Tolyfluanid에 대한 인삼 잿빛곰팡이병균의 감수성 (Sensitivity of Botrytis cinerea Isolated from Infected Leaves of Ginseng to Tolyfluanid)

  • 2005년부터 2006년까지 인삼 재배지에서 분리한 218개의 Botrytis cinerea 균주의 tolyfluanid에 대한 감수성 정도를 조사하였다. Tolyfluanid를 농도별로 첨가한 PDA배지에서 B. cinerea의 균사생장 억제정도를 조사하여 살균제에 대한 B. cinerea 균주들의 감수성 정도를 3 group으로 구분하였다. 분리한 병원균은 감수성 정도에 따라서,$2.0\;{\mu}g\;mL^{-1}$이하인 균주들을 HS group으로, 2.0에서부터 $18.0\;{\mu}g\;mL^{-1}$ 사이에 속하는 균주들을 S group으로, 그리고 $18.0\;{\mu}g\;mL^{-1}$ 이상인 균주들을 LS group으로 구분하였다. 전체 균주 중에서 174개 균주가 S group에 속하였으며, 각각 22개 균주가 HS와 LS group으로 구분되었다. 또한 tolyfluanid의 B. cinerea에 대한 균사생장 억제와 포자발아 억제 효과는 높은 정의 상관 관계를 보였다. 본 연구의 결과는 포장에서 분리한 B. cinerea의 tolyfluanid에 대한 저항성 모니터링의 기준과, 저항성 발현 관리를 위한 기초 자료로 사용될 수 있다.

북방전복, Haliotis discus hannai의 염분 변화에 따른 생리적 반응 (Physiological Responses in Abalone Haliotis discus hannai with Different Salinity)

  • 염분변화에 따른 북방전복의 생리적 반응을 조사하기 위하여 고염분 및 저염분의 농도에 따라 생존율, 삼투질농도, 대사율 및 조직학적 반응을 분석하였다. 염분변화에 따른 북방전복의 생존율은 37 psu에서는 100%였다. 반면 19.8 psu에서 노출 6일째, 12.8 psu 이하에서는 노출 24시간 이내 모두 사망하여 북방전복의 7-days $LS_{50}$은 24.9 psu (20.1-28.2 psu)를 나타내었다. 염분변화에 따른 호흡률은 대조구에 비해 감소하였으며, 반면 암모니아질소 배설률은 증가하였다. 삼투질농도는 염분 26.8 psu이상에서는 1시간 이내 순치하였다. 북방전복 발근육의 조직학적 반응은 염분감소에 따라 혈림프동의 확장, 상피세포의 괴사 및 식세포 증가 등의 현상이 관찰되었다.

  • Since the year 2008, the first demonstration project of the superconducting cable for the KEPCO grid operation has been in progress. To apply the HTS (High Temperature Superconducting) cable system into the commercial network, a new test complex was built in the 154kV Icheon substation and the on site assembly of 22.9kV, 50MVA HTS cable with the cryogenic refrigeration system was finished. A full scale of HTS cable fabricated with 2nd generation wire is to be in service in KEPCO network. In this paper, a series of processes of the installation and operation test are introduced.

수영만에서 분리된 Bacillus cereus Group LS-1 의 세균학적 특성에 관한 연구 (Studies on Bacterial Characteristics of Bacillus cereus Group LS-1 Isolated from Suyeong Bay)

  • 수영만에서 분리된 Bacillus cereus group LS-1 의 형태학적, 생화학적 성상 및 지방산 조성 분석을 통하여 세균학적인 특성을 밝혔다. B. cereus group LS-1 은 면양혈구배지에서 비용혈성의 점조성 집략과 convex 하고 가장자리가 규칙적인 둥근형태로 운동성이 없고 glucose, maltose, sucrose 와 gelatin 을 이용하고 trehalose 와 salicin 을 분해하지 않으며 6.5% NaCl 에서 자라지 않는 Gram 양성의 중심성 아포형성간균으로 표준균주 B. cereus group 과 다소 차이를 나타내었다. 지방산 조성 분석에서 chain 의 길이가 $C_{12}$ 에서 $C_{17}$로 iso $C_{15}$와 iso $C_{13}$의 branched chain 이 우점하는 B. cereus group 의 전형적인 특징을 나타내었으며 $nC^{15}$가 검출되지 않는 B.mycoides GC subgroup B 로 0.312 의 similarity index(SI) 를 지칭하여 다른 연구 결과와도 일치하였다. 한편 API system (API 50 CHB & API 20E) 의 ATB computer profile 에서 "Doubful Profile" 99.7% 의 B. firmus 로 나타내어 큰차이를 나타내었다. 67 개의 biochemical character 로 B. mycoids S-12 는 각각 42%, 42% 59% 와 52% 의 similarity matrix 를 나타내었다. B. cereus group 간의 아주 낮은 similarity 를 나타내어 상당한 차이가 있음을 인식하였다. 따라서 key test 와 지방산 조성등을 종합하여 볼때 B. ceresu group 중의 B. mycoides 로 새로운 biotype 인 것으로 사료되며 지방산 조성 분석으로 동정함이 훨씬 용이하였다.

