Rapid separation of Capsicum annuum L. leaf extract using automated HPLC/SPE/HPLC coupling system (Sepbox system) and identification of α-glucosidase inhibitory active substances (자동화 HPLC/SPE/HPLC 시스템(Sepbox system)을 활용한 고추 잎 (leaf of Capsicum annuum L.) 추출물 분리 및 α-glucosidase 억제 활성 물질 탐색)

  • Phytochemicals include plant-derived natural products that promote and improve the human metabolism and physiological activity, and there is a lot of research to find the value of the molecules is in progress. Likewise, we obtained 288 fractions of Capsicum annuum L. extract in less than 20 h using HPLC/SPE/HPLC coupling experiment through Sepbox system, an effective separation system to search for active substances in natural resources and ensure efficacy and reliability. Therefore, this experiment allowed rapid identification of biologically active molecules from the extract compared to traditional separation processes. Of the above fractions, eight fractions showed the α-glucosidase inhibitory (AGI) activity and subsequent LC-MS analysis revealed one of the active molecules as luteolin 7-O-glucoside. In addition, we proved the increase in AGI activity according to deglycosylation of flavonoid glycoside. Therefore, this study suggests that the Sepbox system can quickly separate and identify active components from plant extract, and is an effective technique for finding new active substances.

Antioxidant and Antiobesity Activity of Natural Color Resources (천연색소 소재의 항산화 및 항비만 활성)

  • This study investigated the antioxidant and antiobesity activity of extract powders from the following natural color resources: Polygonum indigo, Black locust, Cochineal, Catechu, Grape, Tesu flower, Henna, Chrysanthemum, Sandalwood Red, Himalayan Rhubarb, and Madder. Total phenol content was the highest in Catechu extract, at 348.25 mg/g. DPPH, ABTS radical scavenging activity and ferric reducing antioxidant power (FRAP) were also higher in Catechu extract. Bleaching inhibition activity in the ${\beta}$-carotene linoleic acid system was the highest in Black locust extract, as was ${\alpha}$-Glucosidase inhibition activity. ${\alpha}$-Amylase inhibition activity was the highest in Catechu extract. Trypsin inhibition activity of Black locust extract was greater than 60%, and ${\alpha}$- chymotrypsin inhibition activity of Catechu extract was greater than 40%. Lipase inhibition activity was the highest Black locust extract, at 52.73%. Viability of 3T3-L1 cells was not affected by treatment with extracts at concentrations of $1.25{\sim}25{\mu}g/ml$. Lipid accumulation in the 3T3-L1 cells was the lowest following treatment with Catechu extract, at 55.8%, and this extract also inhibited adipocyte differentiation. These results suggest that the Catechu and Black locust extracts have high antioxidant and antiobesity activities and can be useful ingredients in functional foods.

  • In this study, we evaluated the polyphenol content, flavonoid content, antioxidant and biological of 70% EtOH extract and its fractions (dichloromethane, ethyl acetate and water) of the plant Chrysanthemum indicum L. (CI) belonging to family chrysanthemum. antioxidant activity was determined 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS), and superoxide scavenging activity. The biological activity tests included measurement of ${\alpha}$-glucosidase and PTP1B. The results show that content of polyphenol and flavonoid in 70% EtOH of CI 30.20 mg and 31.58 mg, respectively. Among the different samples, ethyl acetate (EA) fraction showed the highest level of polyphenol (87.87 mg) and flavonoid (65.12 mg). The DPPH, ABTS and superoxide dismutase(SOD) antioxidant assay of EA fraction showed 93.84%, 93.38% and 84.68% inhibition respectively. The EA fraction also showed the highest antioxidant activity compared to 70% EtOH extract and other fractions. EA fraction ($20{\mu}g/mL$) showed the highest 30.29% and 99.24%, respectively compared to other samples. The results of this study show that the EA fraction contained more antioxidant and antidiabetic components than 70% EtOH extract and other fractions. In conclusion, we believe that CI may be useful natural antioxidant and functional material.

The Effect of Genistein on Melanin Synthesis and In vivo Whitening (제니스테인의 멜라닌 생성 억제 및 In vivo 미백 효과)

  • The effect of genistein on melanin synthesis was studied using in vitro and in vivo model. Genistein inhibited melanin synthesis in cultured melan-a cells dose dependently. Tyrosinase activity was decreased by genistein treatment in melan-a cells, but genistein did not inhibit tyrosinase directly. Genistein did not affect the expression of tyrosinase in melan-a cells. Genistein inhibited the activity of ${\alpha}$-glucosidase in virtro and the glycosylation of tyrosinase in melan-a cells. The resulting unsaturated glycosylation of tyrosinase makes it unstable and disturb correct transportation. To further clarify the effect of genistein on the melanogenesis, we established UVB-induced hyperpigmentation on the shaved backs of brown guinea pigs. The animals were exposed to UVB radiation once a week for three consecutive weeks. Genistein (1 and 2%) or vehicle alone as a control were then topically applied to the hyperpigmented areas daily. Genistein showed significant lightening effect on the UVB-induced hyperpigmentation in five weeks. Depigmenting effect was prominent in 2% genistein treatment with Fontana-Masson staining. In conclusion, genistein may be a useful agent for skin whitening.

