단백질 분해효소 생산을 위한 Aspergillus oryzae PF균주의 배지조성 (Medium Composition of Aspergillus oryzae PF for the Production of Proteolytic Enzyme)

  • The most favorable nitrogen source for the production of protease by Aspergillus oryzae PF was 2% soybean flour among sodium nitrate, ammonium sulfate, defatted soybean, skim milk, casein, peptone, and yeast extract. The production of protease from A. oryzae PF was higher at the concentration of 2% lactose than at variable concentration of glucose, sucrose, soluble starch, corn starch, potato starch, wheat starch, rice starch, cellulose, and gum arabic. Protease production was affected by the concentration of KH2PO4, Triton X-100, CaCo3, and MgSO4, and it was the highest at the highest at the concentration of 3% KH2PO4, 0.01% Triton X-100, 0.3% CaCO3, and 0.06% MGSO4.

  • 두유응고 효소생산균으로 분리, 보고한 균주 중 생산능이 우수한 Bacillus licheniformis 192균주의 두유응고 효소생산을 위한 최적조건을 검토하였다. 최적배지 조성은 potato starch 1.5, 두유 2.0, 대두박 추출액 10 및 KH2PO4 0.6(pH6.1)였으며 35-4$0^{\circ}C$에서 3일간 배양하였을 때 가장 높은 효소활성을 나타내었다.

Biosynthesis of Eudesmane-type Sesquiterpenoids by The Wood-rotting Fungus, Polyporus brumalis, on Specific Medium, including Inorganic Magnesium Source

  • Fungi, such as the wood-rotting Polyporus brumalis, are excellent sources of pharmaceutically interesting natural products such as sesquiterpenoids. In this study, we investigated the biosynthesis of P. brumalis sesquiterpenoids on modified medium. Ten additional species of white rot fungi were inoculated in medium containing nutrients such as $C_6H_{12}O_6$, $C_4H_{12}N_2O_6$, $KH_2PO_4$, $MgSO_4$, and $CaCl_2$ at $28^{\circ}C$ for 25 days. After 10 days of incubation, eudesmane-type sesquiterpenes, ${\beta}$-eudesmane and ${\beta}$-eudesmol, were only synthesized during the growth phase of P. brumalis. Experiments excluding one nutrient at a time were conducted to determine the effects of inorganic nutrients on sesquiterpene biosynthesis. In conclusion, GC-MS analysis showed that biosynthesis of sesquiterpenes was differentially regulated by inorganic nutrients such as $MgSO_4$, $C_4H_{12}N_2O_6$, and $KH_2PO_4$. We found $MgSO_4$ supplementation to be vital for eudesmane-type sesquiterpene biosynthesis in P. brumalis; nitrogen ($C_4H_{12}N_2O_6$) and phosphate ($KH_2PO_4$) inhibited the synthesis of P. brumalis metabolites. Magnesium is a known cofactor of sesquiterpene synthase, which promotes ${\beta}$-eudesmol synthesis. To mechanistically understand eudesmane-type sesquiterpene biosynthesis in P. brumalis, further research into the genes regulating the dynamics of such biosynthesis is warranted.

  • Poly-${\gamma}$-glutamic Acid(${\gamma}$-PGA)를 다량 생산하는 균주를 우리나라의 전통발효식품인 청국장으로부터 Bacillus subtilis GS-2를 분리하였다. 이 균은 glutamic acid 의존형 균으로, 이 균에 의한 ${\gamma}$-PGA 생산 최적 조건을 검토한 바, 단순배지(L-glutamic acid 2.0%, glucose 1.0%, $NH_4Cl$ 0.5%, $KH_2PO_4$ 0.05%, $MgSO_4$ $7H_2O$ 0.01%, pH 7.0)로 진탕배양(220 rpm) 하였을 때, 배양시간 48시간, 최적온도 $33^{\circ}C$, 그리고 초기 pH 6.5로 나타났다. 영양원으로 glutamic acid 3%, sucrose 3%, $NH_4Cl$ 0.25%, $KH_2PO_4$ 0.15%, $MgSO_4$ $7H_2O$ 0.015%에서 ${\gamma}$-PGA 최대 생산량이 31.0 $g/{\ell}$이었다.

