This study was carried out to isolate saponin compounds from red-ginseng efflux, which was produced during the industrial processing of red-ginseng from fresh ginseng. We isolated crude saponin from the efflux extract (moisture content 35.0%) by using Diaion HP-20 adsorption method. Non-saponin fraction, which was adsorbed on Diaion HP-20 resin, was removed by eluating with $H_{2}O$ and 25% spirit. Then crude saponin was eluated with 95% spirit, continuously. Saponin in the eluated fractions was confirmed by TLC analysis. Crude saponin isolated from red ginseng efflux extract contained 12.10% of saponin. whereas those of white ginseng and red-ginseng were 3.30 and 3.39%, respectively. Ginsenoside contents showed the highest contents kin crude saponin from red ginseng efflux extract. Expacilly, the ginsenoside-$Rb_{1}$ and Re showed the highest contents in red-ginseng efflux extract when compared with those of white ginseng and red ginseng crude saponins. And the other ginsenosides except ginsenoside-$Rb_{1}$ and -Re also showed the highest contents in red ginseng efflux extract. However, the ratio of PD saponin (Panaxadiol saponin: $Rb_{1}+Rb_{2}$+Rc+Rd) to PT saponin (panaxatriol: $Re+Rg_{1}$) showed almost the same level when compared with those of ginseng saponin fractions. Ratio of PD/PT from red ginseng efflux extract was 1.99. Ratios of PD/PT from white ginseng and red ginseng were 1.85 and 1.84, respectively. Saponin purity, which was calculated by ratio percent of total ginsenoside to curde saponin content, was 45.90%. In case of white ginseng and red ginseng, the purities were 35.50 and 36.00%, respectively. However, by PHLC analysis, we confirmed that crude saponin isolated from red ginsengs. It suggested that crude saponin isolated from red ginseng ellux also would be useful component as ginseng saponins.
Crude saponin and ginsenosides in commercial white ginsengs such as six kinds of whole ginsengs, five kinds of tail roots and one undergrade raw ginseng were analyzed. The contents of crude saponin and ginsenosides in whole ginsengs were found to be 2.7 to 4.6% and 1.0 to 1.7%, respectively. On the other hand, those of tail roots were 3.3 to 7.2% and 1.3 to 4.4%, respectively. The content ratio of PD to PT saponin in whole ginsengs showed a little variation as 0.73 to 0.92, while those of tail roots showed a greater variation in the range of 0.72 to 1.75, indicating that tail roots contain higher content of PD saponin than whole ginsengs did.
The content and composition of saponin compounds of Panax species were analyzed according to their species, region and processing type of red and white ginseng. The species employed were Korean-, Chinese-, Japanese red ginsengs, and Korean white ginseng of Panax ginseng, American- and Canadian ginsengs of Panax quinquefolium, and Panax notoinseng. Twelve main saponin components in the ginseng were identified and quantified using TLC and HPLC. All three species had remarkably different content and composition. However, within each species they were similar. Twelve major ginsenosides were determined in P. ginseng, eight in p. quinquefolium, and six in P. notoginseng. Of the components of P ginseng Rf, $Rh_1$, $Rh_2$ and Ra were not detected in P quinquefolium, and $Rb_2$, Rc, Rf, $Rh_2$, Ra and Ro not detected in P. notoinseam. Crude saponin content and protopanaxadiol/protopanaxatriol saponin ratio were compared. They were 4.81~5.24% and 1.27~ 1.45 in p. ginsengs, 7.01~7.25% and 2.12~ 2.15 in p. quinquefolium, 9.80% and 0.99 in P. notoineng. The prosapogenin and sapogenin content were different among the Panax species.
The aim of this study was to determine and compare the preventive effect of Korean White Ginseng and Red Ginseng on oxidative stress in $H_2O_2$-treated HepG2 cells. The roots of ginseng were extracted with 70% methanol and partitioned with butanol to obtain saponin fractions, which have been known as bioactive constituents of ginseng. 2',7'-Dichlorofluorescein diacetate (DCF-DA) assay and malondialdehyde (MDA) content were measured for evaluating intracellular reactive oxygen species (ROS) generation. Also, mRNA expressions and activities of antioxidant enzymes were analyzed to determine the antioxidant activity of saponin or non-saponin fractions of ginsengs. According to DCF-DA assay, $H_2O_2$-induced MDA release and ROS generation were significantly reduced by treatment with saponin fractions of white and red ginseng roots. Also, saponin fractions increased effectively intracellular antioxidant enzyme activities including catalase, glutathione peroxidase and superoxide dismutase in $H_2O_2$-treated HepG2 hepatoma cells. In general, red ginseng was more effective than white ginseng for reducing oxidative stress. These results indicate that administration of red ginseng may certainly contribute relatively stronger than white ginseng to prevent from damaging liver function by oxidative stress.