Inhibitory Effects of Maesaengi (Capsosiphon fulvescens) Extracts on Angiotensin Converting Enzyme and α-Glucosidase (매생이 추출물의 angiotensin converting enzyme 및 α-glucosidase 활성 저해 효과)

  • Physiological activities of hot water (MHW) and 80% ethanol (MEH) extracts from Maesaengi (Capsosiphon fulvescens) were investigated in this study. For the evaluation of antioxidant activities for MHW and MEH, 2,2-diphenyl-1-pic-rylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity were measured. DPPH radical scavenging activity and SOD-like activity of MHW, and MEH were increased weekly in a dose-dependent manner, and were about 10.8, 13.8, 62.4, and 27.1% at 10 mg/ml, respectively. The angiotensin converting enzyme (ACE) inhibitory activities of MHW and MEH were about 5.9% and 49.7% at 1 mg/ml, respectively. The ${\alpha}$-glucosidase inhibitory effect of MHW and MEH were about 1.4% and 67.3% at 1 mg/ml, respectively. To determine the influence of MHW and MEH on alcohol metabolizing activity, the generating activities of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were measured. Facilitating rates of ADH activity by MHW and MEH were increased weekly in a dose-dependent manner and ALDH activities were not detected. Elastase inhibitory activities of MHW and MEH were 75.9% and 51.2% at 10 mg/ml, respectively.

Biological Activity of Omija (Schizandra chinensis Baillon) Extracts (오미자(Schizandra chinensis Baillon) 추출물의 생리활성)

  • Extracts from Schizandra chinensis Baillon (Korean name: Omija) were tested for antioxidant and their inhibitory activities of ${\alpha}-amylase$ and ${\alpha}-glucosidase$. Total contents of phenolics were found as 4.35 mg/g (water extract)${\sim}$6.35 mg/g (60% ethanol extract). Electron donating ability (EDA), ABTS [2,2'-azinobis(3-ethyl-benzothiaznoline-6-sulfonic acid)] radical decolorization, antioxidant protection factor (PF) and thiobarbituric acid reactive substance (TBARS) were measured for the antioxidative activity of the extracts from S. chinensis. The water extract were determined as 97.5% at 200 ${\mu}g/ml$ while the activity of 60% ethanol extract were 96.2% at 200 ${\mu}g/ml$ in EDA. The 60% ethanol extract showed higher antioxidant activity than water extract when evaluated by ABTS radical decolorization, antioxidant PF and TBARS. ${\alpha}-Amylase$ inhibitory activity of water extract was similar with that of 60% EtOH extract. ${\alpha}-glucosidase$ inhibitory activities of water extract (97.4%) was higher than that of 60% ethanol extract (84.5%) at 200 ${\mu}g$/ml. The water extract from S. chinensis did not show an antimicrobial activity against Helicobacter pylori, but the 60% ethanol extract showed high antimicrobial activities such as 23 ${\pm}$ 1.6 mm of clear zone in 200 ${\mu}g/ml$ of phenolics. The result suggest that the water and 60% ethanol extract from S. chinensis will be useful as natural antioxidants and functional foods.

Anti-obese effects of mulberry (Morus alba L.) root bark through the inhibition of digestive enzymes and 3T3-L1 adipocyte differentiation (소화효소 저해 및 지방세포 분화 억제활성에 의한 상백피의 항비만 효능)

  • Anti-obese effects of mulberry (Morus alba L.) root bark was investigated in vitro by measuring its inhibitory effect against 3T3-L1 preadipocyte differentiation and digestive enzymes such as ${\alpha}$-amylase, ${\alpha}$-glucosidase and pancreatic lipase. Ethanol extract of mulberry root bark (MRE) showed the potent inhibitory activities on ${\alpha}$-amylase, ${\alpha}$-glucosidase and pancreatic lipase with $IC_{50}$ values of $7.86{\pm}0.36$, $0.12{\pm}0.03$ and $7.93{\pm}0.11mg/mL$, respectively. Furthermore, MRE significantly suppressed cellular lipid accumulation in 3T3-L1 cells in a dose-dependent manner. To elucidate the mechanism of MRE, we performed qRT-PCR and Western blotting for the expression of genes related with adipogenesis and lipogenesis. Treatment of MRE markedly suppressed the protein expression of $PPAR{\gamma}$, $C/EBP{\alpha}$ and SREBP-1c, as well as FAS and ACC, which are the key transcription factors and metabolic enzymes in adipogenesis and lipogenesis. In addition, qRT-PCR analysis indicated that the anti-adipogenesis effect of MRE might be due to its inhibition at transcription levels. These results demonstrate that MRE can effectively suppress adipocyte differentiation and inhibit key enzymes related to obesity. Our findings suggest that mulberry root bark may have a potential benefit in preventing obesity.

