탕포산(湯泡散)이 삼종(三種)의 각막염(角膜炎) 유발균들에 미치는 영향 (The Experimental Studies on Anti-bacterial Effect of Tangpo-san on Three species of causative bacteria of Keratitis)

  • This experimental study was performed to investigate the effect of herbal eye drops, Tangpo-san and Coptidis rhizoma on Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa keratitis. The following results were obtained by using Minimum inhibition Concentration(MIC) and inhibition Zone. 1. MIC on Staphylococcus aureus in Tangpo-san was 100%, in Coptidis rhizoma was 100% and in Cravit was 0.1% 2. MIC on Staphylococcus epidermidis in Tangpo-san was 100%, in Coptidis rhizoma was 10% and in Cravit was 0.1%. 3. MIC on Pseudomonas aeruginosa in Tangpo-san, Coptidis rhizoma was not showing and in Cravit was 0.1%. 4. The size of inhibition zone on Staphylococcus aureus for Tangpo-san was 13.3mm in $50{\mu}{\ell}$, for Coptidis rhizoma was 26mm in $50{\mu}{\ell}$ and for Cravit was 31mm in $50{\mu}{\ell}$, showing the highest antibacterial effect. 5. The size of inhibition zone on Staphylococcus epidermidis for Tangpo-san was 16mm in $50{\mu}{\ell}$, for Coptidis rhizoma was 25mm in $40{\mu}{\ell}$ and for Cravit was 34mm in $50{\mu}{\ell}$, showing the highest antibacterial effect. 6. The size of inhibition zone on Pseudomonas aeruginosa for Tangpo-san, Coptidis rhizoma was not and for Cravit was 24.7mm in $50{\mu}l$, showing the antibacterial effect. In addition, the results shows that the herbal eye drops, Tangpo-san and Coptidis rhizoma can be used to cure Staphylococcus aureus, Staphylococcus epidermidis keratitis and if further study is performed, the use of herbal eye drops will be valuable and beneficial in the clinical medicines.

In Vitro Antifungal Activities of Amphotericin B, Fluconazole, Itraconazole, Terbinafine, Caspofungin, Voriconazole, and Posaconazole against 30 Clinical Isolates of Cryptococcus neoformans var. neoformancs

  • Aantifungal agents were tested against 30 clinical isolates of Cryptococcus neoformans var. neoformans using the NCCLS method(M27-A2). Posaconazole, itraconazole and amphotericin B had lower MIC than the remaining four antifungal agents. The MIC result for posaconazole was over 220-fold lower active than fluconazole. Fluconazole MICs for most isolates fell within the dose-dependant range. The overall MIC ranges and $MIC_{50}s$ were amphotericin B(0.03-0.25; 0.25), fluconazole(0.5-64; 16), itraconazole(0.015-1; 0.125), terbinafine(0.06->2; 1), caspofungin(8-32; 32), voriconazole(0.015-0.5; 0.25), and posaconazole(0.015-0.25; 0.06 ${\mu}g/ml$), respectively. In conclusion, the $MIC_{50}s$ of these drugs did not exhibit any sign of an upward shift with the exception of fluconazole and tendency cross-resistance between the seven drugs was not observed. We conclude that in vitro resistance to antifungal agents has not significantly changed despite the recent wide-spread use of triazoles for long-term treatment of Cryptococcal meningitis.

다종의 안질환 사용 처방이 각막염 유발균인 Staphylococcus epidermidis에 미치는 영향 (The Experimental Study on Anti-bacterial Potency of Variou Herb-medicine used for eye disease on Staphylococcus epidermidis Keratitis)

