Phenolic Compounds from Japanese Anise (Illicium anisatum L.) Leaves

  • Japanese anise (Illicium anisatum L.) leaves were collected and ground after drying, then immersed with 50 % aqueous acetone for 3 days. After filtration, the extracts were fractionated with n-hexane, chloroform ($CHCl_3$), ethylacetate (EtOAc) and $H_2O$, and then freeze dried after concentration. A portion of EtOAc (3.12 g) and $H_2O$ (6.08 g) soluble fractions were chromatographed on a Sephadex LH-20 column with various aqueous MeOH solution to isolate the compounds. Compound 1 ((+)-catechin) was isolated from EtOAc soluble fraction. Compounds 2 (quercetin), 3 (quercitrin) and 4 (2''-O-rhamnosylvitexin) were isolated from $H_2O$ soluble fraction. For the first time, quercitrin (3) and 2''-O-rhamnosylvitexin (4) of the isolated compounds were obtained from the extracts of japanese anise leaves.

Effects of Red Ginseng Extracts on Hydrogen Peroxide Production of Murine Prtitoneal Macrophages (홍삼추출물이 마우스 복강 대식세포 Hydrogen Peroxide 생산에 미치는 영향)

  • This experiment has conducted to evaluate whether single injection of red ginseng extract including 50% ethanol extract, crude saponin, and lipid soluble fraction can induce oxidative burst of mouse peritoneal macrophages with use of fluorescence spectrophotometer. To optimize conditions of fluorescent spectrophotometry, concentrations of DCFH-DA(2', 7' -dichlorofluorescin diacetate) was 1.6 ${\mu}{\textrm}{m}$ and control oxidative burst by Zymosan A and PMA(phorbol myristate acetate) were 100$\mu\textrm{g}$, 250ng, respectively. Though in vitro macrophages failed to induce increment of H2O2 production, but 50% ethanol extract group induced significant enhancement of H2O2 production when zymosan A triggered oxidative burst. On the other hand, lipid soluble fraction enhanced significantly H2O2 production than that of control group. These findings consisted with the other reports which showed ginsenosides inhibited nitric oxide production and lipid soluble fraction activated colony stimulating factor(granulocyte - monocyte) activity in bone marrow stem cells. As is well known, lipid soluble fraction contains phenol compound, polyacetylene compound and alkaloids. Further study would unravel which component of it can induce H2O2 production of macrophages. Antioxidative Activity of Prunus sargentii Outer Bark Extractives

  • The outer bark of Prunus sargentii was collected, air-dried and extracted with 70% aqueous acetone. Then it was successively partitioned with n-hexane, dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc) and $H_2O$. From the EtOAc soluble fraction, four compounds were isolated by the repeated Sephadex LH-20 column chromatography. The isolated compounds were determined as (+)-catechin (1), (-)-epicatechin (2), taxifolin (3), and neosakuranin (4) by the spectroscopic analysis including $^1H$, $^{13}C$-NMR, and 2D-NMR spectrometers. The antioxidative activities on the isolated compounds and the separated fractions were evaluated by DPPH radical scavenging assay. The crude, EtOAc, and $H_2O$ soluble fractions indicated good antioxidative potential compared to the $CH_2Cl_2$ and n-hexane soluble fractions.

Characteristics of Total Atmospheric Deposition by the Filtration-Sampling Method at Coal-Fired Power Plant Area (여과식 채취방법에 의한 대기오염 총침착물의 특성 -석탄화력발전소 주변지역을 중심으로-)

  • Total(=wet+dry) atmospheric depositions were collected by filtration-sampling method at 17 sampling sites of the coal-fired power plant area from September 1999 to January 2000. The soluble and insoluble fractions of deposition were also measured to investigate a suitability of simplified collection method for a long-term monitoring of total deposition. In the study, the 50% of sampled soluble fractions showed the electric conductivity (E.C.) of below 50 $\mu$S/cm and the 42% of them showed the lower pH than 5.0. The monthly mean fluxes of water soluble ionic components; S $O_4$$^{2-}$, C $l^{[-10]}$ , N $O_3$$^{[-10]}$ , N $a^{+}$, N $H_4$$^{+}$, $K^{+}$, $Mg^{2+}$, $Ca^{2+}$ were 168.4 kg/k $m^2$.month, 100.5 kg/k $m^2$.month, 88.6kg/k $m^2$.month, 31.3kg/k $m^2$.month, 25.6 kg/k $m^2$.month, 13.3 kg/k $m^2$.month, 8.7 kg/k $m^2$.month, 43.1kg/k $m^2$.month, respectively. The mean ionic concentration of all sample(n=79) was 314 $\mu$eq/ι, with contributions of 24.2% and 23.0% by [nss-C $a^{2+}$] and [nss-S $O_4$$^{2-}$]. The ratio of [N $O_3$$^{[-10]}$ ]/[nss-S $O_4$$^{2-}$] and [N $H_4$-C $a^{2+}$] were found to be 0.52 and 0.68, respectively.espectively.

