A Study on Filtration Effect of Anaerobic Digestion Wastewater by Composition of Filtration Layer Materials (퇴비단 여과재 구성에 따른 혐기소화액 여과 효과)

  • This study was carried out to compare the filtration effect of anaerobic digestion waste water by compost filtration bed. Three types of filtration systems were set for filtration test following to composition of bedding materials. Two filtration systems were filled with sawdust, rice-hull and wood-chip. And the other was filled with sawdust and rice-hull. Anaerobic digestion waste water was applied evenly on the surface of the filtration bed with spraying nozzle. The concentration of nitrogen, biochemical oxygen demand (BOD), suspended solids (SS) of anaerobic digestion waste water was decreased by filtration treatment. The total nitrogen content was decreased from 1,313 mg/L to 1,090~1,293 mg/L and BOD concentration was decreased from 529.7 mg/L to 125~162.3 mg/L, respectively. The SS content was decreased from 69~79% by filtration process. In addition, the electrical conductivity (EC) was decreased from 10.22 ds/m to 7.91~8.31 ds/m.

Effect of Combining Wood Particles and Plastic(Polypropylene) Screen on the Physical and Mechanical Properties of Board (목재(木材)파이티클과 플라스틱(폴리프로필렌) 망(網)의 결체(結締) 보오드의 물리(物理) 및 기술적(機械的) 성질(性質)에 미치는 영향(影響))

  • As a way for the effective utilization of pallman chips and sawdusts, these furnish materials were combined with non-woody material of plastic (polypropylene) screen in board manufacturing to improve their weak physical and mechanical properties. The conventional boards were made with conditions of specific gravity 0.40, 0.55, 0.70, and 0.85, resin content 8, 10, 12 and 14%, and number of polypropylene screen 1, 2, 3 and 4, and press-lam boards were also manufactured. The physical and mechanical properties were measured and discussed on thickness swelling, bending modulus of rupture and elasticity, tensile strength, internal bond strength, and screw holding strength. The results obtained at this study were summarized as follows: 1. In thinckness swelling both of pallman chip board and sawdust board were improved by the increase of resin content, and press-lam boards showed lower thickness swelling than conventional boards. 2. Both the modulus of rupture and elasticity were increased with the increase of specific gravity, and press-lam boards showed higher modulus of rupture and elasticity than conventional boards. On the other hand, modulus of rupture was increased with the increase of number of polypropylene screen and resin content whereas these effects in modulus of elasticity was not recognized. 3. Tensile strength was increased with the increase of specific gravity, and the boards combined with polypropylene screen showed higher tensile strength than control boards. Also tensile strength was increased with the increase of number of polypropylene screen, and press-lam boards revealed higher tensile strength than conventional boards. 4. Internal bond strength was increased with the increase of specific gravity, and the boards combined with polypropylene screen were lower in internal bond strength than control boards. Also, the boards combined with odd number of polypropylene screen showed lower internal bond strength than those combined with even number of polypropylene screen. 5. Screw holding strength was increased with the increase of resin content and specific gravity but significant difference was not approved between boards combined with polypropylene screen and control boards. In press-lam boards, pallman chip boards of higher specific gravity but sawdust boards of lower specific gravity showed better screw holding strength than control boards.

Mycelial growth and wood decaying enzymatic activity analysis by various addition rates of oak powder in the liquid spawn of Lentinula edodes (참나무분 첨가에 따른 표고 액체종균의 균체생산 및 효소 활성)

  • This study was carried out to establish a suitable method for liquid spawn production from Lentinula edodes. The optimum production of liquid spawn (OLS) was achieved using soybean meal medium (SMM) with 0.3% of 850 um oak powder and 10-day incubation period and 0.6 vvm aeration volume. OLS showed activities of laccase on ABTS agar plate and carboxymethyl cellulase (CM-cellulase) on CMC agar plate. In case of liquid spawn, fruiting-body development period was delayed approximately 1 day compared to that of sawdust spawn, however, the yield of 153 g per 1.2 kg polypropylene bag was similar to that of sawdust spawn.

