참외 (Cucumis melo L. var makuwa Makino)의 물과 에탄올 추출물의 항산화 및 항균효과 (Antioxidant and Antimicrobial Effects of Extract with Water and Ethanol of Oriental Melon (Cucumis melo L. var makuwa Makino))

  • 본 연구에서는 참외의 부위별 물 및 알코올 추출물의 항산화효과, ${\alpha}$-Glucosidase 저해활성 및 항균효과를 조사하였다. 총 phenol 함량을 측정한 결과 물 추출물에서는 껍질이 151.64 ${\mu}g/g$, 에탄올 추출물에서는 껍질이 224.77 ${\mu}g/g$로 가장 높은 페놀함량을 나타내었다. 총 flavonoid 함량은 껍질이 물 추출물의 경우 45.53 ${\mu}g/g$, 에탄올 추출은 338.37 67.16 ${\mu}g/g$로 가장 높게 나타내었다. 참외의 항산화 효과는 물 추출물의 경우 1%에서 껍질이 25%로 가장 높았으며, 에탄을 추출은 1%에서 껍질이 83.3로 가장 높은 전자공여능을 나타내었으며, 과육과 태좌는 20% 미만의 낮은 활성을 나타내었다. ABTS는 물 추출물은 1%에서 껍질이 79.2%로 가장 높았으며, 과육이 57.6%, 태좌가 74.0%로 50%이상의 활성을 나타내었고, 에탄올추출물은 99.9%로 껍질이 가장 높게 나타났고, 과육과 태좌는 52.1%, 41.2%의 활성을 보였으며, Xanthine oxidase의 저해활성의 경우 1%에서 껍질이 물 추출물이 29.3%, 에탄올추출물이 30.5%의 가장 높은 활성을 보였지만, 비교적 낮은 저해율을 나타내었다. ${\alpha}$-Glucosidase 저해활성을 측정한 결과 1%에서 열수 추출물은 25.9%, 에탄올 추출물은 37.5%의 활성을 보인 껍질이 가장 높은 활성을 보였다. 또한 Streptococcus agalactiae, Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Staphylococcus epidermidis 및 Salmonella typimimum에 대한 항균활성을 평가한 결과 참외의 에탄올 추출물에서 Streptococcus agalactia에서만 껍질, 과육, 태좌에서 각각 15, 13 및 12mm의 clear zone를 나타내었다.

추출온도에 따른 이팝나무 과육 물 추출물의 항산화 및 항노화 활성 (Antioxidant and anti-aging activities of water extracts from Chionanthus retusus flesh according to different extraction temperatures)

  • 민간에서 식용 및 약용으로 사용되고 있으나 최근에는 활용도가 낮은 이팝나무 성숙열매 과육의 산업적 활용 증진을 위하여 상이한 추출조건(실온 $25^{\circ}C$, $80^{\circ}C$, $110^{\circ}C$)에서 물을 용매로 추출된 이팝나무 과육 3가지 추출물에 대한 항산화 및 항노화 활성을 측정하였다. 총 폴리페놀 화합물은 환류추출기를 이용한 80W에서 72.71 mg/g으로 가장 많이 함유하였으며, 플라보노이드 화합물은 가압열수 추출기를 이용 110W 추출물에서 7.608 mg/g을 함유하였다. 수용성 단백질과 환원당의 함량도 110W에서 각각 4.93 mg/g과 46.77 mg/g으로 가장 많이 함유하였다. DPPH radical 소거율은 1.0 mg/mL에서 모든 추출물이 92% 이상의 소거율을 보였으며, ABTS radical 소거활성은 110W가 88.67%였으며, 80W(87.49%)와 유의적 차이는 없었다. 아질산염 소거능은 pH 1.2의 조건에서 110W가 66.16%였으며, pH 3.0에서는 80W에서 37.86%를 나타내었다. SOD 유사활성 및 xanthine oxidase 저해활성에서도 추출온도가 가장 높은 110W에서 각각 14.95%와 59.45%의 활성을 나타내었다. 추출온도에 따른 이팝나무 과육의 미백효과를 알아보고자 tyrosinase 저해율을 측정한 결과, 환류추출기를 이용한 80W 추출물이 35.31%로 비교적 높은 저해율을 나타내었다. Collagenase에 대한 저해효과는 1.0 mg/mL에서 110W가 37.78%로 가장 우수하였으며, elastase 저해활성은 80W 20.39%, 110W 20.33%로 유사한 저해효과를 나타내었다. 모든 실험에서 농도 의존적으로 항산화 및 항노화 활성이 증가하는 경향을 보였으며, 실온보다는 높은 온도에서 추출한 이팝나무 과육 추출물에서 유용성분의 함량과 생리활성이 높게 나타났다. 이는 추출온도가 높을수록 유용성분의 추출이 용이해져 다양한 항산화 및 항노화 활성을 나타내는 것으로 사료되며, 이팝나무 과육은 기능성 식품 및 제품을 위한 천연소재로 활용 가능할 것으로 판단된다.

