This study examines the preliminary license and approval facilitation standards for improvements in quality for childcare centers focusing on the number of children, building space standards, facilities and equipment, and childcare program. Data from 48 caese was collected from childcare centers nationwide in Korea through a questionnaire and building plan. The first step for starting care licensing is to work out the ages and numbers of children within four or five age ranges. The second step is to work out the number of children as small, medium, or large considering staff to child ratios and building size. The Nnext step is to make a choice about childcare service quality classification as minimum, fair, or good, considering space requirements per child for the building, the classroom and the outdoor playground. The next step is to make a choice of space organization relating to service programs, considering the sleeping and eating area, indoor play area, toilet & washing facilities, classroom layouts such as cluster type, double zone type, single zone type, and others. Also, each room and entrance, office, kitchen, storage, laundry, teacher's area, and chilldren's area, need to be checked for space requirements and performances. The last step is to arrange the childcare program with the building and site plan. In conclusion childcare service quality will get better by upgrading of license regulation especially in minimum space requirements per child, corresponding with an increase in GNP and housing area per person. This is needed for childcare licensing in order to determine the quality level of childcare service.
The purpose of this study was to assess the microbiological quality of home-delivered meals during production and delivery for children from low-income families. Production flows from a facility in Seoul that provides home-delivered meals were analyzed and the time-temperature of the food was measured. Microbiological assessment was performed for the production environment, personal hygiene, and food samples at each production and delivery step based on the process approach. It took 2 hours or longer from completion of production to meal delivery. An aerobic colony count (ACC) and coliform were not detected at knives, cutting boards, and dish towels. However, ACC (at pre-preparation, preparation, and packing areas) and coliform (at the preparation area) were detected on the hands and gloves of employees. Air-borne bacterial counts varied according to day and preparation area (ND~6 CFU/plate/15 min). Food temperatures, on the completion of production and meal delivery, fell into temperature danger zones. ACC and coliform counts of raw ingredients did not decrease after pre-preparation (washing and sanitizing) for menus involving food preparation with no cook step. ACC decreased after cooking step for menus of food preparation with cook step, but the ACC of the stir-fried and seasoned dried filefish fillet on the completion of cooking was too numerous to count due to improper heating. The ACC of seasoned young Chinese cabbages (a menu with complex food preparation) increased during delivery (from 2.5 log CFU/ml to 5.0 log CFU/ml). This qualitative assessment of foodborne pathogens revealed that B. cereus was detected in vegetable and meat product menus. These results suggest time-temperature control is necessary during production and delivery and management guidelines during production of home-delivered meals are provided for safe production.
본 연구는 생산 후 3일 이내인 시판두부의 가공공장별, 두부유형별 위생실태를 파악하여 두부에 대한 미생물 규격의 설정 가능성을 검토하며, 가공단계별 총생균수와 대장균군을 분석하여 오염 원인을 규명코자 실시되었다. 시판중인 두부의 위생성은 동일유형이더라도 가공공장마다 차이가 있었고 동일공장의 시료 간에도 차이를 보였다. 총생균수가 $1.0{\times}10^6CFU/g$ 이상을 보인 일부 포장두부에서는 관능적 부패 변질 현상이 관찰되었다. 총 148개 두부시료 중 총생균수와 대장균군이 각각 $1.0{\times}10^5CFU/g$ 이하와 음성에 적합한 시료는 단순포장두부가 32.0%와 12.0%, 포장가열두부가 86.9%와 83.7%인 것으로 조사되었다. 이러한 가공공장에 따른 위생성 차이는 가열과 열처리사이의 공정인 응고, 압착, 성형, 1차냉각, 내포장공정에서 2차오염 정도의 차이에서 기인하는 것으로 생각되었다. 포장두부의 가공공정 중 미생물이 증가하여 각별한 관리가 요구되는 공정으로는 원료대두의 세척 침지와 마쇄, 응고 후 압착 성형, 1차냉각공정이었다. 따라서 보다 위생적인 두부를 가공하기 위해서는 이들 가공공정에서 사용되는 각종 설비나 작업도구, 성형포 및 작업자 등에 대한 세척 및 소독 강화가 절실하게 요구되었다.
