Comparative Feeding Ecology of Sympatric Greenling Hexagrammos otakii and Schlegel's Black Rockfish Sebastes schlegeli in the Jnngbong Tidal Flat, Incheon, Korea (장봉도 갯벌의 쥐노래미(Hexagrammos otakii)와 조피볼락(Sebastes schiegeli)의 섭식생태)

  • The comparative feeding ecology of greenling Hexagrammos otakii and Schlegel's black rockfish Sebastes schlereli populations was investigated in the Jangbong tidal flat near Incheon, Korea. Monthly samples were taken using a modified otter trawl from November 1999 to January 2001. The stomach contents of 124 greenling and 115 Schlegel's black rockfish were analyzed. The diet of H. otakii was dominated by the amphipods Gammaropsis iaponicus, Caprella scaura, Isaeopsis sp. and Monocorophium acherusicum, the shrimps Latreutes mucronatus and Alpheus japonicus, the mysid Neomysis orientalis and the bivalve Mytilus edulis. In contrast, S. schlegeli mainly fed on the mysids N. orientalis, N japonica, and N. awatschensis, the shrimps Exopalaemon carinicauda, A. japonicus and L. mucronatus, and the fish Pholis fangi and unidentified gobiids. A comparison of the frequency and abundance of food items showed that crustaceans (e.g. amphipods, mysids and shrimps) were important foods for both species. Their main dietary components, however differed which is probably due to differences in the spatial distribution of the two species and their prey items. In conclusion, greenling and Schlegel's black rockfish, despite being sympatric on a tidal flat have distinct food habits and low dietary overlap (Schooner's index: 0.1). These results indicate, to some extent, resource partitioning for the maximum utilization of available foods in the tidal flat.

  • The present further survey is carried out to clarify the unidentified kinds of the fruit sucking moths and these damages at Suwon and Jinju fruit growing areas, in order to set up the basis of the control for the harmful moths. It was known that 10 species be recorded up to 1968 11 and, in addition, identified 4 species in 1969, such as Apatele rumicis oriens S., Leucania separate W., Scoliopteryx libatrix L. and Anomis flava F. of the fruit sucking moths in Korea. The damage ratio ib percentage of the noxious moths if $5.1\%$ on grape in Suwon and $11.8\%$ on Pears in Jinju in 1969, although it was $8.9\%$ in Suwon and 3.4\%$ in Jinju in 1968.

Identification and Antibiotic Susceptibility Test of Bacteria from In Vitro Cultures of Pinellia ternata and Tea Plant (반하 및 차나무의 기내배양시 발생하는 세균의 동정 및 항생제 감수성 검정)

  • Contamination of bacterial infection is one of serious problems in in vitro culture system of root crops. From the contaminated tubes over 140 of petiole cultures of Pinellia ternata, a medicinal plant, 4 genera 8species 48 strains of bacteria, including Aeromonas and Pseudomonas, were isolated and identified and another 8 strains were not fully identified. Most of them were motile Gram positive bacteria as in common in early stage of in vitro cultures. Six strains of bacteria, 5 of Gram negative, including Enterobacter, and 1 of Gram positive, were identified from the embryonic axes cultures of tea plant. From the susceptibility test to pre-screened 5 antibiotics, all of the bacteria except for 2 species of Pseudomonas were susceptible to cefotaxime 60∼100mg/L. While 60mg/L erythromycin only was effective to Pseudomonas. Combination of erythromycin 20mg/L and cefotaxime 60mg/L totally suppressed the growth of all bacterial strains tested. Susceptibility test of bacteria from tea embryonic axes cultures showed similar results. Combination of erythromycin 35mg/L and cefotaxime 60mg/L was effective to 15 bacterial strains and partially effective to 1 unidentified.

Isolation and Identification of Fungal Species from the Insect Pest Tribolium castaneum in Rice Processing Complexes in Korea

