Saengmaeksan, a traditional herbal formulation consisting of Panax ginseng, ameliorates hyperuricemia by inhibiting xanthine oxidase activity and enhancing urate excretion in rats

  • Background: Saengmaeksan (SMS) is a traditional Korean medicine composed of three herbs, Panax ginseng, Schisandra chinensis, and Liriope platyphylla. SMS is used to treat respiratory and cardiovascular disorders. However, whether SMS exerts antihyperuricemic effects is unknown. Methods: Effects of the SMS extract in water (SMS-W) and 30% ethanol (SMS-E) were studied in a rat model of potassium oxonate-induced hyperuricemia. Uric acid concentrations and xanthine oxidase (XO) activities were evaluated in the serum, urine, and hepatic tissue. Using renal histopathology to assess kidney function and uric acid excretion, we investigated serum creatinine and blood urea nitrogen concentrations, as well as protein levels of renal urate transporter 1 (URAT1), glucose transporter 9 (GLUT9), and organic anion transporter 1 (OAT1). The effects of SMS on in vitro XO activity and uric acid uptake were also evaluated. The components of SMS were identified using Ultra Performance Liquid Chromatography (UPLC). Results: SMS-E reduced serum uric acid and creatinine concentrations, and elevated urine uric acid excretion. SMS-E lowered XO activities in both the serum and liver, and downregulated the expression of renal URAT1 and GLUT9 proteins. SMS-E reduced renal inflammation and IL-1b levels in both the serum and kidneys. SMS-E inhibited both in vitro XO activity and urate uptake in URAT1-expressing oocytes. Using UPLC, 25 ginsenosides were identified, all of which were present in higher levels in SMS-E than in SMS-W. Conclusion: SMS-E exhibited antihyperuricemic effects by regulating XO activity and renal urate transporters, providing the first evidence of its applicability in the treatment of hyperuricemia and gout.

Scopolamine 유발 기억 손상 마우스 모델에서 국산 산사와 중국산 산사의 항건망 효과 비교 (Comparative Study on the Effects of Korean and Chinese Crataegus pinnatifida on Scopolamine-induced Memory Impairment in Mice)

  • This study was conducted to investigate the cognitive improvement and memory recovery effects of Korean and Chinese Crataegus pinnatifida ethanolic extracts on scopolamine-induced memory impairment in mice. In vivo studies were carried out with mice treated with Korean Crataegus pinnatifida extracts (KCF) and Chinese Crataegus pinnatifida extracts (CCF) in doses of 5 and 50 mg/kg (p.o.) and scopolamine was injected 30 min before the behavioral testing. Antioxidant activity was assessed by 2,2-diphenyl-1-picryl hydrazyl (DPPH) assay and 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay, and acetylcholinesterase inhibition by Ellman's modified method. The chlorogenic acid and hyperoside as marker compounds of KCF and CCF was quantified by ultra-performance liquid chromatography analysis (UPLC). Results showed that KCF was more contained high content of chlorogenic acid and hyperoside than CCF. In addition, KCF was more exerted free radical (DPPH and ABTS) scavenging activity and blocked AChE activity than CCF. In vivo studies also showed that KCF administration has a further improved the memory of scopolamine-treated mice than CCF in Y-maze test, passive avoidance test and Morris water maze test. These results revealed that KCF more prevents scopolamine-induced memory impairments through antioxidant and acethylcholinesterase inhibition effect compared CCF.

Rapid determination and quantification of hair-growth compounds in adulterated products by ultra HPLC coupled to quadrupole-orbitrap MS

  • Recently, a number of adulterated products, which are advertised as hair-growth enhancer have been emerged among those who suffer hair loss disease. For continuous control of illegal products, in this study, a rapid and sensitive method for simultaneous screening of 12 compounds that enhance hair-growth was established to protect public health by ultrahigh-performance liquid chromatography coupled to quadrupole-orbitrap mass spectrometry (UHPLC-Q-Orbitrap-MS). Fragmentation pathways of them were proposed based on $MS^2$ spectral data obtained using the established method. In this analysis, the LODs and LOQs ranged from 0.05 to 50 ng/mL and from 0.17 to 167 ng/mL, respectively. The square of the linear correlation coefficient ($R^2$) was determined as more than 0.995. The intra- and inter-assay accuracies were respective 88-112 % and 88-115 %. Their precision values were measured within 5 % (intra-day) and 10 % (inter-day). Mean recoveries of target compounds in adulterated products ranged from 84 to 115%. The relative standard deviation of stability was less than 12 % at $4^{\circ}C$ for 48 h. The method was employed to screen 14 dietary supplements advertised to be effective for the treatment of hair loss. Some of the products (~21 %) were proven to contain synthetic drugs that promote hair growth such as triaminodil, minoxidil, and finasteride.