Culture of Tricholoma matsutake Mycelium using Solid Matrix (고체 매질을 이용한 송이(松栮)(Tricholoma matsutake)균 배양)

  • This study was conducted to develop optimal solid culture medium for Tricholoma matsutake. As the solid matrix, granitic soil, perlite, vermiculate, pine sawdust and peat moss were compared regarding their effected on mycelial growth. Ergosterol content which is a fungal wall component was used as the growth index of the mycelia. Among the various solid matrixes, the granitic soil, perlite and mixture of the two supported the growth most. Barely flour appeared to be very effective on the stimulating of the mycelial growth when added to the solid matrix. An mixture of the matrix contained an even (1:1:1:1, v/v/v/v) mixture of granitic soil, perlite, vermiculate and pine sawdust. T. matsutake started growth 2 weeks after inoculation and reached stationary growth phase after 8th weeks in the solid matrix mixture. The mycelial density in the solid matrix was 7 times higher than that in fairy-ring soil. In addition, 30~70% water content and 10% humus soil in the solid matrix also supported good growth suggesting that T. matsutake needs humus soil for a nutrient sources. The solid matrix developed in the present study could be used to study physiological characteristics of T. matsutake as well.

  • The mycelial mass of K. mutabilis greatly increased at pH 5.5~6.0 but decreased pH 6.0. The linear mycelial growth wsa mostly supported on sawdust of Quercus accutisima and the mycelial density wsa high on sawdust of Q. accutisima and corn cob. Much mycelial distribution could be showen in ray parenchyma cell and ray tracheid. Severe degradation of ray parenchyma cell was found but little degradation of ray tracheid cell was found. The dry weight loss wsa 5.9% after agar-block test. And the pH wsa acidified from 6.07 to 4.31 and hot water extractives was decreased after degradation of Q. serrata sawdust by K. mutabilis.

  • Experiments were carried out to pursue the availability and the feasibility of utilizable cellulosic materials as substrate for the production of cellulosic single cell protein. The resuluts were obtained as fellows. 1. Effects of carbolydrates as a sole carton source on the growth of Cellulomonas flavigena KIST 321 were examined. The result showed that cellulose and xylose would be most utlizable for cell mass production. 2. Alkaline treated waste papers and clothes resulted in good growth of the organism than intact ones did. However the waste papers as substrate of cellulosic fermentation were not digestible, even if the meterial was treated with alkalies. 3. Rice straw, rape straw and panic grass appeared to be good substrates for the cell mass production. 4. Leaves were proved to be a good substrate for the cell mass production, but wood sawdust was hardly digested by merely alkaline treatment. 5. When cellulosic wastes as the substrate were examined into the concentration of alkaline solution, the result suggested that the best productivity of cell mass from cellulosic materials was obtained on treatment with 0.8∼1.0% NaOH solution. 6. The productivity of cell mass was increased by washing out with water after alkaline treatment of newspaper, pine sawdust, lime sawdust and pine leaf.

Investigation of Harmful Microorganisms and Physiological Characteristics of Mycelia in the Bed-log of Shiitake Strains Showing Low Fruit-body Productivity (버섯 발생이 부진한 표고골목 내부균사의 생리적 특성 및 해균 조사)

  • Attempts were made to investigate the conditions of mycelia of one low-temperature type strain and one high-temperature type strain of shiitake showing poor fruit-body formation in bed-log, and to survey harmful microorganisms formed on the log surface in Gapyung County, Korea. When tested the growing ability of mycelia, the low-temperature type strain showed ca. 1.1% decrease compared with preserved original strain. And, the high-temperature type one showed ca. 8.0% decrease. The growth of isolated mycelia was tested in sawdust medium. The high-temperature type strain showed ca. 10.8% decrease compared with original strain, and the low-temperature type one showed ca. 25.1% decrease. Weight reduction rate was investigated. The high-temperature strain showed ca. 20.1% decrease and the low-temperature one ca. 19.0%. When compared with non-treatment, original high-temperature type strain showed 107.0% decrease, the isolated high-temperature type strain 49.5%, original low-temperature type one 85.4%, isolated low-temperature type one 50.0%. As the results of confrontation culture, the high-temperature type strain and the low-temperature type one were same as the original ones, respectively. And, in the bed-logs, Hypoxylon truncatum, Coriolus versicolor, Inonotus xeranticus, Daedaleopsis tricolor, Graphostroma platystoma, two species of Myxomycetes, Trichoderma sp. Hypoxylon fragiforme, H. howeianum, and Nitschkia confertula were observed as harmful microorganisms, and the bed-logs were not in good condition.

