stress measuring instrument developed by this author. Reliability of this instrument was found to be adequate (Cronbach's Alpha=0.9079). To ascertain the urinary mutagenicity of the experimental group, the mean and the standard deviation of the colonies of Tester strains appearing on the minimal plates were taken and compared differences between two groups. T-test was employed for the significance test of two groups. The physical symptoms were compared between the two groups through the analysis of the nurse' self-reports. The mean and standard deviation of the stress levels of the experimental group were also calculated and were examined through t-test. The results were summarized as follows : 1. The experimental group revealed significantly higher urinary mutagenicity both in the activation method test and the non-activation method test of the tester strains TA98, TA100 and TA1535. In the case of TA1537, two groups showed no difference in the non-activation method test, but the activation method revealed difference. 2. The physical symptoms were also much more frequently reported in the experimental group. 79.3% of the experimental group reported more than 1 kind of physical symptoms. On the other hand, 33.2% of the control group complained of 1 kind of physical symptom. The items with high symptom frequency were 'headache', 'itching sensation', 'corneal congestion', 'skin allergy' 3. The mean score of stress in the experimental group was 2.41(range 1-4). The experimental group showed the stress level above 2.0 in the 14 of 15 items in all. The highest stress level were recorded in the following items in the order quoted, 'I fear that anticancer drug may touch any part of body while handling it.', 'I feel concerned there is no protective countermeasure against anticancer drug handling.', 'I am afraid the anticancer drug handling may produce a fetal loss in the future'.
Pork was cooked using three kinds of instrument [electric pill (EG) for 5min., microwave oven (MW) for 6min. and reheated using a MW] and then extracted with $80\%$ methanol. The Ames test was performed on the methanol extracts, employing the S. typhimurium tester strain, TA100. The methanol extract of cooked pork showed high mutagenicity ion the 5.0 mg/plate without the S9 mix, but a higher mutagenicity was induced with the S9 mix With increasing refrigeration $(4^{\circ}C)$ and freezing $(-18^{\circ}C)$ periods the extracts showed higher mutagenicities and TBA values, and the same results where shown with reheating. Correlations of the mutagenicity (-S9 mix) and rancidity of the pork cooked by EG, according to storage at and $-18^{\circ}C$ and reheated by MW (1 min), were r=0.85, 0.86, 0.98 and 0.83, respectively. When the MW was used for reheating, the refrigeration storage (r=0.98) showed a higher correlation coefficient than for that stored frozen (r=0.83). From the structure of cooked pork, as observed by SEM, many vapor pathways were viewed in the pork reheated using themicrowave oven.

This study was performed to investigate the ameliorating effect of a mixture of extracts from Houttuynia cordata Thunb, Nelumbo nucifera G. (leaf), and Camellia sinensis (seed) (MIX) on acute ethanol-induced hangover in Sprague-Dawley rats. Plasma ethanol and acetaldehyde levels in MIX-treated rats significantly decreased at 3 h and 5 h after acute ethanol administration (25%, 3 g/kg body weight/d) as compared to ethanol-treated rats. Hepatic alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities were significantly higher in MIX-treated rats than in ethanol-treated rats. MIX exhibited high ADH and ALDH activities on direct assays using S9 rat liver fraction for ethanol metabolic enzymes clearance action. These results suggest that MIX could alleviate ethanol-induced hangover symptoms by elevating activities related to hepatic ethanol-metabolizing enzymes against ethanol induced metabolites, and MIX should be further developed to be a new anti-hangover material.
The present investigation is to identify an optimum mix combination amongst 28 different types of artificial lightweight aggregates by pelletization method with aggregate properties. Artificial aggregates with different combinations were manufactured from fly ash, cement, hydrated lime, ground granulated blast furnace slag (GGBFS), silica fume, metakaolin, sodium bentonite and calcium bentonite, at a standard 17 minutes pelletization time, with 28% of water content on a weight basis. Further, the artificial aggregates were air-dried for 24 hours, followed by hardening through the cold-bonding (water curing) process for 28 days and then testing with different physical and mechanical properties. The results found the lowest impact strength value of 16.5% with a cement-hydrated lime (FCH) mix combination. Moreover, the lowest water absorption of 16.5% and highest individual pellet crushing strength of 36.7 MPa for 12 mm aggregate with a hydrated lime-GGBFS (FHG) mix combination. The results, attained from different binder materials, could be helpful for manufacturing high strength artificial aggregates.
The purpose of this study was to measure thermal expansions of dental investments, Biovest(Casting Investment. Dentsply International INC, U.S.A.), Multi-Best (Use for all dental chrome-cobalt alloys, The Ransom & Randolph Co. U.S.A.), Kerr(Inlay Investment. Sybron Kerr, U.S.A.), O. K. (Inlay Investment. Shofu Dental MFG, Co. Japan), Whip-Mix (Cristobalite Inlay Investment. Whip-Mix Corporation. U.S.A.). Thermal expansion of specimens(5mm in diameter and 50mm in length) was measured by a dilatometer at the temperature range from $20^{\circ}C$ to $700^{\circ}C$ by comparing expansion between standardized quartz and experimental specimens with heating rate about $300^{\circ}C$/hr. The following results were obtained. 1. The coefficient of thermal expansion of Biovest was $15{\times}10^{-6}/^{\circ}C$ in the water powder ratio 18/100 and $14{\times}10^{-6}/^{\circ}C$ in the water powder ratio 28/100. Those of Multi-Best were $9{\times}10^{-6}/^{\circ}C$ in the water powder ratio 14/100 and $7{\times}10^{-6}/^{\circ}C$ in the water powder ratio 24/100. 2. The coefficient of thermal expansion of Kerr were $17{\times}10^{-6}/^{\circ}C$ in the water powder ratio 38/100 and $14{\times}10^{-6}/^{\circ}c$ in the water powder ratio 48/100. Those of O. K. were $9{\times}10^{-6}/^{\circ}C$ in the water powder ratio 33/100 and $7{\times}10^{-6}/^{\circ}C$ in the water powder ratio 43/100 3. The coefficient of thermal expansion of Whip-Mix were $14{\times}10^{-6}/^{\circ}C$ in the water powder ritio 40/100 and $12{\times}10^{-6}/^{\circ}c$ Fein the water powder ratio 50/100. Those of Hi-Heat were $11{\times}10^{-6}/^{\circ}c$ in the water powder ratio 28/100 and $10{\times}10^{-6}/^{\circ}c$ in the water powder ratio 38/100.
