Skin Whitening and Skin Immune Activities of the Peptides Isolated from Crassostrea gigas by Ultrasonification Processes (초음파 복합처리를 통한 참굴 펩타이드의 피부미백 및 피부면역 활성)

  • In this study, peptides were isolated from Crassostrea gigas using an ultrasonification process at $40^{\circ}C$. The yield of the peptides was greater than 34%, and their cytotoxicity was found to be less than 22.8% against several cell lines that were treated with the extracts at a dose of 1.0 mg/mL. In addition, the tyrosinase inhibitory and melanin synthesis of the peptides isolated from Crassostrea gigas were also evaluated to determine if they could be used as a potential cosmetic agent. The peptides were found to significantly inhibit the melanin synthesis of the clone M-3 cell line by up to 62.7%. The inhibitory activities of the tyrosinase were observed 34.51% in ascorbic acid, 42.49% in extract with the ultrasonification at $40^{\circ}C$ and 35.37% in $40^{\circ}C$ extract at 1.0 mg/mL concentration, respectively. Finally, when samples were treated with the peptide extracts at a concentration of 0.6 mg/mL, PGE2 expression was significantly decreased. Taken together, these results indicate that Crassostrea gigas may be a source of cosmetic agents capable of improving physiological hyperpigmenting and immuno-modulating skin disorders.

  • The anticancer activities of the extracts from Acanthopanax senticosus Harms, Ephedra sinica Stapf, Rubus coreanus Miq and Artemisia capillaris Thunb were compared according to extract systems. About 70% of the growth of human hepatocarcinoma cancer cell was inhibited in adding 1.0 mg/ml of the water extract from Rubus coreanus Miq with ultrasonification at $60^{\circ}C$. The growth of human normal lung cell was limited to 25% in adding the extracts with ultrasonification at $60^{\circ}C$. The effect of extracts obtained by only water and with ultrasonification on different of human promyelocytic leukemia cells was also observed.

Study on Characteristics of Solubilization for Sewage Sludge Using Electronic Field and Ultrasonification (전기장과 초음파를 이용한 하수슬러지의 가용화 특성 연구)

  • Sludge solubilization using sonification has been increasingly used for sludge volume reduction along with enhancing digestion efficiency during anaerobic biogas production. In this study, either electric field or ultrasonification or in combination with were investigated using three types of sludge (return, excess and mixed at G sewage treatment facility) for the most efficient solubilization. As a the closed loop, 200 L of sludge was continuously passing through the solubilization system at an average flow rate of $0.7m^3/h$, which is equivalent to 3.5 times treated per hour for up to 84 times (24 h). Only implying electric field showed no variation for sCOD/tCOD before and after treatment on sludge solubilization regardless of types of sludge. However, employing the ultrasonic or combined system could both increasingly solubilize sludge with regard to the number of passing-through, which more enhanced by the combined. In addition, VSS/TSS was lowered to in the range of 2 and 6% while its particle size, diameter (0.9) and diameter (0.5) were more minimized than that of raw sludge. For return sludge, ultrasonification was more efficiently facilitated for solubilization, whereas electric field-ultrasonification was more preferably applied for excess and mixed sludge. It is concluded that depending on types of sludge, solubilization system must be selectively applied for the most efficient break-up of them.

Effects of Dandelion (Teraxacum platycarpum) with Various Extracting Method on Antioxidative Capacity, Lipid Metabolism in Diet-induced Obese Rats (초음파추출과 열수추출에 의한 민들레의 항산화 및 지질강하 효과)