간장게장은 우리나라 전통 식품 중의 하나로 생 꽃게를 세척한 후 간장소스를 첨가하고 저온에서 숙성시켜 포장하여 상업적으로 판매하고 있다. 하지만, 간장게장 제조특성 상 열처리를 하지 않기 때문에, 간장게장의 미생물학적 품질을 유지하는데 어려움이 따른다. 따라서 본 연구에서는 원물 꽃게의 초기 미생물 저감화를 위해 여러 살균세척제의 효능을 비교하고 저장 중 간장게장 제품의 미생물학적 품질을 유지하기 위해 키토산의 항균 효능을 평가하였다. 먼저, 상온에서 생 꽃게를 염소수(50 mg/L), 과초산(40 mg/L), acetic acid (5%), lactic acid(5%)에 각각 10분간 침지시켜 일반세균수를 분석하였다. 결과를 살펴보면, 여러 살균세척제 중 5% acetic acid 세척이 생 꽃게에 존재하는 일반세균수를 약 1.5 log CFU/g까지 감소시켜 가장 효과적인 것으로 나타났다. 키토산 효능을 평가하기 위해 현재 상업적으로 제조되고 있는 간장게장(방법 1; 전해수 세척), 5% acetic acid로만 세척된 꽃게(방법 2), 5% acetic acid로 세척된 꽃게에 0.5%(방법 3)와 1%(방법 4)의 수용성 키토산이 첨가된 간장으로 제조된 간장게장을 각각 $4^{\circ}C$와 $12^{\circ}C$에서 최대 30일까지 저장하면서 일반세균수, 대장균군 및 대장균수를 측정하였다. $12^{\circ}C$에서 저장된 간장게장의 일반세균 수는 7일이 지났을 때 약 8 log CFU/g까지 증가하였다. $4^{\circ}C$의 경우, 1% 키토산이 첨가된 군(방법 4)에서 20일 동안 약 2.9 log CFU/g까지만 증가한 것으로 나타나 키토산 무첨가군(방법 1과2)과 0.5% 첨가군(방법 3)(4.2~4.5 log CFU/g)에 비하여 훨씬 효과적이었다. 본 결과에 따라, 생 꽃게를 5% acetic acid로 세척한 후 간장게장에 1% 키토산을 첨가하여 냉장온도에서 저장한다면 간장게장 제품의 미생물학적 품질 향상에 큰 도움이 될 것으로 여겨진다.
전해수로 처리한 생식 원료 백미, 현미, 찹쌀, 보리를 열풍건조 및 동결건조를 실시하여 이화학적 특성 및 기능성 성분의 변화를 조사하였다. 백미, 현미, 찹쌀, 보리의 단백질, 지방, 탄수화물, 회분 등 성분의 함량은 전해수로 처리하여 열풍건조 및 동결건조를 통하여서는 거의 변화가 없는 것으로 나타났다. 백미, 현미, 찹쌀, 보리는 전해수로 처리하여 열풍건조 및 동결건조를 실시한 후 L값과 a값이 변화가 없는 것으로 관찰되었고 백미와 찹쌀의 열풍건조를 실시한 시료가 증가한 것으로 나타났으나 전해수로 처리하고 동결건조를 실시한 시료와 전해수 처리를 하지 않고 동결건조를 실시한 시료는 차이가 없는 것으로 나타났다. 식이섬유의 함량은 열풍친조를 실시한 시료가 증가를 나타냈으나 전해수로 처리한 시료와 처리하지 많은 시료 사이에는 차이가 없는 것으로 나타났다. 백미, 현미, 찹쌀, 보리 중의 niacin 함량과 보리 중의 ${\beta}-glucan$ 함량은 열풍건조 및 동결건조를 실시한 시료가 서로 차이를 보이지만 전해수로 처리하여 동결건조를 실시한 시료와 전해수로 처리하지 않고 동결건조를 실시한 시료 사이에는 차이가 없는 것으로 나타났다. 동결건조를 진행한 시료는 전해수 처리 전후 이화학적 특성과 기능성 성분 함량이 변화가 없는 것으로 나타났다. 결론적으로, 생식 원료 중 곡류인 백미, 현미, 찹쌀, 보리의 전해수 처리 시 이들의 이화학적 특성 중 단백질, 지방, 및 탄수화물, 회분, 및 식이섬유의 함량과 색도에는 영향을 주지 않았고 기능성 물질 중 niacin과 ${\beta}-glucan$의 함량은 전해수 처리 후 열풍건조 시 약간의 영향을 나타냈으나 동결건조 시에는 전혀 영향을 주지 않았으므로 전해수를 이용한 세척을 생식 제조에 충분히 사용이 가능하다고 생각된다.