  • In this study, we build system and techniques of evaluating the interruption performance of the GCB with experimental method. We constructed a simplified synthetic test circuit of which ability is up to 245kV, 50kA BTF test. And We composed a model test circuit breaker with puffer assisted self blasting type GCB. With this circuit breaker, we carried out the experiment of no load and SLF90. During the tests, we measured the several factors such as stroke, pressure, arc temperature, the voltage and current near the current zero and dI/dt, dV/dt. Arc conductivity before 200ns before current zero which is one of the indexes of the thermal recovery of a GCB was measured. With these kinds of measurement, we could estimate the performance of a GCB fundamentally. Futhermore these results were used to adjust the arc modeling with CFD(computational fluid dynamics) and we could increase the plausibility of the analytical method.

Karyotype Analyses of a Rice Cultivar 'Nakdong' and its Four Genetically Modified Events by Conventional Staining and Fluorescence in situ Hybridization

  • Conventional staining and fluorescence in situ hybridization (FISH) karyotypes of the non-genetically modified (GM) parental rice line, 'Nakdong' (Oryza sativa L. japonica), and its four GM rice lines, LS28 (event LS30-32-20-1), Cry1Ac1 (event C7-1-9-1), and LS28 ${\times}$ Cry1Ac1 (events L/C1-1-3-1 and L/C1-3-1-1) were analyzed using 5S and 45S rDNAs as probes. Both parental and transgenic lines were diploids (2n=24) with one satellite chromosome pair. The lengths of the prometaphase chromosomes ranged from 1.50 to $6.30{\mu}m$. Four submetacentric and eight metacentric pairs comprised the karyotype of 'Nakdong' and its four GM lines. One pair of 5S rDNA signals was detected near the centromeric region of chromosome g in both the parental and transgenic lines. The 45S rDNA signals were detected on the secondary constrictions of the satellite chromosome pair in both the parental and transgenic lines. There was no significant difference in chromosome size, length, and composition between 'Nakdong' and its four GM lines. This research was conducted as a preliminary study for chromosomal detection of transgenes in GM rice lines and would be useful for their breeding programs.

Prevalence of Disc Degeneration in Asymptomatic Korean Subjects. Part 3 : Cervical and Lumbar Relationship

  • Objective : There are many cases in which degenerative changes are prevalent in both the cervical and lumbar spine, and the relation between both spinal degenerative findings of MRI is controversial. The authors analyzed the prevalence of abnormal findings on MRI, and suggested a model to explain the relationship between cervical and lumbar disc in asymptomatic Korean subjects. Methods : We performed 3 T MRI sagittal scans on 102 asymptomatic subjects (50 men and 52 women) who visited our hospital between the ages of 14 and 82 years (mean age 46.3 years). Scores pertaining to herniation (HN), annular fissure (AF), and nucleus degeneration (ND) were analyzed. The total scores for the cervical and lumbar spine were analyzed using correlation coefficients and multiple linear regression with various predictive parameters, including weight, height, sex, age, smoking, occupation, and sedentary fashion. Results : The correlation coefficients of HN, AF, and ND were 0.44, 0.50, and 0.59, respectively. We made the best model for relationship by using multiple linear regression. Conclusion : The results of the current study showed that there was a close relationship between the cervical score (CS) and lumbar score (LS). In addition, the correlation between CS and LS, as well as the LS value itself, can be altered by other explanatory variables. Although not absolute, there was also a linear relationship between degenerative changes of the cervical and lumbar spine. Based on these results, it can be inferred that degenerative changes of the lumbar spine will be useful in predicting the degree of cervical spine degeneration in an actual clinical setting.

