Re-Identification of Aspergillus Subgenus Circumdati Strains in Korea Led to the Discovery of Three Unrecorded Species

  • Aspergillus is one of the largest and diverse genera of fungi with huge economical, biotechnological, and social significance. Taxonomically, Aspergillus is divided into six subgenera comprising 27 sections. In this study, 235 strains of Aspergillus subgenus Circumdati (section: Candidi, Circumdati, Flavi, Flavipedes, Nigri, and Terrei) preserved at the Korean Agricultural Culture Collection (KACC) were analyzed and re-identified using a combined dataset of partial b-tubulin (BenA), Calmodulin (CaM) gene sequences and morphological data. We confirmed nineteen species to be priorly reported in Korea (A. neotritici, A. terreus, A. floccosus, A. allahabadii, A. steynii, A. westerdijkiae, A. ochraceus, A. ostianus, A. sclerotiorum, A. luchuensis, A. tubingensis, A. niger, A. welwitschiae, A. japonicus, A. nomius, A. tamarii, A. parasiticus, A. flavi, and A. oryzae). Among the studied strains, three species (A. subalbidus, A. iizukae, and A. uvarum), previously unreported or not officially documented, were discovered in Korea, to the best of our knowledge. We have given a detailed description of the characteristic features of the three species, which remain uncharted in Korea.

Unrecorded Fungi Isolated from Fire Blight-controlled Apple Orchard Soil in Korea

  • To explore fungal diversity in orchard soil where fire-blighted apple trees are buried, we collected soil samples from apple orchards in Chungju, Korea. Fungal isolates were obtained from DG18 agar and identified at the species level based on morphological features and phylogenetic analyses. The colony characteristics and microstructures were examined using a light microscope and a scanning electron microscope after culturing on potato dextrose agar (PDA), malt extract agar (MEA), Czapek yeast agar (CYA), and oatmeal agar (OA) The PCR-amplified products of the ITS1-5.8S-ITS2 region and 28S large subunit of the nuclear ribosomal RNA gene, as well as partial sequences of the β-tubulin, calmodulin, and translation elongation factor 1-α genes were sequenced and analyzed phylogenetically. Seven previously unknown fungal species were explored in Korea. All samples, including Aspergillus aureolatus, Botryotrichum atrogriseum, Dactylonectria novozelandica, Fusarium denticulatum, Paecilomyces tabacinus, Sarcopodium tibetense and Talaromyces stollii, had ascomycetes. Herein, we report their descriptions and features.

Taxonomic Investigations on Korean Higher Fungi(VI) (한국산(韓國産) 고등(高等) 균류(菌類)의 분류학적(分類學的) 연구(硏究)(제6보)(第6報))

  • More than one-hundred-twenty specimens of Basidiomycetes, Ascomycetes and slime molds were collected in Seoul National Polytechnic University Campus during the priod from June 1984 to November 1987 and examined for identity. They were classified into 25 families, 53 genera and 81 species. Among them, one genus and 2 species were confirmed as unrecorded taxa in Korea. The unrecorded genus was genus Hapalopilus and was named 'Band Dal Busut' in vernacular. The unrecorded 2 species were Hapalopilus rutilans and Hygrophorus nitratus. They were named 'No Lan Ban Dal Busut' and 'Jil San Beot Geot Busut,' respectively and their characteristics were here reported.

Wood-Rotting Fungi Collected in Suweon Campus of Seoul National University (1) (서울대학교(大學校) 수원(水原)캠퍼스에서 채집(採集)한 목재부후균(木材腐朽菌)(I))

  • Wood-rotting fungi were collected in Suweon campus of Seoul National University from September 1981 to October 1982. These fungi were classified into 2 orders, 8 families, 22 genera and 34 species. Of these, Gloeoporus adustus (Willd. ex Fr.) $Pil\acute{a}t$ and G. dichrous (Fr.) Bres. were found to be new species that belong to unrecorded genus in Korea. Korean common names of these two new species were designated by the authors. List of the wood-rotting fungi was given with the generic name of host trees on which the fungus was collected.