Surface Temperature of Flooring Board Using Woodceramics with Sending an Electric Current (우드세라믹 통전에 의한 마루판의 표면온도 변화)

  • To examine possibilities to make use of woodceramics for the lower material of hot-floored ondol boards, woodceramics made from sawdust board with Pinus densiflora, put them in a wood frame and concrete block and heated them sending an electric current, and measured changes in surface temperature of a laminated floor board and a plywood floor board. As the sending an electric current to time passed, their surface temperature increased rapidly up to 20 minutes, and then gradually ascended. In terms of the surface temperature in the wood frame, the beginning temperature of a plywood floor board was higher than that of laminated floor one; however, as time passed, a laminated floor board's temperature rose, and after 60 minutes, it showed similar to the temperature of a floor board of lamination. On the other hand, the surface temperature in a concrete block showed that the laminated floor board was higher than the plywood floor board in both early and 60-minute later temperatures. With the lapse of time after switching off, the surface temperature of floor boards slowly dwindled up to 9 minutes, and from that time on, began to drop sharply. In terms of the descent speed of surface temperature, when woodceramics' surface temperature was adjusted at $70^{\circ}C$, the laminated floor board was the highest; when it was at $80^{\circ}C$, the plywood floor board was the highest, resulting in rapid descent speed of heat.

Cultural Characteristics of New Cultivar of Lentinula edodes, Poongnyunko (표고 신품종 풍년고의 배양 특성)

  • In this study, a new cultivar of Lentinula edodes was bred from dikaryotic KFRI 490 and monokaryotic KFRI 536 by Di-mon method. The new breed was named "Poongnyunko" and physiological characteristics was investigated. Its optimal growing temperature was $25^{\circ}C$, and it had thin density. Its colony was colorless and did not form any epithelium. On the $7^{th}$ day of mycelial growth, "Poongnyunko" was 56.6 mm, and showed higher cellulase activity compared with "Sanlim 4-ho"(52.9 mm). On the $5^{th}$ day of mycelial growth, "Poongnyunko" was 58.0 mm, and showed higher laccase activity compared with "Sanlim 4-ho" (55.6 mm). Its pH at a substrate was 6.0. Before cultivation, the pH of sawdust substrate was 4.7-5.5 went down to 3.7-3.9 after 120-day cultivation. After 60 days of cultivation, the change of $CO_2$ was the greatest. During the cultivation, $CO_2$ concentration in a growing bag was 4.67% to 3.90%.

Microscopical Characteristics of Softwood Sawdusts Cultivated with Enokitake (Flammulina velutipes) (1주기 수확을 끝낸 팽이버섯(Flammulina velutipes) 재래용 톱밥배지 분해의 현미경적 특징)

  • This study was carried out to explore for the possibility of recycling the pine wood sawdusts for the substrate for enokitake (Flammulina velutipes) cultivation. The wood species of sawdusts cultivated for enokitake mushroom were identified mostly as hard pine (Pinus spp.). Distribution of enokitake hyphae was restricted to ray parenchymas and tracheids exposed to fungi. Nevertheless, degree of cell wall degradation by enokitake was slight. Light microscopic observation showed the thinning of secondary cell wall in some tracheids. Under polarized microscopy the 1()ss of birefringence was observed only in a few latewood tracheids. All the middle lamella remained intact. The present work showed clearly that pine sawdusts used as substrate for enokitake cultivation held enough cell wall materials for mushroom cultivation. The relative resistance of softwood cell walls against enokitake fungus was also discussed.