산벚나무(Prunus sargentii R.) 수피의 화장품활성에 관한 연구 (A Study on the Cosmeceutical activities of Prunus Sargentii R.)

  • 산벚나무는 장미과에 속하는 것으로, 수피인 화피의 radical 소거능과 항염증 효과가 보고되고 있어, 이를 이용하여 화장품 원료로 이용하고자 생리활성을 검증하였다. 전자공여능은 에탄올추출물 500 ppm에서 92%의 효능을 보여 열수추출물 59% 보다 우수하였고, SOD 유사활성의 경우 열수추출물 1,000 ppm에서 에탄올추출물의 경우는 50%의 활성능을 나타내었다. xanthine oxidase 저해능은 500 ppm의 농도에서 열수추출물의 경우 40%, 에탄올추출물의 경우 60%의 저해능을 나타내었다. 지방산패 억제능 측정의 경우 $Cu^{2+}$에 의한 저해 효과는 열수추출물보다 에탄올추출물의 효과가 높았으며, $Fe^{2+}$에 의한 저해 효과는 열수와 에탄올추출물 모두 500 ppm에서 80% 이상의 저해능을 보였다. 피부미백과 관련된 tyrosinase 저해능의 경우 1,000 ppm에서 열수추출물의 경우는 26%, 에탄올추출물의 경우 20%의 저해능을 보였다. 항염증과 관련된 hyaluronidase 저해능의 경우 500ppm에서 에탄올추출물의 경우 96%의 효능을 보였다. 산벚나무 수피추출물의 피부상재균인 Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli 및 Propionibacterium acne에 대한 저해환형성을 관찰한 결과 열수추출물보다 에탄올추출물의 효능이 더 우수하였다.

비쑥 추출물에 대한 항산화 활성 및 염증억제 효과 (Anti-oxidant Activities and Anti-inflammatory Effects on Artemisia scoparia)

  • We investigated the antioxidant activities such as DPPH radical scavenging capacity, xanthine oxidase inhibitory activity, and superoxide radical scavenging capacity of the aqueous EtOH extract and its solvent fractions of Artemisia scoparia. The ethyl acetate fraction showed high antioxidant activity, compared to positive controls such as ascorbic acid, butylated hydroxy anisole (BHA), trolox, and allopurinol in these assay systems. Moreover, we examined the inhibitory effect of solvent fractions of A. scoparia on the production of pro-inflammatory factors that the nitric oxide (NO), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and prostaglandin E2 $(PGE_2)$ production activated with LPS $(1{\mu}g/ml)$ in murine macrophage cell line RAW264.7. The amounts of protein levels were determined by immunoblottting. Tn the sequential fractions of hexane and dichloromethane inhibited the NO and $PGE_2$ production and the protein level of iNOS and COX-2. These results suggest that A. scoparia may have anti-inflammatory activity through the antioxidant activity and inhibition of pro-inflammatory factors.