This study was conducted to provide basic information on ginseng saponin of dried ginseng radices. In order to achieve the proposed objective ginsenoside compositions of dried ginseng radices extract with 70% ethyl alcohol were examined by HPLC. The total saponin content, the sum of all ginsenosides, showed that Wild simulated ginseng (WSG), White fine ginseng (WFG), Skin White ginseng (SWG), and White ginseng (WG) stood at 2.510%, 1.643%, 0.587, and 0.429%, respectively. WSG in PPD/PPT ratio was highest at 3.190, WFG (1.934), WG (1.600), SWG (1.386) in order. In the content of ginsenoside Rb1, one of the marker compounds of ginseng, WSG (1.095%) showed the highest content, and WFG (0.527%), SWG (0.246%), WG (0.133%) in this order. The content of ginsenoside Rb1 of WSG (1.095%) was 4.5 times higher than SWG (0.246%). WSG (0.230%) showed the highest content in ginsenoside Rg1, a marker compounds of ginseng, followed by WFG (0.180%), SWG (0.141%) and WG (0.086%). The content of ginsenoside Rg1 of WSG (0.230%) was 1.6 times higher than SWG (0.141%).
Total saponins and ether extracts of red and white ginseng were obtained and their effects on blood pressure in cat and their histamine liberating activities in rabbits were measured. 1) Ether extract of red ginseng showed a transient hypotensive effect and subsequently showed a remarkable and persistent hypotensive effect, whereas other three fractions, such as saponin fractions of red and white ginseng and ether extract of white ginseng showed only a initial transient hypotensive effects. 2) Histamine levels liberated into blood after administration of each fractions measured by the bioassay with guinea pig ileum. Ether extract of red ginseng immediately increased histamine contents in plasma but the histamine levels decreased to normal level within 10min in spite of decreased blood pressure was sustained. Although white ginseng saponin lowered blood pressure immediately when it is administered, histamine release was observed after 10min. The results suggest that hypotensive effects of ginseng seems to have no correlation with the histamine liberating activity. Ginseng appears to show hypotensive effect via some other mechanisms.
The present study was carried out to investigate the effects of Panax ginseng saponin extracts on the dexamethasone-induced apoptosis of mouse thymus in vivo and mouse thymocytes in vitro. The saponin fractions of red ginseng (R-SAP) and white ginseng (Wl-SAP) were provided by the Korea Ginseng & Tobacco Research Institute, and the other saponin fraction of white ginseng (W2-SAP) was extracted in our laboratory. 1. The male ICR mice (3~4 wk old; weighing 15$\pm$2 g) were given by each saponin fraction of 5 mg/kg/ day for 4 days, and at one hour after the last treatment, they were injected by deuamethasone (5 mg/kg : DX). The mouse thymus was extracted at 6 hours after DX injection, and they were stained with hematoxylin-eosin reagents and an Apop-Tag kit, respectively, and the thymocytes prepared from it were labelled with anti-mouse FITC-anti-CD4 and anti-mouse PE-anti-CD8 and then analyzed by fluorescence activated cell sorter (FACS). DX-induced reduction of thymus weight was significantly attenuated by W2- SAP but was not affected by other saponin fractions. And DX-induced apoptotic death of thymocytes, appeared in the histologic findings of the thymus, was inhibited by the saponin fractions and the order of these inhibitory potencies was R-SAP》W2-SAP>Wl-SAP. However, in respect of T cell receptors, the differentiation of thymocytes seems not to be changed by treatments with DX or/and the saponin fractions. 2. In the primary thymocyte culture, the DX-induced reduction of thymocyte MTT values was rather greater in RPMI 1640 medium of IWc fetal bovine serum (FBS) or horse serum (HS). In addition, the DX-Induced MTT reduction was significantly inhibited by R-SAP or W2-SAP, in the culture using that medium of 5% FBS or HS. But these saponin fraction did not effected the DX-induced reduction of thymocyte MTT value in primary culture of 10% FBS or 10% HS. These results suggest that R-SAP and some W-SAP fractions may protect thymocyte from stress or glucocorticoisteroid-induced death of them.
The changes in aldehyde dehydrogenase(ALDH, E.C. 1.2.1.3.) activity in neurons and astrocytes isolated from rat brains were investigated after administration of ethanol and Korean red ginseng(Panax ginseng C.A. Meyer) saponln. The cerebral ALDH activity with acetaldehyde and Propionaldehyde was higher in the white matter than in the gray matter. However, using indole-3-a-cetaldehyde and 3,4-dihydroxyphenylacetaldehyde as substrates, there was no significant difference in activity between two regions in cerebrum. In ethanol treated group, ALDH activity with all the substrates in the gray and white matter was lower than in normal group. In ethanol-saponin treated group, the enzyme activity in the white matter remarkably Increased. The ALDH activity in neurons isolated from cerebral cortex in ethanol-treated group was lower than in normal group. In ethanol-saponin treated group, neuronal ALDH activity with propionaldehyde was significantly recovered but not with Indole-3-acetaldehyde. In astrocytes, although the ALDH activity with propionaldehyde in the ethanol-treated group was not changed as compared with normal group, considerable increase in activity was found in ethanol-saponin treated group. These results suggest that Korean red ginseng saponin may protect the neuronal functions from the toxic effects of acetaldehyde derived from ethanol by stimulation of ALDH activity in astrocytes surrounding nerve cells.