  • The red flour beetle, Tribolium castaneum, is one of the most common and economically important pests of stored cereal products worldwide. Furthermore, these beetles can act as vectors for several fungal post-harvest diseases. In this study, we collected T. castaneum from 49 rice processing complexes (RPCs) nationwide during 2016-2017 and identified contaminating fungal species on the surface of the beetles. Five beetles from each region were placed on potato dextrose agar media or Fusarium selection media after wet processing with 100% relative humidity at $27^{\circ}C$ for one week. A total of 142 fungal isolates were thus collected. By sequence analysis of the internal transcribed spacer region, 23 fungal genera including one unidentified taxon were found to be associated with T. castaneum. The genus Aspergillus spp. (28.9%) was the most frequently present, followed by Cladosporium spp. (12.0%), Hyphopichia burtonii (9.2%), Penicillium spp. (8.5%), Mucor spp. (6.3%), Rhizopus spp. (5.6%), Cephaliophora spp. (3.5%), Alternaria alternata (2.8%) and Monascus sp. (2.8%). Less commonly identified were genera Fusarium, Nigrospora, Beauveria, Chaetomium, Coprinellus, Irpex, Lichtheimia, Trichoderma, Byssochlamys, Cochliobolus, Cunninghamella, Mortierella, Polyporales, Rhizomucor and Talaromyces. Among the isolates, two known mycotoxin-producing fungi, Aspergillus flavus and Fusarium spp. were also identified. This result is consistent with previous studies that surveyed fungal and mycotoxin contamination in rice from RPCs. Our study indicates that the storage pest, T. castaneum, would play an important role in spreading fungal contaminants and consequently increasing mycotoxin contamination in stored rice.

Observation and Record of Sea Turtles in Bycatch and Stranding from Jeju Island of Korea (우리나라 제주도에서 관찰된 바다거북의 좌초와 혼획)

  • Bycatch and stranding of sea turtles from Jeju coastal water of Korea were monitered from 2008 to 2009. A total of 41 sea turtles, consisting of 12 greens (Chelonia mydas), 3 loggerheads (Caretta caretta), 1 hawksbill (Eretmochelys imbricata) and 25 unidentified species, were recorded and observed in those area. Reported stranding and bycatch reached to a peak in August. The largest number of bycatch occurred in the coastal waters around Aewol-eup located in northwest area of Jeju Island. Most of the bycatch were taken by set nets and gill nets, which prevailed over the fishing grounds in the coastal waters of the Jeju Island. It is suggested that an appropriate monitoring system be necessary to conserve endangered sea turtle species in Jeju of Korea.

Development of the Large-Scale Oligonucleotide Chip for the Diagnosis of Plant Viruses and its Practical Use

  • A large-scale oligonucleotide (LSON) chip was developed for the detection of the plant viruses with known genetic information. The LSON chip contains two sets of 3,978 probes for 538 species of targets including plant viruses, satellite RNAs and viroids. A hundred forty thousand probes, consisting of isolate-, species- and genus-specific probes respectively, are designed from 20,000 of independent nucleotide sequence of plant viruses. Based on the economic importance, the amount of genome information, and the number of strains and/or isolates, one to fifty-one probes for each target virus are selected and spotted on the chip. The standard and field samples for the analysis of the LSON chip have been prepared and tested by RT-PCR. The probe's specific and/or nonspecific reaction patterns by LSON chip allow us to diagnose the unidentified viruses. Thus, the LSON chip in this study could be highly useful for the detection of unexpected plant viruses, the monitoring of emerging viruses and the fluctuation of the population of major viruses in each plant.

Distribution of Fish Eggs, Larvae and Juveniles around the Youngsan River Estuary (영산강 하구역 주변해역의 난자치어 분포)

  • Distribution of ichthyoplanktons and its relation to environmental factors (temperature, salinity, $chlorophyll\;\alpha$) around the Youngsan River Estuary of Mokpo, southwest coast of Korea were investigated based on the samples collected bimonthly from February to December, 2001. Unidentified fish eggs were most abundant $1,345\;eggs/100m^3$ in August, while eggs of Engraulis japonicus were most abundant $63\;eggs/100m^3$ in June. A total of forty species of fish larvae and juveniles belonging to the twenty families under six orders were caught, with six gobiids, four icefishes, four croakers and three herrings being represented. Fish larvae and juveniles peaked $17,757\;inds./100m^3$ in August, which consisted of $91.02\%$ of gobiids, $3.69\%$ of gunnels and $3.67\%$ of icefishes, and they were most abundant $7,467\;Inds./100m^3$ at St. 7. Cluster analysis based on the individuals of fish larvae and juveniles showed that six months were classified into two groups at the dissimilarity 70, the first was composed of June and August and the second was February and December and April and October were not clustered with any other months. In August when species diversity index was the highest, eight stations were classified into two groups at the dissimilarity 70, the first was composed of St. 1, 3, 4, 5 and 8 and the second was St. 6, 7 and 2. As a result from correspondence analysis, there is no effect of the environmental factors with a density of fish eggs and larvae, but correlation could be found between a density of fish eggs and $chlorophyll\;\alpha.$

