Comparative Study of Dissolution Properties of Immediate-release and Controlled-release Type Vitamin C Tablets (속방형 및 용출조절형 비타민 C 정제의 용출 특성 비교 분석)

  • In this study, a comparative dissolution experiment was conducted between an immediate-release and a controlled-release vitamin C tablet applied with a technology to control the dissolution of vitamin C to maintain the vitamin C level in the human body. In order to confirm the dissolution rate (%) of vitamin C tablets, HPLC determination was conducted based on the dissolution test methods in the 'Korean Pharmacopoeia (No. 2020-88),' 'Guidelines on Specifications of Dissolution Tests for Oral dosage Forms,' and 'Standard and Specifications for Health Functional Foods (No. 2020-63)' from Ministry of Food and Drug Safety (MFDS). In addition, the dissolution pattern between the immediate-release tablet and the controlled-release tablet was comparatively analyzed. The analysis result confirmed that the immediate-release vitamin C tablet was 100% dissolved after 45 minutes, while the controlled-release vitamin C tablet was 100% dissolved after 480 minutes (8 hours). Furthermore, the dissolution rate (%) at 60 minutes was slower than that of the immediate-release vitamin C tablet. Based on these results, this study confirmed that the dissolution rate (%) test and development of controlled-release tablets containing vitamin C as the main component a re possible.

Effects of Temperature and Relative Humidity on Fat Soluble Vitamin Contents in Commercial Vitamin Tablet (저장 온도 및 상대습도가 비타민 정제 중 지용성 비타민 함량의 변화에 미치는 영향)

  • Effects of temperature and relative humidity on fat soluble vitamin contents in two commercial vitamin tablets ('Multivitamin Dandelion' and 'vitamin E Yarrow') were investigated. When they stored at different temperature (25, 35, and $45^{\circ}C$) with cap and stored at low relative humidity (11% RH) without cap, all fat soluble vitamins (vitamin A acetate, vitamin E acetate and vitamin E) were stable during 24 weeks of storage period. However, when they stored at high relative humidity (75% RH and 100% RH) without cap, vitamin A acetate and vitamin E acetate dramatically disintegrated at the early stage of storage. On the other hand, vitamin E gradually decreased during storage. Therefore, stability of some fat soluble vitamins, which exist as acetate form, was highly dependent on relative humidity rather than storage temperature. Water may act as a plasticizer and increased the mobility of molecules in vitamin tablet resulting in relatively fast disintegration of some fat soluble vitamins.

Absorption Enhancer and Polymer (Vitamin E TPGS and PVP K29) by Solid Dispersion Improve Dissolution and Bioavailability of Eprosartan Mesylate

  • The aim of the present study was to improve the solubility and bioavailability of a poorly water-soluble drug in human body, using a solid dispersion technique (hot melt extrusion). The solid dispersion was prepared by cooling the hot melt of the drug in the carrier (Vitamin E TPGS and PVP). The dissolution rate of formulation 1 from a novel formulation prepared by solid dispersion technique was equal to release of formulation 6 (40% of eprosartan mesylate is in contrast to teveten$^{(R)}$) within 60 min (Table 1). The oral bioavailability of new eprosartan mesylate tablet having vitamin E TPGS and PVP K29 was tested on rats and dogs. Of the absorption enhancer and polymer tested, vitamin E TPGS and PVP K29, resulted in the greatest increases of AUC in animals (about 2.5-fold increase in rat and dog). When eprosartan mesylate was mixed with the absorption enhancer and polymer in a ratio of 2.94:2:1, vitamin E TPGS and PVP K29 improved eprosartan mesylate bioavailability significantly compared with the conventional immediate release (IR) tablet Teveten$^{(R)}$ (formulation 7). These results show that solid dispersion using vitamin E TPGS and PVP K29 is a promising approach for developing eprosartan mesylate drug products.

The Effect of Long-term Endurance training and Antioxidant Combined Vitamin Supplementation on blood Antioxidant Enzymes Activity, Lipid Peroxidation and Lipoprotein metabolism (장기간의 지구성 훈련시 항산화 Vitamin 복합 투여가 혈중 항산화 효소 활성도, 지질 과산화 및 지단백 대사에 미치는 영향)

  • This study was to investigate the effect of long-term high intensity endurance training on the activation of antioxidation enzyme activity, lipid peroxidation and lipoprotein metabolism. 15 subjects were divided into, endurance exercise + antioxidation Vitamin supplement(n=5), endurance exercise(n=5), and the control(n=5) groups. The endurance exercise groups(endurance exercise + antioxidation Vitamin supplement and endurance exercise) had 12 week of endurance exercise program. The antioxidation Vitamin supplement group was taken a Vitamin C tablet with 1000mg/day and Vitamin E tablet with 671.14mg/day right after lunch. The results obtained from this study were as follows; 1. Looking at the changes of SOD, Endurance exercise+antioxidation Vitamin supplement group and endurance exercise groups showed the significantly greater decrease in the activation of SOD after 12 weeks of all-out exercise. 2. Looking at the changes of CAT, Endurance exercise+antioxidation Vitamin supplement group revealed subjects tended to increase CAT after all-out exercise although statistically non-significant. Endurance exercise+antioxidation Vitamin supplement group showed the significantly greater increase in the activation of CAT after 12 weeks treatment for all-out exercise. 3. Looking at the changes of GPX, Endurance exercise+antioxidation Vitamin supplement group revealed subjects tended to increase GPX for the rest and after all-out exercise although statistically non-significant. Endurance exercise+antioxidation Vitamin supplement group showed the significantly greater increase in the activation of GPX after 12 weeks treatment for all-out exercise. 4. The MDA change showed the significant decrease after 6 weeks, after 12 weeks for the all-out exercise of Endurance exercise + antioxidation Vitamin supplement group. 5. There was non-significant change in lipoprotein metabolism for the rest and after all-out exercise.