국내산 더덕의 Tangshenoside I과 Lobetyolin 정량분석 (Quantitative Analysis of Tangshenoside I and Lobetyolin from Korean Deoduk (Codonopsis lanceolata))

  • Deoduk (Codonopsis lanceolata) has a complex chemical composition that includes polyphenols, saponins, amino acids, and other unidentified compounds. The contents of tangshenoside and lobetyolin are considered as standard of quality evaluation of Deoduk. In this study, an ultra-performance liquid chromatography (UPLC) method was developed for the quantitative determination of the two marker constituents, tangshenoside and lobetyolin. The methods for determining the standards of quality were validated by measuring their linearity, specificity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy using UPLC. Reversed-phase UPLC analysis was conducted quantitatively to identify individual tangshenoside and lobetyolin in Deoduk extracted with 50% (v/v) aqueous ethanol. We used 21 samples to carry out quantitative analysis of tangshenoside and lobetyolin. Based on their dry weights, the levels of tangshenoside and lobetyolin were 0.36~3.54 mg/g, 0.24~1.29 mg/g, respectively. These results will be valuable as basic data for standardization of Korean Deoduk.

참느릅나무(Ulmus parvifolia) 잎 추출물 분석 및 안정성 평가 (Analysis and stability test of the extracts from Ulmus parvifolia leaves)

  • Ulmus parvifolia 잎의 약리학적 지표 성분을 제시하기 위해 간단하고 신뢰할 수 있는 HPLC 분석방법을 개발하였다. 지표 성분들은 NMR 및 UPLC-QTof-MS 분석에 의해 neochlorogenic acid (trans-5-O-caffeoylquinic acid, 5-CQA)과 chlorogenic acid (trans-3-O-caffeoylquinic acid, 3-CQA)로 구조 동정 되었다. HPLC를 이용하여 두개 화합물을 포함한 잎 추출물의 정성/정량 분석 방법을 평가하였다. 잎 30% 에탄올 추출물과 지표 성분의 안정성 테스트는 6개월 동안 평가되었다. 그러나 안정성 조사 기간 동안 추출물과 지표 성분들 각각의 함량에 대한 유의한 변화는 관찰되지 않았다.

방풍, 식방풍의 대사체 프로파일링을 통한 지표성분 선정 및 분석법검증 (Metabolic profiling and method validation of marker compounds from Saposhnikoviae Radix and Peucedani Japonici Radix)

  • 방풍(Saposhnikoviae Radix, SR) 과 식방풍(Peucedani Japonici Radix, PR)은 각각 다른 기원식물을 가지며 속과 종이 다르지만 한국, 중국, 및 일본에서 대표적인 전통 한약재로 오랫동안 혼용되어 사용되었다. 본 연구에서는 방풍과 식방풍의 판별 마커를 확인하기 위하여 UPLC-QTOF/MS를 이용한 대사체 프로파일링 및 다변량 통계분석을 진행하였다. 그 결과, 5-O-methylvisammioside와 peucedanol을 각각 방풍과 식방풍의 지표성분으로 선정하였으며, UPLC를 이용하여 분석법을 검증하였다. 제안된 방풍 및 식방풍의 지표성분에 대한 분석법 검증은 방풍과 식방풍의 분류와 품질 평가 및 성분 검증을 위한 효과적인 방법이 될 것으로 기대된다.

Pharmacokinetic profiles of levofloxacin after intravenous, intramuscular and subcutaneous administration to rabbits (Oryctolagus cuniculus)

  • Levofloxacin pharmacokinetic profiles were evaluated in 6 healthy female rabbits after intravenous (I/V), intramuscular (I/M), or subcutaneous (S/C) administration routes at a single dose of 5 mg/kg in a 3 × 3 cross-over study. Plasma levofloxacin concentrations were detected using a validated Ultra Performance Liquid Chromatography method with a fluorescence detector. Levofloxacin was quantifiable up to 10 h post-drug administration. Mean AUC0-last values of 9.03 ± 2.66, 9.07 ± 1.80, and 9.28 ± 1.56 mg/h*L were obtained via I/V, I/M, and S/C, respectively. Plasma clearance was 0.6 mL/g*h after I/V administration. Peak plasma concentrations using the I/M and S/C routes were 3.33 ± 0.39 and 2.91 ± 0.56 ㎍/mL. Bioavailability values, after extravascular administration were complete, - 105% ± 27% (I/M) and 118% ± 40% (S/C). Average extraction ratio of levofloxacin after I/V administration was 7%. Additionally, levofloxacin administration effects on tear production and osmolarity were evaluated. Tear osmolarity decreased within 48 h post-drug administration. All 3 levofloxacin administration routes produced similar pharmacokinetic profiles. The studied dose is unlikely to be effective in rabbits; however, it was calculated that a daily dose of 29 mg/kg appears effective for I/V administration for pathogens with MIC < 0.5 ㎍/mL.

