Certain microalgae have been known to use light and various carbon sources to produce carbohydrates, mainly in the form of starch. This is one of the pertinent feedstocks replacing agricultural products for the production of bioethanol by yeast. This study focuses upon dilute acid hydrothermal pretreatments at low cost and high efficiency to compete with current methods, and employs Chlamydomonas reinhardtii UTEX 90 as the feedstock. With dry cells of 5%(w/v), the algal biomass was pretreated with sulfuric acid(1-5%) under temperatures from 100 to $120^{\circ}C$, from 15 to 120 min. As a result, the glucose release from the biomass was maximum at 58%(w/w) after pretreatment with 3% sulfuric acid at $110^{\circ}C$ for 30 min. This method enabled not only starch, but also the hydrolysis of other oligosaccharides in the algal cell in high efficiency. Arrhenius-type of model equation enabled extrapolation of some yields of glucose beyond this range. The pretreated slurry was fermented by yeast, Saccharomyces cerevisiae S288C, resulting in an ethanol yield of 29.2% from algal biomass. This study suggests that the pretreated algal biomass is a suitable feedstock for ethanol production and can have a positive impact on large-scale applied systems.
This research improved the growth potential of Bifidobacterium animalis subsp lactis strain JNU306, a commercial medium that is appropriate for large-scale production, in yeast extract, soy peptone, glucose, L-cysteine, and ferrous sulfate. Response surface methodology (RSM) was used to optimize the components of this medium, using a central composite design and subsequent analyses. A second-order polynomial regression model, which was fitted to the data at first, significantly lacked fitness. Thus, through further analyses, the model with linear and quadratic terms plus two-way, three-way, and four-way interactions was selected as the final model. Through this model, the optimized medium composition was found as 2.8791% yeast extract, 2.8030% peptone soy, 0.6196% glucose, 0.2823% L-cysteine, and 0.0055% ferrous sulfate, w/v. This optimized medium ensured that the maximum biomass was no lower than the biomass from the commonly used blood-liver (BL) medium. The application of RSM improved the biomass production of this strain in a more cost-effective way by creating an optimum medium. This result shows that B. animalis subsp lactis JNU306 may be used as a commercial starter culture in manufacturing probiotics, including dairy products.

Simultaneous saccharification and fermentation (SSF) experiments were carried out with a lignocellulosic biomass. The effects of temperature on enzymatic saccharification and the ethanol fermentation were also investigated. The batch SSF process gave a final ethanol concentration of 10.44 g/l and equivalent glucose yield of 0.55 g/g, which was increased by 67% or higher over the saccharification at 42℃. The optimal operating condition was found to vary in several parameters, such as the transmembrane pressure, permeation rate, and separation coefficient, related to the SSF combined with membrane system (semi-batch system). When the fermentation was operated in a semi-batch mode, the efficiency of the enzymes and yeast lasted three times longer than in a batch mode.
Cultures of Saccharomyces cerevisiae were subjected to the effect of PEF (pulse electric field) and a source of selenium. The culture period after which yeast cells were subjected to PEF treatment was optimized, as was the duration of the exposure. Optimization of the nutrient medium composition in S. cerevisiae cultures resulted in an over 1.8-fold increase in selenium accumulation with relation to cultures on the initial substrate. Optimization of the pH value and of culture duration resulted in selenium accumulation increase by approximately 78%. A significant correlation was found between the accumulation of selenium in yeast cells and its concentration in the culture substrate. The highest accumulation of selenium in the biomass of yeast, approx. $240\;{\mu}g/g$ d.m., was obtained after 15-min exposure to PEF on a 20-h culture. An approx. 50% higher content of selenium in cells was recorded, as compared with the control culture without the application of PEF.

The bed stability of Streamline DEAE (p = 1.2 g/mL) in a 20mm (i.d.) glass expanded bed contactor, and its performance on the recovery of glucose 6-phosphate dehydrogenase (G6PDH) from unclarified yeast homogenate were investigated. A residence time distribution study showed that a stable expanded bed was achieved. The theoretical plate and Bodenstein numbers determined were 25 and 53, respectively. A recovery yield of 87% and purification factor of 4.1 were achieved in the operation using 5% (w/v) biomass concentration feedstock. The performance of the anion exchange EBAC was still considerable good at a biomass concentration as high as 15% (w/v).