The Distribution and Standing Crop of Phytoplankton of Lagoons in the East Coast of Korea (동해안 석호의 식물플랑크톤 분포와 현존량)

  • The distribution and standing crop of phytoplankton were investigated at 12 stations of Songjiho, Ssangho, Maeho and Hyangho as of four lagoons in the east coast from May to November, 2001. It turned out to be total of 164 taxa, and classified as four phylums, four classes, 14 orders, 20 families, 59 genera, 139 species, 22 varieties, two forms and 1 unidentified species by Engler's classification system. Among 104 taxa, 16 taxa including Oscillatoria chlorina were identified as indicators of water pollution and only Cocconeis placentula was the indicator of the clean water. Standing crops of all stations investigated appeared to be relatively high values. Based on the present study upon the distribution and standing crop of phytoplankton, it is regarded as the state of the eutrophication in Songjiho, Ssangho, Maeho and Hyangho as of four lagoons in the east coast.

Studies on the antioxidant Effects of Carthami Flos Extract (홍화(紅花) 추출물의 항산화 효과에 대한 연구)

  • Objective : The objective of this study was to investigate the antioxidative effects of Carthami Flos extract. Methods : Total antioxidant status was examined by total antioxidant capacity(TAC) and total antioxidant response(TAR) against potent free radical reactions. The effect of Carthami Flos extract was examined far details of total phenolic content concentration at which 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical scavenging activity was inhibited, the inhibitory effect on lipid peroxidation, and the effect on reactive oxygen species(ROS) generation. Results : TAC of Carthami Flos extract at the concentration of 5 mg/ml was 1.84 mM Trolox equivalent. 2. TAR of Carthami Flos extract, on the other hand, couldn't be determined due to interference from unidentified compounds. 3. Total phenolic content of Carthami Flos extract at the concentration of 5 mg/ml was 2.01 mM gallic acid equivalent. 4. Concentration of Carthami Flos extract at which DPPH radical scavenging activity was inhibited by 50% was 6.43 mg/ml as compared to 100% by Pyrogallol solution as a reference. 5. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeS04/ascorbic acid. Carthami Flos extract at the concentration of 10 ms/ml slightly but significantly decreased TBARS concentration. The extract continued to prevent lipid peroxidation in a dose-dependent manner. 6. The effect of Carthami Flos extract on reactive oxygen species(ROS) generation was examined using a cell-free system induced by hydrogen peroxide/FeS04. Addition of 1 mg/ml of Carthami Flos extract significantly reduced dichlorofluorescein(DCF) fluorescence. Carthami Flos extract caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that the ektract significantly prevented ROS generation in vitro. Conclusion: : Antioxidant efffcts of Carffami ffor extract seem to be due, at least in part, to the prevention offree radical-induced oxidation, fellowed by inhibition of lipid peroxidation.

A Study on Clustering and Identifying Gene Sequences using Suffix Tree Clustering Method and BLAST (서픽스트리 클러스터링 방법과 블라스트를 통합한 유전자 서열의 클러스터링과 기능검색에 관한 연구)

  • The DNA and protein data of diverse species have been daily discovered and deposited in the public archives according to each established format. Database systems in the public archives provide not only an easy-to-use, flexible interface to the public, but also in silico analysis tools of unidentified sequence data. Of such in silico analysis tools, multiple sequence alignment [1] methods relying on pairwise alignment and Smith-Waterman algorithm [2] enable us to identify unknown DNA, protein sequences or phylogenetic relation among several species. However, in the existing multiple alignment method as the number of sequences increases, the runtime increases exponentially. In order to remedy this problem, we adopted a parallel processing suffix tree algorithm that is able to search for common subsequences at one time without pairwise alignment. Also, the cross-matching subsequences triggering inexact-matching among the searched common subsequences might be produced. So, the cross-matching masking process was suggested in this paper. To identify the function of the clusters generated by suffix tree clustering, BLAST was combined with a clustering tool. Our clustering and annotating tool is summarized as the following steps: (1) construction of suffix tree; (2) masking of cross-matching pairs; (3) clustering of gene sequences and (4) annotating gene clusters by BLAST search. The system was successfully evaluated with 22 gene sequences in the pyrubate pathway of bacteria, clustering 7 clusters and finding out representative common subsequences of each cluster