Analysis of Folate by Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry

  • Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) was used to observe folic acid and its derivatives such as tetrahydrofolate and 5-methyltetrahydrofolate in a vitamin tablet and in foods. Folic acid in a vitamin tablet was determined using angiotensin I as an internal reference. Tetrahydrofolic acid, 5-methyltetrahydrofolic acid, and an oxygenated folate were observed from a human blood sample using graphite plate. The results show that these mass spectrometric methods are useful for quickly obtaining a profile of folates.

  • 포장된 복합비타민의 보관수명을 예측하기 위한 수학모델을 수치해석의 방법 중 finite difference method을 이용한 컴퓨터 프로그램을 개발하였다. 이 컴퓨터 프로그램에는 포장재료의 수분확산계수와 용해도를 비롯한 식품의 수분확산계수 등 다양한 인자들을 포함하고 있다. 포장재료를 통과하는 수분의 물질전달은 G.A.B water sorption isotherm을 기본으로 했다. 본 연구에서는 비정상상태에서의 물질전달로 기본 수학모델링을 구하였다. 본 수학모델과 프로그램의 결과는 실험치와 비교하지 않았고 대신 특정한 경우를 가상하여 이에 존재하는 analytical solutions를 실제로 구하여 본 수학모델과 컴퓨터 프로그램의 신뢰도 측정을 하였다.

  • Effects of temperature and relative humidity on contents of water-soluble vitamins (vitamins $B_1,\;B_2,\;B_3,\;B_6$, and C) of two commercial tablets ("Multivitamin Dandelion" and "Chewable vitamin C") were investigated. When stored at various temperatures (25, 35, and $45^{\circ}C$) with cap, all measured vitamins were stable and degraded very slowly during 24 weeks of storage; low relative humidity (11% RH) without cap also provided stability during storage period. At higher relative humidities (75 and 100% RH), contents of all water-soluble vitamins, except vitamins $B_2\;and\;B_3$, decreased significantly at early storage period. These results showed that stability of water-soluble vitamins is highly dependent on relative humidity rather than storage temperature.

  • An analytical method using capillary electrophoresis (CE) was developed for quantitation of water soluble vitamin contents in various vitamin products. The method includes the optimization of separation of 11 water soluble vitamins changing the micellar concentration and pH of running buffer, applied voltage and sample preparation. Best resolution was obtained with 25 mM phosphate buffer (pH=8.0) containing 50 mM sodium dodecyl sulphate (SDS) as micellar phase. At optimum condition, water soluble vitamins were determined in orange juice and vitamin products such as vitamin C pulvis, vitamin injection, coated multivitamin tablet. The quantitative analysis of water soluble vitamins with CE was suitable for quality control of pharmaceutical products with sound reproducibility.

Vitamin C Tablet Assay by Near -Infrared Reflectance spectrometry

  • When a drug is prepared in a tablet, the active component represents only a small portion of the dosage form. The other components of the formulation include materials to assist in the dissolution, antioxidants, coloring agents and bulk fillers. The tablets are tested using approved testing methods usually involving separation and subsequent quantification of the active component. Tablets may also be tested by near-Infrared Reflectance spectrometry (NIRS). In the present study, based on NIRS and multivariate calibration methods, a novel and precise method is developed for direct determination of ascorbic acid in vitamin C tablet. Two different tablet formulations were powdered in three different sizes, 63-125 ${\mu}{\textrm}{m}$, and examined. Spectral region of 4750-4950 $cm^{-1}$ / was used and optimized for quantitative operations. Partial least squares (PLS) and multiple linear regression (MLR) methods were performed for this spectral region. The results of optimized PLS and MLR methods showed that reproducibility increase with decreasing grain size and standard error of calibration (SEP) of less than 1% w/w of ascorbic acid and a correlation coefficient of 0.998 can be achieved. The PLS method showed better results than MLR. Seven overdose and underdose samples (prepared in the laboratory to match marketed products) were tested by proposed and iodometric standard methods. A correlation between NIRS predicted ascorbic acid values and iodomet.ic values was calculated ($R^2$=0.9950). Finally, the direct analysis of individual intact tablets in their unit-dose packages (Blistering in aluminum and PVC foils) obtained from market were also carried out and a correlation coefficient of 0.9989 and SEP of 0.931% w/w of ascorbic acid were achieved.

A Studies on Nutririon of Aged patients in hospital (노인 입원환자들의 영양섭취 실태에 관한 조사연구)

  • This survey was conducted to investigated the status of nutritional consumption and relationship of nutrition of patients in hospitals from 1998.9.1 to 1998.9.10. Calorie intake of male patients was lower than RDA but that of female patients were higher than RDA except female younger than 64 years old. Protein intake of male patients was 130% higher than RDA, and that of female patients was 150% more than RDA. Calcium intake was lower than RDA in male patients but higher than RDA in female patients except older than 75 years. Iron intake was higher than RDA in all patients except in male patients older than 75 years. while vitamin B$_1$and vitamin C intake was higher than RDA in all patients but vitamin A and niacin intake was lower than RDA. The rich patients intake enough iron through animal foods and stabilized patients intake vitamin B$_2$ more than RDA. All patients were recognized the importance of protein and vegetable for health and they were recognized also careful choosing of foods for health. Positive relationships were showed between intake of nutritional tablet and nutritional status, between less vegetable intake and less intake of calorie, fat, sugar and fiber and between appetite and vitamin B$_1$ and vitamin C. The result showed that the shorter period staying in hospital, the more intake of sugar, vitamin A and vitamin C.