  • Objective : This experimental study was performed to investigate the effect of Herbe-medicine used for eye disease(Sean-tang, Jinpi-san, Tangpo-san, Copitdis Rhizoma) on Staphylococcus epidermidis keratitis. Methods : After administering various herbal eye drops on Staphylococcus epidermidis, I measured MIC and the size of inhibition zone. MIC was measured by dropping from 20 to $50{\mu}{\ell}$ according to density. Anti-bacterial potency was measured by the size of inhibition zone with change of volume under 2 days culture condition. Also continuous anti-bacterial potency of herbal eye drops was measured by the size of inhibition zone according to 2 days and 6 days culture each under the $50{\mu}{\ell}$ condition. Results : 1. MIC on Staphylococcus epidermidis in Sean-tang, Jinpi-san was 1%, $50{\mu}{\ell}$ 2. MIC on Staphylococcus epidermidis in Tangpo-san, Coptidis rhizoma was 10%, $50{\mu}{\ell}$ 3. MIC on Staphylococcus epidermidis in Cravit was 0.1%, $20{\mu}{\ell}$. 4. Under the 2 days culture condition, the size of inhibition zone on Staphylococcus epidermidis by volume for Sean-tang was 10.0mm in $50{\mu}{\ell}$, Jinpi-san was 16.0mm in $50{\mu}{\ell}$, Tangpo-san was 17.5mm in $50{\mu}{\ell}$, Coptidis rhizoma was 31.0mm in $50{\mu}{\ell}$ and Cravit was 34mm in $50{\mu}{\ell}$, showed the highest anti-bacterial potency. 5. Under the $50{\mu}{\ell}$ condition, the size of inhibition zone on Staphylococcus epidermidis by 2 and 6 culture days for Sean-tang was 47.5mm in 6days, Jinpi-san was 36.0mm in 6days, Tangpo-san was 45.0mm in 6 days and Cravit was 48.0mm in 6 days, which showed the highest anti-bacterial potency. 6. Under the $50{\mu}{\ell}$ culture condition, the size of inhibition zone on Staphylococcus epidermidis by 2 and 6 culture days for Coptidis rhizoma was 31.0mm in 2 days and 6 days, which showed the same anti-bacteria1 potency. Conclusions : The present author think that Herbe-medicine used for eye disease can be used to cure Staphylococcus epidermidis keratitis and if further study is performed, the use of the Herbe-medicine used for eye disease will be valuable and beneficial in the clinical medicines.

살모넬라 균주들에서 열처리에 의한 항생제 내성 연구 (Antibiotic Resistance of Salmonella spp. After Exposure to Mild Heat Treatment)

  • Salmonella spp. 11 strains에 대해 저온 열처리(50℃) 3, 6, 9분 후 MIC값을 측정하여 항생제 내성을 알아보았다. Chloramphenicol에 대해 대조군과 열처리한 strains 대부분에서 감수성(S)이 있는 것으로 나타났고, 열처리한 strains의 MIC값은 대조군과 비교하였을 때 유지되거나 감소하였다. Ciprofloxacin에 대해 대조군과 열처리한 strains는 대부분 감수성(S)이 있거나 중간(I)을 나타냈다. Tetracycline은 모든 strains에서 감수성(S)이 있는 것으로 나타났으며, S. Gaminara BAA 711에 대해 열처리 후 MIC값이 증가하였다. Gentamicin에 대해 대조군 strains들에서 감수성을 나타낸 strains가 3 strains, 중간을 나타낸 strains 2 strains, 내성을 가진 strains가 6 strains였으며, 이 중 S. Heidelberg ATCC 8326는 MIC값을 측정했을 때 대조군에서 MIC값이 8 ㎍/mL로 MIC break point가 중간이었으나, 3분과 9분 열처리 후 MIC값이 16 ㎍/mL로 증가하여 break point가 내성을 나타냈다. 본 실험결과 Salmonella spp. 11 strains에 대해서 저온 열처리 후 열내성 효과에 의한 항생제 내성을 알아봤을 때 ciprofloxacin에서 S. Montevideo BAA 710을 3, 6분 열처리한 경우, gentamicin에서 S. Enteritidis 109 D1을 3분 처리한 경우와 S. Heidelberg ATCC 8326을 3, 9분 처리한 경우, tetracycline에서 S. Gaminara BAA 711을 6, 9분 처리한 경우 MIC값이 증가하였다. 후속 연구를 통해 Salmonella spp. strains에 대해 열처리 후 열내성 효과를 나타내는 병원성 유전자의 특성에 대한 지속적인 연구가 필요하다.