  • The simulation results of the third harmonic conversion with 1.064 um Nd :YAG laser using polarization mismatch method are reported. The third harmonic conversion efficiency was over 65% for the Gaussian incident pulse of $300{\sim}400MW/cm^2$ peak intensity, and over 80% for $3{\sim}4GW/cm^2$ peak intensity pulse. The dependence of the third harmonic conversion efficiency' on the variation of incident polarization angle ${\theta}_p$ and angular discrepancy ${\Delta}{\theta}$ of phase matched angle ${\theta}_m$ is discussed.

  • Cellulose를 효과적으로 분해하는 효소를 생산하는 미생물인 strain No. 33 균주를 토양으로부터 분리하였다. 본 균주의 배양상등액을 2.0%의 CMC가 함유된 agar 평판배지에 paper disc법을 이용하여 loading 후 cellulose 분해활성을 검토하였을 때, disc 주위로 노란색의 환이 형성되는 것을 확인할 수 있었다. 본 균주를 동정하기 위하여 전자현미경으로 그 형태를 관찰한 결과 본 균주는 간균의 형태였으며, 생리학적 특성을 검토한 결과 Bacillus속으로 동정되었다. 이에 본 균주를 Bacillus sp. No. 33으로 명명하였다. 본 균주는 1.0% glucose, 3.0% yeast extract, 0.5% $KH_2PO_4$, 0.02% $MgSO_4{\cdot}7H_2O$가 함유된 pH 7.0의 배지에서 $30^{\circ}C$로 39시간 동안 진탕배양하였을 때 최대 균체생육을 나타내었다. 효소생산은 4.0% CMC, 2.0% yeast extract, 0.5% $KH_2PO_4$, 0.04% $MgSO_4{\cdot}7H_2O$, pH 7.0의 조건에서 $30^{\circ}C$, 42시간 동안 진탕배양하였을 때 최대 효소생산을 나타내었다.

플라스크배양에서 버들송이버섯 균사체 배양에 관한 연구 (A Study on the Mycelial Growth of Agrocybe aegerita in Flask Culture.)

  • 버들송이버섯의 균사체 대량 생산의 기초자료를 얻을 일환으로 균사의 영양생장에 필요한 적합한 조건을 연구하고자 균사생장을 위한 최적 배양 조건 및 영양원을 조사한 결과는 다음과 같다 5종의 공시배지 중 ME 배지에서 버들송이 버섯의 균사생장 및 밀도가 가장 양호하였으며, 균사생장최적 온도는 $25^{\circ}C$이고, 최적 pH는 5.5이며 최적 배양일수는 12일이었다. 버들송이 버섯의 균사생장을 위한 최적 배지조성은 탄소원에서는 다당류인 dextrin이었으며, 최적 탄소원인 dextrin의 적정농도는 3% (w/v)였다. 질소원에서는 유기태 질소원인 yeast extract이었고, yeast extract 최적농도는 2% (w/v)였으며, 최적 무기염류는 MgSO$_4$와 KH$_2$PO$_4$이고, 최적 농도는 MgSO$_4$0.05% (w/v), KH$_2$PO$_4$0.15 (w/v)를 혼합하였을 때 가장 많은 생산량을 보였다. 기본배지인 ME 배지와 본 연구에서 얻어진 최적배지로 액체 배양하여 균체량을 비교한 결과 기본배지는 배양 12일째 최대 건조 균체량 7.59 g/l을 얻었으며, 최적배지는 배양 12일째 17.6 g/l의 최대 건조 균체량을 얻어 기본배지 보다 최적배지의 균체량이 훨씬 높은 균사체 생산수율을 보였다.