SWAT과 SATEEC 모형을 이용한 토양유실량 비교 (Comparison of Soil Loss Estimation using SWAT and SATEEC)

  • Soil erosion is a natural process and has been occurring in most areas in the watershed. However, accelerated soil erosion rates have been causing numerous environmental impacts in recent years. To reduce soil erosion and sediment inflow into the water bodies, site-specific soil erosion best management practices(BMPs) need to be established and implemented. The most commonly used soil erosion model is the Universal Soil Loss Equation(USLE), which have been used in many countries over 30 years. The Sediment Assessment Tool for Effective Erosion Control(SATEEC) ArcView GIS system has been developed and enhanced to estimate the soil erosion and sediment yield trom the watershed using the USLE input data. In the last decade, the Soil and Water Assessment Tool(SWAT) model also has been widely used to estimate soil erosion and sediment yield at a watershed scale. The SATEEC system estimates the LS factor using the equation suggested by Moore and Burch, while the SWAT model estimates the LS factor based on the relationship between sub watershed average slope and slope length. Thus the SATEEC and SWAT estimated soil erosion values were compared in this study. The differences in LS factor estimation methods in the SATEEC and SWAT caused significant difference in estimated soil erosion. In this study, the difference was -51.9%(default threshold)${\sim}-54.5%$(min. threshold) between SATEEC and non-patched SWAT, and -7.8%(default threshold)${\sim}+3.8%$(min. threshold) between SATEEC and patched SWAT estimated soil erosion.

The characteristics of bovine satellite cells with highly scored genomic estimated breeding value

  • Background: The grading of Hanwoo (Korean native cattle) is based on four economic traits, and efforts have been continuously made to improve the genetic traits associated with these traits. There is a technology to predict the expected grade based on the 4 economic genetic SNP characteristics of Korean cattle calves using single nucleotide polymorphism (SNP) technology. Selection of highly proliferative, self-renewing, and differentiating satellite cells from cattle is a key technology in the cultured meat industry. Methods: We selected the Hanwoo with high and low-scored of genomic estimated breeding value (GEBV) by using the Hanwoo 50K SNP bead chip. We then isolated the bovine satellite cells from the chuck mass. We then conducted comparative analyses of cell proliferation, immunocytochemistry, qRT-PCR at short- and long-term culture. We also analyzed the differentiation capability at short term culture. Results: Our result showed that the proliferation was significantly high at High scored GEBV (Hs-GEBV) compared to Low scored GEBV (Ls-GEBV) at short- and long-term culture. The expression levels of Pax3 were significantly higher in Hs-GEBV bovine satellite cells at long-term culture. However, there were no significant differences in the expression levels of Pax7 between Hs- and Ls-GEBV bovine satellite cells at short- and long- term culture. The expression levels of MyoG and MyHC were significantly high at Ls-GEBV bovine satellite cells. Conclusions: Our results indicated that selection of bovine satellite cells by Hanwoo 50K SNP bead chip could be effective selection methods for massive producing of satellite cells.

Survival of the Ark Shell, Scapharca subcrenata and Physiological and Histological Changes at Decreasing Salinity

  • We examined physiological and histological responses related to the survival, oxygen consumption, excretion, and O/N ratio of the ark shell, Scapharca subcrenata, as a result of salinity changes. The 20-day $LS_{50}$ (median lethal salinity) at $15^{\circ}C$ was 13.87 practical salinity units (psu; confidence limits 10.30-18.74 psu), whereas the 14-day $LS_{50}$ at $25^{\circ}C$ was 12.59 psu (confidence limits 8.03-18.16 psu). In conditions of decreasing salinity, the osmolarity of individuals acclimated within 5 h above 26.4 psu but required more than 60 h below 13.2 psu. Oxygen consumption and ammonia excretion rates varied irregularly as salinity decreased. The O/N ratio was 19 and 27 at water temperatures of $15^{\circ}C$ and $25^{\circ}C$, respectively, but decreased to 1-10 as salinity declined. The effects of decreasing salinity were observed in the histological changes to each organ of S. subcrenata. As salinity decreased, cilia fell off, the epithelial layer underwent necrosis and vacuolation, the connective tissue layers of the mantle and visceral mass were destroyed, and hemocytes increased in the gills. The results of this study could prove important in investigating causes of mass mortality and managing shellfish aquaculture farms.