  • 배 경 : 현재 moxifloxacin은 기존의 quinolone 제제들에 비하여 항결핵효과가 크다고 알려지면서 다제내성 결핵에 많이 사용되어 왔지만 moxifloxacin과 기존의 quinolone간의 교차내성에 대한 보고가 없는 상황에서 기존의 quinolone에 내성인 균주에 대한 moxifloxacin의 효과를 예측하기 어려웠다. 이에 본 연구를 통하여 moxifloxacin과 ofloxacin간의 교차내성을 알아봄으로써 ofloxacin 내성 균주에 대한 moxifloxacin의 항 결핵 효과를 알아보고자 하였다. 방 법 : 2003년 3월부터 2004년 3월까지 폐결핵 치료를 위해서 국립마산병원에 방문 또는 입원하였던 환자들의 객담에서 분리된 M. tuberculosis isolate에 대한 약물감수성 검사상 ofloxacin에 대한 본원 임상 검사실의 내성기준 농도인 $2.5{\mu}g/m{\ell}$의 농도에 내성을 보인 균주 37개와 감성을 보인 균주 16개를 대상으로 하여 ofloxacin 및 moxifloxacin 각각에 대한 MIC를 비례법을 이용하여 조사하였다. 결 과 : Ofloxaicn 감성균주들에 대한 $MIC_{50}$$MIC_{90}$은 ofloxacin에서 모두 $1.25{\mu}g/m{\ell}$, moxifloxacin에서는 각각 $0.31{\mu}g/m{\ell}$$0.63{\mu}g/m{\ell}$였다. ofloxacin 내성균주들에 대한 $MIC_{50}$은 ofloxacin에서 $10{\mu}g/m{\ell}$ 이상, moxifloxacin에서 $5{\mu}g/m{\ell}$였으며 $MIC_{90}$은 ofloxacin과 moxifloxacin 모두에서 $10{\mu}g/m{\ell}$이상의 값을 보였다. 약물 농도 $2.5{\mu}g/m{\ell}$을 기준으로 한 moxifloxacin의 ofloxacin에 대한 교차 내성률은 67.6%로 나타났다. 결 론 : moxifloxacin은 $10{\mu}g/m{\ell}$의 농도에서 ofloxacin 내성균주의 82.4%(28/34)에서 항결핵 효과가 있었으나 ofloxacin 내성을 포함하는 다제내성 결핵환자에게 사용하기 위해서 좀 더 많은 연구가 필요할 것으로 사료된다.

Under the 2 days culture condition, the size of inhibition zone on Staphylococcus aureus by volume for Sean-tang was 12.0mm in $50{\mu}{\ell}$, Jinpi-san was 19.0mm in $50{\mu}{\ell}$, Tangpo-san was 15.0mm in $50{\mu}{\ell}$, Coptidis rhizoma was 32.7 mm in $50{\mu}{\ell}$ and Cravit was 31mm in $50{\mu}{\ell}$, Coptidis rhizoma showed the highest anti-bacterial potency. 5. Under the $50{\mu}{\ell}$ condition, the size of inhibition zone on Staphylococcus aureus by 2 and 6 culture days for Sean-tang was 12.0mm, Jinpi-san was 19.0mm, Tangpo-san was 15.0mm, Coptidis rhizoma was 32.7mm and Cravit was 31.0 mm, which showed sameness anti-bacterial potency in 2 days and 6 days. Conclusions : The present author think that the herbal eye drops can be used to cure Staphylococcus aureus keratitis and if further study is performed, the use of the herbal eye drops will be valuable and beneficial in the clinical medicines.

서울에서 시판중인 굴의 세균학상 오염과 분리된 비브리오속 균의 항생제 내성 양상 (Antibiotic Resistance of Vibrio spp. and Bacterial Contamination of Commercial Oysters in Seoul, Korea)

  • The purpose of this investigation was to measure the level of bacterial contamination of oysters, and observe antibiotic resistance pattern of Vibro spp. found in oysters. For this experiment, 100 oysters were collected from 100 markets around seoul area from October, 2004 to January, 2005. Bacterial contaminations of the oysters were confirmed by performing the method of standard plate count and desoxycholate lactose agar plate. Total plate counts were $2.7\times$$10^{2}$$\sim$$1.2\times$$10^{5}$ cfu/g. Coliform group were $1.0\times$$1.0^{1}$$\sim$$3.2\times$$10^{5}$ cfu/g. Results have shown that Vibrio spp. was present in $64\%$ of the 100 sampls. Antibiotic resistance pattern of Vibrio spp. showed that $MIC_{50}$ of chloramphenicol was 0.2 g/mL, $MIC_{90}$ was 25 $\mu$g/mL, and $MIC_{50}$ of tetracycline was < 0.05 $\mu$g/mL, $MIC_{90}$ was 25 $\mu$g/mL, and $MIC_{50}$ of ciprofloxacin was 0.01 g/mL, $MIC_{90}$ was 10> $\mu$g/mL. Three of the six strains were identified as Vibrio alginolyricus.