Monascus anka albidus의 적색색소 생산 (Red Pigment Production from Monascus anka albidus)

  • Monascus anka albidus 균사를 쌀가루 7%, $NH_4NO_3$ 0.15%, $KH_2PO_4$ 0.05% 및 $MnSO_4$ 0.1%, 조성의 초기 pH 6.0으로 조절한 배지에 접종하여 $30^{\circ}C$에서 5일간 진탕 배양 하였을 때 가장 높은 색소 생산을 보였다. 특히 탄소원으로써 찹쌀가루가 제일 좋았으며 동시에 질소원으로써 $NH_4NO_3$, 인산염으로는 $KH_2PO_4$가 가장 우수하였으며 최적 C : N율은 18 : 1로 나타났다. 상기 조건에서 색소 생산은 플라스크에서 2.6mg/ml을, 그러나 5l 발효조에서 약 70%인 1.8mg/ml을 생산하였다.

Comparison of Various Single Chemical Extraction Methods for Predicting the Bioavailability of Arsenic in Paddy Soils

  • The Codex Committee of Contaminants in Food (CCCF) has been discussing a new standard for arsenic (As) in rice since 2010 and a code of practice for the prevention and reduction of As contamination in rice since 2013. Therefore, our current studies focus on setting a maximum level of As in rice and paddy soil by considering bioavailability in the remediation of As contaminated soils. This study aimed to select an appropriate single chemical extractant for evaluating the mobility of As in paddy soil and the bioavailability of As to rice. Nine different extractants, such as deionized water, 0.01 M $Ca(NO_3)_2$, 0.1 M HCl, 0.2 M $C_6H_8O_7$, 0.43 M $HNO_3$, 0.43 M $CH_3COOH$, 0.5 M $KH_2PO_4$, 1 M HCl, and 1 M $NH_4NO_3$ were used in this study. Total As content in soil was also determined after aqua regia digestion. The As extractability of the was in the order of: Aqua regia > 1 M HCl > 0.5 M $KH_2PO_4$ > 0.43 M $HNO_3$ > 0.2 M $C_6H_8O_7$ > 0.1 M HCl > 0.43 M $CH_3COOH$ > deionized water > 1 M $NH_4NO_3$ > 0.01 M $Ca(NO_3)_2$. Correlation between soil extractants and As content in rice was in the order of : deionized water > 0.01 M $Ca(NO_3)_2$ > 0.43 M $CH_3COOH$ > 0.1 M HCl > 0.5 M $KH_2PO_4$ > 1 M $NH_4NO_3$ > 0.2 M $C_6H_8O_7$ > 0.43 M $HNO_3$ > 1M HCl > Aqua regia. BCF (bioconcentration factor) according to extractants was in the order of : 0.01M $Ca(NO_3)_2$ > 1 M $NH_4NO_3$ > deionized water > 0.43 M $CH_3COOH$ > 0.1 M HCl > 0.43 M $HNO_3$ > 0.2 M $C_6H_8O_7$ > 0.5 M $KH_2PO_4$ > 1 M HCl > Aqua regia. Therefore, 0.01 M $Ca(NO_3)_2$ ($r=0.78^{**}$) was proven to have the greatest potential for predicting As bioavailability in soil with higher correlation between As in rice and the extractant.

Serratia sp. KH-95에 의한 적색 색소 생산 및 배양학적 특성 (Production of Red Pigment by Serratia sp. KH-95 and its Cultural Properties)

  • Optimal media and cultural conditions for the production of prodigiosin-like pigment were established using Serratia sp. KH-95. Glucose and phosphate(K2PO4) stimulated the cell growth, but inhibited the production of pigment at concentration levels of above 10 g/L and 2.0 g/L, respectively. Addition of soy been oil or rice oil to the production medium accelerated cell growth up to more than 2-3 times, but the production of prodigiosin increased about 15-20% in spite of the good cell growth. The effect of pH on the production of pigment was investigated in a 5 liter-bioreactor. When the pH of culture broth was maintained below 8.0, most of pigment was attached to the surface of cells. When the pH of culture broth was above 8.5, however, about 70% of total pigment was suspended in the supernatant of the broth. The cell growth and production of pigment were inhibited at dissolved oxygen concentration of below 10% of air-saturation.