  • This study was conducted to investigate the effects of dandelion (Teraxacum platycarpum) extracts obtained by only water and with ultrasonification on antioxidative system and lipid metabolism in high cholesterol-fed rats. Five groups of rats were given high cholesterol diets for 8 weeks. The control group received without dandelion extracts and the other four groups received with one of dandelion extracts for 4 weeks respectively ; TP-N-1(100 mg/kg/day of Teraxacum platycarpum water extract), TP-N-2(200 mg/kg/day of Teraxacum platycarpum water extract), TP-S-1(100 mg/kg/day of Teraxacum platycarpum water-ultrasonification extract), TP-S-2(200 mg/kg/day of Teraxacum platycarpum water-ultrasonification extract). The results are summarized as follows; The hepatic and plasma TBARS levels significantly decreased in the dandelion extracts groups compared to those of no treatment group. Especially the group TP-N-2 was comparatively best among those. TP-N-2 groups had significantly higher levels of glutathione peroxidase (GSH-Px) and catalase activities. There was no significant difference between dandelion extracts groups and no treatment group in SOD levels. In plasma triglyceride level, plasma FFA level, TP-S-2 group had significantly lower levels than that of the other groups. In plasma glucose levels, dandelion extracts group were similar to those of normal rats. Plasma total cholesterol levels significantly decreased in the TP-S-2 group compared to those of the other groups. HDL levels were also significantly higher than those of the other groups. Compared with those of no treatment group, dandelion extract groups had significantly higher levels of LDL. In liver total cholesterol level, TP-S-2 groups had significantly lower levels than that of the other groups. Compared with those of no treatment group, dandelion extracts groups had significantly lower levels of liver triglyceride, but especially the TP-S-2 group showed comparatively the best significant effect among those. TBARS, triglyceride, LDL, FFA levels significantly decreased in TP-S-2 groups compared to the other four groups. HDL levels was also significantly higher than the other four groups. According to the above result, it could be suggested that ultrasonic extraction have the upper hand in lipid metabolism and water extraction have the advantage of antioxidative system.

Comparison of Anti-Skin Wrinkle Activities of Aronia melanocarpa Extracts by Extraction Methods (추출방법에 따른 아로니아 추출물의 주름 개선 효능 비교)

  • This study was to investigate anti-skin wrinkle effect of aronia extracts by extraction processes. The 70% ethanol extract by ultrasonification process (UEE) showed highest DPPH scavenging activity of 90.4% in order water extract (WE), 70% ethanol extract (EE) was measured 84.2%, 84.3%. collagen production measured $245ng/m{\ell}$ from UEE. WE was measured $53.5ng/m{\ell}$ and EE was measured $224.4ng/m{\ell}$. MMP-1 production was observed lowest $22.5pg/m{\ell}$ at UEE. MMP-1 production of EE was $34.6pg/m{\ell}$ and WE was $102.3pg/m{\ell}$. These results were found the highest antioxidant and anti-wrinkle effect at UEE among three processes. It was also confirmed that anti-skin wrinkle activities of the aronia extract was strongly correlated with anti-oxidant activities due to high amounts of antocyanins in the extract.

Optimal Extract Condition for the Enhancement of Anticancer Activities of Artemisia princeps Pampanini (강화 사자발쑥의 항암활성 증진을 위한 추출조건의 최적화)

  • Extractions of Artemisia princeps Pampanini were obtained by using water and ethanol at different temperatures ($60^{\circ}C,\;100^{\circ}C$) with or without ultrasonification process (40 kHz). Yield of ultrasonificated extracts were about 20% higher than that of control group. Cytotoxicity of all conditions through adding 1.0 mg/$m{\ell}$ was below 37%, and treated with ultrasonification group was lower than the other group, about $5{\sim}8%$. $100^{\circ}C$ water extract with ultrasonification was higher anticancer activities as maximum 73% and higher selectivities at concentrations over 0.8 mg/$m{\ell}$. The extracts treated with ultrasonification were higher anticancer activities than the control. Densitometric analysis of bcl-2 revealed that extracts of high anticancer activity had low density. This results suggest that expression of bcl-2 protein by adding of Artemisia princeps Pampanini extracts relative to taking cancer. To conclude, optimum condition for efficient extraction of Artemisia princeps Pampanini is using water with ultrasonification at over $60^{\circ}C$ below $100^{\circ}C$.