One-step reduction/dyeing method was optimized for wool dyeing with natural indigo dye in this study. The effects of reduction/dyeing conditions including dye temperature and time, the pH of bath, concentration of dye, and reducing agent on dye uptake and color were investigated. The dye uptake was higher with no addition of alkali. Dyeing was carried out through the use of only sodium hydrosulfite in the bath. The maximum dye uptake was obtained at $60^{\circ}C$ for 30min and the dyed fabrics appeared in the PB Munsell color range. Dye uptake improved with the increase of a natural indigo dye concentration with the same sodium hydrosulfite concentration. At a higher dye uptake, the fabric color became more purplish and the maximum absorption shifted from 660nm to 620nm. Color reproducibility was reliable with a color difference in the range of 0.41~1.43. Regardless of color strength, washing and dry cleaning fastnesses were good with a 4/5 rating, and fastnesses to rubbing and light were acceptable with a 3/4~4 rating.
The Jun and Fos families of eukaryotic transcription factors form heterodimers capable of binding to their cognate DNA enhancer elements. We are interested in searching for inhibitors or antagonists of the binding of the Jun-Fos heterodimer to the activator protein-1 (AP-1) site. The basic-region leucine zipper (bZIP) domain of c-Fos was expressed as a fusion protein with glutathione S-transferase, and allowed to form a heterodimer with the bZIP domain of c-Jun. The heterodimer was bound to glutathione-agarose, to which were added radiolabeled AP-1 nucleotides. After thorough washing, the gel-bound radioactivity was counted. The assay is faster than the coventional electrophoretic mobility shift assay because the gel electrophoresis step and the autoradiography step are eliminated. Moreover, the assay is very sensitive, allowing the detection of picomolar quantities of nucleotides, and is not affected by up to 50% dimethylsulfoxide, a solvent for hydrophobic inhibitors. Curcumin and dihydroguaiaretic acid, recently known inhibitors of Jun-Fos-DNA complex formation, were applied to this Jun-GST-fused Fos system and revealed to decrease the dimer-DNA binding.
Hobun(Oyster shell White) is a traditional material used as extender and white pigment from ancient times. The production method of it, however, has been discontinued. We have studied the traditional production method of Hobun by weathering oyster shell, which is one of the traditional ways for preparing Hobun. Reproduction study of manufacturing method of the discontinued traditional material is an important accomplishment of our research. Also this study provides solid background knowledge to stabilize the production and supply of Hobun for the cultural asset repairing materials. The result can be summarized as follows: The production process of Hobun by weathering method takes 5 steps - (1) weathering shells ${\rightarrow}$ (2) washing ${\rightarrow}$ (3) pulverization ${\rightarrow}$ (4) separating fine powder by submerging in water ${\rightarrow}$ (5) drying. The major aim in step (1) is to eliminate organic impurities. In the step (4), the fine particles smaller than $25{\mu}m$ are separated by extracting the supernatant from stirred suspension after heavy particles are submerged. Also, the soluble inorganic impurities can be eliminated through the powder submerge in 15 times water and stirring the suspension 6 hours and changing the water 3~4 times. The final products have high quality with 94.03, 0.52, 2.05 for L, a, b, less than $25{\mu}m$ particle size, fine resistance for discoloration by light and environmental pollution and good workability.
본 연구는 아크릴 폐수의 COD제거를 위한 전터리 공정 및 전 처리된 아르릴폐수를 한외여과 중공사형 모듈과 역삼투 나권형 모듈에 적용하여 적용압력과 온토변화에 따른 한외여과막과 역삼투막의 분리특성을 고찰하였다. 또한, 장시간 연속운전으로부터 한외여과 및 역삼투 막의 세척 적정시간을 결정하고, 운전시간에 따른 막오염을 고찰하였다. 14시간을 기준으로 한외여과 막의 플럭스가 급격히 떨어지고 있으며, 역삼투 막도 비슷한 경향을 나타내고 있다. CaO 및 모래여과장치를 거친 처리수를 중화처리 하여 UF/RO공정에 적용한 결과 COD 및 TDS가 배출허용 기준치 이하로 제거됨을 알 수 있었다.
The low melting PET yarn has sheath/core structure: sheath portion consists of low melting point PET and core portion is regular PET. Dyeing properties of fabric made from low melting PET yarn were investigated at different dyeing temperatures. It was found that the exhaustion yield on the low melting PET fabric was higher than on regular PET fabric. The total K/S value of the dyed low melting PET fabric increased as heat setting temperature increased above $150^{\circ}C$ because the sheath portion of the low melting yarn melted. Although fastness to light of the low melting PET fabric was similar to regular PET fabric, fastness to washing was inferior to regular PET fabric by 1 grade.
