Effects of Dietary Fiber Extracted from Pumpkin (Cucurbita maxima Duch.) on the Physico-Chemical and Sensory Characteristics of Reduced-Fat Frankfurters

  • In this study, we investigated the effects of reducing fat levels from 30% to 25, 20, and 15% by substituting pork fat with water and pumpkin fiber (2%) on the quality of frankfurters compared with control. Decreasing the fat concentration from 30% to 15% significantly increased moisture content, redness of meat batter and frankfurter, cooking loss, and water exudation, and decreased fat content, energy value, pH, and lightness of meat batter and frankfurter, hardness, cohesiveness, gumminess, chewiness, and apparent viscosity. The addition of 2% pumpkin fiber was significantly increased moisture content, yellowness of meat batter and frankfurter, hardness, cohesiveness, gumminess, chewiness, and apparent viscosity, whereas reduced cooking loss and emulsion stability. The treatment of reduced-fat frankfurters formulated with 20 and 25% fat levels and with pumpkin fiber had sensory properties similar to the high-fat control frankfurters. The results demonstrate that when the reduced-fat frankfurter with 2% added pumpkin fiber and water replaces fat levels can be readily made with high quality and acceptable sensory properties.

Comparison of Seed Viability Among 42 Species Stored in a Genebank

  • This study was conducted to compare seed viability among 42 species after ten years of storage in the midterm storage complex ($4^{\circ}C$, 30-40% RH) at the National Agrobiodiversity Center (NAC) Korean genebank maintained by the Rural Development Administration (RDA), Republic of Korea and to suggest the relative seed longevity and suitable monitoring intervals. The germination data from initial tests and after ten years of storage were compared to measure changes in viability during storage. The decline in seed viability varied greatly among seeds from -11.5% for Triticum sp. to 80% for melon. Coriander, crowndaisy, safflower, cosmos, Chinesebellflower, waxgourd, melon, castorbean, Welch-onion, hollyhock, wild barley, and tallfescue showed significant decreases in viability of 34.2%, 73.4%, 36.5%, 30.0%, 40.2%, 71.3%, 80.0%, 65.9%, 45.5%, 51.4%, 53.0%, and 33.5%, respectively. Gardenpea, soybean, perilla, onion, wild rice, Italian-ryegrass, and pepper showed a 15-30% decline in viability, while the viability of morningglory, adzukibean, maize, and Capsicum sp. decreased by 15% to 5%. Chicory, radish, Chinese-cabbage, bottlegourd, watermelon, cucumber, pumpkin, Cucurbita sp., groundnut, kidneybean, clubwheat, sesame, wheat, Triticum sp., rice, barley, orchardgrass, buckwheat, and wild tomato showed changes in viability of <5%. The changes in storage viability also varied within families. The wild types of rice and barley showed rapid viability loss and presented different aspects from cultivars. Since seed viability of species, classified as index 1 or 2, showed germination losses >15% after ten years of storage, a viability test should be conducted with five year intervals, while species with germination loss of <15% (in index 3 or 4) can be retested at ten year intervals.

감자와 단호박에서 분리한 Verticillium tenerum의 동정 (Identification of Verticillium tenerum Isolated from Potato and Pumpkin in Korea)

  • 2003년 4월부터 11월에 수집한 감자와 단호박 시료에 발생하는 진균을 조사한 결과, 지금까지 우리나라에서 발생기록이 없었던 Verticillium tenerum이 검출되었다. 이 균은 강원 평창과 전북 김제지역에서 수집한 감자괴경 시료와 연천지역에서 수집한 단호박 과실 및 종자 시료에서 검출되었다. V. tenerum의 분생포자경은 주로 $3{\sim}5$개의 피알라이드를 윤생으로 형성하였다. 피알라이드는 플라스크형으로 가늘었으며 무색이었고, 크기는 $10{\sim}26{\times}2{\sim}5\;{\mu}m$이었다. 분생포자는 난형 또는 장타원형으로 크기는 $2.5{\sim}7.5{\times}2{\sim}4\;{\mu}m$였고 slimy head를 형성하고 연한 적갈색을 띠었다. 그리고 감자와 호박 시료 위에 적갈색을 띠는 독특한 균총을 형성하였다.