의치환자에서 Candida albicans분리와 amphotericin B 및 miconazole에 시험관내 감수성 (Isolation of Candida albicans from Denture Patients and in Vitro Activities of Amphotericin B and Miconzole)

  • Candida albicans is now well recognized among the denture stomatitis patients. The broth macrodilution test is the most widely used technique for antifungal susceptibility testing. The purpose of this study was to determine the C. albicans carrier rate of the denture patients in Iksan, chonbuk. To determine the C. albicans carrier rate of denture patients, culture were made from 227 sample taken in Iksan, Chonbuk during July 1997 to August 1997. Also activities of amphotericin B and miconzole against isolates of denture patients of C. albicans were tested by broth macrodilution test using RPMI medium 1640. The results were as follows : First C. ablicans was isolated from 6.6% of denture patients samples and the frequency of isolation fo C. albicans was highest(50%) in the age group of 71-year-old to 80-year-old denture patients. Second, against C. albicans, the MIC range of amphotericn B was $0.06{\sim}0.25{\mu}g/ml$. MIC50 and MIC90 were $0.13{\mu}g/ml$ and $0.25{\mu}g/ml$, respectively. Third, the MIC range of miconazole was $10-{\ge}20{\mu}g/ml$ and MIC50 and MIC90 were $20{\mu}g/ml$ and ${\ge}20{\mu}g/ml$, respectively. It was concluded from this study that C. albicans acrriages from healthy denture individuals only over 60-year-old ages were isolated, they remain susceptible to amphotericin B and not rarely resistant to miconzole.

한국과 일본의 담수어에서 분리되는 운동성 aeromonads의 표현형적 특성과 약제감수성 (Phenotypic characteristics and antimicrobial susceptibilities of motile aeromonads isolated from freshwater fish in Korea and Japan)

  • 본 연구에서는 우리나라와 일본의 담수어에서 분리된 운동성 aeromonads 7균주와 American Type Culture Collection (ATCC)에서 분양받은Aeromonas hydrophila 4균주의 표현형적 특성을 API20E와 APIZYM 방법으로 평가하고, 7종류의 항생제에 대한 최소 성장 억제 농도 (minimum inhibitory concentrations; MICs) 를 측정하였다. API20E 시험 결과 시험한 모든 균주는 (n=7) 운동성 aeromonads로 동정되었다. API20E시험에서lysine decarboxylase와mannitol, rhamnose, amygdalin, arabinose를 포함한 4종류의 carbohydrates의 산 생성은 균주에 따라 다른 반응을 나타내었다. APIZYM 시험을 이용한 효소 활성능을 평가한 결과, 모든 시험된 균주가valine arylamidase, cystine arylamidase, α-chymotrypsin, α-galactosidase, β-glucuronidase, α-glucosidase, α-mannosidase, α-fucosidase반응에서 음성 반응을 나타내었으나, 비록 그 효소 활성의 강도에서 차이는 있었으나alkaline phosphatase, esterase-lipase, leucine arylamidase, β-galactosidase, N-acetyl-β-glucosaminidase 모든 균주에서 양성 반응이 나타났다. 최소 성장 억제농도를 시험한 결과, 시험된 모든 균주는 ampicillin sodium (MIC>100㎍/ml) 에 내성을 가지며 chloramphenicol (MIC≤1.6㎍/ml) 감수성을 나타내었다. 그러나 1998년 이후에 분리된 3균주 (AC9804, AC0202, GMA0361)는 tetracycline (MIC=50㎍/ml) 모두 저항성이 있었으며, AC9804는 oxolinic acid (MIC=12.5㎍/ml), GMA0361는 kanamycin sulfate (MIC>100㎍/ml)와 streptomycin sulfate (MIC>100㎍/ml)에도 저항성을 나타내었다.

Postantibiotic Effects and Postantibiotic Sub-MIC Effects of Antibiotics on Fusobacterium nucleatum and Porphyromonas gingivalis

  • Postantibiotic effects (PAE) refer to suppression of the bacterial growth following limited periods of exposure to an antibiotic and subsequent to the removal of the antibiotic agent. Fusobacterium nucleatum and Porphyromonas gingivalis are Gram-negative anaerobic bacteria associated with several periodontal diseases. In this study, postantibiotic effects (PAE), postantibiotic sub-MIC effect (PA SME) and sub-MIC effect (SME) of antibiotics on F. nucleatum ATCC 25586 and P. gingivalis W50 were investigated. The PAE was induced by 10X the MIC of antibiotic and antibiotic was eliminated by washing. The PA SMEs were studied by addition of 0.1, 0.2 and 0.3X MICs during the postantibiotic phase of the bacteria, and the SMEs were studied by exposition of the bacteria to antibiotic at the sub-MICs only. Amoxicillin, doxycycline and tetracycline induced PAE for F. nucleatum ATCC 25586 and P. gingivalis W50. But metronidazole and penicillin induced PAE for only F. nucleatum ATCC 25586. Metronidazole and doxycycline induced PA SME and SME for both species of anaerobic bacteria used in this study. The PA SME values for both strains were substantially longer than the SME values. The present study showed the existence of PAE, PA SME and SME for various antibiotics against F. nucleatum ATCC 25586 and P. gingivalis W50.