Baker's yeast was disrupted in a 1.4-L stainless steel horizontal bead mill under a continuous recycle mode using 0.3 mm diameter zirconia beads as abrasive. A single pass in continuous mode bead mill operation liberates half of the maximally released protein. The maximum total protein release can only be achieved after passaging the cells 5 times through the disruption chamber. The degree of cell disruption was increased with the increase in feeding rate, but the total protein release was highest at the middle range of feeding rate (45 L/h). The total protein release was increased with an increase in biomass concentration from 10 to $50\%$(w/v). However, higher heat dissipation as a result of high viscosity of concentrated biomass led to the denaturation of labile protein such as glucose 6-phosphate dehydrogenase (G6PDH). As a result the highest specific activity of G6PDH was achieved at biomass concentration of $20\%$(ww/v). Generally, the degree of cell disruption and total protein released were increased with an increase in impeller tip speed, but the specific activity of G6PDH was decreased substantially at higher impeller tip speed (14 m/s). Both the degree of cell disruption and total protein release increased, as the bead loading increased from 75 to $85\% (v/v)$. Hence, in order to obtain a higher yield of labile protein such as G6PDH, the yeast cell should not be disrupted at biomass concentration and impeller tip speed higher than $20\%(w/v)$ and 10 m/s, respectively.
단세포 단백질 자원으로 이용 가능한 균주로 알려진 Candida utilis ATCC 42416 균체를 생산하기 위하여 무를 파쇄하여 얻은 무즙을 배지로 균체배양 실험을 하였다. 무즙의 수용성 고형물과 총 당량은 각각 5.0~8.8 Brix와 3.5~6.5%였다. C. utilis ATCC 42416 균주를 무즙에 접종하여 $30^{\circ}C$에서 200rpm으로 진탕 배양하였을 때 24시간 이내에 증식이 끝났으며, 단위 소모당에 대한 최대 균체 생산량은 당농도 1% 회석 무즙에서 L당($5\;\times\;DCRI$) 21.5g의 건조균체를 생산하였다. 5배 회석무즙에 약간의 영양소 glucose(2.0%), yeast extract(0.2%), peptone(0.2%), ammonium sulfate(0.2%) 및 $KH_2PO_4(0.2%)$ 등을 각각 보강하여도 증식 속도, 균체생산 및 세포 단백질 함량 등에 차이가 거의 없어서 무즙자체가 C. utilis ATCC 42416 균체 생산에 좋은 기질이었음을 알 수 있다.
분리된 효모에 의한 aromatic ring compound의 전환반응 실험에 앞서 phenol 자화능에 대한 환경 인자의 영향을 조사하기 위하여 phenol을 유일한 탄소원으로 사용하여 분리된 효모균을 30시간, 3$0^{\circ}C$에서 진탕배양하여 균체증식량을 측정하였다. 1) pH3.5~4.5 온도 35$^{\circ}C$에서 가장 자화능이 우수하였으며 phenol의 처음 농도 1000ppm 까지 는 쉽게 이용할 수 있었고 2000ppm에서는 5 일간 배양시켰을 때 비로소 균체증식이 나타났다. 2) pH 5.0, 배양온도 3$0^{\circ}C$에서 본 균의 maximum specific growth rate는 0.27hr-1 이었으며, phenol 을 1000ppm씩 단계적으로 첨가하면서 phenol 소비율에 대한 균체수율 정수 Y값을 구한 결과 Y=3.2였다. 3) Yeast extract를 필수영양원으로 첨가할 때 0.005~0.01% 농도에서 가장 효과적이었으며, 또 산화환원소 효소 조해제인 HgCl$_2$, phenylhydrazine 을 명명 $10^{-5}$M, $10^{-3}$M을 배지내에 첨가하면 분리된 효모균의 증식을 거의 볼 수 없었다. 4) Phenol의 자화에 명종 실소원의 효과를 비교 해 본 결과 urea가 가장 효과적이었고, 이 효모균은 phenol 외에 catechol, resorcinol, benzidine 을 자화할 수 있다
목질계 바이오 매스 전처리에 사용되는 ionic liquid는 전처리 후 100% 회수되지 않아 잔존하는 ionic liquid의 독성이 직접적으로 미생물 균주의 생육에 나쁜 영향을 미쳐 에탄올 발효의 수율 및 생산성을 저해하는 문제를 가지고 있다. 본 연구에서는 ionic liquid에 저해를 받지 않으며 높은 ethanol 생산 효율을 가진 균주를 얻고자 유도적 돌연변이 유발 실험을 진행하였다. 선별된 돌연변이 균주 D452-B2와 D452-S3는 3% [EMIM][Ac]가 포함된 배지에서 glucose 소비속도는 $4.5g{\cdot}l^{-1}{\cdot}h^{-1}$와 $4.4g{\cdot}l^{-1}{\cdot}h^{-1}$로 모균주인 S. cerevisiae D452-2 균주에 비해 6배 가량 증가하였으며, ethanol 생산성은 각각 $1.99g{\cdot}l^{-1}{\cdot}h^{-1}$와 $2.0g{\cdot}l^{-1}{\cdot}h^{-1}$로 27배 가량 증가하였다.
전분질로부터 효모균체를 직접 생산하기 위하여 분리 동정된 전분이용성 효모, Sp. holsaticus FRI Y-5의 기본적인 배양조건에 대하여 검토하였다. 이 균주는 산소를 절대적으로 요구하며, 배지의 초기 pH가 6.9일 때 최대균체량 7.2 g/l, 균체수율 0.27, 비성장속도 $0.14hr^{-1}$로 가장 좋은 생육현상을 보여 주었고 배양온도는 $23^{\circ}C$가 적온이었다. 균체생육의 활성화 에너지 Ea는 17.33 kcal/mole로 산출되었다. 검토된 13종의 질소원 가운데 $(NH_2)_2CO$가 3일 배양후 균체량이 10.69 g/l로 가장 많았으며 이때 균체수율은 0.451이었다. $SO_4\;^{-2}$염으로서 $Mg^{+2},\;K^+$ 및 $Zn^{+2}$와 유기영양원으로서 yeast extract도 균체증식에 좋은 영향을 주는 것으로 판명되었다.