Human Immunodeficiency Virus Type I에 대한 수종 식물 추출물의 억제활성 검색 (Screening of Some Plant Extracts for Inhibitory Effects on HIV-1 and Its Essential Enzymes)

  • In order to elucidate the relationship between anti-HIV-1 enzyme activity and inhibition of HIV-1 replication by natural sources, extracts from some plants using the foods and oriental medicines were tested for inhibitory effects on the viral replication, reverse transcriptase (RT), protease and ${\alpha}-glucosidase$. In the anti-RT test, water extracts of Ficus carica (leaf), Houttuynia cordata (aerial part) and Ixeris tamagawaensis (aerial part) showed more than 79% inhitibion at a concentration of $100\;{\mu}g/ml$. The protease and ${\alpha}-glucosidase-inhibiting$ samples in the screening were water extract of Syringa dilatata (leaf) and methanol extract of Hibiscus syriacus (leaf and stem), which showed more than 40% inhibition at a concentration of $100\;{\mu}g/ml$. In the primary anti-HIV-1 test, water extracts of Equisetum arvense (aerial part), Hibiscus syriacus (leaf), Ixeris tamagawaensis (aerial part) and Pueraira thunbergiana (leaf) showed the potent inhibition against HIV-1 induced cytopathic effects.

First Report of Cucumber mosaic virus Infecting Pinewood Coneflower (Rudbeckia bicolor) in Korea

  • A virus isolate causing symptoms of yellow mosaic, fern leaves, malformation and plant necrosis on Rudbeckia bicolor was prevalent around Pyeongchang area in Korea. The causal virus was identified as Cucumber mosaic virus (CMV) using characteristics from biological, serological and molecular analyses and named as CMV-Rb. CMV-Rb caused mosaic on Nicotiana benthamiana, N. tabacum, Capsicum annuum, and Lycopersicon esculentum. However, typical local lesions did not develop on inoculated Pisum sativum, Cucurbita moschata, Datura stramonium and Tetragonia expansa plants. Full-length genome sequences of CMV-Rb RNAs 1, 2 and 3 were obtained using 12 primer pairs by RT-PCR analysis. The genome of CMV-Rb RNA segments 1, 2, and 3 consists of 3363nt, 3049nt, and 2214nt in length, respectively. In order to ascertain their taxonomic identity, nucleotide and the deduced amino acid sequence analyses RNAs 1, 2 and 3 of CMV-Rb isolates were conducted with previously reported sequences of CMV strains and/or isolates. CMV-Rb RNAs showed about 90 to 99% sequence identity to those of subgroup I strains suggesting that CMV-Rb is more closely related to CMV isolates belong to subgroup I. To our knowledge, this is the first report of CMV on Rudbeckia bicolor in Korea.

최소가공 단호박 (Cucurbita maxima Duchesne) 제품의 저장 중 품질 특성 (Quality Characteristics of Minimally Processed Sweet-pumpkin during Storage)

  • 각종 전처리 및 진공포장이 최소가공한 단호박 제품의 품질 특성에 미치는 영향을 조사하기 위하여 ascorbic acid, citric acid, NaCl, MgC1$_2$및 이들의 혼합조성물(1:1:1:1)을 슬라이스와 채썰기로 최소가공한 단호박에 처리하여 진공 포장한 후 저온저장 하였다. 최소가공 처리에 따른 저장 중 중량감소율은 모든 처리구에서 증가하는 경향을 나타내었으며 슬라이스 처리시 보다 채썰기한 경우 더욱 크게 나타났다. 한편 ascorbic acid 처리구는 전 시료 중에서 가장 큰 중량감소율을 나타내었으며 저장 10일경에 가장 현저한 중량감소율을 보였다. 갈변저해 효과는 슬라이스의 경우 ascorbic acid와 NaCl 및 MgC1$_2$로 처리한 구가, 채썰기의 경우 혼합조성물과 ascorbic acid가 가장 효과적이었다. 저장 중 과육의 경도는 증감을 반복하고 있으나 결론적으로 저장 후기에는 저장 초기에 비해 경도가 감소하였으며 절단 형태에 따라 다른 양상을 나타내어, 슬라이스의 경우 전처리를 행한 경우 오히려 경도가 증가하는 것을 볼 수 있으나 채썰기한 경우 전처리를 행할 경우 경도가 감소가 현저하였다. 최소가공 단호박의 호흡 양상을 살펴보면 이산화탄소의 발생량은 citric acid를 처리한 경우 가장 많았으며 이는 산소 소모량도 가장 높게 나타났다. 또한 ascorbic acid의 경우 저장 초기에는 호흡률이 높았으나 저장 후기로 갈수록 오히려 완화되는 경향을 나타내었다.