작약감초탕의 추출용매에 따른 성분 분석 및 항피로 효능 비교 (Comparison of Ingredient Quantities and Anti-Fatigue Effects of Jakyakgamcho-Tang according to Extraction Solvent)

  • Objectives : Jakyakgamcho-tang has been used as analgesic and spasmolytic for muscle pain. It has reported to anti-diabetes, anti-inflammation, and neuro-protective effects. A clinical study of muscle fatigue and pain improvement effect of Jakyakgamcho-tang has been reported with increasing frequency of clinical use. However, the anti-fatigue effect of Jakyakgamcho-tang in animal model has not been studied. In this study, we compared anti-physical fatigue effects of water and 30% ethanol extract of Jakyakgamcho-tang. Methods : The amounts of components contained in water and 30% ethanol extract of Jakyakgamcho-tang were compared by ultra-performance liquid chromatography. Anti-physical fatigue effects were evaluated using weight-loaded forced swimming test. We also investigated the effects of Jakyakgamcho-tang on the change of fatigue parameters by blood biochemical analysis. Results : The relative amount of components of Jakyakgamcho-tang were about 19-53% higher in the 30% ethanol extract than in the water extract. In the swimming test, 30% ethanol extract showed a significantly greater anti-fatigue effects than conventional water extract of Jakyakgamcho-tang. The 30% ethanol extract of Jakyakgamcho-tang improved the exhausted swimming time (54%), and serum lactate dehydrogenase (48%) and lactic acid (60%) levels compared with water extract of Jakyakgamcho-tang. Conclusions : These results showed that differences in the amounts of components by different extraction were associated with differences of anti-fatigue effect of Jakyakgamcho-tang. Thus, the 30% ethanol extraction method could be applied to dose-reducing formulations of traditional herbal medicines.

Ethanol extract of Sinsun-yukza-hwan, a Korean medicinal prescription, promotes hair growth in C57BL/6 mice, an alopecia animal model

  • Objectives : In Korean medicine, a prescription of Sinsun-yukza-hwan (Shenxian-liuzi-wan, SSY) has been used in clinic for treatment of alopecia via oral. This study was performed to determine transdermal effects of the ethanol extract from SSY on hair growth and -related gene expressions in mice. Methods : We analyzed index compound, 5-hydroxy-methyl-2-furaldehyde (HMF), in SSY extract by ultra performance liquid chromatography (UPLC). 6 weeks old C57BL/6 mice with removed hair were used as an alopecia animal model. Mice were divided into 3 experimental groups including normal (3 water: 1 ethanol: 2 polyethylene glycol mixture as a vehicle), SSY extract and 5% minoxidil (as a positive control), treated groups. SSY was applied topically on the hair-shaved skin of C57BL/6 mice every day for 15 days. The color, thickness and density of hair were monitored every $5^{th}$ day by naked eye, photograph and phototrichogram using folliscope. Also hair growth-associated gene expressions were measured by immunoblotting assay. Results : Hair density of minoxidil or SSY-treated group was significantly increased compared to that of vehicle application on the $15^{th}$ day, respectively. And hair thickness of minoxidil and SSY groups was increased compared to that of vehicle treated group on the $15^{th}$ day, respectively. Induction of insulin-like-growth factor 1(IGF-1) and vascular endothelial growth factor (VEGF) were also significantly accelerated by SSY extract compared to those of vehicle-applied group. Conclusions : These results provide scientific evidence to support the potent multi-application of SSY as a cosmeceutical material for promoting hair growth.

Specific urinary metabolites in canine mammary gland tumors

  • The identification of biomarkers that distinguish diseased from healthy individuals is of great interest in human and veterinary fields. In this research area, a metabolomic approach and its related statistical analyses can be useful for biomarker determination and allow non-invasive discrimination of healthy volunteers from breast cancer patients. In this study, we focused on the most common canine neoplasm, mammary gland tumor, and herein, we describe a simple method using ultra-high-performance liquid chromatography to determine the levels of tyrosine and its metabolites (epinephrine, 3,4-dihydroxy-L-phenylalanine, 3,4-dihydroxyphenylacetic acid, and vanillylmandelic acid), tryptophan and its metabolites (5-hydroxyindolacetic acid, indoxyl sulfate, serotonin, and kynurenic acid) in canine mammary cancer urine samples. Our results indicated significantly increased concentrations of three tryptophan metabolites, 5-hydroxyindolacetic acid (p < 0.001), serotonin, indoxyl sulfate (p < 0.01), and kynurenic acid (p < 0.05), and 2 tyrosine metabolites, 3,4-dihydroxy-L-phenylalanine (p < 0.001), and epinephrine (p < 0.05) in urine samples from the mammary gland tumor group compared to concentrations in urine samples from the healthy group. The results indicate that select urinary tyrosine and tryptophan metabolites may be useful as non-invasive diagnostic markers as well as in developing a therapeutic strategy for canine mammary gland tumors.