Antioxidant Activity of the Extractives from the Needles of Picea abies Karsten (독일가문비(Picea abies Karsten) 잎 추출성분의 항산화 활성)

  • The dried needles (1.5 kg) of Picea abies Karsten were ground, extracted with acetone-$H_2O$ (7:3, v/v), concentrated, and fractionated with a series of n-hexane, methylene chloride, ethyl acetate and water on a separatory funnel. Each fraction was freeze dried, then a portion of ethyl acetate soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-n-hexane mixture as eluents. The isolated compounds were identified by cellulose TLC, $^1H$-, $^{13}C$-NMR, FAB and EI-MS. (+)-catechin (compound I), (-)-epicatechin (compound II), kaempferol-3-O-${\beta}$-D-glucopyranoside (compound III), 4-hydroxyacetophenone (compound IV) were isolated from the ethyl acetate soluble fraction and (+)-catechin (compound I), protocatechuic acid (compound V) were isolated from the $H_2O$ soluble fraction of P. abies needle. The antioxidative activities of each fraction and the isolated compounds were tested by DPPH radical scavenging method, and EtOAc soluble fraction, (+)-catechin and (-)-epicatechin showed similar values to ${\alpha}$-tocopherol and BHT as controls.

Phenylethanoid Glycosides from Seeds of Paulownia coreana

  • Seeds of Paulownia coreana were collected, extracted with acetone-$H_2O$ (7 : 3, v/v), concentrated under reduced pressure and successively fractionated with n-hexane, methylene chloride, ethyl acetate and water on a separatory funnel. The $H_2O$ soluble fraction was chromatographed on a Sephadex LH-20 column using aqueous methanol and ethanol-hexane as washing solvents. Two isomeric phenylethanoid glycosides, verbascoside (1) and isoverbascoside (2), and one epimeric phenylethanoid glycoside, campneoside II (3), were isolated and their structures were elucidated on the basis of chemical and spectroscopical data.

Inhibitory Effect of the Root of Coptis japonica on Catecholamine Biosynthesis in PC12 Cells

  • The effect of the root of Coptis japonica (COPT), both dichloromethane soluble $(CH_2Cl_2)$ and insoluble $(H_2O)$ fractions, on catecholamine contents and tyrosine hydorxylase (TH) activity in PC12 cells was investigated. $(CH_2Cl_2){\;}and{\;}H_2O$ fractions showed 21 and 53% inhibitions on dopamine content, respectively, at a ocncentraction of 40 .mu.g/ml in medium : the $(H_2O)$ fraction proveided a grateer inhibitory effect. The TH activity was reduced by the treatment of COPT ($(H_2O)$ fraction). These results suggest that COPT has an inhibitory effect on the catecholamine biosynthesis by the reduction of TH activity in PC12 cells.

Biolagical Activity on Extracts of Japanese Anise(Illicium Anisatum L.) Leaves and Twigs (붓순나무 잎과 가지의 추출물에 대한 생리활성 평가)

  • Japanese anise (Illicium anisatum L.) leaves and twigs were extracted with 50 % aqueous acetone three times. After filtration, the extracts were fractionated with n-hexane, chloroform, ethyl acetate and $H_2O$, and then freeze dried after condensation. Then antioxidation and antiviral activity were evaluated on each fractions. In the antioxidative activities, the results indicated high activity in the EtOAc soluble fraction of the leaves and the EtOAc and $H_2O$ soluble fractions of the twigs. It showed much higher antioxidative value compare to the controls, BHT and ${\alpha}$-tocopherol. In the antiviral activities, the all fractions were negative effects in HRV 1B and EV 71, but good in Influenza PR8. The activities of the crude extracts of the leaves and twigs showed more than 80% activity at the concentration of $10{\mu}g/mL$ and $50{\mu}g/mL$, respectively, and the activities of the EtOAc and $H_2O$ soluble fractions were close to 80%. Based on the above results, the extracts of Japanese anise may be applied for one of the natural biomass sources that can be used as an antioxidant and an antiviral substance.

Anti-oxidant Effect of the Ethyl Acetate Soluble Fraction of Sophorae Radix in H9c2 Cells (심근세포에서 고삼 에틸 아세테이트 분획의 항산화 효과)

  • To test the cytoprotective effect of sophorae radix (SR) against hydrogen peroxide (H₂O₂)-induced cytotoxicity, we investigated the cell viability using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay in the presence of ethyl acetate subfractions of SR water extracts In H9c2 cells. And to clarify the cytoprotective mechanism of SR extracts, we evaluated the cellular glutathione (GSH) contents in the presence of subfraction 1, 2, 3, and 4 of SR ethyl acetate soluble fractions. Among 1 -12 subfractions of SR ethyl acetate soluble fractions, 1, 2, 3 and 4 subfractions have an efficacy inhibiting the cytotoxicity induced by H₂O₂ in H9c2 cells. Also, the protective effects of 1, 2, 3 and 4 subfractions of SR ethyl acetate soluble fractions resulted from the anti-oxidant effects. These results suggest that ethyl acetate soluble fractions of SR water extracts is effective in the prevention of H₂O₂-induced cytotoxicity and 1, 2, 3 and 4 subfraction of ethyl acetate soluble fractions possess the anti-oxidant component.

Cytoprotective Effect of Organic Solvents Extracts of Sophorae Radix in H9c2 Cells (심근세포에서 고삼 유기용매 추출물의 항독성 효과)

  • To test the cytoprotective effect of sophorae radix (SR) against hydrogen peroxide (H₂O₂)-induced cytotoxicity, we investigated the cell viability using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay in the presence of methylene chloride, n-butanol, ethyl acetate and water soluble fraction of SR water extracts in H9c2 cells. These results were obtained as followed; H₂O₂ decreased the cell viability of H9c2 cells in a dose dependent manner. Cells pretreated with SR water extracts were protected the H₂O₂-induced decrease of viability in H9c2 cells. Among organic solvents fractions of SR water extracts, ethyl acetate soluble fractions of SR protected the decrease of viability induced by H₂O₂ in H9c2 cells. These results suggest that ethyl acetate soluble fractions of SR water extracts is effective in the prevention of H₂O₂-induced cytotoxicity.