Nucleotide Sequence of Coat Protein Gene of Kyuri Green Mottle Mosaic Virus Isolated from Zucchini

  • The coat protein (CP) gene of kyuri green mottle mosaic virus zucchini strain (KGMMV-Z) isolated from zucchini (Cucurbita pepo) in Chonfu, Korea in 1999 was sequenced by the reverse transcription and polymerase chain reaction with degenerate and generate primers originated from tobamoviruses. The degenerate primers were very effective in amplification of KGMMV-Z CP region. The KGMMV-Z CP gene consisted of 486 nucleotides and had the same nucleotide length compared with those of cucurbit-infecting tobamoviruses. KGMMV-Z CP gene shared 43.8, 44.2, and 44.4% nucleotide sequence similarity with the CP gene of cucumber green mottle mosaic virus watermelon strain (CGMMZ-W), CGMMV-KW1, and CGMMV-SH, respectively, whereas three CGMMV strains among themselves showed 98.6-99.6% nucleotide similarity. The deduced amino acids of KGMMV-Z CP gene were 161 amino acid residues with the molecular weight of 17,181 daltons. The first 24 codons of KGMMV-Z CP gene corresponded to the sequences of the N-terminal amino acid of the viral capsid protein. The amino acid sequences of KGMMV-Z CP had 45.3% similarity compared with those of three CGMMV strains. However, the amino acid sequences of CGMMV strains were identical. These results showed that two cucurbit-infecting tobamovirus members, KGMMV-Z and CGMMV were genetically distantly related.

Resistance of Cucumber Grafting Rootstock Pumpkin Cultivars to Chilling and Salinity Stresses

  • Grafting using a pumpkin (Cucurbita sp.) rootstock is an effective way to improve cucumber (Cucumis sativus) resistance to a combination of chilling and salinity stresses. We evaluated the tolerance of 15 pumpkin cultivars to chilling, salinity, and combined stresses at the germination and seedling stages. Selected plant characteristics, including germination rate, germination potential, germination index, plant height, stem thickness, fresh weight, and dry weight, were analyzed. We used the unweighted pair group method with arithmetic mean for cluster analyses to determine the stress tolerance levels of the pumpkin cultivars. The 15 cultivars were divided into three clusters: tolerant, moderately tolerant, and susceptible to stress treatments. The stress tolerances of all cultivars were variable in the germination and seedling stages, and most cultivars were not tolerant to individual treatments of chilling or salinity stresses at both stages. These results suggest that identifying suitable cultivars for use as rootstock during cucumber grafting should involve the evaluation of stress tolerance during different growth stages. Additionally, cultivars tolerant to chilling stress may not be tolerant to salinity stress; therefore, the choice of pumpkin rootstock should depend on where the grafted plant will be grown. Cultivars tolerant to a combination of chilling and salinity stresses may be useful as rootstock for cucumber grafting. Our findings may serve as reference material for choosing appropriate pumpkin rootstocks for cucumber grafting.

Effects of Pumpkin (Cucurbita moschata Duch.) Leaf Ethanolic Extracts on Lipid Oxidation and Microbial Activity in Refrigerated Raw Ground Pork

  • This study was conducted to evaluate the antioxidant effects of pumpkin leaf extracted using a 50% ethanol on ground pork during storage. The pumpkin leaf extracts were added at concentrations of 0.05 (PE-0.05), 0.1 (PE-0.1), and 0.2% (PE-0.2) to ground pork, and 0.05% of ascorbic acid (As-0.05) was added as a control. Each sample was collected after 1, 4, 7, and 10 d of storage and the pH, total viable counts (TVC), conjugated dienes (CD), free fatty acids (FFA), and thiobarbituric reaction substance (TBARS) values were measured. The pH of the pork samples decreased until day 7, and then increased thereafter, except for the control and PE-0.05 sample. Lower CIE $a^*$ values were observed for pork samples containing PE relative to As-0.05 at increasing storage time (p<0.05). The addition of PE decreased the TVC, CD, FFA and TBARS values levels in the ground pork when compared to the control during 10 d of storage. These results indicate that PE can produce notable effects on meat products, such as inhibiting lipid oxidation and discoloration.

국내 닭의장풀에서 분리된 오이모자이크바이러스 분리주들의 외피단백질 유전자와 병징 다양성 비교 (Comparative Analysis of Genetic Variation of Cucumber Mosaic Virus from Commelina communis in Korea)

  • 춘천과 충주에서 퇴록현상과 모자이크 병징이 뚜렷한 닭의 장풀을 채집하여 바이러스 분리 동정하였다. 해당 바이러스는 RT-PCR과 RFLP 분석결과 CMV로 동정되었으며, 닭의장풀 CMV 분리주들은 서로 다른 서브그룹으로 존재하고 있는 것이 확인되었다. 외피단백질유전자의 염기서열을 분석한 결과, 각 CMV 분리주들은 CMV-Fny와 비교 하였을 때 CMV-Co와 96.3%, CMV-Co2는 96.3%, CMV-Co3는 95.9%의 유사성을 나타내었다. 또한 기주 반응 분석 결과 닭의장풀 분리주들은 대조바이러스(CMV-Fny)와 달리 C. pepo에서 전신 괴사병징이 특징적으로 나타났으며 Physalis angulata과 N.rustica의 경우 닭의장풀 분리주 간에 병징의 차이를 보였다. 이들 결과를 통해, 국내 닭의장풀에서 분리된 CMV들에서 유전적, 생물학적 다양성이 있는 것으로 확인되었다.