백작약 열수추출물의 피부노화 억제 효과 (II) -주름형성 억제 효과를 중심으로- (Inhibitory Effects of Peonia japonica Water Extract on Skin Aging (II) - Focussed on Inhibitory Effects of Wrinkle Formation-)

  • To investigate the inhibitory effects of Peonia japonica water extract(PJWE) on skin wrinkle formation, skin wrinkles were induced by both the irradiation of UVB and the application of squalene monohydroperoxide to the backs of hairless mice for 4 weeks. And at the same time experimental materials were applied topically. Wrinkles for the control (C) group were formed as a pattern of deep furrows and thick crests. Whereas wrinkles for the positive control (PC, 0.01% retinoic acid) and experimental(E, PJWE) groups were formed as a pattern of shallow furrows and thin crests, which were similar to that of the normal(N) group. Collagen and elastic fibers in dermis of the PC and E groups were almost intact with a regular arrangement, which were similar to those of the N group. The activity of xanthine oxidase, the free radical generating enzyme, was significantly lower in the E group than the C and PC groups. The activities of superoxide dismutase and catalase, the free radical scavenging enzymes, were much higher in the E group than the C and PC groups and similar to the N group. As for the amount of matrix metalloproteinase-3(MMP-3) expression, PC and E groups were significantly lower than the C group. Therefore, PJWE could be very effective natural herbal material for the inhibition or improvement of wrinkle formation in hairless mice skin.

S-Thiolation and Oxidation of Glycogen Phosphorylase b and Peroxidation of Liposome Initiated by Free Radical Species

  • The relationship of S-thiolation and oxidation of glycogen phosphorylase b and peroxidation of phosphatidyl choline liposome by xanthine oxidase (XOD), 2,2'-azobis(2-amidinopropane) hydrochloride (AAPH), and 2,2'-azobis(dimethylvaleronitrile) (AMVN)-generated free radicals was investigated, Glycogen phosphorylase b was S-thiolated in the presence of glutathione and oxidized in the absence of it by XOD, AAPH and AMVN. In XOD-initiated reaction, the rates of S-thiolation and oxidation of phosphorylase were very similar and addition of liposome to the reaction mixture showed little inhibition of the modifications. In AAPH-initiated reaction, the rate of oxidation was higher than that of S-thiolation and addition of liposome increased oxidation of the protein but had no effect on S-thiolation. In AMVN-initiated reaction, S-thiolation was higher than oxidation and addition of liposome increased S-thiolation remarkably but showed no effect on oxidation. The effect of liposome on modifications of protein in AAPH and AMVN reaction seemed to be caused by certain reactive degradation products or intermediates of liposome by free radical attack. Peroxidation of liposome was not observed in XOD-initiated reaction. Liposome was gradually peroxidized by AAPH reaction. The peroxidation was inhibited by addition of GSH and phosphorylase. Peroxidation of liposome by AMVN was extreamly fast, and was not affected by GSH and phosphorylase.

감초와 향신료 물추출물의 향균 및 항산화능 (The Antibacterial and Antioxidative Activity of Licorice and Spice Water Extracts)

  • The purpose of this research related to the development of natural preservatives, in which licorice and spices (clove, fennel fruit and Chungyang green pepper) were extracted with distilled water, and the extracts were tested for their antibacterial and antioxidative activities. The polyphenol contents of the water extracts from licorice, clove, fennel fruit and Chungyang green pepper were 17.4, 21.4, 6.6, and 0.9 mg/g, respectively. The water extracts from licorice and clove demonstrated antibacterial activity against S. aureus. The electron donating abilities (EDA) of the water extracts from the licorice and other spices ranged from 60 to 88% at 1,000 ppm; the highest value was for the licorice followed by fennel fruit, clove, and green pepper. The xanthine oxidase inhibition ratio (XOD) of the extracts ranged from 28 to 50% at 1,000 ppm, where the highest value occured in the cloves, followed by fennel fruit, green pepper, and licorice. The superoxide dismutase (SOD)-like activity ranged from 33 to 53% at 1,000 ppm, and the highest value was for the licorice followed by cloves, fennel fruit, and green pepper. The nitrite scavenging abilities (NSA) at 1,000 ppm of the clove and fennel fruit water extracts were 95% and 65% at pH 1.2, respectively. The NSAs of the extracts were highest at pH 1.2, and decreased with increasing pH. Considering all the obtained results, we have concluded that licorice and spice extracts can be used as natural preservatives in the development of health foods.

산벚나무 수피추출물의 생리활성 및 페놀성 화합물 분리 (Biological Activity Investigation, and Phenol Compounds Isolation from Barks of Prunus Sargentii R.)