Anti-Diabetic, Alcohol Metabolizing Enzyme, and Hepatoprotective Activity of Acer tegmentosum Maxim. Stem Extracts (산겨릅나무 줄기 추출물의 항당뇨, 알코올 대사 효소 및 간 보호 활성)

  • This study was carried out to investigate the antidiabetic, alcohol metabolism, anti-inflammatory, and hepatoprotective effects of Acer tegmentosum extracts (ATE). A. tegmentosum has been traditionally used as a folk medicine to treat hepatic disorders. The antioxidative activities of ATE were measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide (SOD) assay. DPPH radical scavenging and SOD activities of ATE were about 89% and 82.9% at $0.5{\mu}g/mL$, respectively. Alcohol dehydrogenase and acetaldehyde dehydrogenase activities were 118.0% and 177% at 2 mg/mL, respectively. ${\alpha}-Glucosidase$ inhibitory activity of ATE was 75% higher at $50{\mu}g/mL$ and remarkably increased in a dose-dependent manner. Nitric oxide productions in macrophage RAW 264.7 cells stimulated by lipopolysaccharide was reduced to 16.7% by addition of ATE at 1 mg/mL. ATE showed significant protective effects against tacrine-induced cytotoxicity in Hep G2 cells at $100{\mu}g/mL$. Based on our results, we conclude that ATE may be used as a major pharmacological agent and anti-diabetic, anti-hepatitis, and anti-inflammatory remedy.

Biological Activities of Hot Water Extracts from Euonymus alatus Leaf (화살나무 잎 열수추출물의 생리활성)

  • This study was performed to investigate the biological activities of roasted (RE) and non-roasted (NRE) hot water extract samples from Euonymus alatus leaf by measuring DPPH radical scavenging, total polyphenol content, hydroxyl radical scavenging, superoxide dismutase (SOD)-like activity, anti-inflammatory activity, and inhibitory effects on ${\alpha}-amylase$ and ${\alpha}-glucosidase$ in vitro. The $IC_{50}$ values fur DPPH free radical scavenging activity of the NRE and RE hot water extracts were $19.1{\mu}g/mL$ and $21.9{\mu}g/mL$, respectively, and their total polyphenol contents were estimated as 9.6 mg/g for NRE and 10.6 mg/g for RE. Both NRE and RE scavenged the hydroxyl radical in a concentration-dependent manner, but their activities were lower than that of BHA. It was also shown that SOD-like activity was dependent on NRE and RE concentration, and the SOD-like activity of NRE was slightly higher than that of RE. The highest SOD-like activity obtained from NRE was 20% at a concentration of 3 mg/mL. Neither NRE nor RE seemed to have an effect on ${\alpha}-amylase$ and ${\alpha}-glucosidase$ inhibition. Finally, the hot water extracts of NRE and RE significantly decreased the concentration of LPS-induced NO in RAW 264.7 cells, indicating anti-inflammatory activity.

Anti-diabetic and Anti-oxidative Effects of Opuntia humifusa Cladodes (천년초 선인장 줄기의 항당뇨 및 항산화 효과)

  • To investigate the anti-diabetic and antioxidant effects of Opuntia humifusa cladodes, O. humifusa cladodes powder was extracted with 75% ethanol and fractionated with various solvents. Among the extracts fractionated with various solvents, ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibitory activities of O. humifusa cladodes were highest ($89.94{\pm}1.15%$ and $29.01{\pm}3.03%$, respectively) in the ethyl acetate fraction. Further, total phenolic and flavonoid contents of the ethyl acetate fraction were the highest ($196.02{\pm}5.26$ and $114.00{\pm}10.03{\mu}g/mg$, respectively). DPPH and ABTS radical scavenging activities increased according to the concentration of O. humifusa cladodes extract, and those of the ethyl acetate fraction were the highest. Ferric reducing antioxidant powers of chloroform and the ethyl acetate fraction were higher than those of other fractions. Overall, the ethyl acetate fraction of O. humifusa cladodes showed the highest anti-diabetic and antioxidant effects. Results indicate that O. humifusa cladodes powder has potential as a useful ingredient with anti-diabetic and antioxidant effects.