Analysis on Patent Trends in Nonthermal Processing Technologies for Medicinal Herbs (한약재 가공 기술의 특허 동향 연구 - 비가열 가공 기술을 중심으로 -)

  • The purpose of this study was to analyze the patent application trend in the processing technology for medicinal herbs. Recently, in processing technology for medicinal herbs, experimental researches have frequently been published through papers in journals. However, the research results about the patent area were fewer than the others. We tried to analyze the patent application trend in nonthermal processing technologies for medicinal herbs by country as Korea, Japan, U.S.A. and Europe. The detailed technologies consisted of pulsed electric field, oscillatory magnetic field, intense pulsed light, ultrasonification, high hydrostatic pressure, microwave, radiation, Ohmic heating, and supercritical extraction. As a result we found that patents of nonthermal processing technologies has been growing steadily in quantity from 1980s and growing quickly since 2000s. The number of patent in Korea is larger than others as making up 70% in that whole. The number of patent in ultrasonification field was larger than others in portfolio analysis. Patent application trend in nonthermal processing technologies for ingestion occupies high share compared to other usage applications. In conclusion, patent trends of nonthermal processing technologies for medicinal herbs belong to the period in the development.

Antioxidative Activity of Kalopanax pictus Shoot Extracted Using Different Extraction Methods (추출 방법이 개두릅 추출물의 항산화 활성에 미치는 영향)

  • The antioxidative activity of Kalopanax pictus shoots extracted using five different methods, stirrer extraction (SE), ultrasonification extraction (USE), vacuum extraction (VE), reflux extraction (RE), and reflux extraction after ultrasonification extraction (RUE), was evaluated to determine the most effective extraction method. RUE showed the highest yield of extract, and total polyphenol and flavonoid content. Additionally, the RUE extract showed higher antioxidative activity than that of the other extracts based on DPPH and ABTS free radical scavenging (90.85%, 99.83%), reducing power activity (OD 0.95, 700 nm), nitrite scavenging activity (55.46%), and ferrous ion chelating effect (45.12%).

Physiological Activities of Rubus coreanus Miq. Extracts Using Different Extraction Methods (추출방법에 따른 복분자 추출물의 생리활성)

  • This study was carried out to evaluate biological activities concerning extracts according to extraction methods from unripened fruit of Rubus coreanus Miq. The extraction methods were HWE (hot water extraction for 4 hr at $100^{\circ}C$), SFE (extraction for 3 hr at $40^{\circ}C$ under 300 bar, 100% of $CO_2$ fluid), USE (ultrasonification extraction for 4 hr at $50^{\circ}C$ with water), USE+HWE (hot water extraction for 2 hr at $100^{\circ}C$ after ultrasonification process for 2 hr), VE (vacuum extraction for 4 hr at $90^{\circ}C$ under 0.9 bar with water). VE extract showed the highest contents of total polyphenol ($178.78{\pm}3.79\;mg/g$) and total flavonoid ($40.93{\pm}0.68\;mg/g$). $IC_{50}$ values of DPPH radical scavenging activity, linoleic acid peroxidation inhibition activity and LDL (low density lipoprotein) oxidation inhibition activity of HWE extract showed the lowest $35.39{\pm}0.25{\mu}g/mL$, $12.61{\pm}0.31{\mu}g/mL$ and $1.31{\pm}0.02{\mu}g/mL$ among other all extracts, respectively. $IC_{50}$ values of ${\alpha}$-glucosidase inhibitory activities of VE and HWE extracts showed lower $14.34{\pm}0.20{\mu}g/mL$ and $15.83{\pm}0.20{\mu}g/mL$ than those of other extracts, respectively. Specifically, HWE and VE extracts have relatively better biological activities than other extracts; these could be potentially used as a bioactive source for health functional foods.