  • Prunus sargentii of Rosaceae familiy, has been reported to have radical scavenging activity and anti-inflammatory effect. On these facts, this research was conducted to evaluate pharmaceutical activities of the bark extracts P. sargentii. Free radical scavenging activities of fraction (Fr) -5${\sim}$10 isolated from P. sargentii was higher than 80% respectively at 10ppm. The superoxide dismutase (SOD)-like activity of Fr-5, 9 were about 97, 84%, respectively at 1,000 ppm. Xanthine oxidase inhibitory effect of Fr-9, 10 were about 75, 78%, respectively at 1,000 ppm. The tyrosinase inhibitory effect related to skin-whitening was 72, 68% in Fr-2, 9 isolated from Prunus sargentii R. at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 98% for Fr-8 at 500 ppm. Isolation of the methanol soluble fraction from P. sargentii yielded two major phenol compounds, (-)-epicatechin and taxifolin. The structure of the compound was certainly determined by chemical analyses, as well as NMR and Mass spectroscopy. The present study was carried out in a search for new cosmetic material from the bark from P. sargentii. and, (-)-epicatechin and taxifolin were isolated as active principles. So P. sargentii R. methanolic extracts may be used for the cosmetic material.

노회(蘆薈)(알로에), 자화지정(紫花地丁)의 항산화, 항염증, 주름, 미백에 미치는 영향 (Effects of Aloe and Violae herba Extract on the Anti-oxidant, Anti-inflammatory, Anti-wrinkle and Whitening)

  • Objective : This study was performed to assess the effects of Aloe and Violae herba extracts on skin disease and skin beauty. Methods : Anti-oxidant effects were measured by the scavenging for DPPH radical, xanthine oxidase activity. Anti-inflammatory effects were examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-$\alpha$, NF-kB, COX-2, MAP kinase. The skin wrinkle formation effects were measured by collagenase and elastase activities. The whitening effects were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Aloe and Violae herba extracts showed high radical scavenging activity. 2. In an anti-inflammatory test, Aloe and Violae herba extracts strongly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from the RAW 246.7 macrophage cells. Aloe and Violae herba extracts also inhibited the LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effects of Aloe and Violae herba extracts on macrophage activation were via the inhibition of NF-kB, evidenced by transient transfection assay. Furthermore, Aloe and Violae herba extracts weakly inhibited the activation of Jun-N-terminal kinase(JNK) but they did not have any effects on p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Aloe extract strongly inhibited collagenase and elastase, whose activity are tightly related with the wrinkle formation. 4. In the skin whitening assay, Aloe and Viloae herba extracts weakly inhibited tyrosinase activity, however, it was not statistically significant. Besides they did not have any effects on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : This study show that Aloe and Violae herba extracts may play a significant role in skin disease and skin beauty.

침향 추출물의 면역조절 및 생리활성 분석 (Evaluation of Immunomodulatory and Biological Effects of Aquilaria crassna Extracts)

  • Objectives : Aquilaria crassna is a traditional herbal medicine, which is used to treat allergies, diabetes, neurological diseases. Recently, Aquilaria crassna extracts have been reported in anti-bacterial and anti-inflammatory activities. In this study, various solvents fraction of Aquilaria crassna were investigated on various physiological activities. Methods : According to the polarity, the solvents fraction of Aquilaria crassna were confirmed through TLC, and the activities of the extracts were confirmed in anti-diabetes, anti-obesity, whitening, anti-gout, and anti-inflammation. Results : TLC results showed that ACM and ACM/E have similar patterns and most of the components were transferred to ACM/E. Treatment with ACM and ACM/E fraction were significantly decreased the generation of NO in lipopolysaccharide (LPS)-stimulated macrophage cells. Analysis of biological activities such as α-glucosidase, protein tyrosine phosphatase (PTP1B), tyrosinase, xanthine oxidase (XO) and pancreatic lipase inhibition, showed that ACM and ACM/E have more inhibitory effects than other fractions. Conclusions : Therefore, the results of the present study clearly demonstrate that Aquilaria crassna and its constituents might be beneficial in the prevention or treatment of immune-regulating effects.