Two Unrecorded Enophytic Fungi Isolated from Root of Ligusticum chuanxiong in Korea: Pithomyces chartarum and Plectosphaerella niemeijerarum (토천궁의 뿌리에서 분리된 2종의 국내 미기록 내생균: Pithomyces chartarum and Plectosphaerella niemeijerarum)

  • Endophytic fungal strains were isolated from the sterilized roots of a medicinal plant, Ligusticum chuanxiong and identified based on morphological characteristics and molecular analysis of internal transcribed spacer, large subunit rDNA, and beta-tubulin DNA regions. Our results confirmed the presence of Pithomyces chartarum and Plectosphaerella niemeijerarum in the fungal strains. To the best of our knowledge, the fungal strains have not been reported in Korea. In this report, we describe morphological characteristics and phylogenetic trees of these endophytic fungal strains.

  • In the present study, we isolated endophytic fungal strains from the rhizoids of the moss Funaria hygrometrica. The isolated strains were identified based on morphological characteristics and analysis of the internal transcribed spacer (ITS) and large subunit (LSU) rDNA sequence regions. Consequently, we confirmed the presence of two endophytic fungal species, Curvularia protuberata and Didymella anserina, which have not been reported in Korea previously. Here, we describe the morphological characteristics and molecular analysis results of these fungal species.

  • In this study, we isolated endophytic fungal strains from the rhizoid and the leaf of Mnium heterophyllum and Hypnum plumaeforme, respectively. The isolated strains were identified based on morphological characters and molecular analysis of the internal transcribed spacer, large subunit rDNA, β-tubulin, and RNA polymerase II subunit regions. From the results, we confirmed two endophytic fungal species, Cercophora thailandica, and Biscogniauxia petrensis, which to the best of our knowledge, have not yet been reported in Korea. We further describe the morphological characteristics and the results of the molecular analyses of these isolated fungal strains.

Notes on Some Unrecorded Species of Arbuscular Mycorrhizal Fungi Collected from Rhizospheres of Plants in Korea (식물 근권토양에서 분리한 수지상균근균)

  • In this study, arbuscular mycorrhizal fungi (AMF) were isolated and cultivated from rhizospheres of various plants in Korea. Five species belonging to four genus were identified based on spore morphology and 18S rDNA sequence analysis: Acaulospora cavernata Claroideoglomus luteum, Diversispora aurantium, D. trimurales, Rhizophagus irregularis. These fungal species have not been cultivated or reported to date in Korea, Morphological description and phylogenetic analysis for each species were presented. This result could be useful for research of AMF diversity in Korea.

  • We isolated endophytic fungi from the leaves of woody plants inhabiting the Ulleungdo Island and Yeosu in South Korea. The fungal strains were identified based on their morphological characteristics in conjunction with phylogenetic analysis, which was performed using sequences of the internal transcribed spacer, large subunit rDNA, and β-tubulin DNA. We confirmed the presence of Neofusicoccum mangiferae and Thyronectria cucurbitula, two endophytic fungal species that had not been previously recorded in Korea. In this report, we describe the morphological characteristics of these fungal strains and results of the DNA-based phylogenetic analysis.

Notes on Five Unrecorded Endophytic Fungi Isolated from Coniferous Leaves and Orchid Roots in Korea (한국에 서식하는 침엽수의 잎과 난초과 식물의 뿌리에서 분리한 5종의 국내 미기록 내생균)

  • We collected leaves of Pinus koraiensis and Thuja koraiensis and roots of Bletilla striata from various sites in Korea. The leaf and root samples were surface-sterilized and endophytic fungi were isolated. Fungal isolates were identified based on their morphological characteristics and a phylogenetic analysis of internal transcribed spacer regions, large subunit regions, and the ${\beta}$-tubulin gene. Consequently, we identified five species of endophytic fungi, namely Colletotrichum simmondsii, Fusarium sterilihyphosum, Diatrypella pulvinata, Ochroconis globalis, and Sphaeria chrysosperma. These species have not been previously reported in Korea and we report them here with descriptions and illustrations.
